        Background : Sorghum (Sorghum bicolor) is the fifth most important crop in the world. Although it is known as a crop with many medicinal action, Studies on sorghum breeding are inferior to other crop. Therefore, it is necessary to cultivate excellent varieties of sorghum which is stable and has excellent physiological activity. So, this study was conducted to investigate the total phenolic content and the total flavonoid content of the sorghum accessions to select the sorghum accessions with rich antioxidants. The results of this study can be used as basic data for future breeding. Methods and Results : The seeds for study were supplied from the Center for Agricultural Genetic Resources of National Institute of Agricultural Sciences. The seeds were extracted with 100% methanol and concentrated at 45℃. To determine antioxidant, we measured the total phenolic content and total flavonoid content. The highest total phenolic content showed in the accession from IT No. 155552 (55600.34 ± 17.78 ㎎·GAE/g) and the total flavonoid content showed the highest in IT No. 143716 (116563.20 ± 149.39 ㎎·QE/g). On the other hand the lowest total phenolic content showed in the accession from IT No. 135777 (20.14 ± 4.29 ㎎·GAE/g) and the total flavonoid content showed the lowest in IT No. 152910 (701.25 ± 5.53 ㎎·QE/g). Conclusion : Among 300 sorghum accessions, we analysed the total phenolic contents and total flavonoid contents.

The seeds were extracted with 100% methanol and concentrated at 45℃. To determine antioxidant activity, we measured the DPPH radical scavenging ability and ABTS radical scavenging ability. The highest DPPH radical scavenging activity showed in the accession from IT No. 158264 (RC50 : 5968.71 ± 4986.24 ㎍/㎖) and the ABTS radical scavenging activity showed the highest in IT No. 143744 (RC50 : 1558.47 ± 273.21 ㎍/㎖). On the other hand the lowest DPPH radical scavenging activity showed in the accession from IT No. 155497 (RC50 : 3.26 ± 20.06 ㎍/㎖) and the ABTS radical scavenging activity showed the lowest in IT No. 152910 (RC50 : 3.31 ± 20.10 ㎍/㎖). Conclusion : Among 300 sorghum accessions, we analysed the antioxidant activities of DPPH and ABTS.

        Background : Sorghum is a major cereal food crop used in many parts of the world. It has been grown on a subsistence level by farmers, under various conditions of environmental stresses in the semi-arid tropics of Africa and Asia. This crop has received significant attention because of its ability to lower cholesterol in the blood, and its anti-dementia, antioxidant, and antimicrobial properties. It is possible to develop a functional and commercially viable sorghum variety by using superior cultivars of sorghum. The objective of this study was to construct a database of superior sorghum accession. Methods and Results : We used 250 sorghum accessions collected from different geographical bioregion in Korea. We determined various growth characters including germination rate and ear length of these accessions. To determine the antioxidant capacity, we measured the DPPH radical scavenging activity, ABTS scavenging activity, total phenolic contents, and total flavonoid contents. 189 accession showed higher germination (> 80%) than other accessions. Higher DPPH radical scavenging activity was observed in 11-SB-078 (RC50: 1.89 ± 2.88 ㎍/ ㎖), and higher ABTS scavenging activity was recorded in 11-SB-116 (RC50: 35.48 ± 2.42 ㎍/㎖) than other accessions. The ear length ranged from 15 ㎝ to 48 ㎝, the total phenolic contents ranged from 3 ㎎·GAE/g to 77 ㎎·GAE/g, and total flavonoid contents ranged from 0.09 ㎎·QE/g to 1.07 ㎎·QE/g. Conclusion : Among 250 sorghum accessions, we selected 10 with both superior agronomic characters and higher functional food quality.

        This study was conducted to determine the optimal dose of gamma-ray for mutation breeding in sorghum (Sorghum bicolor M.). Gamma-rays irradiated to dry seeds with various doses (0 to 1000 Gy) at Korea Atomic Energy Research Institute. Lethal dosage (LD50) was approximately 256 Gy. Significant decreases in growth characteristics (plant height, tiller number and fresh weight) were observed by dose of increased over 300 Gy. Reduction doses (RD50) was approximately 363 Gy in plant height. We also conducted comet assay to observe nucleus DNA damage due to gamma irradiation. In comet assay, a clear difference was identified over 100 Gy treatments. With increasing doses of gamma-ray in the range of 100 to 400 Gy, the rate of head DNA was decreased significantly from 93.52% to 67.57%. The most of the Sorghum cells were severely damaged in the integrity of DNA by gamma-ray. These data provide valuable information when the optimal dose should be chosen for purpose of mutation breeding program of Sorghum.

        본 연구에서는 붉은장목수수(Sorglum Bicolor M.) MeOH 추출물과 분획물들의 다양한 생리활성을 비교하기 위하여 항산화(DPPH assay, 환원력, 총 페놀 및 플라보노이드 함량), 항당뇨(α-Glucosidase 저해활성 및 α-Amlyase 저해활성), 항암(MTT assay) 활성을 관찰 하였다. 항산화활성의 경우 EtOAc fraction이 높은 총 페놀과 플라보노이드 함량을 나타내었으며, DPPH assay와 reducing power 결과 역시 EtOAc fraction과 MeOH fraction이 positive control로 사용한 α-tocopherol보다 우수한 활성을 나타냈다. α-Glucosidase, α-amlyase 저해능 평가 결과 D.W. fraction이 가장 높은 활성을 보였으며 n-BuOH fraction과 MeOH extract도 활성을 나타내었다. 암세포인 AGS, HT29, HCT116세포주에 대한 세포독성 연구인 MTT assay에서는 EtOAc, n-BuOH, D.W. fraction을 처리한 처리구에서 독성을 보였으며 이중 n-BuOH fraction이 모든 세포주에서 농도 의존적으로 세포독성을 가지는 것을 확인할 수 있었다. 연구결과 붉은장목수수의 추출물 EtOAc fraction의 항산화활성, D.W. fraction의 항당뇨활성, n-BuOH fraction의 암세포에 대한 독성과 관련된 화합물의 분리 및 구조규명에 관한 연구가 필요 할 것으로 사료되어지며, normal cell에 대한 MTT assay를 진행하여 항암활성에 관한 연구를 진행하여 붉은장목수수 추출물을 이용한 다양한 건강기능식품과 의약품 개발을 통해 건강 증진과 질병으로부터 보호 받을 수 있을 것으로 기대한다.
