In the field of reproductive medicine, assessment of sperm motility is a key factor for achieving successful artificial insemination, in vitro fertilization, or intracellular sperm injection. In this study, the motility of boar sperms was estimated using real-time imaging via confocal microscopy. To confirm this confocal imaging method, flagellar beats and whiplash- like movement angles were compared between fresh and low-temperature-preserved (17℃ for 24 h) porcine sperms. Low-temperature preservation reduced the number of flagellar beats from 11.0±2.3 beats/s (fresh sperm) to 5.7±1.8 beats/s and increased the flagellar bending angle from 19.8°±13.8° (fresh) to 30.6°±15.6°. These data suggest that sperm activity can be assessed using confocal microscopy. The observed motility patterns could be used to develop a sperm evaluation index and automated confocal microscopic sperm motility analysis techniques.

Mammalian spermatogenesis occurs in a precise and coordinated manner in the seminiferous tubules. One of the attempts to understand the detailed biological process during mammalian spermatogenesis at the molecular level has been to identify the testis specific genes followed by study of the testicular expression pattern of the genes. From the subtracted cDNA library of rat testis prepared using representational difference analysis (RDA) method, a complimentary DNA clone encoding type III member of a DnaJ family protein, DnaJC18, was cloned (GenBank Accession No. DQ158861). The fulllength DnaJC18 cDNA has the longest open reading frame of 357 amino acids. Tissue and developmental Northern blot analysis revealed that the DnaJC18 gene was expressed specifically in testis and began to express from postnatal week 4 testis, respectively. In situ hybridization studies showed that DnaJC18 mRNA was expressed only during the maturation stages of late pachytene, round and elongated spermatids of adult rat testis. Western blot analysis with DnaJC18 antibody revealed that 41.2 kDa DnaJC18 protein was detected only in adult testis. Immunohistochemistry study further confirmed that DnaJC18 protein, was expressed in developing germ cells and the result was in concert with the in situ hybridization result. Confocal microscopy with GFP tagged DnaJC18 protein revealed that it was localized in the cytoplasm of cells. Taken together, these results suggested that testis specific DnaJC18, a member of the type III DnaJ protein family, might play a role during germ cell maturation in adult rat testis.

In a 12-week experiment, the rock bream, Oplegnathus fasciatus, and olive flounder, Paralichthys olivaceus, were investigated to determine the effects of starvation on their physiological parameters. The protein and DNA contents of the starved fish were significantly higher than the initial values and those of the fed fish. The RNA contents and RNA/DNA ratios of the fed fish were significantly higher than those of the other groups (P<0.05). The hematocrit, hemoglobin, red blood cells (RBC), and mean corpuscular volume (MCV) of the fed rock bream were significantly higher than at baseline (P<0.05), whereas the mean corpuscular hemoglobin concentration (MCHC) of the fed fish was lower than at baseline (P<0.05). The hematocrit, hemoglobin, RBC, and MCHC of the starved group were significantly lower than the baseline values, whereas the MCV of the starved group was significantly higher than the baseline value (P<0.05). No significant difference in alanine aminotransferase was observed between the fed fish and baseline, whereas the starved fish value was significantly higher than the baseline value (P>0.05). There were no significant differences in cortisol levels. However, the glucose level in the fed group was significantly higher than the baseline level and that in the starved group was significantly lower than the baseline level (P<0.05).

Present study aimed to determine the effect of ‘bitter melon’, a popularly used fruit in Bangladesh and several other Asian countries, on high-fat-diet-induced type 2 diabetes. To investigate the effect, ethanol extract from bitter melon (BME) as a dietary supplement with mouse chow was used. BME was found to significantly attenuate the high-fat diet (HFD) -induced body weight and total fat mass. BME also effectively reduced the insulin resistance induced by the HFD. Furthermore, dietary supplementation of BME was highly effective in increasing insulin sensitivity and reducing hepatic fat and obesity. These results indicate that BME could be effective in attenuating type 2 diabetes and could therefore be a preventive measure against type 2 diabetes.

