최근 정원에 대한 관심이 증가함에 따라 자생 식물을 활용 하기 위한 연구가 진행되고 있다. 참식나무(Neolitsea sericea (Blume) Koidz).는 관상용뿐만 아니라 목재로도 이용할 수 있 다. 따라서 이 연구는 참식나무를 정원 식물 소재로 활용하기 위한 대량 증식 기술 개발을 위하여 종자의 발아 특성과 저장 특성에 대해 알아보고자 진행하였다. 2022년 11월 2일 제주 지역에서 채집한 종자를 실험에 사용하였다. 종자의 내·외부 형태 관찰을 한 결과, 종자가 탈리되는 시점에 배가 완전히 발 달한 형태를 가지고 있기 때문에 형태적휴면(morphological dormancy, MD)이 없음을 확인할 수 있었다. 수분흡수 실험 을 통하여 참식나무 종자는 72시간 이후 40% 정도의 수분흡수 율을 보여 내과피의 불투성에 의한 물리적휴면(PY, physical dormancy)이 없는 것으로 판단하였다. 채종 후 곧바로 무가 온 온실에 파종하면 이듬해 5월에 약 60%가 발아하였다. 저 온습윤처리 0, 2, 4, 8, 12주 후 25/15°C에 배양한 결과 최 종 발아율은 각각 0%, 0%, 21.1%, 32.6%, 34.7%였다. GA3 를 0, 10, 100, 1000mg·L-1 농도로 처리한 후 25/15°C에서 배양한 결과, 최종 발아율은 각각 0, 14.5, 22.7, 42.2%였다. 따라서 본 실험의 결과 8주 이상의 저온 처리 또는 GA3 처리 를 통하여 어느 정도 휴면을 타파시킬 수 있었다. 이러한 결과 를 통하여 종자는 intermediate PD를 가지는 것으로 판단 된다. 일부 발아하지 않은 종자는 deep PD를 가지고 있는 것 으로 판단하였다. 후숙 처리 실험의 결과 4주 만에 수분 함량 이 2% 이하로 감소하였고 종자는 전혀 발아하지 않았다. 따라 서 종자의 저장 특성 중 recalcittrant 특성을 지닌 난저장성 종자로 보인다.
A new Hibiscus syriacus ‘Danna’ was developed at the National Institute of Forest Science in 2013. It was initially derived from an artificial cross in 2004, between H. syriacus ‘Hanŏl’, a seed parent, a Korean local cultivar with vigorous growth and H. syriacus ‘Hanŏl-tanshim’, as a pollen parent, with long bloom period. ‘Danna’ was preliminarily selected as ‘KFRI-HH03’ for the aesthetic and stable quality of its unique color of its flower and rapid growth in elongation in 2008. Trial evaluation was conducted for 6 years (2008 - 2013), meanwhile, all investigated traits were retained in rooted cuttings originated from it. ‘Danna’ had a bright pink flower with very small red eye zone. Its flower diameter is 111.3 mm; length of red eye spot on petals is 13.8 mm. Leaves were 71.3 mm long and 43.1 mm wide, and petiole length was 14.2 mm. Flowering period of the cultivar investigated in 2013 was 81 days. It was registered as a new cultivar at the Korea Seed & Variety Service in 2015 (Grant Number 5756). Its tree habit shows vigorous and fast growth and is upright, so it can be used as an ornamental tree or a street tree.
A full genomic DNA microarray technique was employed to investigate the effects of Dongchunghacho on aortal and hepatic gene expression in apolipoprotein E knockout mice fed a high-fat/high-cholesterol diet. Male 8- week - old ApoE-/- mice were randomly divided into two groups, control(high cholesterol group; HC) and supplementation of Dongchunghacho (SD). All of the mice were fed a high-fet/high cholesterol diet with or without Dongchunghacho supplemented by 1% for 6 weeks. At first, lipid profile of the Dongchunghacho was measured by biochemical analysis. No differences were observed in serum triglyceride and total cholesterol levels between the two groups. Antigenotoxic effect of the Dongchunghacho was measured by the single cell gel electrophoresis assay (Comet assay) and quantified as % fluorescence in tail. Dongchunghacho supplementation decreased significantly leukocytic DNA damage and also there was a tendency of reduction in hepatic DNA damage in Dongchunghacho group compared with the control group. In up regulated genes in liver and aorta of the mice, genes with 0 to 2- fold difference in expression level between the two group (HD and SD) was very much more in liver than in aorta, on the contrary, those with 2-fold to 16-flod difference increased greatly rather in aorta than in liver. Also, almost the same results were observed in down regulated genes in liver and aorta between the two groups. These results suggested that supplementation of Dongchunghacho might be helpful in preventing leukocytic DNA damage induced by high fat diet, and has a more crucial roles in aortal gene expression.
