The effect of heat treatment and vacuum conditions on the textural properties and electrochemical performance of commercially available activated carbons (ACs) was investigated. The AC after post-heat treatment was characterized by nitrogen adsorption–desorption, X-ray diffraction, Raman spectroscopy, X-ray photoelectron spectroscopy, and Fourier-transform infrared spectroscopy measurements. The ACs treated under vacuum conditions exhibit a higher specific surface area and micropore surface area than those treated under nitrogen atmospheric pressure without significantly affecting the graphite structure of the AC. Under 800 °C temperature and vacuum conditions (AC-V800), the AC with the highest Brunauer– Emmett–Teller surface area of 1951.9 m2 g−1 (16.4% improvement relative to that of the original AC (1677.2 m2 g−1)) was obtained. This is attributed to the removal of oxygen-containing functional groups and volatile matters in the carbon by thermal treatment under vacuum conditions. Consequently, the electric double-layer capacitor using ACs treated under vacuum conditions (1 kPa) at 800 °C (AC-V800) shows considerably improved electrochemical performance in terms of higher specific capacitance and better cycling stability at a high working voltage (3.1 V), compared to the nitrogen-treated and commercial ACs.
South Korea has over 0.38 million of managed honey bee (Apis cerana) colonies before 2009 years ago, which produce the highest quantity of honey in the Korea; however, almost colony (90%) were collapsed by Korean Sacbrood Virus (KSBV) in South Korea. Korean Sacbrood Virus (KSBV) is the pathogen of A. cerana Sacbrood disease, which poses a serious threat to honeybee A. cerana, and tends to cause bee colony and even the whole apiary collapse. Colony collapse of A. cerana was first reported on the Pyeong-Chang of the South Korea in 2009. Several scientists and governments has been tried research for cure the sacbrood disease in A. cerana colony by medicines and management techniques. Unfortunately, The sacbrood disease dosen’t improve. So, we were developed a better breed of A. cerana for resistance of sacbrood virus by selection and then artificial insemination. A. cerana breeding technique was first successful applied with A. cerana in Korean. Queens was grafted from sacbrood resistance line and then it was growing in sacbrood disease colony that was survived 100%. Altogether selected 18 queens were artificially inseminated and 2,000 drones of A. cerana in Korea was used to evaluate amount of semen collection. We are select two scabrood resistance A. cerana line (R and H). R line be used for rearing the Queen. Drone was reared in H line colony. The RH hybrid were not infected sacbrood virus even spread sacbrood virus (2×106). RH colonies have very excellent hygienic behavior, brood, and sacbrood disease resistance activity.
PURPOSES : Thermal cracking (also called low-temperature cracking) is a serious stress for asphalt pavement, especially in eastern South Korea, the northern USA, and Canada. Thermal cracking occurs when the level of thermal stress exceeds the corresponding level of low temperature strength of the given asphalt materials. Therefore, computation of thermal stress is a key factor for understanding, quantifying, and evaluating the level of low-temperature cracking resistance of asphalt pavement. In this paper, two different approaches for computing thermal stress on asphalt binder were introduced: Hopkins and Hamming’s algorithm (1967) and the application of a simple power-law function. All the computed results were compared visually; then the findings and recommendations were discussed.
METHODS: Thermal stress of the tested asphalt binder was computed based on the methodology introduced in previous literatures related to viscoelastic theory. To perform the numerical analysis, MATLABTM 2D matrix-correlation and Microsoft Excel visual basic code were developed and used for the function fitting and value-minimization processes, respectively.
RESULTS : Different results from thermal stress were observed with application of different computation approaches. This variation of the data trends could be recognized not only visually but also statistically.
CONCLUSIONS: It can be concluded that these two different computation approaches can successfully provide upper and lower limits (i.e. boundaries) for thermal stress prediction of a given asphalt binder. Based on these findings, more reliable and reasonable thermal stress results could be provided and finally, better pavement performance predictions could also be expected.
We performed a survey for flavivirus infection and distribution of Aedes albopictus that known as Zika and Dengue virus vector using black–light trap and BG-sentinel trap around urban area in Korea. Mosquitoes were collected in 27 cities during March to November (twice a month) year 2016. Total numbers of mosquitoes collected 102,102 including 19 species 8 genera during collecting period. Total 21,467 Ae. albopictus was collected that 20,961(24.3%) by BG-sentinel trap and 506 (3.2%) by Black-light trap in urban area. Trap index(trap/night) of Ae. albopictus was showed highest in Hamyang (TI:992.3) and lowest in Taebaek (TI:0.3) there was only collected by Black-light trap. A total of 894 pools from all collecting Ae. albopictus were performed a Flavivirus detection. Flavivirus was not detected during study period. This study may provide basic information for surveillance of imported diseases (include Zika virus) and vectors in Korea.
