Spasticity, a frequently encountered symptom in patients with upper motor neuron (UMN) syndrome, poses a significant challenge, negatively affecting function, activity, and social engagement. Despite the acknowledged benefits of exercise in the rehabilitation of UMN syndrome, therapy sessions often trigger an unwelcome increase in muscle stretch reflex activity, resulting in considerable muscle tension despite improvements in function and activity levels. Despite the recognized benefits of exercise in UMN syndrome rehabilitation, there's often an undesirable rise in muscle stretch reflex activity during therapy, leading to considerable muscle tension despite improvements in function and activity levels. The challenge lies in identifying effective strategies that enhance function, activity, and participation while curbing excessive muscle tension caused by heightened stretch reflex activity. Spasticity significantly disrupts the daily lives of affected individuals and presents substantial challenges for caregivers. However, existing methods for measuring and evaluating spasticity have their limitations and are susceptible to errors. This article describes both established and innovative methods utilized for quantitative spasticity assessment and management of spasticity, with the overarching goal of improving the definition of spasticity and identifying assessment techniques suitable for clinical application.
식이 영양에 의한 충분한 영양분 섭취는 치매에 의한 뇌 인지 기능의 저하의 위험성을 줄일 수 있는 중요한 수단이다. 설포라판은 뇌 인지 기능 개선효과가 있는 것으로 알려져 있는 영양 성분으로, 설포라판이 다량으로 포함된 브로콜리 추출물은 인지 기능 개선 효과가 좋을 것으로 기대된다. 본 연구에서는 제주도 탈염 용암해수 재배로 제조한 브로콜리 추출물이 뇌 인지 기능 개선 효과를 가지고 있는지 시험하 였다. 제주도 용암해수는 유용 미네랄 (아연, 바나듐, 게르마늄)이 풍부해서, 용암 해수에서 재배한 작물은 유용미네랄 함량이 높은 것으로 알려져 있다. 제주도 탈염 용암 해수를 사용해서 재배한 브로콜리 추출물의 화학 조성 중 설포라판과 글루코라판 성분을 분석하기 위해 LC-Q-orbitrap 질량 분석기를 사용하였고, 설포라판의 정량 분석을 위해 HPLC 를 사용하였다. 브로콜리 추출물의 신경 세포 사멸 억제 효과와 항 염증 효과를 시험하기 위해 SH-SY5Y 세포를 사용한 실험을 수행하였고, 시냅스 가소성 촉진 효과를 시험하기 위해 SH-SY5Y 세포에서 시냅스 가소성 관련 단백질의 발현 변화와 아세틸콜린 분해 효소의 활성 변화를 측정하였다. 이러한 실험들을 수행한 결과 탈염 염지하수로 재배한 브로콜리 추출물은 신경 세포 사멸 억제 효과와 항염증 효과가 있음을 확인 하였고, 시냅스 가소성 관련 단백질 발현을 증가 시키고 아세틸콜린 분해 효소의 활성을 억제해서 시냅스 가소성을 증가 시키는 효과가 있는 것을 확인 하였다. 이 상의 결과들은 제주도 탈염 용암해수로 재배한 브로콜리 추출물이 치매에 의한 뇌 인지 기능 저하를 억제하는 좋은 식이 영양분으로 사용될 수 있는 가능성을 제시하였다.
Background: Lumbar disc herniation (LDH) causes neurological symptoms by compression of the dura mater and nerve roots. Due to the changed in proprioception inputs that can result in abnormal postural pattern, delayed reaction time, and changed in deep tendon reflex.
Objective: To investigate the effects of lumbar stabilization exercises on motor neuron excitability and neurological symptoms in patients with LDH.
Design: Randomized Controlled Trial (single blind)
Methods: Thirty patients with LDH were recruited; they were randomly divided into the balance center stabilization resistance exercise group (n=15) and the Nordic walking group (n=15). Each group underwent their corresponding 20- minute intervention once a day, four times a week, for four weeks. Participants’ motor neuron excitability and low back pain were assessed before and after the four-week intervention.
