        본 연구는 붉바리 어미의 사육환경에 따른 난질의 변화를 난의 생화학적 분석을 통하여 난질 에 영향을 미칠 수 있는 요소를 구명하기 위하여 수행하였다. 그 결과 육상수조에서 사육하는 것보다 해상가두리에서 사육할 때 부상률, 수정률, 난 발생 생존율 및 부화율이 우수하였다. 그 리고 해상가두리에서 생산한 수정란의 유리아미노산의 함량이 높았다. 즉, 해상가두리에서 사 육관리할 때 난질이 우수한 수정란을 생산할 수 있고, 난질이 우수할수록 유리아미노산의 함 량이 높았다.

        본 연구는 붉바리 배란유도를 위해 다양한 호르몬과(ovaprim, pimozide, LHRHa, HCG)과 LHRHa 의 농도별 효과를 조사하였다. LHRHa는 50 μgkg-1, 100 μgkg-1, 150 μgkg-1, 200 μgkg-1의 농 도로 처리하였다. 호르몬은 등 근육에 주사하였으며, 조사 결과 LHRHa 단독, 그리고 LHRHa와 Pimozide를 혼합하여 투여했을 때 가장 효과적이었으며, 두 실험구 사이의 차이는 거의 없는 것으로 나타났다. 따라서 LHRHa를 단독으로 사용하는 것이 가장 효율적인 것으로 판단된다. 그리고 LHRHa를 다양한 농도로 처리하여 배란유도 효과를 조사한 결과 100 μgkg-1의 농도로 투여했을 때 가장 우수한 것으로 조사되었다.

        붉바리(Epinephelus akaara) 종자생산 시 기형 발생에 의한 손실이 크지만 기형어에 대한 생물 학적 정보는 많지 않다. 본 연구에서는 부화 후 96일 붉바리 치어를 정상 그룹과 두 유형의 기 형 그룹(머리, 턱)으로 나누어 형태형성과 연관된 4개의 주요 유전자(insulin like growth factor 1: IGF-1, bone morphogenic protein 4: BMP4, peroxisome proliferator-activated receptors γ: PPARγ, matrix Gla protein: MGP) 발현을 조사하였다. 각 그룹에서 뇌, 간 및 근육을 잘라낸 다 음 total RNA를 추출한 후 real-time PCR을 사용하여 유전자 발현 차이를 비교하였다(n=20). 부화 후 96일 붉바리 치어에서 IGF-1과 BMP4 유전자는 기형 그룹의 뇌와 간에서 정상 그룹과 비교하여 유의한 발현 차이를 나타냈다(p <0.05). 반면에 PPARγ와 MGP 유전자는 어떤 조직 에서도 정상 그룹과 기형 그룹 사이에 유의한 발현 차이를 보이지 않았다. IGF-1과 BMP4 유전 자는 치어 단계의 붉바리 기형 상태와 관련되어 있는 것으로 보인다.

        To determine the optimum water temperature (a temperature which minimizes metabolic activities and stress in fish) for long distance transportation of red-spotted grouper (Epinephelus akaara), by evaluating the behavior, survival and physiological and hematological responses to variable water temperature conditions (9, 12, 15, 18 and 21℃) for 48 hours. Fish exposed to 9℃ died at 48 hours exposure, but those exposed to 12, 15, 18 and 21℃ treatment groups all survived. Fish in the 15, 18 and 21℃ exposure groups exhibited normal swimming, while those exposed to 12℃ were observed to be stationary at the bottom of the tank. The plasma cortisol and glucose concentration were higher in fish exposed to 12℃ than at other temperature conditions. The fish in the 12 and 15℃ groups had significantly higher hematocrit (Ht) and hemoglobin (Hb) than those in the 18 and 21℃ groups (P<0.05). Levels of aspartate aminotransferase (AST), NH3 and osmolality showed a pattern similar to the levels of plasma cortisol, while alanine aminotransferase (ALT) and total protein did not significantly vary among the experimental groups. These results indicate that the optimum water temperature for long distance transportation of the red-spotted grouper is 15℃.

