Background : Excess alcohol consumption can lead to alcoholic liver damage and gastritis. Korean thistle (Cirsium japonicum var. maackii) has been reported that it’s administration protective from liver from lipid accumulation and toxicity. Also, Korean dandelion (Taraxacum coreanum Nakai) has been traditionally used for treating various inflammation-related diseases including ethanol-induced gastritis. In this study, we aimed to determine whether the combination treatment of Korean thistle and Korean dandelion could regulate alcoholic liver damage and gastritis in mice. Methods and Results : Seven-week-old mice were treated with ethanol (5 g/㎏ of body weight) by gavage every 12 h for a total of three doses to induce acute liver damage and gastritis. Ethanol extract of Korean thistle, Korean dandelion or combination was gavaged simultaneously with ethanol for three doses. The results showed that the combination treatment of Korean thistle and Korean dandelion significantly inhibited alcohol-induced increase of serum alanine transaminase and aspartate transaminase levels, liver weight, and hepatic caspase-3 expressions. Hepatic histopathological changes induced by alcohol were also remarkably improved by the combination treatment. The combination treatment inhibited alcohol-induced Morphological changes of gastritis and attenuated the expressions of pro-inflammatory protein, such as inducible nitric oxide synthase. Conclusion : The results of our study suggest that the combination treatment of Korean thistle and dandelion may simultaneously alleviate acute alcohol-induced gastritis and liver damage.

Background : It is necessary to develop a diagnostic tool for the precision agriculture according to the change of agriculture environment. In particular, medicinal crops are often cultivated in poor areas such as mountains and valleys, and are susceptible to abnormal climates. Therefore, there is a need for a method that can diagnose the growth easily and with time. We conducted a study to investigate the possibility of remote sensing for 10 species of medicinal crops using unmanned aerial vehicles and multispectral images. Methods and Results : This study was conducted in 0 - 4 times continuously cultivated field with 10 species of medicinal crops (Angelica gigas, A. koreana, A. tenuissiman, A. dahurica, A. acutiloba, A. polymorpha, Ledebouriella seseloides, Lithospermum erythrorhizon, Rhemannia glutinosa). Growth characteristics such as plant height, number of leaves, leaf length, leaf width, stem diamanter and number of stems were investigated between July 10 and July 16. The reflectance map of red (660 ㎚) and NIR (790 ㎚) was obtained using a UAV equipped with a multi-spectral sensor and NDVI [Normalized Difference Vegetation Index] was obtained with a Pix4D mapper. As the number of times of continuos cultivation increased, the growth and NDVI tend to decreased. 6 species of Angelica genus were positively correlated with growth characteristics such as plant height, leaf length, leaf width, and stem diameter and NDVI. Otherwise, there was no significant relationship between growth characteristics and NDVI in Ledebouriella seseloides, Lithospermum erythrorhizon, and Rhemannia glutinosa. This is because the difference in the growth according to time of continuos cultivation was not significant and the canopy structure tended to overlap with the upper and lower leaves. Conclusion : NDVI is widely used as a useful tool for evaluating the canopy productivity and is highly correlated with the above-ground growth characteristics of 6 species of Angelica spp. It is necessary to develop the growth index suitable for the each species and to enhance the validity of growth index by examining the correlation under various cultivation conditions.

Background : Turmeric (Curcuma longa L.) and black pepper (Piper nigrum L.) have traditionally been used as Asian medicinal and culinary herb. Curcumin, a major compound of turmeric, has been known to have antitumor activity. However, curcumin is bioavailable because it is rapidly metabolized and released from the body. Therefore, the addition of adjuvants such as piperine, a potent inhibitor of drug metabolism, is one of the ways to increase the bioavailability of curcumin. Methods and Results : The yields of turmeric and black pepper ethanolic extracts (TM and BP) are 18.2 and 8.2% (w/w), respectively. The EC50 values of A549 and NCI-H292 cells exposed to TM were 77.8 ㎍/㎖ and 92.0 ㎍/㎖, respectively. No significant cytotoxicity was observed up to the 400 ㎍/㎖ in the A549 and NCI-H292 exposed to BP. Based on the central composite design, the co-treatment of TM and BP enhanced the cytotoxicity of A549 and NCI-H292 cells. The optimal combination concentration (optimal EC50 value) of TM and BP calculated by the response surface methodology assay were 48.5 and 241.7 ㎍/㎖. The conbination index assay confirmed that the cytotoxic effect at optimal combinatino concentration was due to the synergistic effect. Conclusion : We hypothesized that co-treatment of TM and BP enhanced cytotoxicity more than single treatment of TM against lung cancer cells, and cell death at this time may synergetic cytotoxicity effects associated with curcumin metabolism.

