Background : The Production of high quality ginseng seedling become increasingly important in the cultivation of ginseng because the quality of ginseng seedling is closely related to yield of ginseng. However, it has become difficult to produce high quality ginseng seedling due to environmental instability such as an increase in the occurrence of abnormal climate and pests, and the difficulty in obtain new place to cultivate ginseng seedling. This study was carried out to analyze the growth characteristics of ginseng seedling according to application of fermented compost during ginseng seedling cultivation Methods and Results : In order to evaluate the effect of fermented compost on the growth of ginseng seedling, fermented compost containing antagonistic microorganism was prepared and applied 300kg per 10a to planned place for ginseng cultivation. After seeding of the ginseng seeds, the growth of ginseng was analysed. The pH, EC, organic matter, P2O5 and NO3-N of the compost treated group were 6.1, 2.75 ds/m, 18.97 g/㎏, 1,202 ㎎/㎏ and 287.4 ㎎/㎏, respectively, while those of non-treated group were 6.6, 0.25 ds/m, 14.67 g/㎏, 1,055 ㎎/㎏ and 8.77 ㎎/㎏, respectively. As a result of analyzing the growth characteristics of ginseng seedling, the growth of aboveground part was not significantly different between the treated and non-treated group. The root length and root diameter were 13.0 ± 1.50 ㎝ and 5.5 ± 0.27 ㎜, respectively, which were not significantly different from non-treated group. However, root weight was 1.20 ± 0.14 g, which is 1.4 times higher than that of control. Conclusion : It was confirmed that the quality of ginseng seedling was improved by compost application. These results suggested that fermented compost could be used as a material for nutrients management in ginseng seedling cultivation.

Lectins belong to the pattern-recognition receptors (PRRs) class and play important roles in the recognition and elimination of pathogens via the innate immune system. Recently, it was reported that lily-type lectin-1 is involved when a pathogen attacks in the early immune response of fish. However, this study is limited to information that the lectin is involved in the innate immune response against viral infection. In the present study, the lily-type lectin-2 and -3 of Oplegnathus fasciatus (OfLTL-2 and 3) have been presented to be included B-lectin domain and two D-mannose binding sites in the amino acid sequence that an important feature for the fundamental structure. To investigate the functional properties of OfLTLs, the tissue distribution in the healthy rock bream and temporal expression during early developmental stage analysis are performed using quantitative real-time PCR. OfLTL-2 and 3 are predominantly expressed in the liver and skin, but rarely expressed in other organ. Also, the transcripts of OfLTLs are not expressed during the early developmental stage but its transcripts are increased after immune-related organs which are fully formed. In the challenge experiment with RBIV (rock bream iridovirus), the expression of OfLTLs was increased much more strongly in the late response than the early, unlike previously known. These results suggest that OfLTLs are specifically expressed in the immune-related tissues when those organs are fully formed and it can be inferred that the more intensively involved in the second half to the virus infection.

Background : Some of invasive plants, which were introduced from foreign countries, have caused problems in Korea. Invasion of these invasive plants in the ecosystem threatens the habitat of endemic species, reducing biodiversity, and causing a disturbance in the ecological system. Hypochaeris radicata L. (Asteraceae), the most invasive plants in Korea, particularly in Jeju Island, invade farmland, and autochthonous forest, establishing monocultures and modifying the ecosystem structure. This invasive species has become a serious environmental problem because they displace the indigenous plant species. This study was conducted to evaluate the antioxidantive effects of ethanolic extracts from different parts (root, stem, seed and leaf) of the invasive exotic species Hypochaeris radicata L. Methods and Results : The aim of present study was to estimate the total phenolic and flavonoid contents and to investigate in vitro antioxidant potential of ethanolic leaf, root, seed, and stem extracts of the Hypochaeris radicata. Antioxidant activity was assessed by using 2,2-diphenyl-1-picryl-hydrazyl (DPPH) assay, reducing power activity, [2,2-azino-bis(3-ethylbenzthiazoline-6-sulphonic acid)] ABTS+ assay and ferrous ion chelating activity. The total phenolic and flavonoid contents were also determined and expressed in gallic acid and quercetin equivalent respectively. The results of the study indicate that the ethanolic extracts of the leaf, root, seed, and stem of H. radicata posses significant scavenging activity against DPPH (21.25% for leaf, 34.98% for root, 60.76% for seed and 45.25% for stem at 250 μg/ml each) and ABTS+ radical scavenging activity (14.85% for leaf, 17.40% for root, 35.91% for seed and 24.70% for stem at 250 μg/ml each), reducing power activity (0.178 absorbance at 300 μg/ml for leaf, 0.211 absorbance at 300 μg/ml for root, 0.447 absorbance at 300 μg/ml for seed, 0.276 absorbance at 300 μg/ml for stem). The free radical scavenging and antioxidant activities may be attributed to the presence of adequate phenolic (gallic acid content is 361.92.98 μg/g in leaf, 356.59μg/g in root, 719.72 μg/g in seed and 512.08 μg/g stem) and flavonoid compounds (219.52 μg/g in leaf, 75.67μg/g in root, 281.39 μg/g in seed and 215.66 μg/g stem). This study revealed that the ethanolic extracts of both leaf, root, seed and stem of H. radicata has demonstrated significant antioxidant activity. Conclusion : In conclusion, the present study has demonstrated that Hypochaeris radicata seed ethanol extracts are rich in phenolics and have a strong antioxidant activity and a radical-scavenging action in all of the tested methods. This suggests that Hypochaeris radicata is a good source of natural antioxidants.