The PCR analysis was performed on DNA samples extracted from a total of 20 individuals using six oligonucleotides primers. The author accomplished clustering analyses to reveal the Euclidean genetic distances among four clam populations from Gochang, Seocheon, Taean and Anmyeon of the Korean peninsula. The oligonucleotides primer OPA-08 generated 5 unique loci to each population, approximately 550 bp and 600 bp, respectively, in the MCS population. Especially, the primer OPA-20 generated 15 unique loci to each population, which were identifying each population, approximately 400 bp, 750 bp and 800 bp, in the MCT population. Individuals from MCG clam population (0.637±0.227) exhibited higher bandsharing values than did individuals from MCG clam population (0.402±0.115) (P<0.05). The dendrogram obtained by the six oligonucleotides primers indicates four genetic clusters: cluster 1 (MCG 01, 02, 04 and 05), cluster 2 (MCS 06, 07, 08, 09 and 10), cluster 3 (MCT 11, 12, 13, 14 and 15) and cluster 4 (MCA 16, 17, 18, 19, 20 and MCG 03). Among the twenty clam individuals, the shortest genetic distance that displayed significant molecular differences was between individuals 14 and 15 from the MCT population (genetic distance = 0.094), while the longest genetic distance among the twenty individuals that displayed significant molecular differences was between individuals MCG no. 01 and MCG no. 02 (genetic distance = 0.687). Comparatively, individuals of MCS clam population were fairly closely related to that of MCT clam population, as shown in the hierarchical dendrogram of Euclidean genetic distances.

The purpose of this study is to investigate the differences fatty acids and hormonal parameters in the spawning and non-spawning season between the diploid and induced triploid Far Eastern catfish, Silurus asotus. The measured triploids were produced by cold shock for 50 min at 4℃ in May 2014, the spawning season of diploid was in May, and the nonspawning season was designated in January. Estradiol and testosterone and gonadosomatic index of diploid were higher than those of induced triploid in spawning season (P<0.05), and those of diploid in spawning season were higher than nonspawning season. On the other hand, thyroid stimulating hormone and thyroxine of induced triploid were higher than those of diploid in spawning season (P<0.05). Erythrocyte count of diploid was higher than that of induced triploid in spawning season and non-spawning seasons. Mean corpuscular volume and mean corpuscular hemoglobin of induced triploid were higher than those of diploid in both seasons (P<0.05). Percentages of total saturated fatty acids and n-3 polyunsaturated fatty acids of induced triploid were higher than those of diploid in spawning season, but those of diploid were higher in non-spawning season (P<0.05). Percentages of total mono unsaturated fatty acids and total n-6 polyunsaturated fatty acids of diploid were higher than those of induced triploids in spawning season, while those of induced triploid in non-spawning season were higher (P<0.05). Therefore, induced triploids in the spawning season tend to concentrate on growth and lipid-synthesization, whereas, diploids concentrate on reproduction and gonadal maturation rather than on growth. In non-spawning season, growth and lipid- synthesization were not significantly different between diploid and induced triploid.

This study was conducted to observe egg and larvae morphological development of carp to obtain basic data for resource conservation and taxonomic research. Brood carp used in the research (total length 67.3-75.5 cm, average 71.0±3.45 cm) were bred in a circular rearing aquarium (600×300×100 cm) using a running water system from January to July, 2015. Breeding water temperature was maintained at 23.0-25.0℃(average 24.0℃). Fertilized carp eggs were translucent and globular, and their size was 1.75-1.89 mm (average 1.82±0.06 mm). Blastoderms formed 10 min after fertilization and reached the two-cell stage 30 min after fertilization. Then, the embryo turned dark and exhibited melanophores, and blood started flowing from the heart across the egg yolk at 42 hrs and 50 min after fertilization. Hatching began 70 hrs and 26 min after fertilization larvae emerged through the egg membrane, starting from the head. The length of prelarvae immediately after hatching was 5.23-5.38 mm (average 5.31±0.11 mm) the mouth and anus were closed, and the pectoral fin was formed. Postlarvae at 18 days after hatching had a total length of 11.9-13.9 mm (average 12.9±1.40 mm), separate anal fin and back membranes, and fin ray. Juveniles fish at 35 days after hatching had a total length of 29.9-30.2 mm (average 30.1±0.13 mm), with the body covered with scales, and the same number of fin rays, color, and shape as their broodstork.

