Crocin is a carotenoid that may protect cells against oxidative stress by scavenging free radicals particularly superoxide anions. It has been reported that oocyte maturation is influenced by the free radicals generated during in vitro culture (IVC) process. The objective of study was to examine the effect of crocin in in vitro maturation (IVM) medium as an antioxidant on oocyte maturation and embryonic development after parthenogenesis (PA). Cumulus-oocyte complexes (COCs) were collected from ovaries of prepubertal gilts. The basic medium for IVM was medium-199 containing 10% pig follicular fluid, cysteine, pyruvate, epidermal growth factor, kanamycin, insulin, and hormones. Oocytes were treated for 44 hours with crocin at 0, 25, 50, and 100 μg/ml during IVM. Oocytes reached the metaphase II stage were induced for PA and cultured for 7 days in porcine zygote medium-3. Nuclear maturation of oocytes was not influenced by various concentrations of crocin (89.0, 87.3, 84.3, and 94.1% for control, 25, 50, and 100 μg/ml crocin, respectively). IVM oocytes treated with 50 μg/ml crocin showed a higher (P<0.05) intraoocyte glutathione (GSH) contents than untreated oocytes (1.00 vs. 1.29 pixels/oocyte). Blastocyst formation of PA embryos treated with 50 (42.9%) and 100 μg/ml crocin (43.8%) was significantly higher (P<0.05) than oocytes treated with 25 μg/ml crocin (30.5%) but not different from that (35.2%) of untreated oocytes. In summary, crocin increases cytoplasmic maturation in terms of intraoocyte GSH content which may be beneficial for later embryonic development by protecting from harmful effect of reactive oxygen species. Further studies are needed to determine whether the beneficial effect of crocin treatment during IVC would be shown in embryonic development after in vitro fertilization and somatic cell nuclear transfer.

Loop-mediated isothermal amplification (LAMP) is a rapid, specific, cost-effective detection method by amplifying nucleic acid under isothermal conditions. In this study, we used LAMP for detection of Hamiltonella defensa that lives as a facultive endosymbiont of whitefly ‘Bemisia tabaci’. We designed the Hamiltonella-specific primers by targeting 16S ribosomal RNA gene and validated the specificity of one primer set. To find the optimum temperature for our primer set, the LAMP reaction was held at the temperature, 60℃, 62℃ and 65℃. As a result, 62℃ was the optimum reation temperature for LAMP reaction. Specificity of primer set was tested by the reaction to both Trialeurodes vaporariorum and B. tabaci. After the whole procedure, the amplicons by LAMP were visualized by adding SYBR Green to the reaction tube.

Cotton aphid, Aphis gossypii Gloever is one of the major pests on a wide range of economically important crops in the world. The sustained use of chemical insecticides to control the aphid has led to the emergence of resistant strains to numerous used insecticides. As an alternative strategy entomopathogenic fungi have been used as part of integrated pest management program to control aphid, especially insecticide-resistance population. In particular, Beauveria bassiana-based commercial bio-insecticide has been used to reduce the pest population under greenhouse conditions in various countries. In this study, we investigated the control efficacy of a prototype of commercial mycopesticide using an B. bassiana (wettable powder) against cotton aphid on potted cucumber plant in greenhouse conditions.

Differential gene regulation is crucial for development. Indianmeal moth is a global pest of stored and processed food products. Here, we determined the whole developmental expression patterns of 13 genes (shsp, hsp70, grp78, and hsp90), ecdysone receptor (EcR), ultraspiracle (USP), hemolin, β-1,3-glucan recognition protein (βgrp), prophenoloxidase (ProPO), superoxide dismutase (SOD) and thioredoxin peroxidase (Tpx), and two hexamerin storage protein genes (SP1 and SP2) related to growth, stress, metabolism, and host defense. We also studies effects of Bracon hebetor envenomation on these gene expressions to explore their role in host regulation. We found unexpected transcriptional peaks especially hsps which were high in egg and adult stages. This study provides comprehensive understanding about development and parasitoid regulation at molecular level.

The bluetongue is a serious vector-borne viral disease that affect wild and domestic ruminants. It is transmitted by midges belonging to the genus of Culicoides which count more than 1350 species. Since 1998, the disease has been spread in Europe and North Africa in 2000 including Algeria, while Korea reported serotype 1 of the virus on 2015. To know further about the existence and distribution of Culicoides species in Algeria, adult midges were collected from 17 different regions in Algeria from 2009 to 2015. The Interactive Identification Key of Culicoides (IIKC) has been used, followed by molecular identification by the sequencing of partial COI gene. At least 21 genetically different species have been identified. Among them, at least 5 potential virus vector species are present in Algeria. This study provides important insights for the understanding of genetic diversity of Culicoides and the existing potential of bluetongue spread in Algeria.

