Sloan-Kettering virus gene product of a cellular protooncogene c-Ski is an unique nuclear pro-oncoprotein and belongs to the Ski/Sno proto-oncogene family. Ski plays multiple roles in a variety of cell types, it can induce both oncogenic transformation and terminal muscle differentiation when expressed at high levels. The aim of the present study was to locate Ski protein in rat ovaries in order to predict the possible involvement of Ski in follicular development and atresia. First, expression of c-Ski mRNA in the ovaries of adult female rats was confirmed by RT-PCR. Then, ovaries obtained on the day of estrus were subjected to immunohistochemical analysis for Ski and proliferating cell nuclear antigen (PCNA) in combination with terminal deoxynucleotidyl transferase- mediated dUTP nick end-labeling (TUNEL). Ski was expressed in granulosa cells that were positive for TUNEL, but negative for PCNA, regardless of the shape and size of follicles. Expression of Ski in TUNEL-positive granulosa cells, but not in PCNA-positive granulosa cells, was also verified in immature hypophysectomized rats having a single generation of developing and atretic follicles by treatment with equine chorionic gonadotropin (eCG). These results indicate that Ski is profoundly expressed in the granulosa cells of atretic follicles, but not in growing follicles, and suggest that Ski plays a role in apoptosis of granulosa cells during follicular atresia.

Ski protein is implicated in proliferation/differentiation in a variety of cells. We had previously reported that Ski protein is present in granulosa cells of atretic follicles, but not in preovulatory follicles, suggesting that Ski has a role in apoptosis of granulosa cells. The alternative fate of granulosa cells other than apoptosis is to differentiate to luteal cells, however, it is unknown whether Ski is expressed and has a role in granulosa cells undergoing luteinization. Thus, the aim of the present study was to locate Ski protein in the rat ovary during luteinization to predict the possible role of Ski. In order to examine the expression pattern of Ski protein along with the progress of luteinization, follicular growth was induced by administration of equine chorionic gonadtropin to immature female rat, and luteinization was induced by human chorionic gonadtropin treatment to mimic luteinizing hormone (LH) surge. While no Ski-positive granulosa cells were present in preovulatory follicle, Ski protein expression was induced in response to LH surge, and was maintained after the formation of corpus luteum (CL). Though Ski protein is absent in granulosa cells of preovulatory follicle, its mRNA (c-ski) was expressed and the level was unchanged even after LH surge. Taken together, these results demonstrated that Ski protein expression is induced in granulosa cells upon luteinization, and suggested that its expression is regulated post-transcriptionally.

Magnetars are neutron stars possessing a magnetic field of about 1014-1015 G at the surface. Thermodynamic properties of neutron star matter, approximated by pure neutron matter, are considered at finite temperature in strong magnetic fields up to 1018 G which could be relevant for the inner regions of magnetars. In the model with the Skyrme effective interaction, it is shown that a thermodynamically stable branch of solutions for the spin polarization parameter corresponds to the case when the majority of neutron spins are oriented opposite to the direction of the magnetic field (i.e. negative spin polarization). Moreover, starting from some threshold density, the self-consistent equations have also two other branches of solutions, corresponding to positive spin polarization. The influence of finite temperatures on spin polarization remains moderate in the Skyrme model up to temperatures relevant for protoneutron stars. In particular, the scenario with the metastable state characterized by positive spin polarization, considered at zero temperature in Phys. Rev. C 80, 065801 (2009), is preserved at finite temperatures as well. It is shown that, above certain density, the entropy for various branches of spin polarization in neutron matter with the Skyrme interaction in a strong magnetic field shows the unusual behavior, being larger than that of the nonpolarized state. By providing the corresponding low-temperature analysis, we prove that this unexpected behavior should be related to the dependence of the entropy of a spin polarized state on the effective masses of neutrons with spin up and spin down, and to a certain constraint on them which is violated in the respective density range.

Lettuce was a leaf vegetable with short growing term and cold-tolerant plant. We grew it the whole year. The price of lettuce is the most expensive from July to September. But damages by lots of insect pests appear to greenhouse grown lettuce in summer season. In this study, we observed biological controls of western flower thrips(Franklinella occidentalisi) by the predator mites, Amblyseius swirskii and Neoseiulus cucumeris in lettuce greenhouse from July 7 to September 10. Hypoaspis aculerfer larvae and adults were released 30 individuals per m2 at the first one time as lettuce was planted on the greenhouse. We checked the density of F. occidentalis every 10 days on the 30 lettuce leaves. When the population of F. occidentalis reached 1~5 individuals per a leaf, A. swirskii larvae and adults were released 35 individuals per m2 at 1~3 times and Neoseiulus cucumeris larvae and adults also were released 150 individuals per m2 at 2~3 times in each lettuce greenhouse. During the high temperature season, Amblyseius swirskii was more effective biological controls of F. occidentalis than Neoseiulus cucumeris.

