Background : Root diseases caused by Cylindrocarpon destructans and Fusarium solani decrease the yield and quality of ginseng. Cylindrocarpon root rot is a major disease caused by replanting failure in ginseng garden. Methods and Results : Solarization was done in the infested soil of the greenhouse for summer season (from July 24 to Autumn 31, 2014) after putting green manure (Sudan grass) and calcium cyanamide (CC) into the soil. Mycelium and conidia of C. destructans died at 4 0℃ after 15 hours, and 45℃ after 5 h, but it did not die at 35℃ after 15 h. Those of C. destructans died after keeping it for 2 hours daily at 40℃ for 9 days, and 45℃ for 8 days, but did not die at 38℃ for 9 days. Maximum soil temperature was 55.4℃ in 5 cm depth, 48.7℃ in 10 cm, 44.7℃ in 15 cm, 42.5℃ in 20 cm, and 31.9℃ in 30 cm by putting green manure into the soil and solarization. Reduction of sudan grass increased electrical conductivity (EC), organic matter, P2O5, K, and Mg, while decreased pH, NO3-N, and Na. Addition of calcium cyanamide and urea gave a negative effect on the growth of ginseng because EC and NO3-N were increased excessively than the optimal range. Solarization using green manure mixed with CC was the most effective in decreasing soil-borne pathogens of 2-year-old ginseng. But the root disease that occurred between single treatment of sudan grass and the treatment mixed with calcium cyanamide showed not a significant different. Addition of calcium cyanamide showed the decrease of root weight because leaves were dead early by a excessive increase of EC and NO3-N. Conclusion: Soil disinfection using green manure and solarization in greenhouse was effective in inhibiting root rot, however, it did not completely kill the soil-borne pathogens.

Background : Angelica gigas is a monocarpic biennial or short lived perennial plant. A. gigas, also called Dang Gui or Korean Angelica, is a major medicinal herb used in Asian countries such as Korea, Japan and China. In Korea, we are using the roots of A. gigas, but, they are using Angelica sinensis in China and Angelica acutiloba in Japan to obtain many active constituents. The biggest problem in the using of A. gigas would be the confusion with A. acutiloba or A. sinensis. These three plants can't be distinguished by appearance. And the constituent ratios of the three plants are different. This confusion can cause an accident or the pharmaceutical effects do not meet the expectations. In this study, we developed chloroplast SSR markers that can distinguish A. gigas, A. acutiloba and A. sinensis. Methods and Results : We collected A. gigas, A. acutiloba and A. sinensis. and extrated DNA using CTAB method. The DNA was diluted to 10 ng/㎕ and kept -20℃. We designed the primer sets using CLC Main Workbench based on chloroplast DNA SSR region of A. gigas. PCR were performed on the three angelica plant samples (in 5 repeat). Conclusion : We made five primer sets from five SSR regions of A. gigas cpDNA. All of the primer sets amplified the amplicon effectively. Two of the 5 primer sets had polymorphism. We can distinguish A. gigas, A. acutiloba, and A. sinensis using the 2 primer sets

Background : Codonopsis lanceolata is used as a natural medicine or vegetables. It originates in East Asia such as Korea, Japan and China. C. lanceolata roots contain various chemical compounds including saponins like Panax ginseng. Although C. lanceolata are cultivated in different regions of South Korea, no variety has been developed. Therefore, it is necessary to develop discriminating methods such as molecular markers in C. lanceolata species. Methods and Results : To find simple sequence repeat (SSR) markers, we sequenced C. lanceolata genomic DNA using Illumina HiSeq 2000 System. A total of 250,455 putative SSR loci were obtained, and 26,334 non-redundant primers were designed to amplify these SSRs. Di-nucleotied repeats were the most abundant SSR reapeats, accounting for 89.53% (23,578) of primer designed SSRs. Tri-nucleotide, tetra-nucleotide and penta-nucleotide accounted for 8.44% (2,223), 1.3%, (348) 0.2% (55), respectively. Tri-, tetra-, and penta-nucleotide (total of 2,626 SSRs) were investigated in silico to identify polymorphism between individuals. Consequently, 573 SSRs showed polymorphism. Forty genomic SSR markers were tested in 16 C. lanceolata plants for determination of PCR amplification and polymorphism. From these primers, 27 (67.5%) amplified products and the average polymorphism information content (PIC) value was 0.52. Conclusion : We development 27 SSR markers from C. lanceolata using NGS, and it could be used for breeding of new varieties in the future.