Chinese cabbage, a traditional food source in Asia, has been highlighted due to its functional aspects. In particular, breakdown products of glucosinolates (GSLs) that contribute to the unique flavor and fragrance of Brassicaceae vegetables and act in plant defense mechanisms has been known as an anti-cancer agent. The objective of this study was to investigate optimal condition to grow Chinese cabbage with focusing on enhancing glucosinolates contents. The main tools for our analysis were correlation analysis and principal component analysis (PCA), and they were applied into a data obtained from Chinese cabbage grown in the plant factory. The data included glucosinolate contents and growth conditions (day, temperature, humidity, and EC). As a result of correlation analysis, day showed the largest significance with glucosinolates, followed by EC and temperature. The results of PCA showed that three principal components were able to explain more than 75% of variations in glucosinolate contents caused by different growth conditions. The first principal component (PC1) was mainly represented by day, EC, and temperature, while the main factors in the second (PC2) and the third (PC3) components were represent humidity, and , respectively. In conclusion, glucosinolate contents in Chinese cabbage were largely affected by day, EC, and temperature during the growth.

The belief that honey bee venom (BV) can be used to treat certain immune-related diseases, such as arthritis and rheumatic conditions, goes back to antiquity. A growing number of reports have demonstrated that BV contains at least 18 pharmacologically active components, including phospholipase A2 (PLA2). Recent research has shown that bee venom PLA2 (bvPLA2) induces protective immune responses against several diseases including asthma, Parkinson’s disease, and drug-induced organ inflammation. However, the antiviral properties of bvPLA2 have not been well investigated. Hence, we examined the potential inhibitory effects of bvPLA2 and its possible mechanism of action against a broad panel of pathogenic viruses in vitro. Pre-treatment with bvPLA2 significantly inhibited the replication of vesicular stomatitis virus (VSV), coxsackie virus (H3), enterovirus-71 (EV-71), herpes simplex virus (HSV) and Adenovirus (AdV) dramatically. However, bvPLA2 did not show antiviral activity against Influenza A virus (PR8) and Newcastle disease virus (NDV). Such inhibitory effects were explained by blocking of the attachment of the virus to cells upon bvPLA2 treatment. Additionally, we observed that Heparan sulfate (HS) has an inhibitory effect on the attachment of HSV to the cell surface dose dependently, which was inconsistent with bvPLA2 treatment. These findings suggest that bvPLA2 has an inhibitory effect on the replication of diverse viruses by blocking their attachment to the cell surface and could be a promising source of natural antiviral agents.

Determined according to urban parks and green urban planning, urban planning facilities are established in accordance with the “National Land Planning and Utilization Act.” The designation of land use is accompanied by certain restrictions, and urban planning facilities are provided for cases that are based on the expropriation or use of land property rights, such as the result of enforcement. The property rights and time of certain individuals may even be sacrificed to ensure the enforcement of a project, but it is uncertain whether the Constitutional Court ruling on the Constitution was administrative or opportunistic with regard to the practice bar. This study is based on case studies, and consideration is given to working out a long-term solution to the problem of urban parks owned by local governments, and the budget reality. In terms of presenting evidence, 248 response measures will be provided among the as-yet non-constructed parks in the city of Daejeon, where a park is likely to lose its qualification analysis and assessment system to cope with the indicators elicited for the sake of prioritization. According to these new evaluation criteria, the priority order of long-term unexecuted urban parks in the city of Daejeon is as follows. Gayang Birae, Guam, Daesa, Sajeong, Hodong, and Hoedeok neighborhood park were evaluated in the order.

Aneurysmal bone cyst (ABC) in maxilla is a rare and benign lesion but shows extensive bony destruction, occasionally accompanied with secondary osseous lesions, i.e., central giant cell granuloma, ossifying fibroma, fibrous dysplasia, etc. As the pathogenesis of ABC has not been clearly defined, ABC is diagnostically challenged due to its variable histological features. A 17-year-old boy showed a huge radiolucent lesion at right anterior maxilla, which was accidentally found in routine dental-radiological examination for orthodontic treatment. He had no medical history of systemic disease, and did not remember any traumatic experience on his right anterior maxilla. The radiolucent lesion involved periapical area from right central incisor to right first premolar, and was clinically diagnosed as odontogenic keratocyst. During surgical operation a cyst-like sac was enucleated with severe hemorrhage. In the histological observation the thick fibrous sac showed no lining epithelium, and its luminal side disclosed multiple aneurysmal spaces which were shrunken and almost obliterated. The fibrous sac itself was hyperplastic with abundant vascular channels, and produced fibromatous thickening associated with ossifying trabecular bones. This fibro-osseous tissue was hamartomatous, which was not directly connected and organized with marrow bone of maxilla. Finally, the present case was diagnosed as secondary type ABC differentially from traumatic bone cyst (TBC), odontogenic cyst, and central reparative granuloma. And it was presumed that the hamartomatous proliferation of fibro-osseous tissue in the cystic sac of ABC could produce the swelling pressure effect in the bone marrow similar to the overgrowth of central giant cell granuloma, ossifying fibroma, fibrous dysplasia, etc., in the secondary type ABC.