Background : Ginseng seeds are harvested in immature stages and take 3 - 4 month to be germinated following cold stratification. Improvement of germination speed can be a valuable traits to be characterized for economically important medicinal plant. Modulation of cell wall compositions is also considered as another agricultural trait when the crops can be processed for foods and forages. Here we show the functional characterization of ginseng pPLAIIIβ focused on its possible values on germination and modulation of cell wall compositions. Methods and Results : Patatin-related phospholipase AIII family genes were identified from ginseng EST clones and further confirmed by PCR. Clone showing the highest homology with pPLAIIIβ was overexpressed in model plant Arabidopsis, and it displayed dwarf plants. qPCR analysis showed that pPLAIIIβ expressed in all organs of 2-years-old ginseng. Overexpression of ginseng pPLAIIIβ also displayed apparently enlarged seeds as well as altered germination speed in early stage compared to the control. Chemical stainings and direct quantification of lignin and cellulose were performed to understand the link of cell elongation patterns and cell wall compositions. Conclusion : Overexpression of ginseng pPLAIIIβ inhibited longitudinal cell elongation in all tested organs except seeds which is enlarged in both directions than the control. Shorter root length is related with auxin responsive genes and its dwarf morphological phenotype is resulted in altered cell wall compositions.

Background : Cytochrome P450 enzymes belong to the superfamily of monooxygenases that are found in all living organisms with diversity in their reaction chemistry. These P450 enzymes are being used extensively by plants in their defense mechanism. Tetracyclic triterpenes reported as major ginsenoside constituents in Panax ginseng, formed by multiple hydroxylation of cytochrome P450. CYP P450 enzymes is effective in metabolising both natural and xenobiotic compounds. In this study, two ginseng CYP genes (PgCYP76C and PgCYP736A) were functionally characterized by overexpression in heterologous plant system. Methods and Results : In Arabidopsis thaliana and Panax ginseng, planta transformation was generated by floral and cotyledon dipping method using Agrobacterium tumefaciens (C58C1) individually. Spatial and temporal patterns of gene expression were analyzed by qRT-PCR against abiotic and biotic stresses. Herbicide (chlorotoluron) tolerance was performed at seedling stage by in vitro assay to check ectopic expressions. PgCYP736A expression were more abundant in leaves compared to roots and stem of 2-years-old ginseng, where as PgCYP76C gene was found to be highly expressive in rhizome, roots and leaves. Differential expressions were observed in response to abiotic stresses of two ginseng CYP genes, both expressed higher in response to NaCl and SA (salysilic acid) in ginseng. Transgenic Arabidopsis plants overexpressing PgCYP76C gene showed small, rounded leaves and reduced height. This phenotype was found to be caused by down regulation of gibberellin biosynthesis-related genes. Another function revealed tolerance towards herbicides that belongs to phenyl urea. Conclusion : This study indicates the functional roles of CYP genes in plant growth and stress responses. Studied genes also can be used as a tool to make plants herbicide tolerant. Further investigation might focus on it’s function over terpene biosynthesis and defence mechansim against (a)biotic stress.

Background : The 1,2-unsaturated PAs, reported to be widely present in medicinal plants belonging to Asteraceae, Boraginaceae, and Fabaceae, cause hepatotoxicity and genotoxicity in humans and animals. Hence, there is a need for an analytical method that allows these dangerous plant toxins to be determined. In this study, we developed a method that can be used for the rapid and accurate determination of nine toxic PAs in medicinal plants using ultra-pressure liquid chromatography–electrospray ionization–quadrupole–time-of-flight mass spectrometry (UPLC-ESI-Q-TOF). Methods and Results : The compounds were eluted onto a C18 column with 0.1% formic acid and acetonitrile, and separated with good resolution within 11 min. all analytes was characterized by its precursor ions generated by ESI-Q-TOF and fragment ions produced by collision-induced dissociation (CID), which were used as a reliable database. The proposed analytical method was verified with reference to the ICH guidelines. The proposed UPLC-ESI-Q-TOF method was applied to four medicinal plants, and lycopsamin, echimidine, senkirkine and senecionine were detected by matching with reference standard, and additional six PAs were tentatively identified though chemical profiling. In addition, the QuEchERS method was the most efficient in comparison with methods like hot water and methanol in extraction efficiency of pyrrolizidine alkaloids. Conclusion : The our proposed method can determine PAs rapidly and accurately in medicinal plants and will be utilize as an important data for other researchers who need analytical information of PAs.