Background : Osteoclasts as multinucleated cells originate from hematopoietic monocyte/ macrophage precursor cell, shows the bone absorption through the commitment, differentiation, fusion, and bone resorption stages by regulation of M-CSF and RANKL. It has been reported a significant negative correlation between the increase of oxidative stress and the bone density, and when RANKL reaction to the osteoclasts precursor cells is mainly generated ROS is due to increased activity of NADPH oxidase1 (NOX1), and these ROS act as a factor which promotes osteoclasts differentiation. Thus, RANKL signaling process is important that excessive osteoclast formation and differentiation inhibited through the regulation of each step. Methods and Results : F3570 ethanol extract showed relatively high activity at in-vitro antioxidant activity. F3570 water extract inhibited ROS generation in RAW 264.7 cells stimulated with H2O2 and RANKL, even at low concentrations. The inhibitory effect of osteoclast differentiation on F3570 water extract was confirmed that shown through NF-κB pathway, MAPK pathway including ERK and JNK. F3570 ethanol extract is considered to be regulated by the p38 MAPK and the other signaling pathway. Also, F3570 both water and ethanol extract were significantly reduced gene expression such as TRAP, calcitonin receptors and integrin β3 of RANKL-induced mature osteoclast in the bone resorption stage. Conclusion : Through this study, F3570 extract revealed an outstanding inhibitory effect and signaling mechanisms in osteoclast differentiation induced by RANKL. These results suggest that F3570 is bone diseases associated with aging or osteoporosis caused by menopause in an aging society is expected to be a superior candidate for the treatment or the prevention

Background : Korean Red Ginseng has been reported to have various anti-virus, anti-tumor, anti-inflammation and immune-regulating activity. However, Korean Red Ginseng Crude Saponin (KRGCS) effect on Pulmonary inflammation has been insufficient research. We have investigated the effect of KRGCS on pulmonary inflammation in mouse model. Methods and Results : Pulmonary inflammation induced by cigarette smoke condensates (CSC) and lipopolysaccharide (LPS) in BALB/c mice. 100uL (ratio 1:1) of CSC (4mg/mL) and LPS (100ug/mL) solution instilled with intratracheal injection once per a week for 3 weeks in mice. KRGCS was also administered orally a dose of 10mg/kg and 25mg/kg for 3 weeks. KRGCS treatment groups decreased the number of inflammatory total cells in lung tissues and bronchoalveolar lavage fluid (BALF) and the number of neutrophils in BALF dropped more than negative control groups. Additionally, KRGCS reduced the absolute number of CD4+ and CD8+ cells in BALF, and the number of CD11b+ Gr-1+cells in the lung and BALF. Furthermore, we found that KRGCS declined significantly the level of the cytokine chemokine (C-X-C motif)-1 (CXCL-1), Macrophage inflammatory protein-2 (MIP-2), and tumor necrosis factor alpha (TNF-α) in BALF. Conclusion : These results that KRGCS has an inhibitory effects on pulmonary inflammation such as COPD, may be a candidate of effective herbal pharmaceuticals for the cure of lung diseases.

Background : Lycium ruthenicum Murr. was known as black wolfberry. It belongs to Solanaceae. In order to elucidate biological activities and functional substances of Lycium ruthenicum Murr., the methanol extract of Lycium ruthenicum Murr. harvested from Qaidam Besin. area was prepared and was studied. Methods and Results : Antioxidant, anticancer, anti-inflammatory and antidiabetic activities of the methanol extract of Lycium ruthenicum Murr. were examined using diverse in vitro bioassays and functional substances were analyzed by HPLC and LC-MS/MS. The methanol extract of Lycium ruthenicum Murr. showed strong DPPH and ABTS radical scavenging activities and also showed strong SOD-like activity. To examine the effects of the methanol extract of Lycium ruthenicum Murr. for the expression of antioxidant enzymes, superoxide dismutase (SOD) and catalase (CAT), RT-PCR was performed. SOD expression was strongly increased by the methanol extract of Lycium ruthenicum Murr. CAT expression was also increased with dose-dependent manner. The methanol extract of Lycium ruthenicum Murr. showed inhibitory effects against α-glucosidase and α-amylase, and also inhibited NO production. Anticancer effects of the methanol extract of Lycium ruthenicum Murr. assay were examined with five human cancer cell lines. The potent anticancer activity exhibited in HepG2 hepatoma cells. As a result of main component analysis by HPLC, chlorogenic acid and rutin were identified as the major flavonoids of Lycium ruthenicum Murr.. In addition, by LC-MS/MS, petunidine-3-O-rutinoside (trans-p-coumaroyl)-5-O-glucoside was determined as a major anthocyanin of Lycium ruthenicum Murr.. Conclusion : Lycium ruthenicum Murr. showed various biological activities and could be used as a food and a functional material to enhance human health. Further study to identify an unknown flavonoids of Lycium ruthenicum Murr. would be needed.