Background : The Aralia cordata and Dendropanax morbifera, belonging to the family Araliacea, is a perennial herbaceous species. Although its beneficial effects for several chronic diseases are well-known, the role of its effects on chronic intestinal inflammation, colon carcinogenis are limited. Inflammatory bowel diseases (IBD) are chronic inflammatory disorders of gastrointestinal tract. It has been reported that IBD is associated with colon cancer which is one of the most common types of cancer diagnosed and a major cause of cancer-related mortality. Methods and Results : The present study hypothesized that Araliacea family would exert a protective effect on inflammation and dextran sulfate sodium (DSS)-induced colitis mouse model. Effect of Aralia cordata extracts (ACE) and Dendropanax morbifera leaf extracts (DMLE) on pro-inflammatory markers, mitogen-activated protein kinase signaling (MAPKs), activation of nuclear factor κB (NF- κB) in THP-1 human monocyte and HT-29 colon epithelial cell model were investigated. In addition, colon inflammation inhibitory effects of ACE and DMLE on the characteristics of in DSS-induced colitis model. Results showed that ACE, DMLE attenuated the severity of DSS-induced colitis, as assessed by disease activity index scores, shrinkage of colon length, and histopathologic changes and infirmatory cytokine concentration. Furthermore, ACE treatment significantly suppressed the colon carcinogenesis in AOM/DSS induced colon cancer model. The apoptosis induction abilities of the DMLE were studied by analyzing the expressions of Bcl2 family protein, caspase, Annexin V/PI staining and DNA fragmentation. According to our results, DMLE significantly induced cell death in SW-480 cells. Apoptosis was determined by cell morphology and electrophoresis of DNA fragmentations. Furthermore, treatment with DMLE also induced the increase in caspase activity, pro-apoptotic proteins (Bax and Bad) and the decrease in anti-apoptotic proteins (Bcl-2 and Bcl-xL). In addition, DMLE induce cell cycle arrested at the G1/S phase in SW-480 cells and exhibited significant inhibition on the expressions of G1/S phase specific CDK, cyclin and up regulation of CDK inhibitors. Conclusion : Taken together, these results provide evidence that ACE and DMLE prevention colon cancer by regulating inflammation, colitis, and colon carcinogenesis which indicates its benefit for colon health.

Previously we observed that human adipose-derived stem cells (hADSCs) could form aggregation during culture in the presence of human serum (HS). In the present study, we have examined if the aggregation might result from the cell migration and analyzed the difference of cell adhesivity after culture in various conditions. When cells were cultured in fetal bovine serum (FBS) alone, there was no morphological change. Similarly, cells pretreated with FBS for 1 day or cultured in a mixture of FBS and HS showed little change. In contrast, cells cultured in HS alone exhibited formation of cell-free area (spacing) and/or cell aggregation. When cells cultured in FBS or pretreated with FBS were treated with 0.06% trypsin, almost cells remained attached to the dish surfaces. In contrast, when cells cultured in HS alone were examined, most cells detached from the dish by the same treatment. Treatment of cells with forskolin, isobutylmethyl xanthine (IBMX) or LY294002 inhibited the formation of spacing whereas H89 or Y27632 showed little effect. When these cells were treated with 0.06% trypsin after culture, most cells detached from the dishes as cells cultured in HS alone did. However, cells treated with IBMX exhibited weaker adhesivity than HS alone. Based on these observations, it is suggested that HS treatment might decrease the adhesivity and induce three-dimensional migration of hADSCs, in the latter of which cAMP signaling could be involved.

