Purpose: Introduction of the LEGO Mindstorms EV3 as a low cost research tool for various studies and to show how study tools could be built using the LEGO with ease. Methods: To show what the LEGO Mindstorms EV3 could do for studies, programming method was described and some example devices used previous papers were reconstructed; EX1 - Selection of left right device for Stoet(2010), EX2 - Time expectation device for Kim and Ryu(2015), and EX3 - Four digits random number generator for Huh and Lee(2015). Time delay of each example device was tested to evaluate effectiveness of the LEGO Mindstorms EV3 as research device. Results: Combination of single execution structure and button switch on the EV3 brick showed 1 ms error of time measurement, and combination of single execution structure and touch sensor showed 10 ms error of time measurement. 1 ms error of time measurement is same accuracy achieved by Windows based computer system.
본 연구는 전남 목포시에 위치한 H 노인복지관 주간보호센터를 이용하는 80~86세의 뇌질환 노인 7명을 대상으로 원예치료 프로그램을 실시하여 자아존중감, 감정균형, 얼굴표정에 미치는 영향을 알아보고자 하였다. 3월 28일부터 8월 21일까지 매주 1회 1시간씩 총 20회에 걸쳐 실시하였고, 자아존중감, 감정균형 척도, 스마일 척도 평가지를 이용하여 원예치료이 효과를 조사하였다. 자아존중감은 대상자 7명 전부 향상되었으며, 원예치료 프로그램 실시 전 평균 25.9에서 실시 후 평균 33.0으로 높아졌고, 통계적으로도 유의성을 나타냈다. 감정균형은 프로그램 실시 전에는 평균 13.6이었으나 실시 후에는 평균 16.7점으로 높아져 원예치료 프로그램이 정서 안정에 효과적이었음을 알 수 있었고, 통계적으로도 유의성을 나타냈다. 스마일 척도의 변화를 매 회기마다 조사한 결과 프로그램 실시 후 얼굴표정이 밝아졌고, 전체 평균이 실시 전 4.3에서 실시 후 4.8로 향상되었다. 따라서 꽃장식과 압화작품 만들기와 같은 원예치료 프로그램은 뇌질환 노인들의 자아존중감을 향상시키고, 정서적 안정에 효과적임을 알 수 있었다.
An efficient plant regeneration system of Gentiana scabra through somatic embryogenesis was established. Leaves and roots of seedlings of Gentiana scabra excised after germination were cultured on MS basal medium with 2,4-D, NAA or BA. Embryogenic callus was obtained on MS medium with 0.5 mg/L 2,4-D alone or 0.1 mg/L 2,4-D combimation with 1.0 mg/L BA after 45 days of culture. These embryogenic calli gave rise to somatic embryos, which subsequently developed into plantlets on MS medium without PGRs. Also, shoots were effectively differentiated from embryogenic callus when root segments were cultured on MS medium supplement with 0.1 mg/L 2,4-D and 1.0 mg/L BA. Shoots were effectively rooted on MS medium without PGRs. In vitro flowers were formed from plantlets cultured on MS medium with 5% sucrose after 60 days of culture.
We studied a extraction and degradation of alginate from seaweed-stems using microorganism DS-02. DS-02 has a maximum growth rate at 30℃ and the enzyme has a maximum activity of alginate extraction at 35℃. The yield of alginate extraction using DS-02 is about 16.0% for 3.0 hour and molecular weight of the alginate decreased to about 1/8 of initial value after 24 hour extraction. Alginate extraction method by DS-02, compared with general alkali-extraction method, has an advantage of decreasing the molecular weight of alginate during extraction.
We studied a storage of waste-brown seaweed at room temperature and degradation of alginate in seaweed by microorganism DS-02.
The seaweeds, mixed with 5.0 wt% DS-02 and sealed in vinyl package without any other treatment, could be stored longer than 1 year without spoilage at room temperature.
During the storage process, the alginate of seaweed was decomposed by enzyme of DS-02 and the molecular weight of alginate decreased to about 1/10 of initial quantity. DS-02 growed as fast as it had maximum weight after 24 hour culture and it's enzyme had a maximum activity of alginate degradation at 40℃. The seaweed sample became particles in DS-02 culture solution and the M.W of alginate decreased to about 1/10 of initial value after 24 hour decomposition. The effect of alginate degradation with DS-02 was similar to that of degradation with 3.0 M HCl solution for 24 hour.
