A viral histone H4 (=CpBV-H4) is encoded in a polydnavirus, Cotesia plutellae bracovirus, and symbiotically associated with an endoparasitoid wasp, C. plutellae. It has an extended N-terminal tail consisting of 38 amino acid residues, compared to the host H4 and this extended N-terminal tail has been postulated to play a crucial role in an epigenetic control of gene expression. The (SSH) suppression subtractive hybridization analysis was analyzed in transcriptome by short-read sequencing technology. The SSH analysis provided several target and nontarget genes of a viral histone H4. In this study, we analyzed the effect CpBV-H4 on the expression of two target genes serpins and histone lysine N-methyl transferase. Transient expression of CpBV-H4 by microinjecting recombinant expression vector to non parasitized larvae of Plutella xylostella showed that it was expressed up to 70 h. Under this transient expression condition, we analyzed the effect of CpBV-H4 on the expression of target genes by RT-PCR at different time points. Interestingly, the CpBV-H4 significantly inhibited the expression of target genes after 44 h, while the truncated CpBV-H4 deleting the N-terminal tail did not show the inhibitory activity.
본 연구는 수수× 수수 교잡종의 톤백 및 소포장 사일리지의 부위에 따른 사료가치 및 품질에 미치는 영향을 구명하기 위하여 천안에위치한 국립축산과학원 자원개발부에서 수행되었다. 수수× 수수 교잡종 SS405는 5월 초순에파종한 다음 출수기에 수확하여 사일리지로 조제하였다. 톤백 사일리지의 조단백질 함량은소포장 사일리지에 비해 약간 감소되었으나 통계적인 차이는 보이지 않았다. 그러나 NDF 및TDN 함량에서는 톤백과 소포장 사일리지 사이에 유의적인 차이가 나타났다(p<0.05). 수수×수수 교잡종의 톤백 및 소포장 사일리지는 젖산균의 첨가유무에 상관없이 비슷한 함량을 보였으며 사일리지 부위별 조단백질, NDF, ADF및 TDN 함량도 차이를 보이지 않았다. 사일리지 부위별 조단백질, NDF, ADF, TDN 함량 및in vitro 건물소화율에서는 차이를 보이지 않았다. 톤백 곤포사일리지의 젖산 함량은 소포장사일리지보다 감소하였으나 유의적인 차이는나타나지 않았다. 그러나 톤백 곤포사일리지의초산 함량은 소포장 사일리지보다 유의적으로감소하였다(p<0.05). 또한 사일리지 부위별 젖산, 초산 및 낙산 함량은 큰 차이를 보이지 않았다. 따라서 본 연구의 결과를 요약하면 수수×수수 교잡종 소포장 및 톤백 사일리지의 부위별 사료가치와 품질은 부위에 따라 영향을 받지 않았다.
This study was carried out to investigate the productivity and nutritive values of SSH under Double-cropping SSH followed by whole crop barley (SSFWB) or by Hairy vetch (SSFHV) in paddy land. A field study was conducted at Baeksan-myeon, Kimje, Jeonbuk province in Korea on clay loam soil for 3 years. All plots were arranged in completely randomized design with three replicates. The yield of SSH in SSFWB treatment was higher t㏊n t㏊t of SSFHV (P<0.05). All nutrients values of SSH were not influenced by the cropping system. However, all nutrients of SSH in SSFWB treatment were just a little increase, compared with those of SSFHV.
The SSH100-10 bacterial strain, which exhibits strong antifungal (anti-mold and anti-yeast) activity, was isolated from traditional korean soysauce aged 100 years. The strain was identified as Bacillus velezensis based on Gram-staining, the biochemical properties and 16S rRNA gene sequence determination. B. velezensis SSH100-10 showed strong proteinase activity and NaCl tolerance, but did not produce enterotoxin. Two-antifungal compounds from B. velezensis SSH100-10 were purified using SPE, preparative HPLC, and reverse phase-HPLC. The purified antifungal compounds were identified as C14 and C15 iturin through MALDI-TOF-MS and amino acid composition analysis. The stability characteristics of the antifungal compounds after temperature, pH, and enzyme treatments suggested that B. velezensis SSH100-10 produced more than two antifungal compounds; pH-stable C14 iturin A and C15 iturin A, and unidentified pH-unstable compounds. The results suggested that B. velezensis SSH100-10 can be used in soybean fermentation as a starter. Moreover it has potential as a biopreservative in the food and feed industry and as a biocontrol agent in the field of agriculture.