Background : Oxidative stress-related iflammatory mechanisms may play an important role in the pathogenesis of cisplatin-induced nephrotoxicity. Safflower (Carthamus tinctorius L.) seed is a crude drug rich in serotonin derivatives to exhibit several biological activities, including antioxidant, anti-inflammation and anti-cancer effects. The purpose of this study was to determine the renoprotective effects of Safflower seed, using cisplatin-induced nephrotoxicity mice. Methods and Results : Safflower seed was orally administered at a dose of 100 and 200 mg/kg body weight for 5 days before the intraperitoneal injection of cisplatin 20 mg/kg body weight, and the effects were compared with those of vehicle-treated cisplatin administered to control and normal mice. In the serum and kidney, renal function parameters reactive oxygen species and expression of protein related to oxidative stress, DNA damage, inflammation and apoptosis were examined. Safflower seed treatment attenuated serum BUN, createinine and renal oxidative stress through reduction of reactive oxygen species and increase in the protein expression level of catalase. Safflower seed reduced renal protein expression of p-p38 and p-JNK (mitogen-activated protein kinase pathway), pro-apoptotic factors (such as Bax and caspase 3) and nuclear factor-kappa B-targeting pro-inflammatory cyclooxygenase-2 and inducible nitric oxide synthase. In addition, Safflower seed treatment led to significantly attenuated histological damage in the kidney. Conclusion : These renoprotective effects of Safflower seed were achieved through attenuation of oxidative stress and its sensitive protein expression associated with inflammation and apoptosis in cisplatin-induced nephrotoxicity mice.

Background : Panax ginseng C.A.Meyer is a perennial herb and its growth potential keep relatively high at least for the early 3 years old. If the transplanted ginseng roots of 2 or 3 years old grow well, a period of cultivation and the disease management cost could be reduced for the production of 6 years old ginseng roots. Methods and Results : The rooting and growth of a transplanted ginseng root could be affected by the age of a transplant. Therefore, the experiments were conducted to examine the influences of the age factor on the rooting and growth of transplanted ginseng roots. The ginseng roots with different plant ages in growth years (1, 2, and 3 years old), used in this study, were obtained by raising in a seedling production field near the Punggi Ginseng Experiment Station. The transplants were continuously grown under the same conditions after transplanting. The number of lateral or branched roots at the root-growing period after transplanting was basically higher in originally older plant ages compared to younger plants: the numbers of secondary and tertiary branched roots of the 1 year-old transplants were 8, 12, respectably; 2 years old: 12, 20; 3 years old: 12, 28. Those did not show any difference in both overground and underground growing, in compared to conventional cultivations with roots of a year old. The plants grew well without anthracnose, spotting disease, and high temperature injury. Conclusion : The conventional cultivation period of above 5 or 6 years for the raw material of red ginseng might be shorten and also the cost of disease control could be decreased, by transplanting of 2 or 3 years old roots raised at a seedling nursing field.

Background : Cylindrocarpon root rot, caused by Cylindrocarpon destructans and Fusarium solani, is a major disease which lead to replanting failure in ginseng garden. Chlamydospores of C. destructans, generated by poor environmental condition, can survived more than ten years in soil without host plant, ginseng. Density of soil pathogens gradually decreased as the progress of time since ginseng was harvested. Methods and Results : Soil chemical properties was analyzed from soil samples of 43 regions in farmer’s ginseng garden to study the variation of content and the correlation among inorganic contents. Soil samples of 24 regions was also analyzed to study correlation between progressed-year after ginseng harvest and soil chemical properties. Variation of soil chemical properties in descending order was Fe, Zn, P2O4, NO3, Mn. The content of Fe and Zn were great variation among inorganic chemicals of soil of farmer’s field. Electrical conductivity to induce physiological demage in excessive concentration showed highly significant positive correlation with the content of NO3 and K. As the progress of year after ginseng harvest, the content of organic matter and zinc was increased, while pH and Na were decreased in farmer’s field to cultivate ginseng. There were highly significant positive correlation between the progress of year after ginseng harvest and zinc content in farmer’s field to cultivate ginseng. Ratio of root rot of 2-year-old ginseng showed significant positive correlation with K content, and negative correlation in experimental field cultured by six rotation crops for one year. Conclusion : Root rot by soil pathogens was closely related with the content of potassium and zinc in soil chemical properties.

