우리 나라 재래종중(在來種中) 내용성분(內容成分) 면(面)에서 바람직한 향끽미(香喫味) 물질(物質)을 함유(含有)하고 있는 것으로 알려진 향초(香草)를 대상(對象)으로 이식시기(移植時期)가 향끽미(香喫味) 물질(物質)에 미친 영향(影響)을 구명(究明)하여 얻어진 결과(結果)는 다음과 같다. 전(全)알카로이드 함량(含量)은 7월(月) 5일(日) 이식구(移植區)가 4.16%로 가장 높고 5월(月) 20일(日) 이식구(移植區)는 1.37%로 가장 낮았으며, 니코틴 함량(含量)은 전(全)알카로이드 함량(含量)과 같은 경향(傾向)을 보였다. 전당(全糖) 함량(含量)은 4월(月) 20일(日) 이식구(移植區)가 12%로 가장 높았고 이것보다 이식(移植)이 빠르거나 늦을 수록 감소(減少)되는 경향(傾向)이었다. 석유(石油)에텔추출물(抽出物)은 4월(月) 5일(日) 이식구(移植區)가 5.77%로 가장 높았고 이식시기(移植時期)가 늦을 수록 감소(減少)하였다. 전당(全糖) 석유(石油)에텔추출물(抽出物) 전질소(全窒素)는 4월(月) 20일(日) 이식구(移植區)에서 가장 높았으며 이식시기(移植時期)가 빠를 수록 지방산(脂肪酸), malic산(酸)과 citric산(酸) 함량(含量)은 감소(減少)되었다. 이식시기(移植時期)가 빠를 수록 2.3-methylbutanoic산(酸), 3-methylpentanoic산(酸)의 함량(含量)이 증가(增加)되었다.
Nicotine is a major component of tobacco alkaloids. Nicotine is synthesized via putrescine N-methyl transferase (PMT), quinolinate phosphoribosyl transferase (QPT) and nicotine synthase. In this study, we down regulated the QPT activity using RNAi technique. RNAi vector was constructed by amplifying 500 bp region of QPT and inserted to pFGJJ374 vector in forward and reverse directions. We transferred the vector to Agrobacterium tumefaciens (LBA4404) and transformed Nicotiana tabacum (NC82). The nicotine contents of the transformant were significantly decreased by the transformation. The QPT gene expression of the transformant was decreased to 1/40 - 1/218 of the control from the quantitative RT-PCR. We made homogeneous transformed lines by segregation of the transformants. The low nicotine contents of the homogeneous lines were steadily maintained over the generations. The transformed lines could be used for low nicotine cigarette manufacturing.
Phenylalanine ammonia-lyase (PAL), a key enzyme of the phenylpropanoid biosynthesis pathway, is activated by a number of developmental and environmental cues. The coding region of the NtPAL4 gene was 2,154 bp in length, and its deduced protein was composed of 717 amino acids. Sequence analysis of NtPAL4 cDNA from tobacco (Nicotiana tabacum L.) revealed high structural similarity to PAL genes of other plant species. The NtPAL4 gene exists as a single copy in the tobacco plant, and its transcripts were strongly expressed in flowers and leaves. NtPAL4 expression was significantly induced in response to NaCl, mannitol, and cold treatments, but it was not induced by abscisic acid (ABA). NtPAL4 expression decreased gradually after treatment with ABA and H2O2; however, NtPAL4 transcripts accumulated after treatment with methyl viologen (MV). Our results suggest that the NtPAL4 gene may function in response to abiotic stresses.

Presowing treatements to seed are reasonable for elevating the seedling production efficiency. This experiment was carried out to determine the effects of GA3 [0 (water) : 0.01 mM] and light qulaity (red : white) simultaneously treated before sowing on seedling emergence and growth of tobacco (Nicotiana tabacum L.) cv NC 82 and Burley 21. Red or white light was illuminated 12 hours a day while seeds of the cultivars were imbibed with either water or GA3 0.01 mM for 3 days. Seedling emergence was measured up to 20 days after sowing but morphological characters and dry weights were done 40 days. The seedling emergence rate of cv NC 82 was more affected by light quality and GA3 pretreatments than that of cv Burley 21. Light quality more influenced the rate than GA3 while red light or water imbibition enhanced the rates of the two cultivars compared to the other treatement. Although mean plant height and leaf number per seedling were nearly equal between the treatements, red light declined leaf area per seedling of cv Burley 21 of which seeds were imbibed in water but white light decreased the areas of both cultiars of which seeds were soaked in GA3solution. Shoot dry weight was greater in cv NC 82 but less in cv Burley 21 pretreated with red light, and root dry weight showed the reverse result in white light, meaning that presowing light and GA3 treatments could be changed the seedling development and growth of tobacco.
