Background: The grading of Hanwoo (Korean native cattle) is based on four economic traits, and efforts have been continuously made to improve the genetic traits associated with these traits. There is a technology to predict the expected grade based on the 4 economic genetic SNP characteristics of Korean cattle calves using single nucleotide polymorphism (SNP) technology. Selection of highly proliferative, self-renewing, and differentiating satellite cells from cattle is a key technology in the cultured meat industry. Methods: We selected the Hanwoo with high and low-scored of genomic estimated breeding value (GEBV) by using the Hanwoo 50K SNP bead chip. We then isolated the bovine satellite cells from the chuck mass. We then conducted comparative analyses of cell proliferation, immunocytochemistry, qRT-PCR at short- and long-term culture. We also analyzed the differentiation capability at short term culture. Results: Our result showed that the proliferation was significantly high at High scored GEBV (Hs-GEBV) compared to Low scored GEBV (Ls-GEBV) at short- and long-term culture. The expression levels of Pax3 were significantly higher in Hs-GEBV bovine satellite cells at long-term culture. However, there were no significant differences in the expression levels of Pax7 between Hs- and Ls-GEBV bovine satellite cells at short- and long- term culture. The expression levels of MyoG and MyHC were significantly high at Ls-GEBV bovine satellite cells. Conclusions: Our results indicated that selection of bovine satellite cells by Hanwoo 50K SNP bead chip could be effective selection methods for massive producing of satellite cells.
This study aims to investigate the effects of ankle foot orthosis(AFO) on the activities of tibialis anterior(TA), soleus(Sol), rectus femoris(RF) and biceps femoris(BF) during stairs descending. The activities of TA, Sol, RF and BF were initially measured while descending 4 stairs without using AFO. The activities of the same muscles were then measured again while descending 4 stairs while using AFO. Wilcoxon signed-rank tests were used to analyze the results in order to examine the differences between the with using AFO and without using AFO. Although the activities of TA, Sol and RF were relatively lower while using AFO than without using AFO, only the differences in Sol and RF activities were significant(p<0.05). The activity of BF was relatively higher while using AFO compared to the activity of BF observed without using AFO. However, difference was not significant(p>0.05). Conclusion of this study was observed since AFO’s ground reaction force absorption during stairs descending reduced the need to use So and RF that is related to shock absorption. BF activity was increased with AFO than without during standing forward to correct the trunk stability.
This study was conducted to observe egg and larvae morphological development of carp to obtain basic data for resource conservation and taxonomic research. Brood carp used in the research (total length 67.3-75.5 cm, average 71.0±3.45 cm) were bred in a circular rearing aquarium (600×300×100 cm) using a running water system from January to July, 2015. Breeding water temperature was maintained at 23.0-25.0℃(average 24.0℃). Fertilized carp eggs were translucent and globular, and their size was 1.75-1.89 mm (average 1.82±0.06 mm). Blastoderms formed 10 min after fertilization and reached the two-cell stage 30 min after fertilization. Then, the embryo turned dark and exhibited melanophores, and blood started flowing from the heart across the egg yolk at 42 hrs and 50 min after fertilization. Hatching began 70 hrs and 26 min after fertilization larvae emerged through the egg membrane, starting from the head. The length of prelarvae immediately after hatching was 5.23-5.38 mm (average 5.31±0.11 mm) the mouth and anus were closed, and the pectoral fin was formed. Postlarvae at 18 days after hatching had a total length of 11.9-13.9 mm (average 12.9±1.40 mm), separate anal fin and back membranes, and fin ray. Juveniles fish at 35 days after hatching had a total length of 29.9-30.2 mm (average 30.1±0.13 mm), with the body covered with scales, and the same number of fin rays, color, and shape as their broodstork.
The influence of triploidization on histological characteristics of retina, trunk kidney, liver and midgut tissue, and comparative analysis of cell cycle and expression of cell cycle protein from tail fin and gill tissue in Far Eastern catfish, Silurus asotus were analyzed. In the infertile triploid fish, the nucleus and/or cell size of secondary proximal tubule cells of trunk kidney, hepatocyte and midgut epithelium are much larger than those of the corresponding cells in the diploid fish (P<0.05). However, triploid tissue showed fewer number of outer nuclear layer in retina and nuclei in secondary proximal tubule of trunk kidney than those for diploid tissue. The mean percentages of the G1-, the S- and the G2+M-phase fractions were 92.5%, 3.2% and 4.3% in tail fin tissue of diploid, and 93.4%, 2.6% and 4.0% in those of triploid, respectively. There were no significant differences in the percentages of each cell cycle fraction between diploid and triploid. The differences of cell cycle between tail fin tissue and gill tissue were statistically significant in diploid and triploid (P<0.05). Also, the differences between diploid and triploid were statistically significant in tail fin tissue and gill tissue (P<0.05). Results from this study suggest that some characteristics in the triploid exhibiting larger cell and nucleus size with fewer number of cell than diploid can be used as an indicator in the identification of triploidization and ploidy level in Far Eastern catfish.
A new dark pink colored standard rose (Rosa hybrida Hort.) cultivar, 'Global Pink' was developed by the cross between 'Nobless' and `Belami' at the Jeollanamdo Agricultural Research and Extention Services (JARES) through seedling and line selection in 2003 and characteristic evaluations during 2004~2006. 'Global Pink' showed Red purple group color (RHS No. 65B), very weak fragrance, and standard flower with good harmony between ray florlet and flower center. Its yield was 134 stems/m2 averaged over three years (2004-2007). It showed 10 days vase life and was resistant to powdery mildew. This cultivar was registered as a commercial cultivar in 2007. It can be cultured in greenhouses in Korea and will be released to the farmers in 2009.