Background : Panax ginseng Meyer, the king of Oriental medicine is a geophytic perennial shrub. P. ginseng possess long juvenile period which spans for at least 3-years before it could attain reproductive competence. To explore the significance of flowering time genes we isolated, characterized the Flowering Locus T (FT) gene and named as PgFT1 and PgFT2 to understand their role in ginseng floral transition. Methods and Results : By using extensive cloning and bioinformatics analysis we identified two FT homologs from ginseng a. The deduced PgFT sequences has a considerable homology to established FT homologs with the essential amino acids of a florigen function conserved. The tissue specific expression profiling by Real-time qPCR from the juvenile (2-years-old) and adult (4-year-old) ginseng plants have shown a spatial and temporal expression pattern, furthermore PgFT transcripts exhibited photoperiod dependent oscillation as well. Besides, PgFT1 and PgFT2 have shown an age specific expression pattern, in particular the transcripts were concentrated to the roots of P. ginseng. The ectopic expression of PgFT homologs in Arabidopsis thaliana induce precocious flowering by altering several floral pathway integrators, in addition PgFT1 transgenic Arabidopsis seedlings had an increased root length while, the influence of PgFT2 in transgenic seedlings apart from flowering is as yet to be revealed. Conclusion : Based on our results, we alleged the spatial and temporal expression of PgFT homologs could be significant in the flowering time transition of ginseng and their age specific expression pattern suggests the roots of ginseng are influential in determining the reproductive competency. On top of it, results from transgenic studies in Arabidopsis claimed that PgFT1 might involve in the underground organ development. Albeit, their molecular regulation, developmental role and the manipulation of PgFTs in ginseng flowering warranties further investigation.

Background : Panax ginseng is an important crop based on its pharmaceutical efficacy in the Asian countries, it cultivated in Korea is usually harvested after 4 to 6 years of cultivation, whereas various abiotic stresses lead to a reduction of its quality. One of them is the Aluminum (Al) accumulation. Plant growth promoting rhizobacteria (PGPR) can play a role in plant health and growth and has been considered as a new trend for supporting the growth of many crops in heavy metal occupied areas. Accordingly, this study aimed to utilize PGPR to enhance the resistance of the ginseng plant against Al stress. Methods and Results : The screening of several PGPR isolated from ginseng cultivate soil had been carried out by in vitro and in planta studies to confirm the plant growth promoting traits. Amongst, the strain Rhizobium panacihumi DCY116T was registered as one novel bacteria species and found to have IAA, siderophores production and phosphate solubilization traits, heavy metal resistance activity, and antifungal activity by in vitro test. Furthermore, in the perennial plant ginseng, fifteen minutes dipping of 108 CFU/㎖ DCY116T was enough to prime 1.5-years-old ginseng seedlings against given Al stress. Ginseng seedlings treated with DCY116T showed higher biomass, active compounds (like ginsenoside, proline, total phenolic, sugars) than non-bacterized Al-stressed seedlings. DCY116T treatment induced Al stress related genes, ROS scavenging genes, ABA biosynthesis genes and root hair formation genes which prevent the Al stress. Conclusion : R. panacihumi DCY116T can be used to prime ginseng seedlings and induced its Al resistance. Also, it can promote the root hair formation. Based on these results, strain DCY116T can be as a potential plant growth promoting bacterium used in the future for the cultivation of ginseng or other kind of crop/ plant growth under Al stress.

Backgound : Inflammatory bowel diseases (IBDs) are chronic disorders that are characterized by intestinal epithelial inflammation and injury. Currently, the most employed therapies are antibiotics and anti-inflammatory drugs; however, the side effects limit long-term effectiveness. Methods and Results : We evaluated the impact of glucose-lysine Maillard reaction products (Glc-Lys MRPs) on colitis, induced in rats by an administration of 5% dextran sulfate sodium (DSS) in drinking water. Glc-Lys MRPs ameliorate DSS-induced colitis, as determined by a decrease in disease. index activity, colon weight/length ratio, nitric oxide levels in serum, recovery of body weight loss, colon length and serum lysozyme levels. Furthermore, Glc-Lys MRPs increase the glutathione content and the activity of glutathione peroxidase, superoxide dismutase and catalase, and inhibit lipid peroxidation and myeloperoxidase activity in colon tissues. In particular, Glc-Lys MRPs suppress the mRNA level of the inflammatory cytokines and nuclear factor-κB in colon tissues. Conclusion : This study suggests the potential of Glc-Lys MRPs in preventing or treating IBDs.