경기도 8개 지역에서 2010년부터 2012년 동안 식균성인 노랑무당벌레의 발생기주를 조사한 결과, 흰가루병에 감염된 12종의 식물에서 관찰 이 되었다. 특히 가장 밀도가 높았던 배과원에서 노랑무당벌레는 7월 상순부터 11월 상순까지 발견되었다. 식균성인 노랑무당벌레의 장내에서는 흰가루병 균사나 포자 외에 다른 먹이의 흔적이 발견되지 않았고, 알과 번데기를 제외한 전 발육단계에서 균을 섭식하는 특성을 볼 때 절대적 식균성 곤충으로 생각된다. 25℃에서 오이 흰가루병균을 섭식한 노랑무당벌레의 발육기간은 알, 유충, 번데기, 성충이 각각 3.9, 10.4, 4.1, 37.7일 이었고, 발육단계별 오이 흰가루병 섭식량은 45.6, 144.4, 372.2, 628.1, 473.7 mm 2로 4령, 성충, 3령, 2령, 1령 순으로 많았다. 본 연구를 통해 노랑무당벌 레의 오이 흰가루병에 대한 섭식능력을 바탕으로 향후 유용 토착천적으로써 대량사육기술, 저독성 약제 선발 등 작물 흰가루병 종합방제기술(IPM) 에 대한 연구가 필요하리라 사료된다.
Muscle satellite cell (SC) is responsible for postnatal muscle growth, repair, and regeneration. Satellite cell is an im-portant source of multi-potent stem cell process and differentiation into adipogenic, myogenic, and osteoblastogenic. The objective of this study was to identify alter of transcriptome during differentiation in porcine satellite cell and to elevated transcriptome at different stages of postnatal development to gain insight into the differences in differ-entiated PSC. We used RNA-seq technique to investigate the transcriptomes during differentiation in pig muscle. Sequence reads were obtained from Illumina HiSeq2000. Differentially expressed genes (DEG) were detected by EdgeR. Gene ontology (GO) terms are powerful tool for unification among representation genes or products. In study of GO biological terms, functional annotation clustering involved in cell cycle, apoptosis, extracellular matrix, phosphoryla- tion, proteolysis, and cell signaling in differences stage. Taken together, these results would be contributed to a better understanding of muscle biology and processes underlying differentiation. Our results suggest that the source of DEGs could be better understanding of the mechanism of muscle differentiation and transdifferentiation.
Satellite cells were derived from muscular tissue in postnatal pig. Satellite cell is an important to growth and development in animal tissues or organs. However, the progress underlying induced differentiation is not clear. The aim of this study was to evaluate the morphologic and the transcriptome changes in porcine satellite cell (PSC) treated with insulin, rosiglitazone, or dexamethasone respectively. PSC was obtained from postnatal muscle tissue. In study 1, for study the effect of insulin and FBS on the differentiated satellite cells, cells were cultured at absence or presence of insulin treated with FBS. Total RNA was extracted for determining the expression levels of myo-genic PAX3, PAX7, Myf5, MyoD, and myogenin genes by real-time PCR. Myogenic genes decreased expression levels of mRNA in treated with insulin. In study 2, in order to clarify the relationship between rosiglitazone and lipid in differentiated satellite cells, we further examined the effect of FBS on lipid accumulation in the presence or absence of the rosiglitazone and lipid. Significant differences were observed between rosiglitazone and lipid by FBS. The mRNA of FABP4 and PPARγ increased in rosiglitazone treatment. In study 3, we examined the effect of dexame-thasone on osteogenic differentiation in PSC. The mRNA was increased osteoblasotgenic ALP and ON genes treated with dexamethasone in 2% FBS. Dexamethasone induces osteoblastogenesis in differentiated PSC. Taken together, in differentiated PSCs, FABP4 and PPARγ increased to rosiglitazone. Whereas, no differences to FBS and lipid. These results were not comparable with previous reports. Our results suggest that adipogenic, myogenic, and osteoblasto-genic could be isolated from porcine skeletal muscle, and identify culture conditions which optimize proliferation and differentiation formation of PSC.