        Skeletal deformities are significant problems that affect the growth and commercial value of fish reared in hatcheries. However, studies of bone metabolic process related to skeletal deformities are limited. We investigated the potential correlation between bone deformities and plasma calcium, phosphorus, and estradiol-17β levels in reared red spotted grouper (Epinephelus akaara) juveniles. We collected E. akaara frys from private farms at 110, 140, 180 and 300 days after hatching (DAH), and classified the normal and deformed fish by observing their external shape and inner frame by soft X-ray. We also analyzed the calcium, phosphorous, and estradiol-17β levels in their plasma. A comparison between normal and deformed fish, indicated that calcium and estradiol-17β levels were higher in deformed fish than in the normal at 180 and 300 DAH. The level of phosphorus was also higher in deformed individuals than in normal fish, but only at 300 DAH. These results suggest that skeletal deformities are associated with increases in plasma calcium, phosphorus, and estradiol-17β levels.

        In fish, light (photoperiod, intensity and spectra) is main regulator in many physiological actions including body growth. The present study was to investigate the effect of light spectra on the body growth in red spotted grouper Epinephelus akaara. Experiment performed from December, 2016 to March, 2017 and fish (n=200, body weight 6.9±0.1, total length 7.5±0.1) was reared under different light spectra (blue, green, red and white) for 15 week under 12 hour light and 12 hour dark photoperiod and natural water temperature (13.6~16.7℃). We examined the mRNA expression levels of growth hormone (GH) in the brain with pituitary and photoreceptors mRNA expression in the retina. Red spotted grouper of BW was no different in each for the four groups (light spectra of blue, green, red and white) to five week of rearing period, but BW of rearing blue light spectra group fish was significantly increased in ten week of rearing period. After that, in 15week of rearing period, BW of four group fish was no showed difference respectively. The total length (TL) of fish was no differed in four experimental groups under the experimental duration respectively. The gh mRNA was more highly expressed in rearing blue light spectra group fish than other experimental groups in 15 weeks. The short wavelength sensitive opsin (sws) mRNA was more increased in rearing blue light spectra group fish than other groups in 15 weeks. The middle wavelength sensitive opsin (mws) and rod opsin mRNA expression was no differed in four experimental groups under all experimental durations respectively. The long wavelength sensitive opsin (lws) mRNA expression was more increased in rearing red light spectra group fish than other groups in 15 weeks. Our present results suggest that somatic growth of red spotted grouper is induced under blue light condition under low water temperature duration. Also, these results are thought to be affected of the gh and sws mRNA expression blue wavelength. However, in this experiment we did not confirm the correlation between GH and photoreceptor. Therefore, further studies are needed to clarify the correlation of the light spectra and photoreceptor with body growth in red spotted grouper.

        Red spotted grouper, Epinephelus akaara, is a popular aquaculture species and a protogynous hermaphrodite. Induction of artificial sex change at the time of primary sex differentiation is of interest but has not been successful due to the lack of necessary basic information. To find out the potential neuroendocrine influence on the primary sex differentiation, the expression of key genes in the brain was investigated during the formation of ovarian cavity. Expression of cyp19a1b, esr1, gnrhr1, fsh, lh and cga in the brain was positively associated with the formation of ovarian cavity, showing gradual increase as the formation proceeds. However, the expression of gnrh1 was suppressed during the early part of the ovarian cavity formation, signifying potential hypothalamic influence on the primary sex differentiation in this species.

        The objective of the current study was to determine acute plasma stress responses in two size groups of juvenile Epinephelus akaara (average body weight: 8.4±2.1 and 3.3±0.6 g; 150 and 120 days after hatch, respectively) exposed to abrupt salinity drops (from 34 practical salinity unit, PSU seawater to 18, 10 PSU (experiment 1) or 26, 18, 10 PSU (experiment 2), respectively). Plasma glucose, glutamic oxalate transaminase, glutamic pyruvate transaminase, red blood cell counts, and gill histology were determined during 72 h exposure. Significantly increased plasma glucose, glutamic oxalate transaminase levels, and red blood cell counts were observed in fish exposed to 18 or 10 PSU. Histological changes, such as hyperplasia and lifting of epithelium in the gill secondary lamellae, were also observed in fish exposed to 18 or 10 PSU at 72 h post-drop. E. akaara exposed to sudden salinity drops to 18 or 10 PSU still seems to undergo the primary adjustment phase before fish reaches a new homeostasis, whereas fish exposed to 26 PSU seems to mount osmotic changes. Therefore, the no observed adverse effect levels for 72 h acute salinity challenge was 26 PSU in our study, and salinity drop to 18 PSU and below can possibly cause acute adverse effect, in which fish could be vulnerable to additional stresses such as a temperature changes or handling stress.