This study was carried out to obtain basic information for possibility of oral vaccine in carrot using Agrobacteruim -mediated transformation system. The epitope region of porcine epidemic diarrhea virus (PEDV) spike gene which is classified as a member of the Coronaviridae and causes an acute enteritis in pigs was successfully expressed in carrot (Daucus carota) using the Agrobacterium-mediated transformation system. Hypocotyl segments of in vitro germinated plantlets were infected with Agrobacteriun tumefaciens LBA 4404 harboring PEDV spike gene. Embryogenic callus (EC) was induced on MS selection medium with 1 mg/L 2,4-D, 50 mg/L kanamycin and 300 mg/L cefotaxime after 45 days of culture. Subcultured ECs on MS selection medium without 2,4-D were converted to somatic embryos (SE) of various stage; globular, heart and torpedo stage. Putative transgenic embryos were selected on MS medium with 50 mg/L kanamycin and 300 mg/L cefotaxime. Regenerated plantlets from transformed SE were induced on MS medium containing 50 mg/L kanamycin after 30 days of culture. Genomic PCR confirmed the integration of PEDV spike gene into nuclear genome of carrot and northern blot analysis demonstrated the expression of PEDV spike gene in transgenic carrot.
Effective alginate extraction from waste-brown seaweeds was studied. The waste-brown seaweeds almost consist of stems of seaweeds. Alginate could be obtained from the stems as well as leaves of seaweed. Ultrasonic vibration(47kHz) facilitated filtering step in the alginate extraction process. Among various alkalies for alginate extraction, NaHCO3 was most appropriate concerning use of dealginates. The yield of alginate extraction using NaHCO3 2wt% solution was 19.3% at 60℃. The dealginates from NaHCO3-extraction process have been found most suitable food for red-worms.
We compared the long-term metabolic effects of equal amounts of carbohydrate from potato, rice and buckwheat on glycemic indices and blood lipids in healthy subjects. Nine healthy volunteers-2 men and 7 women were studied. All subjects ate diets based on the same-7-day rotating menu differing only in that the major source of carbohydrate (about 50% of daily total calories) came either from buckwheat, rice or potato. The study was conducted with a triple crossover design over three 7 day periods. On the morning of the 8th day, fasting blood was drawn from each subject to determine serum glucose, insulin, triglycerides, total and HDL-cholesterol. Subjects were then asked to eat breakfast with their respective carbohydrate within a 20 min period. Blood samples were drawn at 30, 60, 120 and 180 min after the start of breakfast to determine glucose and insulin levels. At 30 min the glucose response to the rice meal(7.15mmol/L) and potato meal(6.71mmol/L) were greater than the response to the buckwheat meal(5.855mmol/L) (P < 0.05). The mean area under the glucose response to the curve following the rice meal was greater than that following the buckwheat meal(P < 0.05). The insulin responses to the potato and rice meals at 30 and 60 min were greater than those to the buckwheat meal (P < 0.05). The mean area under the serum insulin response curve after the rice meal was greater than of buckwheat. Blood lipids, uric acid and glycosylated hemoglobin were not affected by the three meals. The study shows that the buckwheat meal has more beneficial effects on glycemic indices than either the rice meal or potato meal in healthy subjects.
Transgenic Petunia hybrida cv. Rosanpion was produced by Agrobactepium tumefaciens LBA4404 harboring a binary vector pBI 121 containing β-glucuronidase (gus) and neomycin phosphotransferase (nptII). For genetic transformation, leaf discs were precultured on MS medium supplemented with 0.5 mg/L NAA and 1.0 mg/L BA (MNB) for 2 days and cocultured for 15 mins with A. tumefaciens. For selection of transformant, leaf discs were transferred to fresh MNB containing 50 mg/L kanamycin and 500 mg/L cefotaxime. Eighteen plants were regenerated and four were confirmed by PCR for detection of gus and nptII gene integrated into the nuclear genome of petunia ‘Rosanpion’. Using this transformation system, we expect that transgenic petunia ‘Rosanpion’ incorporating a useful gene can be produced.
An efficient regeneration system was established by using in vitro plantlets of germinated seedlings from different cultivars of lettuce (Lactuca sativa cv. Chongchima, Chongchuckmyun, Jeokchima, Jeokchuckmyun). Shoot formation were observed from all cultivars on MS medium supplemented with 0.1 mg/L NAA and 0.5 mg/L BA. In all cultivars, when cotyledon was cultured, the number of shoot per explant was more greater than that hypocotyl and leaf disc were cultured. Shoot formation rate (91.7%) was high in a cotyledon culture of cultivar, Chongchukmyun. The growth of multiple shoots derived from the cultivar, Chongchukmyun, was most effective on medium containing 0.5 mg/L BA and 1.0 mg/L GA3. When shoots were transferred on MS medium without plant growth regulators, roots were effectively differentiated. Rooted plantlets were acclimated on pots for further propagation.