Background : Adenophora triphylla var. japonica Hara. is used to cure internal organ, which is called to important one of the five “Sam (ginseng)” oriental medicinal herbs included Panax ginseng, Salvia miltiorrhiza, Sophora flavescens, Scrophularia buergeriana. Korea imported 24 ton 129 thousand dollar for medicinal herbs and 23 ton 29 thousand dollar for food at 2013. This study was carried out to investigate the effect of several organic compost on the growth and root yield of A. triphylla var japonica with organic fertilizer. Methods and Results : Seeds were collected from Adenophora triphylla var. japonica Hara. at last year in nursery of Department of Herbal Crop Research, NIHHS, RDA. Four types organic fertilizer (germiculture (GG), granular mix oil cake (CJ), mixed organic (DJ), plant oil cake (TG)), chemical fertilizer (CF) and non-fertilizer (NF) were treated. It was fertilized on soil before 2 month of planting. Seeds A. triphylla var. japonica were sowed in tray at early March and then were grown during 2 month. It were planted in furrowed nursery at early with 30 × 10 cm planting distance and black vinyl covering condition. Soil chemical property, survival rate, aerial part and root growth characteristics and yield were investigated at harvest time. For main composition analysis, 10 mg of standard (b-sitosterol, lupenone) was taken, 80% MeOH was added to dissolve in 10 ㎖ flask. 0.5, 1, 2, 4 ㎖ was taken from dissolved solution and then was added to 10 ㎖ of 80% MeOH. It was filtered through 0.45 ㎛ filter and then 5 μℓ was taken to make standard solution. Analytical calibration curve was measured to diluted solution within sample concentration. 2 g of ground sample was taken, 40㎖ of 80% MeOH was added to extract by ultrasound sonication during 60 minutes and then it was filtered through 0.2 ㎛ filter to analyze component content. Conclusion : GG and DJ were the most suitable for organic cultivation of denophora triphylla var. japonica. Content of beta-sitosterol showed the highest value in DJ treatment and the lowest value in CF. Content of lupenone showed the highest value in DJ and the lowest value in CJ.

Background : The roots of Curcuma longa Radix is used to make herbal medicine, which is registrated in the Korean Pharmacopoeia. This species is a perennial herb, and is distributed throughout tropical and/or sub tropical regions. High temperature and humidity with small difference between daytime and nighttime is the finest environment condition in this species. Recently, demand for C. longa has been expanded to cultivation area as central region from southern region. Therefore, this study aimed to investigate the effect of harvest time on the growth, yield characteristics and main composition contents in C. longa. Methods and Results : Curcuma longa rhizomes were harvested from farmland in Jindo, Jeollanam-do in end October, early November, middle November, end November, early December, middle December and end December 2015. Harvested rhizome were dried in hot air drier, and it was investigated as growth characteristics and yield. Contents of curcumin, demethoxycurcumin, bisdemethoxycurcumin was analyzed from dried powder of rhizome. The analysis column was used to Phenomenex, Kinetex 2.6um F5 100A 100*2.1mm. 10 mg of standard was taken, 80% MeOH was added to dissolve in 10 ㎖ flask. 0.5, 1, 2, 4 ㎖ was taken from dissolved solution and then was added to 10 ㎖ of 80% MeOH. It was filtered through 0.45 ㎛ filter and then 5 μℓ was taken to make standard solution. Conclusion : The root weight significantly increased from 376.1 g in end October to 695.9 g in end December as 1.8 double. Curcumin contents differed significantly among harvest times in end November and early December. Demethoxycurcumin increased from early November to end November, and Bisdemethoxycurcumin increased to the early November.