Muscular satellite cell (SC), which is stem cell of postnatal pig, is an important for study of differentiation into adipogenesis, myogenesis, and osteoblastogenesis. In this study, we isolated and examined from pig muscle tissue to determine capacity in proliferate, differentiate, and expression of various genes. Porcine satellite cells (PSC) were isolated from semimembranosus (SM) muscles of 90∼100 days old pigs according to standard conditions. The cell proliferation increased in multi-potent cell by Masson’s, oil red O, and Alizarin red staining respectively. We per-formed the expression levels of differentiation related genes using real-time PCR. We found that the differentiation into adipocyte increased expression levels of both fatty acid binding protein 4 (FABP4) and peroxisome proliferator- acti-vated receptor gamma (PPARγ) genes (p<0.01). Myocyte increased the expression levels of the myosin heavy chain (MHC), myogenic factor 5 (Myf5), myogenic regulatory factor (MyoD), and Myogenic factor 4 (myogenin) (p<0.01). Osteo-blast increased the expression levels of alkaline phosphatase (ALP) (p<0.01). Finally, porcine satellite cells were indu-ced to differentiate towards adipogenic, myogenic, and osteoblastogenic lineages. Our results suggest that muscle satellite cell in porcine may influence cell fate. Understanding the progression of PSC may lead to improved strat-egies for augmenting meat quality.
Propylea japonica is small ladybug(approximately 4-5mm) and is met with everywhere in Korea. When there was no aphid, the survival rate of P. japonica was 0% among another instar bugs and 26.7~32.8% among same instar bugs after 48 hours. When there were sufficient aphids, the cannibalism of P. japonica was low, so the survival rate of P. japonica was 78.6~95.8% among another instar bugs and 80.6~100% among same instar bugs after 48 hours. In the comparison of the number of eggs at different oviposition site, P. japonica preferred the corrugated cardboard. In experiment using T-tube, the rate of decoy was 81% at cotton aphid compared with pepper leaf and 62% at cotton aphid compared with artificial diet. In the artificial diet using shrimp and chicken liver, the developmental period from egg to adult was 17days and the survival rate from egg to adult was 66.7% but the vitality was not good. These data suggest that P. japonica can be used effective natural enemy for control of aphids and is needed to research about artificial diet, mass rearing system and control effect in field.
The present study investigated the effects of follicle stimulating hormone (FSH) and human chorionic gonadotrophin (hCG) on the nuclear maturation of canine oocytes. Oocytes were recovered from mongrel female ovaries in various reproductive states; follicular, luteal or anestrous stage. Oocytes were cultured in serum-free tissue culture medium (TCM)-199 supplemented with various concentrations of FSH (Exp. 1: 0, 0.5, 1.0 or 10 IU) or hCG (Exp. 2: 0, 0.5, 1.0 or 10 IU) or both (Exp. 3: 1 IU FSH + 1 IU hCG) for 72 hr to determine the effective concentration of these hormones, and to examine their combined effect. After maturation culture, oocytes were denuded in PBS containing 0.1% (w/v) hyaluronidase by gentle pipetting. The denuded oocytes were stained with 1.9 μM. Hoechst 33342 in glycerol and the nuclear state of oocytes was evaluated under UV light. More (p<0.05) oocytes matured to MII stage when follicular stage oocytes were supplemented with 1 IU FSH (6.2%) compared with the control, 0.1 or 10.0 IU FSH (0 to 1.2%). Significantly higher (p<0.05) maturation rate to MII stage was observed in follicular stage oocytes supplemented with 1.0 IU hCG (7.2%) compared with the control or other hCG supplemented groups (0 to 1.5%). However, the combination of FSH and hCG did not improve the nuclear maturation rate of canine oocyte (2.4 %) compared with FSH (6.2%) and hCG alone (7.2%). In conclusion, FSH or hCG alone significantly increased the maturation of canine oocytes to MII stage.
Steel structures with added viscoelastic dampers are analysed to investigat their behavior under earthquake excitation. The direct integration method, which produces exact solution for the non-proportional or non-classical damping system, is used throughout the analysis. The results from modal strain energy method are also provided for comparison. Then a new analytical a, pp.oach, based on the rigid floor diaphragm assumption and matrix condensation technique, is introduced, and the results are compared with those obtained from direct integration method and modal strain energy method. The well known phenomenon, that the effectiveness of the viscoelastic dampers depends greatly on the location of the dampers, is once again confirmed in the analysis. It is also found that the modal strain energy method generaly underestimates the responses obtained from the direct integration method, especially when the dampers are placed in only a part of the building. The proposed method turns out to be very efficient with considerable saving in computation this and reasonably accurate considering the reduced degrees of freedom.
Background : This study was development of moisture tolerance and high-yielding Rehmannia glutinosa cultivar. Methods and Results : Segang is developed by the medicinal crop breeding team of National Institute of Horticulture and Herbal Science(NIHHS), Rural Development Administration(RDA), during the period from 2005 to 2015. The reproduction of Rehmannia glutinosa has been accomplished mainly by vegetative propagation with its seedlings have many variants. The cultivar was selected from seedling of Jihwang 1(check variety). The plant type of Segang is some rising from ground. Regional yield trials conducted at three site from 2014 to 2015. The root yield of Segang was 21.1ton per hectare, which was increased 12% compared with Jihwang 1. Also, Segang has higher catalpol content and dried root ratio compared with Jihwang 1. Conclusion : Segang is a moisture tolerance and high-yielding Rehmannia glutinosa cultivar.