Background : Glycyrrhiza uralensis Fischer is one of most important medicinal (Glycyrrhizae radix) herbs in traditional oriental medication and they are also important commercial products used worldwide in sweetening and flavoring. They contain the similar compounds, such as the triterpenoid saponins and flavonoids. Therefore, it is necessary to develop more efficient and sustainable methods for the production of G. uralensis Fischer and its medicinal constituents. This study was accomplished to investigate the effect of medium composition on in vitro propagation and plantlet regeneration from nodal explants of G. uralensis Fischer, and to establish a reliable protocol for micropropagation. Methods and Results : Young and actively growing stem segments were excised from adult plants of new cultivar ‘manju’. They were cut into a 1cm nodal segments with single node after sterilization, and cultivated in the different medium supplemented with various plant growth regulators for two weeks. For shoot multiplication, one-node stem segments, approximately 1 ㎝ in length, were taken from in vitro derived shoots and subcultured. After four weeks of culture, the efficacy of each medium on shoot proliferation and growth was determined, and these shoots were transferred onto medium with different auxin hormones for rooting. Conclusion : After seven to ten days of culture, shoots began to emerge from axillary buds. They showed a vigorous growth and elongation in multiplication medium. During culture period, in vitro cultured plantlets showed significantly different responses to the respective medium with different plant growth regulators. Our experiments confirmed that in vitro growth and multiplication of palntlets could depend on its reaction to the different medium composition, and this micropropagation techniques could be a useful system for healthy and vigorous plant production.

Background : Saliva miltiorrhiza is a perennial medicinal crop of Lamiaceae and has been reported to have various functionalities such as improvement of cardiovascular function and antihypertensive. However, domestic cultivation technique in Korea has not been established yet. Seed production system is required to increase propagation efficiency and to establish breeding basis for stable production. This experiment was carried out to determine the optimal date of the S. miltiorrhiza. Methods and Results : S. miltiorrhiza was seeded in April of 2017 and 2018. The seeds were harvested at 5-day intervals based on the flowering period of 2018. Number of peduncles, flowers, and seeds were investigated at each period. And germination rate was also investigated. The optimum seed harvest time was estimated by relationship between the seed production and days after flowering period (DAF) or cumulative temperature. The flowering rate and amount of seed of 2-years plant was higher than 1-year plant. As DAF increased, number of flowers, fertility rate, and the amount of seeds increased. At 20 DAF, 1-year, and 2-years plant respectively produced 2.2 g and 25.2 g of seed. Seed production (SP) and DAF or accumulated temperature (AT) had a quadratic relation, SP (g) = -0.1052DAF2 + 3.7396DAF - 12.347 (R2 = 0.828) and SP (g) = -0.0002AC2 + 0.1727AC - 9.146 (R2 = 0.853). The maximum amount of seed was estimated to be 20.9 g at 17.8 DAF or 21.3 g at 353℃. The seed germination rate was increased until 20 DAF with 62.0%. The amount of seed and germination rate were decreased at 25 DAF. Conclusion : The seeds of S. miltiorrhiza can be produced the highest amount with high viability on the 18th day after flowering period in 2-years after planting.

Background : Due to changes in climate and cultivation conditions, the growth monitoring is an essential factor in improving crop productivity. With the recent development of image analysis technology incorporating ICT, it has become possible to constantly monitor the crop growth. As a medicinal crop specialized in Gyeongsangbuk-do, Korea, Cnidium officinale Makino was examined for the possibility of growth diagnosis through image analysis for stable production. Methods and Results : The IP camera was installed at 2.5 m height in experiment field. The RGB image of every 06:00 was captured from July 1 to July 30 and used for analysis. The captured images were analyzed using the image analysis tool, Image J. The greeness was estimated by the average value of the green histogram. The canopy size was determined by the color range (red: 0-255, green: mean value-255, blue: 0-255) and was calculated as the ratio of pixels number of the entire image to those of the selected area. The growth temperature during investigation period was measured by Hobo MX2300. High temperature, excess of 28℃, was compared to stress response such as decrement of canopy size. The greeness and the canopy size are respectively represented by the quadratic function greeness = -0.0722GD2 + 6248.9GD – 1e + 08 (GD, growing day; R2 = 0.46) and canopy size = -0.0462GD2 + 3996.7GD – 9e + 07 (R2 = 0.93). From July 11, it began to exceed the growth limit temperature of 28℃, and the canopy size began to decrease from this period. Between the canopy size (C) and the accumulated temperature exceeding 28℃, there was a negative correlation, C = -0.13ATEC + 56.75 (R2 = 0.87) during the decreasing period. Conclusion : Extraction of color information in Cnidium officinale Makino using RGB image should be preceded by standardized setting, but it is considered to be useful tool for analyzing the change of quantitative characteristics over time. In the future, it is necessary to make a comparative study with the actual growth rate in the image diagnosis.