Background : Yam absorbs nutrition and water through water absorptive root generated from head of rhizome. Round-shaped yam (RSY, Dioscorea opposita) and tropical round-shaped yam (TRSY, Dioscorea alata)are more sensitive to drought because of shorter head of rhizome and shallower root compared to long yam or short yam. This study was conducted to elucidate the effect of irrigation on RSY and TRSY yam at 205 in Institute of Bioresources Research, GBARES. Methods and Results : Tuber of RSY and TRSY were cut as 40 g, and dust-coating sterilized with lime. Seedlings were grown in heating wire installed seed bed from 25. March to 16. April. Experimental field were fertilized 2,000 kg compost, 34 kg N2, 28 kg P2O5, and 28 kg K2O. All amount of compost and P2O5 treated as basal fertilizer. N2 and K2O treated 14 and 10 kg of basal fertilizer, and 20 and 18 kg of additional fertilizer, respectively. Tillage, installation of drop-watering tapes, and covering black PE film with 60×25 cm spacing holes on 120 cm row were conducted. 20 L per m2 was irrigated 7 days interval except rainy day from middle of May to late of September. RSY had high emergence rate in early stage due to irrigation, while TRSY had no difference. Irrigation was not effective on total number of tuber but number of marketable tuber (over 200 g). Marketable tuber yield of RSY according to irrigation was increased 89% as 1,147 kg per 10 a. And tuber with irrigation was 73 g heavier. Tuber yield and marketable tuber yield of TRSY as affected by irrigation were respectively 2,611 and 1,715 kg per 10 a compared to control, 1,462 and 428 kg. And irrigated tuber was 66 g heavier. Conclusion : TRSY had more effective on irrigation than RSY. Both of yam had significant increased marketable tuber yield due to irrigation. Therefore RSY and TRSY are necessarily irrigated because of those absorptive characteristics, short head of tuber and shallow root.

Background : The effect of ridge-up bed cultivation and stem training method on the plant growth and yield of Cynanachum willfordii was investigated. Methods and Results : Domestic variety of Cynanachum willfordii was tested at Chungbuk ARES under the following conditions. Nonsupporting (custom) and I-form supporting was used for stem training method, and ridge height was set as 30 (custom) and 60 cm (high ridge). Planting spacing was adjusted to 30 (interrow spacing) × 20 cm (intrarow spacing), and other major management was followed the method of standard cultivation for medicinal crop. Investigated result from leaf characteristics, leaf length was longer in high ridge cultivation (HRC) as 11.1 cm than custom cultivation (CC) as 10.6 cm. Leaf width is proved to be 12.8 (HRC) > 11.2 cm (CC). Leaf number is proved to be 294 (HRC) > 254/plant (CC), with higher number of 44/plant at HRC and weight/10 leaves were more heavy at HRC (4.9 g) than that of CC (2.6 g). It was more fruit setting at HRC over 15/plant. According to the stem training method and ridge height from nonsupporting cultivation, main root was 4.0 (CC), higher than that of HRC over 0.5/plant. However, root length was more longer in HRC (28.6) than that of CC (25.0 cm). Main root diameter was also showed more thicker pattern in HRC. From staking cultivation, root number of HRC was 7/plant, it was recorded more 3/plant than that of CC, and it was also same pattern in main root length and root diameter. By the standard of commercial root, yield of living roots in nonsupporting cultivation were 59.0g/plant (HRC), it was more heavy over 10.4g/plant than that of CC. In staking cultivation, HRC were recorded as 74.2g/plant, more heavy 6.9g/plant than that of CC. Yield from I-form support stem training and ridge-up bed cultivation of HRC was higher approxmiately 52.7% than that of CC. Conclusion : Overall growth by high ridge cultivation of I-form support was good and yield of HRC was also increased over 52.7% than that of CC.

Background : This study was performed to know the effect of organic fertilizers on the growth and root characteristics of Platycodin grandiflorum Ridix. Methods and Results : As basal fertilizer were treated before transplanting Platycodin grandiflorum Ridix with chemical fertilizer(Super21, 21-17-17, Namhae Chemical), mixed organic fertilizer, bacterial cultures and fermented oil cake, manure in each trial of nitrogen based on 3 ㎏/10a criteria. To measure the fresh weight and length, thickness and the number of fine root of the Platycodin grandiflorum Ridix roots harvested in October. Root length appeared in the mixed organic fertilizer and chemical fertilizer were 25.3 and 24.0 ㎝ longest, oil cake was the shortest 15.3㎝. Diameter at the thickest showed 26.6 ㎜ in chemical fertilizer, the following phrase appeared bacterial cultures and mixed organic fertilizer application. The number of fine-root was investigated in mixed organic fertilizer and chemical fertilizer most pieces 20.0 and 17.0. Fresh weight, which is directly related to yield were mixed organic fertilizer to 55.7 g the highest in the number of fine-root and the longest root length. Conclusion : For the increasing the yield that Pharmacological effect, organic fertilizer is effective to increase the number of fine-root high which high contented with saponins.