Background : There are about 30 species of Adenophora triphylla which grow wild in Korea. but they are very similar so that it is difficult to distinguish with the naked eye. Therefore, it is separated by conformational differences such as leaf shape and flower size. However, these morphological distinction is not useful because even in the same type there are differences by a growth phase or the environment. It is difficult to identify and classify Adenophora triphylla because the phenotypes between plant individuals are diverse greatly even within the same type. For this reason, we will investigate the flowering characteristics of Adenophora triphylla and this information will be used as the basis for the breeding line. Methods and Results : We used the second year Adenophora triphylla in Herbal Crop Research Division, Department of Herbal Crop Research in Eumseong. We Investigated flowering characteristics from June 2015 to November 2015. The 34 individuals of 134 individuals were died by pests and diseases. Depending on the flowering and fruition characteristics were classified into 5 populations, except for the early and late maturing individuals. Conclusions : The flowering period of Adenophora triphylla is 1 month from mid-July to mid-August, and it takes 15 days from the flowering season to the 70% of flowering. It usually takes about 60 days from flowering to fruition, and the fruition is finished in mid-October.
Development of the central nervous system (CNS) occurs normally in mammalian fetus despite lower temperature in the brain region than in the heart. To investigate the effects of temperature niche on the neural differentiation of stem cells in vitro, P19 embryonic carcinoma (EC) stem cells and N2a neuroblastoma stem cells were induced to undergo neural differentiation by retinoic acid and LiCl, respectively. The cells were analyzed for the expression of neural marker genes during 12 days differentiation. Although there were Map2 and NCAM expressions in both groups, no clear difference was found. Similarly, expression patterns of Tuj1 and NF-M were not different in both groups, showing more intensive staining patterns at day 12 than those at days 4 and 8, respectively. However, more cells expressed GFAP markedly at day 12 in 37℃ group. There was little expression of the above markers in N2a cells during differentiation except for Ngn2 and Tuj1. It was found that Ngn2 was expressed more intensely at days 6 and 9 in 33℃ group. Tuj1 expression showed a similar pattern to those of P19 EC cells. RT-PCR analysis also showed that the expressed transcripts did not quite different in both groups, although they were different among the days of differentiation. Thus, it appears that neural differentiation occurs normally with a slight delay and probably less cell death in the cells at 33℃ than that at 37℃.
Chemoresistance is one of the main problems to treat different kinds of cancers or cancer cells. Therefore, it is necessary to find out the strategies to make the cancer cells sensitive to chemotherapy along with optimal dosage of drugs. We examined sensitivity of MCF7 cells through pretreating with an epigenetic modulator, azacytidine (AzaC) to doxorubicin (Dox). The cells were treated with 5 and 10 mM of AzaC for a week, subsequently with 50, 100 and 500 nM of doxorubicin for 24 and 48h. It was found that pretreatment of AzaC significantly enhance the sensitivity of MCF7 cells to Dox, inducing cell death. After 24h 15% cells underwent apoptosis in 500 nM dox treatment group while 23.4% cells death occurred in AzaC pre treatment group. After 48h MCF7 cells treated with Dox showed 19.0% cell death while AzaC sensitized cells showed 50.0% cells death when exposed to 500 nM of Dox for 48h. Western blot analysis showed the upregulations in the expression of bax, caspase-3, caspase-9 and p53 in AzaC-sensitized MCF7 cells treated with Dox as compared to those treated with only Dox. There was no clear indication for pro-apoptosis genes in the cells treated with individual drugs. These results showed that pretreatment with the epigenetic modulator significantly increased the sensitivity of MCF7 cells to Dox. Therefore it is concluded that demethylation event might enhance the activity of DNA intercalating agents to induce DNA damage in breast cancer cells.
Neutropenic enterocolitis (NE), the most serious gastrointestinal complication, has been reported as a clinical syndrome that occurs in the setting of disease- or chemotherapy-induced neutropenia. Complications of NE include bowel necrosis with perforation, fistula, stenosis, massive bleeding, abscess formation, and pneumatosis intestinalis (PI). Most physicians recommend initial conservative management with bowel rest, intravenous fluids, total parenteral nutrition, broad-spectrum antibiotics, and normalization of neutrophil counts. Surgical intervention is recommended in the event of obstruction, perforation, persistent gastrointestinal bleeding despite correction of thrombocytopenia and coagulopathy, or clinical deterioration. We experienced a patient whose abdominal computed tomography scan showed pneumoretroperitoneum, intramural gas in the colon, and inferior vena caval gas. Her condition improved after treatment with granulocyte colony-stimulating factor and broad-spectrum antibiotics. We report on this case along with a review of the literature.