An experiment was conducted to investigate the annual reproductive cycle of a Korean endemic species, Acheilognathus majusculus, from Jeokseong-myeon located in Seomjin River. The reproductive cycle is examined histologically regarding water temperature and day length of the habitat, the gonadosomatic index (GSI), the female ovipositor length index (OLI), monthly variation in egg diameter distribution, and developmental characteristics of female and male gonads. The maximum GSI was found in 19.21±2.32 and 6.90±0.53 for female and male respectively when water temperature (14℃) and day length (11.1hr) began to rise. On the other hand, the minimum level was reached during August (1.87±0.67 for female and 0.88±0.50 for male). No samples represent with measurable ovipositor between September and November, while the longest ovipositor length index was in April (79.68±4.69%). We compared and calculated the stages of testis and ovary development process in order to determine the germ cell development characteristics and the reproductive cycle. According to the result, we classified the female Acheilognathus majusculus reproductive cycle into four stages: Ripe (April) and spawning phase (May to June), degenerative phase (July), growing phase (August to December), and mature phase (January to March). The annual reproductive cycle of male Acheilognathus majusculus was categorized into five stages viz. Ripe and spawning phase (May to June), degenerative phase (July to August), resting phase (September to November), growing phase (December to February), and mature phase (March to April).

Surgical castration (also known as orchidectomy, ORX) has been frequently performed to avoid uncontrolled breeding. However, it has some serious disadvantages. Several laboratories have developed chemical castration methods, using bilateral intratesticular injection (BITI) of simple chemical solutions. The present study was undertaken to compare the effects of ORX and of hypertonic saline BITI on the androgen-sensitive tissues such as pituitary and hypothalamus. Serum testosterone (T) levels of ORX animals and hypertonic saline BITI animals (SAL) after 4 weeks of the manipulations exhibited significantly drops as compared with the levels of intact animals (Intact:ORX:SAL = 7.74± 1.31:1.34±0.19:1.28±0.18 ng/ml, p<0.001). Both ORX and BITI method induced similar stimulatory effects on the pituitary gonadotropin subunits and hypothalamic KiSS-1 gene expressions. In contrast, the effects of ORX and hypertonic saline BITI on hypothalamic GnRH gene expression were different from these gene expressions, shown an inverse relationship between the two groups (Intact:ORX:SAL = 1:0.45±0.06:1:2.07±0.41:1.51±0.37 AU; ORX, p<0.001; SAL, p< 0.05). In conclusion, we provided evidence that hypertonic saline BITI method has equivalent efficacy of T depletion to surgical castration in rats. The present study suggests the hypertonic saline BITI could be a promising substitute to conventional surgical castration.