Results: There were significant differences in all variables within each group (p<.05). There were significant differences between the experimental and control groups in the changes of upper motor neuron excitability and pain (p<.05), but not in the changes of lower motor neuron excitability and Korean Oswestry Disability Index.
Conclusion: Lumbar stabilization exercises utilizing concurrent contraction of deep and superficial muscles improved low back function in patients with LDH by lowering upper motor neuron excitability than compared to exercises actively moving the limbs. Lumbar stabilization exercises without pain have a positive impact on improving motor neuron excitability.
Neuron-specific enolase (NSE), a brain specific isoenzyme of the glycolytic enzyme, is characterized by its consistent occurrence in the cytoplasm of mature neurons. The purpose of the present study was to investigate the expression of NSE in the developing tongue of Korean native goats. The tongues of the fetuses were removed from 2- to 4-year-old female Korean native goats by caesarean section performed under general anesthesia. The expression of NSE in the developing tongue of goat fetuses (60, 90, 120, and 150 days) was studied using immunohistochemistry. In 60-day-old fetuses, NSE-immunoreactivity (IR) exhibited weak appearance in lamina propria of the basal portion and the apical epithelia of the tongue. In 90-day-old fetuses, NSE-nerve fibers were extended in the core part of connective tissue, and primordia of the taste buds was moderately positive. In 120-day-old fetuses, NSE-IR was strongly expressed in taste buds and gustatory nerve fibers. In neonates, the taste buds of vallate papillae were strongly positive for NSE, and development of nerve fibers was synapsed with connective tissue of well innervated taste buds. These results indicate that NSE expressions were associated with the sign of nerve innervation in prenatal development of goat tongues and NSE may be a useful neuronal marker to understand the development of gustatory nerve innervations.
Phytophagous insects detect volatile compounds produced by host and non-host plants, using species-specific sets of olfactory receptor neurons (ORNs). To investigate the relationship between the range of host plants and the profile of ORNs, single cell recordings were carried out to identify ORNs and corresponding active compounds in female Uraba lugens (Lepidoptera: Nolidae), an oligophagous eucalypt feeder. Based on the response profiles to 39 plant volatile compounds, 13 classes of sensilla containing 40 classes of ORNs were identified in female U. lugens. More than 95% (163 out of 171) of these sensilla contained 16 classes of ORNs with narrow response spectra, and 62.6% (107 out of 171) 18 classes of ORNs with broad response spectra. Among the specialized ORNs, seven classes of ORNs exhibited high specificity to 1,8-cineole, (±)-citronellal, myrcene, (±)-linalool and (E)-β-caryophyllene, major volatiles produced by eucalypts, while nine other classes of ORNs showed highly specialized responses to green leaf volatiles, germacrene D, (E)-β)-farnesene and geranyl acetate that are not produced by most eucalypts. We hypothesize that female U. lugens can recognize their host plants by detecting key host volatile compounds, using a set of ORNs tuned to host volatiles, and discriminate them from non-host plants using another set of ORNs specialized for non-host volatiles. The ORNs with broad response spectra may enhance the discrimination between host and non-host plants by adding moderately selective sensitivity. Based on our finding, it is suggested that phytophagous insects use the combinational input from both host-specific and non-host specific ORNs for locating their host plants, and the electrophysiological characterization of ORN profiles can be used for predicting the range of host plants in phytophagous insects.
Stem cell therapy is undoubtedly the most promising therapeutic approach for neurological disorders. Adipose tissue is ubiquitous and it can be easily harvested in large quantities under local anesthesia with little patient discomfort, making adipose tissue into the ideal large-scale source for research on clinical applications. In this study we monitored the neuronal cell differentiation potential of human adipocyte in the following condition; i) N2 medium containing 200 uM ascorbic acid (AA) and/or 10 uM flavonoid (F) and ⅱ) N2 medium containing AA and/or 10 ng/ml brain derived neurotrophic factor (BDNF) and/or, 200 ng/ml sonic hedgehog (SHH) plus 100 ng/ml fibroblast growth factor (FGF) 8. Adipose stem cells were cultured in above described differentiation condition for three weeks. RT-PCR analysis demonstrated that the mRNA levels of neuronal cell markers in differentiated adipose stem cells. Under the culture condition using N2 medium containing AA, the expression level of nestin (neural progenitor marker) m- RNA was high in all groups, while those of Neuro D, and LEP and FABP4 (adipocyte marker) mRNA were significantly decreased. Also, the addition of BDNF or SHH+FGF8 in N2 medium containing AA enhanced the neural cell differentiation from adipose stem cells, the expression level of Map2 (mature neuron) mRNA was increased, and that of TH (dopaminergic neuron marker) mRNA was high. In addition, we confirmed that the flavonoid addition has effect on the increase of Map2 expression. These results demonstrate that our designed culture condition has effect on the neural cell differentiation of adipose stem cells and this stimulatory effect may be further enhanced by transplantation.