Background : Liriope platyphylla Wang et. Tang is a perennial plant and its tuberous root is used as herbal medicine. Liriope platyphylla is grown throughout a year from April to April of next year. Recently, the production was lowered because of severe drought/rainfall and injury by successive cropping. The ridge height affected the growth of shoots and roots as affected by drought or rainfall. It is necessary to establish the cultivation techniques for stable quantity per unit area and good tuber’s quality. This experiment was carried out to select a cultivar of good tuber quality and to determine the ridge height for high-yielding. Methods and Results : It were investigated the characteristics of roots and tubers depending on different ridge heights in four varieties of Liriope platyphylla Wang et. Tang. There was not significantly different in shoot growth by different ridge heights. The tubers distribution in less than 10cm soil depth at the surface was decreased about 6.6% in the 15cm ridge height compared to 5cm. So, the mean distributed position of tubers was deepened at higher ridge. The cultivars that tubers were mainly distributed in the 10cm depth from surface was Seoungsu and Cheongyang natives’s lines. The no. of tubers per a plant and the ratio of marketable tubers were increased about 13.2% and 20.1% in the 15cm ridge height compared to 5cm. Considering the tuber sizes and ratio of marketable tubers, Cheongshim and Maekmundong 1 has good quality than Seoungsu or Cheongyang natives’s lines. The tuber yield increased about 19.5% in the 15cm ridge height compared to the 5cm. So, high ridge was advantaged to increase the yield. Conclusion : From the above results, the quality and quantity of tubers of Liriope platyphylla Wang et. Tang was good in the higher ridge. We recommended Cheongshim cultivar because of the large tuber sizes and high ratio of marketable tubers.

Background : Liriope platyphylla Wang et. Tang is a species of Liliaceae and its tuber is used as medicinal herbs. This medicinal plant is mainly cultivated in Cheongyang and Buyeo of Chungcheongnam-Do and Miryang of Gyeongsangnam-Do. It is necessary to establish a cultivation techniques for stable production to solve the problems that are found on farms. These farm surveys were carried out to obtain the basic data on soil environment and cultivation conditions at main producing areas of Chungnam-Do in Liriope platyphylla Wang et. Tang. Methods and Results : The surveys were investigated from 37 farmers in Cheongyang and 32 farmers in Buyeo who cultivate Liriope platyphylla Wang et. Tang. There were slightly differences in the local characteristics between Cheongyang and Buyeo. The rate of successive cropping for more than 3 years were 19% at Cheongyang and 41% in Buyeo. The reason may be due to the a small-owned field. It has grown 28% at Cheongyang and 59% at Buyeo in paddy fields. The injury by successive cropping that farmers argued were the increasing pest/disease occurrence, degradation of tuber quality and yield reduction. Crops planted after the harvest of L. platyphylla were mostly Pepper or soybean in upland. It is necessary to reduce the injury of successive cropping. The results of 112 soil analysis data were classified by 8 soil series. There are many acidic soil, lack of organic matter, excess phosphate. However, it did not analyzed the influences on the growth and yield of the crop as affected by different soil conditions. Conclusion : From the above results, it is necessary to develop cultivation techniques on the reduction of injury of successive cropping, such as cropping system, seedling transplanting cultivation, cultivation methods for good agricultural practices(GAP) and soil improvement.

In animal development, the mechanisms by which localized factors and organelles in egg cytoplasm were exactly distributed into each daughter cell are essential for formation of various cell types. During ascidian Halocynthia roretzi embryogenesis, ooplasmic mitochondria were mainly segregated into muscle and neural precursor cells. At the 32-cell stage, localized mitochondria in the B6.2 blastomeres were preferentially distributed into the B7.4 muscle precursors compared with the B7.3 mesenchyme/ notochord precursors. When the B6.2 blastomeres were isolated from the early 32-cell stage embryos and then allowed to divide 2 times of cell division, the resultant partial embryos showed symmetric distribution of mitochondria, and the partial embryos were composed of equal size cells. In normal development, cell fates of the B7.3 blastomere were correlated with the unequal cleavage of B6.2 lineage cells that normally occurs in the next two-cell division stages to produce a large B8.5 mesenchyme and a small B8.6 notochord cell. Mitochondria are distributed asymmetrically in both cells. When embryos were treated with FGF receptor inhibitor SU5402 and MEK inhibitor U0126 between the 32-cell and the early 64-cell stages, the resultant embryos showed equal cleavage pattern and symmetric distribution of mitochondria in daughter cells of the B6.2 blastomeres. However, blocking of Nodal and Notch signaling did not affect the cell division pattern and mitochondrial distribution in the B6.2 lineage blastomeres between the 32-cell and 110-cell stages. Therefore, it is likely that FGF/MEK signaling is involved in asymmetric distribution of mitochondria and unequal cleavage of the B6.2 lineage blastomeres in ascidian embryo.