Direct communication between neighboring cells through connexin (Cx)-based gap junction is a crucial biolo– gical manner to regulate functions of a tissue consisting of multi-cell types. The present research evaluated expressional changes of Cx isoforms in the caput epididymis of adult rat exposed to estradiol benzoate (EB) or flutamide (Flu) at the early postnatal age. A single subcutaneous administration of EB at a low-dose [0.015 g /kg body weight (BW)] or a high-dose (1.5 g/kg BW) or Flu at a low-dose (500 g/kg BW) or a high-dose (5 mg/kg BW) was performed to an animal at 1 week of age. Quantitative real-time PCR analysis was employed to determine expressional changes of Cx isoforms. The transcript levels of Cxs30.3 and 37 were decreased by a low-dose EB treatment, while decreases of Cxs31, 31.1, 32, 40, and 45 transcript levels were observed with a low-dose EB treatment. The treatment of a high-dose EB resulted in expressional reduction of Cxs30.3, 31, 31.1, 37, 40, 43, and 45. The Flu treatment at a low dose caused increases of Cxs26, 37, and 40 transcript levels but decreases of Cxs31.1, 43, and 45 transcript levels. Increases of Cxs30.3, 31, 37, and 40 mRNA amounts were induced by a high-dose Flu treatment. However, exposure to a high-dose Flu produced expressional decreases of Cxs31.1, 32, and 43 in the adult caput epididymis. These observations suggest that exposure to EB or Flu at the neonatal period could lead to aberrant expression of Cx isoforms in the adult caput epididymis.

Gonadotropins are heterodimers consisting an alpha chain (Cgα) and a beta chain. Interestingly, presence of complicated LH-β transcripts in rat testis was accidently found; testicular LH-β transcripts were confined in seminiferous tubules to spermatids, and the translated products were localized in the elongated spermatids. We hypothesized that mouse testis has potential to produce the tissue specific LH-β with similar structure to the rat testicular forms. To verify our hypothesis, we examined the adult mouse (ICR) testis using RT-PCR and immunohistochemistry. The PCR revealed the presence of the identical products in the reactions for three LH subunit types. The expected product sizes for mouse Cgα and LH-β known as pituitary type were 224 bp and 503 bp, respectively. The testicular type LH-β products were produced by a primer set based on the rat sequences, with unexpected size of 800 bp. Sequencing revealed that the proximal and distal parts (2-82 and 661- 773 bp, respectively) were homologous to rat testicular LH-β cDNA, and middle part (83-660 bp) was a unique mouse-specific region. Both Cgα and LH-β positive signals were in the round and elongated spermatids and mature sperms, and the LH-β signals were more intense. In conclusion, our study demonstrated that the presence and localization of the LH subunits in mouse testis. Further studies will be needed to understand the precise structure and function of mouse testicular LH.

While adipose-derived stem cell-conditioned medium (ADSC-CM) has been demonstrated to promote skin wound healing, the mechanism regulating this effect remains unelucidated. In this study, we aimed to investigate the role of Ell3 in the wound healing activity of ADSC-CM. In vitro analysis revealed that Ell3 suppression in ADSCs impairs the promotive activity of ADSC-CM on the proliferation and migration of mouse embryonic fibroblasts (MEF) and normal human dermal fibroblasts (NHDF). Consistently, the expression of MMP family genes, which regulate cell proliferation and migration, was significantly suppressed in MEF and NHDF treated with siEll3-transfected ADSC-CM. Proinflammatory cytokines, such as interleukin-1 and interleukin-6, were highly expressed in MEF treated with siEll3-transfected ADSC-CM. The wound healing activity of siEll3-transfected ADSC-CM was significantly lower than that of the control in vivo. Our results suggest that Ell3 may contribute to the inhibition of inflammatory response during skin wound healing.

With the development of the third-generation gene scissors, CRISPR-Cas9, concerns are being raised about ethical and social repercussions of the new gene-editing technology. In this situation, this article explores the legislation and interpretation of the positive laws in South Korea. The BioAct does not specify and regulate 'gene editing' itself. However, assuming that genetic editing is used in the process of research and treatment, we can look to the specific details of the regulations for research on humans as well as gene therapy research in order to see how genetic editing is regulated under the Bio- Act. BioAct differentiates the regulation between (born) humans and embryos etc. and the regulation differ entirely in the manner and scope. Moreover, due to the fact that gene therapy products are regarded as drugs, they fall under different regulations. The Korean Pharmacopoeia Act put stringent sanctions on clinical trials for gene therapy products and the official Notification "Approval and Examination Regulations for Biological Products, etc." by Food and Drug Safety Administration may be applied to gene editing for gene therapy purposes.