It failed occasionally to take a reasonable emergence rate since tobacco (Nicotiana tabacum L.) seeds were planted on late Jan. showing lowest temperature. This experiment was done to measure the effect of GA3 (concentration ; period) , light quality (red ; white ; dark) during or after its treatment, daily irradiation hours( 0 ; 8 ; 12 ;16) and germination termperature (20 or 10 ℃ ocnstant ; 20/10℃ alternating) on the germination rate. Red and white light given during grmination showed no differences between the other daily irradiation hours except that 8 hours red light delayed germination although their 12 hours irradiation had the gratest rate. The rate was increased with increased concentration to GA3 0.01 mM or increased imbibition period to 3 days although the rate of cv. NC 82 was less than that of cv. Burley 21 in the case of dark imbibition of GA3 but daily 12 hours irradiation during germination. Light quality forced during GA3 imbibition eliminated such effect of GA3 shown in the darkness so that only light quality pretreatment and termperature during germinition were affected on the rate. The germination rate of thecultivars was decreased in the order of red, white light, darkness meaning that it was highly influenced by the light quality during GA3 treatment. Regardless of GA3 or light quality treatment,on the other hand, the rate was greater in 20 ℃ constat than 10℃ constant and 20/10℃ alternating germination temperature having similar germinative patterns.
연초(N. tabacum L.)의 세포질 웅성불임 F1 식물체와 N. africana의 종간교배에 의한 웅성불임 반수체식물의 육성 및 특성을 조사한 결과는 다음과 같다. 1. (CMS NC82 burley 21) F1 과 N. africana의 종간교배에 의한 세포질 웅성불임 반수체 식물의 출현빈도는 삭당 0.5주로 나타났다. 2. 종간교배으로 얻은 웅성불임 반수체식물 중 잎과 줄기의 색이 green(황색종)과 yellow(버어리종)의 분리비는 3 : 1로 나타났다. 3. 반수체 식물 중 잎과 줄기의 색이 yellow(버어리종 type)인 개체에 TMV저항성을 검정한 결과 저항성과 이병성의 비는 1 : 1로 나타났다. 4. (CMS MC82 Burley 21)F1 과 N. africana의 종간교배으로 세포질 웅성불임 isogenic Burley 21 반수체 식물의 육성이 가능하였고 이 반수체 식물의 염색체를 배가함으로써 여교배육종법에 비하여 조기에 세포질 웅성불임 Burley 21을 육성할 수 있을 것으로 생각된다.
연초의 고향품종 육성의 기초결과를 얻기 위하여 향기성분과 관련이 있는 DVTs와 cis-abienol의 유전양상을 조사하였던 바 얻은 결과는 다음과 같다. 1. TLC 방법은 연초 분리집단에서 DVTs와 cis-abienol의 분석 및 이들 성분을 함유한 계통선발에 매우 유용하게 이용될 수 있었다. 2. DVTs와 cis-abienol의 유전양상이 단일우성인자에 의하여 지배되었고 서로 독립적으로 유전을 하였다. 3. DVTs오 cis-abienol의 유전양상이 단일우성인자에 의하여 지배됨으로 연초 고향 품종 육성에 있어서 이들 형질의 도입은 쉬울 것으로 생각된다.
The effect of growth regulators (NAA, BA and GA_3) and light (blue, red and far-red) on the changes in total protein and thylakoid membrane protein pattern of callus from intergeneric protoplast fusion between Nicotiana tabacum and Solanum nigrum were investigated. When the callus were irradiated with different wavelengths of light, blue and red light accelerated the synthesis of total proteins and thylakoid membrane proteins. Particularly, red light led to an increase in the protein synthesis compared to blue light. When the callus were subjected to various combinations of growth regulators, NAA+GA_3 and NAA-BA treatments induced remarkable increase of total proteins and thylakoid membrane proteins accumulation, particulary in the combination of NAA+GA_3 NAA+GA_3 treatment with irradiation of red light showed highest value in the accumulation of total proteins and thylakoid membrane proteins. We conclude that simultaneous application of red light and NAA+GA_3 treatment may induce synergistic effect in the synthesis of total proteins and thylakoid membrane proteins.
Burley 종 연초재배에 있어서 석회시용의 타당성을 구명하고자 Burley 21을 공시품종으로 석회무시용구, 황산칼슘(Ca : 35kg/10a), 탄산칼슘(Ca : 35kg/10a) 농용석회(pH 6.5 조정량) 시용구를 두고 수량, 엽중 무기 및 유기성분을 분석한 결과는 다음과 같다. 1. 석회무시용구에 비해 황산칼슘 시용구는 토양 및 엽중 무기성분 함량에서 대차를 보이지 않았으나, 탄산칼슘과 농용석회시용구는 Ca, mg 함량이 높아지고 Al, Mn, Fe 함량은 감소하였다. 2. 담배 수량은 석회무시용 < 황산칼슘 < 탄산칼슘, 농용석회순으로 나타났으나, kg 당 가격은 차이가 없었다. 종자 생산량은 무시용 < 농용석회 < 황산칼슘, 탄산칼슘순으로 증가하였다. 3. 무시용구에 비해 황산칼슘시용구는 건조엽의 암모니아 태질소, 비수용성알칼리 계수가 높았으며, 탄산칼슘시용구는 수용성 및 비수용성알칼리 계수, 향기성분 함량이 높았으나, 단백태질소, 석유에텔추출물 함량은 낮았다. 농용석회시용구는 질산태질소, 수용성 및 비수용성알칼리계수, 향기성분 함량은 높았으나, 단백태질소 암모니아태질소 니코틴 석유에텔추출물 함량은 낮았다. 석회시용에 의한 엽중 화학성분의 변화는 장, 단점이 상쇄되어 품질에 큰 영향을 미치지 않을 것으로 생각된다. 따라서 토양 pH를 미산성 (pH 6.5내외)으로 유지시킬 수 있는 석회 시용이 필요할 것이다.
