        This study was investigated the synergistic antibacterial effects from the combination of sulfamethazine (SMZ) and tiamulin (SMT) against Salmonella Typhimurium (S. Typhimurium) and Actinobacillus pleuropneumoniae (A. pleuropneumoniae) isolated from pigs. Minimum inhibitory concentrations (MICs) and fractional inhibitory concentration (FIC) index were determined for 10 bacteria from 13 swine farms in Gyeongsangnam province by the National Committee for Clinical Laboratory Standards broth microdilution procedure. Antimicrobial agents tested included SMZ, tiamulin and SMT. Against S. Typhimurium and A. pleuropneumoniae, the MIC ranges of SMZ, tiamulin and SMT were 125-500, 15.625-62.5 and 15.625-62.5 μg/mL, respectively. In addition, the FIC index ranges of SMT against S. Typhimurium and A. pleuropneumoniae were 0.19-0.38 and 0.19-0.62, respectively. In conclusion, SMT has a high antibacterial activity against S. Typhimurium and A. pleuropneumoniae, then the combination may be applied for the treatment of swine diseases caused by S. Typhimurium and A. pleuropneumoniae.

        Oxidation of erythromycin, sulfamethazine and sulfathiazole by ozone was experimentally investigated to see the effects of background water quality such as ultrapure water, humic acid and biologically treated wastewater and water temperature on the removal rate, consequently to provide design information when the ozone treatment process is adopted. Initial concentration of the antibiotics was spiked to 10 μg/l and ozone dose was 1, 2, 3, 5, 8 mg/l. While the removal rate of erythromycin under ultrapure water background by ozone oxidation was over 99%, that under humic acid and biologically treated wastewater background was markedly reduced to the range of 59.8%~99% and 17.0%~99%, respectively. When water temperature is decreased from 20℃ to 4℃, the removal rate is reduced from the range of 17.0%~99% to the range of 9.4%~97.4% under biologically treated wastewater background. The effects of background and temperature on the removal rate of sulfamethazine and sulfathiazole were similar to erythromycin, but the degree was different. Therefore, it is concluded that the background of water to be treated as well as water temperature should be taken into consideration when the design factor such as ozone dose is determined to meet the treatment objective in the ozone treatment process.

        Antibiotics polymer prepared by chemical bonding and simple blending of antibacterial into polymers have attracted much interest because of their long-lasting and antibacterial activity. Antibiotics polymer can significantly reduce losses associated with dissolution, photolytic decomposition and volatillization. Further more, increased efficiency safety and selectivity are additional benefits which may be realized. In this study, Antibiotics polymer was synthesized by chemical reaction of polyacrylic acid with sulfamethazine by N,N'-dicyclohexylcarbodiimide(DCC) method. Antibacterial susceptibility was determined against Streptococcus pyrogenes[gram(+)] and Esherichia coli.[gram(-)] using a standardized disc test. As a result, the synthetic antibiotics polymer exhibited the broad susceptibilty against Streptococcus pyrogenes and Esherichia coli. Especially, the antibiotic effect of antibacterial polymer against Gram negative(Esherichia coli) was much stronger than that against Gram positive(Streptococcus pyrogenes).

        Sulfonamides, a therapeutically important group of antimicrobial drugs, are widely used to treat and prevent the acute systemic and skin infections in dairy cattle. They also pose an economic hazard through inhibition of growth of dairy starter cultures. This study was carried out to compare four screening methods for detection of sulfamethazine in milk and determine the positive milk sample by HPLC method. Sulfamethazine was used to spike at five levels of sulfamethazine. The Lac-Tek test and CharmII test were also consistent better than TTCII test and Delvo SP test in sulfamethazine detection. Analysis probabilities of obtaining a positive response with TTCII test and Delvo SP test assay at 50 ppb sulfamethazine level in milk samples were only 14%, 42% each. Whereas using the Lac-Tek test and CharmII test would have resulted in 100% identification of the five levels. Determination of sufamethazine using the HPLC method in the spiked milk were 10.64, 19.30, 30.76, 38.83 and 50.23 ppb, respectively.

        To control the maximum residue level (MRL) for sulfamethazine (SMZ) residues in pork tissue, a microbial inhibition method is a regulatory screening assay method in Korea. Microwell plate-based competitive enzyme immunoassay (ELISA) kit is available for routine screening of SMZ residues in pork tissue. One ELISA kit is evaluated. Phosphate buffer extracts of samples fortified with SMZ at 0, 1, 5, and 10 ng/g were used in a recovery teat of the kit. Market pork samples were assayed by the kit. Recovery of sulfamethazine was 104% at 10 ng/g. Intraassay variations and interassay variations for the kit were 7.70% and 5.76%, respectively. Concentration causing 50% inhibition of color development compared with blanks was 16.4ng. The violative pork samples with over MRL (0.1 ug/g) was 4 of 32 cases (12.5%) by used ELISA kit. This result indicates a possibility of the ELISA kit for screening test of SMZ residues in pork tissue, and still needs a comfirmatory assay for mandatory purposes.

        A screening method has been developed for detecting sulfamethazine(SMZ) contamination of meat or feeds by using horseradish pero×idase (HRP) labeled protein A (Prot A-HRP)and an indirect competitive enzyme-linked immunosorbent assay(ELISA). The assay is based on competitive binding of guinea pig anti-SMZ with SMZ in sample and SMZ-gelatin con$lt;jugate(SMZ.GEL) followed by the uptake of prot A-HR,P onto polystyrene microwell plate coated with SMZ.GEL. Percent binding B/Bo × 100) was calculated from the absorbance in the absence (BO) and presence (B) of SMZ. By the sandard curve prepared by plotting log(SMZ) vs percent binding of each lmown reference solution, the detection limit was 1.0 ppb or leas. Cross reaction with sulfadimethoxine, sulfaguaniding, sulfamerazine, sulfamthoaypyridazine, sulfanilamide, aulfisomidine and aulfisoxazole were not observed. But sulfamerazine crosareacted in the test. The EC-50 value (concentration causing 50% inhibition of color development compared with blank) of sulfamerazine was 2.0 ppm. Further quality control will make the ELISA system ideal for the detection of SMZ in meat or feeds.