This research aims to examine the effects of national culture and leadership style on safety performance in bulk shipping companies. Survey data collected from 322 respondents working in dry bulk carriers was used, a multiple regression analysis was conducted to analyze the influence of national culture and leadership styles (i.e. transformational, passive management, and contingent reward) on safety attitude and safety behavior. The results indicate that national culture dimensions such as power distance, uncertainty avoidance, collectivism, and long-term orientation had a positive influence on safety behaviour. Long-term orientation had a positive influence on safety attitude, whereas masculinity had a negative influence on safety attitude of seafarers. Specifically, this research found that transformational leadership had a positive influence on safety attitude and safety behaviour of seafarers. Moreover, practical implication from the research findings to improve ship safety in dry bulk shipping were discussed.

Various insect pests and plant disease can outbreak in a field. For the effective control of pests and plant diseases during crop cultivation, farmers simultaneously or sequentially spray various eco-friendly agricultural materials (EFAM), chemical pesticides and microbial control agents on the same fields. It was reported that many agrochemicals are harmful to entomopathogenic fungi, especially some fungicides with broad spectrum activity that are routinely applied for the control of plant diseases. In addition, some pesticides may antagonize the potential insecticidal activity and efficiency of entomopathogenic fungi. Therefore, sometimes the utilization of fungal entomopathogen in forestry and agricultural production is limited because of the undesirable interference from some fungicides and pesticides. There is little research that examines the compatibility of these EFAMs with entomopathogenic fungi and the influence of EFAMs on the control efficacy of mycopesticides. We conducted a study of influence of pretreated eco-friendly agricultural materials on control efficacy of Isaria javanica isolate against sweet potato whitefly.

Entomopathogenic fungi are natural enemies of insect pests and contribute to the natural regulation of their host populations. These fungal group are often used as active ingredients for microbial insect pest control. In addition, the potential antimicrobial effect by entomopathogenic fungi including Beauveria bassiana, Lecanicillium spp., and Isaria fumosorosea have recently been reported against fungal plant pathogens. Dual microbial control effects with entomopathogenic fungi against both aphids and cucumber powdery mildew had reported in Canada. In our previous studies we conducted bioassay with entomopathogenic fungi to develop dual microbial control agent which can control both aphid and fungal plant disease. We selected an Beauveria bassiana isolate which has high dual control effects against both cotton aphid, Aphis gossypii and sclerotinia rot, Sclerotinia sclerotiorum. In this study, we have tested the dual control efficacy of the B. bassiana isolate against cotton aphid and sclerotinia rot on whole potted cucumber plants. We found that the B. bassiana isolate protected the plant from cotton aphid and sclerotinia rot under laboratory condition.

Tetranychus urticae is extremely hard to control by conventional acaricides due to its rapid development of resistance to nearly all arrays of acaricide. As an alternative control measure of acaricide-resistant mites, RNA interference (RNAi)-based method has recently been suggested. A double-stranded RNA (dsRNA) delivery method using multi-unit chambers was established and employed to screen the RNAi toxicity of 42 T. urticae genes. Among them, the dsRNA treatment of coatomer I (COPI) genes, such as coatomer subunit epsilon (COPE) and beta 2 (COPB2), resulted in high mortality [median lethal time (LT50) = 89.7 and 120.3 h, respectively]. The transcript level of the COPE gene was significantly (F3,9 = 16.2, P = 0.001) reduced up to 24% following dsRNA treatment, suggesting that the toxicity was likely mediated by the RNAi of the target gene. To identify the deferentially expressed gene upon dsRNA ingestion, RNA-seq was employed to compare the transcriptional profiles between mites fed dsEGFP and dsCOPB2. Approximately 928 of genes were up- or down-regulated significantly (P < 0.05) compared to control and 182 genes were commonly responded to the treatment of both dsRNAs. Those dsRNA-responsible genes were mainly categorized into metabolic enzymes, transporters and secretory proteins. Further study would be necessary to elucidate the roles of dsRNA-responsible genes in mite’s dsRNA uptake and defense.