Somatic cell nuclear transfer in cattle has limited efficiency in terms of production of live offspring due to high incidence of fetal failure after embryo transfer to recipients. Such low efficiency of cloning could possibly arise from abnormal and poorly developed placenta. In the present study the placental proteome in late pregnancy established from in vitro fertilization (IVF) and nuclear transfer (NT) was analysed. Proteome alternation was tested using two-dimensional polyacrylamide gel electrophoresis (2-DE) and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI- TOF). Comparing placenta from NT embryos to those from IVF counterparts, significant changes in expression level were found in 18 proteins. Of these proteins 12 were not expressed in NT placenta but expressed in IVF counterpart, whereas the expression of the other 6 proteins was limited only in NT placenta. Among these proteins, cytokeratin 8 and vimentin are considered to be involved in regulation of post-implantation development. In particular, cytokeratin 8 and vimentin may be used as makers for placental development during pregnancy because their expression levels changed considerably in NT placental tissue compared with its IVF counterpart. Data from 2-DE suggest that protein expression was disorientated in late pregnancy from NT, but this distortion was eliminated with progression of pregnancy. These findings demonstrate abnormal placental development during late pregnancy from NT and suggest that alterations of specific placental protein expression may be involved in abnormal function of placenta.
Background : This study was carried out to distribute superior cultivars to farmers by analyzing growth characteristics of ginseng and effectively set the target for breeding by investigating annual agronomic characteristics of ginseng. Methods and Results : As a plant materials, Chunpoong (CP), Yunpoong (YP), Gopoong(GP), Gumpoong (GU) and Sunun (SU) were transplanted in Eumseong (ES), Geumsan (GS) and Yeoncheon (YC) on March 2013. Cultivation and management were conducted on the basis of the standard guidelines. According to stem and petiole by region, YC represent the largest stem diameter, leaf length and petiole length of cultivars, followed by those in ES and those in GS. Leaf growth of cultivars was overall the best in YC. Although CP had long and narrow leaves in all regions, YP and SU had short and narrow leaves. Root weight of all cultivars was the best in YC, but there was no significant differences by them. Growth of CP, YP and GP was generally good in ES. Growth of cultivars was relatively even in GS, but it was poor compared to cultivars which were cultivated in ES and YC. Conclusion : There were considerable differences in root growth of cultivars. However, this results suggest that CP of 4-year-old is produced stably in some regions because it had a small coefficient of variation.
Background : Recently, there is a urgent demand for development of new varieties with enhanced resistance to various biotic/abiotic stresses. In order to develop ginseng varieties with such traits, systematic breeding program and comprehensive field studies are prerequisite. Methods and Results : ‘Cheonmyeong' seeds were collected in 2008 from the farmer field of Buyeo. Physiological investigation and propagation were conducted from 2009 to 2011. It was given the name 'Eumseong No. 8 through the observed yield trial from 2012 to 2013 and local adaptability was carried out from 2014 to 2015. All phenotypes including agronomic characteristics, seed yield, and physiological response to biotic/abiotic stresses were investigated according to the ginseng GAP and UPOV guidelines. Yellow-red leaf and pink berry at maturing stage were observed. The time of emergence, flowering and berry maturity of the ‘Cheonmyeong’ were faster than those of ‘Chunpoong’. Stem length of ‘Cheonmyeong’ was shorter than that of ‘Chunpoong’, whereas stem diameter was thicker than that of ‘Chunpoong’. Main root length was shorter but main root diameter is thicker than that of ‘Chunpoong’. Number of seeds of ‘Cheonmyeong’ was fewer than that of ‘Chunpoong’ but 1,000-seeds weight and stratification rate were higher than those of ‘Chunpoong’. The yield performance of this variety was 661 kg/10 a in local adaptability test for two years, which is 22% higher than that of ‘Chunpoong’. ‘Cheonmyeong’ showed strong resistance to phytophthora blight, mulberry mealybug and nematode and moderate resistance to alternaria blight. ‘Cheonmyeong’ did almost not occur yellow spot of aerial part and rusty skin of root, show moderate resistance at high temperatures. Conclusion : Our study demonstrated that ‘Cheonmyeong’ is an ideal variety with heavier root weight and enhanced stress resistance and contribute will enhance biotic/abiotic stress resistance and increase the farmers' income.
Lonicera caerulea var. edulis is a rare species found in some alpine region of Korea. Genetic variation in L. caerulea var. edulis has been investigated by examining 161 individuals from six natural populations: Mt. Seorak 1, Mt. Seorak 2, Mt. Jeombong, Mt. Bangtae, Mt. Gyebang, Mt. Halla. The mean genetic diversity for all the six populations was 0.25 (S.I.). The highest genetic diversity was found in Mt. Seorak (S.I.=0.3158) and the lowest was in Mt. Gyebang (S.I.=0.1047). Comparatively low level of genetic diversity was observed (Ae=1.25, P= 64.6%, S.I.=0.25), which is a typical pattern for rare tree species. AMOVA showed exceptionally large proportion of genetic variations both for among populations (34.69%) and within populations (65.31%). Excluding Mt. Gyebang, the genetic variation among and within population was 18.71% and 81.29% respectively. The UPGMA dendrogram based on genetic distance is not suitable for geographic relationship. Genetic distance of Mt. Gyebang was most distant from the other populations. Excluding Mt. Gyebang, the genetic identities among the five populations were 0.95 to 0.97, which is very high similarity level of genetic identity. This low level of genetic variations and the lack of site in nature indicates that L. caerulea var. edulis demanded a serious conservation.