Bicuculline is one of the most commonly used GABAд eceptor antagonists in electrophysiological research. Because of its poor water solubility, bicuculline quaternary ammonium salts such as bicuculline methiodide (BMI) and bicuculline methbromide are preferred. However, a number of studies have shown that BMI has non-GABAд eceptor-mediated effects. The substantia gelatinosa (SG) of the trigeminal subnucleus caudalis (Vc) is implicated in the processing of nociceptive signaling. In this study, we investigated whether BMI has non-GABA receptor-mediated activity in Vc SG neurons using a whole cell patch clamp technique. SG neurons were depolarized by application of BMI (20M) using a high Cℓ⁻ipette solution. GABA ( 30-100μM) also induced membrane depolarization of SG neuron. Although BMI is known to be a GABAд receptor antagonist, GABA-induced membrane depolarization was enhanced by co-application with BMI. However, free base bicuculline (fBIC) and picrotoxin (PTX), a GABAд and GABAс receptor antagonist, blocked the GABA-induced response. Furthermore, BMI-induced membrane depolarization persisted in the presence of PTX or an antagonist cocktail consisting of tetrodotoxin (Nα+ nnel blocker),AP-5 (NMDA receptor antagonist), CNQX (non-NMDA receptor antagonist), and strychnine (glycine receptor antagonist). Thus BMI induces membrane depolarization by directly acting on postsynaptic Vc SG neurons in a manner which is independent of GABAд receptors. These results suggest that other unknown mechanisms may be involved in BMI-induced membrane depolarization.
Sanguisorbae radix (SR) from Sanguisorba officinalis L. (Losaceae) is widely used in Korea and China due to its various pharmacological activity. The present study aims to investigate the effect of the methanol extract of SR on amyloid β Protein(25-35) (Aβ (25-35)), a synthetic 25-35 amyloid peptide, -induced neurotoxicity using cultured rat cortical neurons. SR, over a concentration range of 10-50 μg/ml, inhibited the Aβ (25-35) (10 μM)-induced neuronal cell death, as assessed by a 3-[4,5-dimethylthiazole-2-yl]-2,5-diphenyl-tetrazolium bromide (MTT) assay and the number of apoptotic nuclei, evidenced by Hoechst 33342 staining. Pretreatment of SR (50 μg/ml) inhibited 10 μM Aβ (25-35)-induced elevation of cytosolic calcium concentration ([Ca2+]c), which was measured by a fluorescent dye, fluo-4 AM. SR (10 and 50 μg/ml) inhibited glutamate release into medium induced by 10 μM Aβ(25-35), which was measured by HPLC, and generation of reactive oxygen species. These results suggest that SR prevents Aβ (25-35)-induced neuronal cell damage in vitro.

Continuous depth data are often required in applications of both onboard systems and maritime simulation. But data available are usually discrete and irregularly distributed. Based on the neuron network technique, methods of interpolation to the charted depth are suggested in this paper. Two algorithms based on Levenberg-Marquardt back-propaganda and radial-basis function networks are investigated respectively. A dynamic neuron network system is developed which satisfies both real time and mass processing applications. Using hyperbolic paraboloid and typical chart area, effectiveness of the algorithms is tested and error analysis presented. Special process in practical applications such as partition of lager areas, normalization and selection of depth contour data are also illustrated.