        The effect of sudden changes of water temperature (WT) on the survival rate and physiological responses of the red spotted grouper (Epinephelus akaara) were examined by manipulating WT control system for 9 days. Experimental condition was divided in two different regimes at low (from 10°C to 4°C, decreased 1℃/d) and high (from 28°C to 34°C, increased 1°C/d) WT. Survival rate of experimental fishes were observed, and determined the changes of hematological characteristics by analyzing plasma levels of cortisol, glucose, total protein, and electrolytes (Na+, Cl–, K+). No mortality was observed until low WT 6°C (144 h) and high WT 32°C (96 h), and 100% mortality was observed at low WT 4°C (216 h) and high WT 35°C (171 h). Plasma levels of cortisol and glucose increased rapidly as decreasing WT, and the loss of swimming ability and respiration response was observed at low WT 7°C and high WT 34°C conditions.

        We observed the osteological development of larval and juvenile red spotted grouper (Epinephelus akaara) in order to generate data for the assessment of skeletal deformities and to inform phylogenetic systematics research. Larvae and juveniles were obtained from a aquafarm in Muan-gun, Jeolla-namdo Province, Korea. The average water temperature at the time of breeding was 23.0°C and average water salinity was 33.0 psu. Freshly hatched fish larvae had not undergone any ossification, but ossification of the parasphenoid bone, which forms the base of the cranium, occurred as the juveniles reached an average body length (BL) of 2.49 mm. At the same time, ossification of the preopercle and opercle occurred in the operculum, and ossification of the maxilla, which forms the upper jaw, and the dentary bones, which form the lower jaw, began. In addition, ossification of the vertebra occurred by formation of 7 vertebral centra and the neural spine in the abdominal vertebra. When the juveniles reached an average (BL) of 5.22 mm, ossification of the nasal, lateral ethmoid, and alisphenoid bones occurred in the cranium; ossification of the endopterygoid and metapterygoid bones began in the palatine region; and ossification of the hypohyal and interhyal bones occurred in the hyoid arch. At an average (BL) of 20.9 mm, ossification of the basisphenoid bone in the cranium and the suborbital bone in the orbital region occurred. Ossification of the vertebra then occurred by the formation of long pairs of ribs from the third to the ninth abdominal vertebrae, completing osteological development.

        This study was conducted in order to examine the egg development in red spotted grouper, Epinephelus akaara and the morphological development of its larvae and juveniles, and to obtain data for taxonomic research. This study was conducted in June 2013, and 50 male and female fish were used for the study. One hundred μg/kg of LHRHa was injected into the body of the fish for inducing spawning, and the fish were kept in a small-sized fish holder (2×2×2 m). Eggs were colorless transparent free pelagic eggs, 0.71–0.77 mm large (mean 0.74±0.02 mm, n=30), and had an oil globule. Hatching started within 27 h after fertilization. Pre-larvae that emerged just after hatching were 2.02–2.17 mm in total length (mean 2.10±0.11 mm), their mouth and anus were not opened yet, and the whole body was covered with a membrane fin. Post-larvae that emerged 15 days post hatching were 3.88–4.07 mm in total length (mean 3.98±0.13 mm), and had a ventral fin with two rays and a caudal fin with eight rays. Juveniles that were formed at 55 d post hatching, were 31.9–35.2 mm in total length (mean 33.6±2.33 mm), with red color deposited over the entire body, and black chromophores deposited in a spotted pattern. The number of fin rays, body color, and shape were the same as that in the adult fish.