More than 300 Miscanthus accessions as a potential bioenergy crop were collected in Korea and their morphological traits were investigated at various growth stages. Among morphological traits, stem growth habit, the presence of awn in spikelet, and autumn new shoot are the most important key traits enabling to cluster Miscanthus accessions into M. sinensis and M. sacchariflorus groups. Miscanthus sinensis has bunch stem growth habit and awn in spikelet, and produces autumn new shoot, while M. sacchariflorus has scattering stem growth habit with no awn in spikelet and does not produce autumn new shoot. Interestingly, we found several Miscanthus accessions showing intermediate morphological traits. 7 M. sinensis accessions showed morphological traits similar to M. sacchariflorus and 17 M. sacchariflorus accessions showed morphological traits similar to M. sinensis. Flow cytometry and chromosome counting finally revealed 5 Miscanthus hybrids, suggesting that they are resulted from natural hybridization between M. sinensis and M. sacchariflorus. Therefore, these Miscanthus hybrids can be used to understand genetic recombination between these two Miscanthus species and our understanding may support future efforts for breeding new Miscanthus variety with high biomass productivity and environmental adaptability.

To access the physiological response of rice to different types of ionizing radiation, rice seeds were exposed to gamma-ray and ion beam radiation. Exposure to ionizing radiation dramatically decreased the shoot length compared with non-irradiated plants. Fluorescence-activated-cell-sorting (FACs) was used to measure DNA contents. There were significant correlations of dose-dependent between irradiated plant and non-irradiated plant. The radicals induced by the ionizing radiation in the plant could be observed by electron spin resonance (ESR). It was confirmed that the number of free radicals in cell was greatly increased all irradiated plants than non-irradiated plant. A significant positive correlation was shown between ionizing radiation dose and signal intensity. In order to determine the Genetic diversity, AFLP analysis was conducted with the irradiated plant and non-irradiated plant. Based on band patterns, the cluster analysis was conducted to evaluate the genetic variation by using the UPGMA (Unweighted Pair Grouping Method of Averages). Genetic diversity of irradiated plants by low dose ion beam was the closest non-irradiated plant and irradiated by high dose gamma-ray was the furthest from non-irradiated. We describe the detailed methods of ionizing irradiation and discuss its applications in genetic research as well as plant breeding.

Ionizing radiation affects gene expression from plant genomes. To monitor the genome-wide transcriptional changes induced by three types of ionizing radiation, we used the rice RNA sequencing to identify genes that are up- or down-regulated by gamma rays (GAs), proton (PRs) and ion beams (IBs). The Oryza sativa jacalin-like lectin domain containing proteins (OsJAC1) gene was highly induced by GAs, PRs and IBs. OsJAC1 was selected based on the expression patterns of a genome-wide dataset of RNA sequencing. Many jacalin-related lectin genes have been shown to be associated with disease resistance, biotic and abiotic stress signaling. Therefore, we studied its expression pattern in response to different abiotic stress and phytohormone treatments. The expression patterns of OsJAC1 under two different abiotic stress conditions (salt and heat stress) and phytohormones (salicylic acid and methyl jasmonate) were examined. The transcripts of OsJAC1 were significantly induced in response to abiotic stress conditions, including salt and heat treatments. In addition, it was induced in response to the salicylic acid and methyl jasmonate treatments, respectively. To investigate the sub-cellular localization of OsJAC1, the gene was expressed as a fusion protein tagged with GFP, in tobacco leaf epidermis and examined under confocal microscope. The OsJAC1 was clearly localized at the nucleus. These results provide critical insights into the molecular functions of the rice jacalin-like lectin domain containing proteins as receptors of external signals.

Proline has been shown to accumulate in plant under various type of stresses. In our previous study, changes in cold hardiness and proline content showed contrasting patterns during a constant deacclimation. This study was performed to investigate the proline accumulation and related gene expression in response to repeated deacclimation and reacclimation in peach cultivar ‘Daewol’. Proline content was analyzed using the ninhydrin method and related gene expressions were examined using quantitative real-time RT-PCR. Proline contents of ‘Daewol’ increased during the repeated deacclimation treatments. Interestingly, during the twice deacclimation, expressions of P5CS (Δ1-pyrroline-5-carboxylatesynthase) constantly decreased, whereas expressions of P5CR (Δ1-pyrroline-5-carboxylatereductase) increased. Expressions of OAT (ornithine-δ-aminotransferase) indicated up- and down- pattern in response to repeated deacclimation and reacclimation. Our results indicated that proline responds positively to higher temperature in the shoots of peach cultivar ‘Daewol’ and expressions of both P5CS and P5CR genes could show contrasting patterns during the deacclimation. Moreover, our results suggest that ornithine pathway could serve as an alternative pathway in proline synthesis process during deacclimation in peach.