Background : Korean ginseng is an important cash crop in Asian countries. However, plant yield is reduced by pathogens. Among the Ilyonectria radicicola-species complex, I. mors-panacis is responsible for root-rot and replant failure of ginseng in Asia. It is essential to find a way to reveal the existence of the pathogen before starting cultivation. Therefore, qRT-PCR method is developed to detect and quantify the pathogen in ginseng soils. Methods and Results : In this study, species specific Histone H3 primer set is developed for the quantification of I. mors-panacis. The primer set was applied on DNA of other microbes to evaluate its sensitivity and selectivity on I. mors-panacis DNA. Sterilized soil samples artificially infected by the pathogen in different concentrations were used to evaluate the ability of the primer set to detect the pathogen population in the soil DNA. Finally, the pathogen was quantified in many natural soil samples. The designed primer set was found to be sensitive and selective to I. mors-panacis DNA. In artificially infected sterilized soil samples, the estimated template using qRT-PCR was positively correlated with the pathogen concentration in soil samples (R2=0.94), disease severity index (R2=0.99), and colony forming unit (R2=0.87). In the natural soils, the pathogen was recorded in the most of fields produce bad yields with the range of 5.82 ± 2.35 to 892.34 ± 103.70 pg/g of soil. Conclusion : According to the presented results, the proposed primer set is applicable for estimating soils quality before ginseng cultivation. This will help in the disease management and crop protection in the future.

Background : Panax ginseng C.A.Meyer is a perennial herb and its growth potential keep relatively high at least for the early 3 years old. If the transplanted ginseng roots of 2 or 3 years old grow well, a period of cultivation and the disease management cost could be reduced for the production of 6 years old ginseng roots. Methods and Results : The rooting and growth of a transplanted ginseng root could be affected by the age of a transplant. Therefore, the experiments were conducted to examine the influences of the age factor on the rooting and growth of transplanted ginseng roots. The ginseng roots with different plant ages in growth years (1, 2, and 3 years old), used in this study, were obtained by raising in a seedling production field near the Punggi Ginseng Experiment Station. The transplants were continuously grown under the same conditions after transplanting. The number of lateral or branched roots at the root-growing period after transplanting was basically higher in originally older plant ages compared to younger plants: the numbers of secondary and tertiary branched roots of the 1 year-old transplants were 8, 12, respectably; 2 years old: 12, 20; 3 years old: 12, 28. Those did not show any difference in both overground and underground growing, in compared to conventional cultivations with roots of a year old. The plants grew well without anthracnose, spotting disease, and high temperature injury. Conclusion : The conventional cultivation period of above 5 or 6 years for the raw material of red ginseng might be shorten and also the cost of disease control could be decreased, by transplanting of 2 or 3 years old roots raised at a seedling nursing field.

A variety of genetically modified (GM) crops have been developed in Korea. In these crops, the resveratrol-enriched transgenic rice plant has moved ahead to generate the dossier for regulatory review process required for commercialization of GM crop. The resveratrol-enriched transgenic rice plant could be released to farmers for cultivation after national regulators have determined that it is safe for the environment and human health. Here we developed a PCR-based DNA marker based on flanking sequences of transgene for the discrimination of zygosity in resveratrol-enriched transgenic rice plant. This DNA marker will be useful for identifying of resveratrol-enriched transgenic rice plant, and can also be use to estimate transgene movement occurred by pollen transfer or seed distribution.

Clubroot is a devastating disease caused by Plasmodiophora brassicae and results in severe losses of yield and quality in Brassica crops including Brassica oleracea. Therefore, it is important to identify resistance gene for CR disease and apply it to breeding of Brassica crops. In this study, we applied genotyping-by-sequencing (GBS) technique to construct high resolution genetic map and mapping of clubroot resistance (CR) genes. A total of 18,187 GBS markers were identified between two parent lines resistant and susceptible to the disease, of which 4,103 markers were genotyped in all 78 F2 plants generated from crossing of both parent lines. The markers were clustered into nine linkage groups spanning 879.9 cM, generating high resolution genetic map enough to refine reported reference genome of cabbage. In addition, through QTL analysis using 78 F2:3 progenies and mapping based on the genetic map, two and single major QTLs were identified for resistance of race 2 and race 9 of P. brassicae, respectively. These QTLs did not show collinearity with CR loci found in Chinese cabbage (Brassica rapa) but roughly overlapped with CR loci identified in cabbage for resistance to race 4. Taken together, genetic map and QTLs obtained in this study will provide valuable information to improve reference genome and clubroot resistance in cabbage.
