Oyster mushrooms including of P. ostreatus, P. eryngii, P. pulmonarius and P. cornucopiae are one of the famous mushrooms for foods in Korea. RAPD were carried out using 14 of oligoprimers to analyze the phylogenetic relationship among 57 strains of 32 Pleurotus species. Most of species formed the minimum clade with strains within species and was divided respectively species. Therefore clade was separated well in accordance species. Pleurotus species formed again clade to be added in close related to other species, and were discriminated by sixteen clades with each representative species including high similarity groups. Sixteen clades were composed representative species according to each clade. There were clade I of P. pulmonarius(P. sajor-caju, P. opuntiae, P. sapidus), clade II of P. eryngii(P. fuscus var. ferulae, P. fossulatus), clade III of P. ostreatus(P. ostreatus var. columbinus, P. spodoleucus, P. floridanus), clade IV of P. florida, clade V of P. djamor(P. flabellatus, P. incarnates, P. salmoneo-stramineus), clade VII of P. populinus(P. subareolatus), clade VIII of P. cystidiosus(P. cystidiosus var. formosensis), clade X of P. dryinus(P. dryinus var. pometi), clade XIV of P. cornucopiae(P. citrinopilieatus, P. euosmus), and clade XV of P. australis. These species were representative species each clades. Five species, P. ulmarius(clade VI), P. griseus(clade IX), P. calyptratus(clade XI), P. lampas(clade XII), P. smithii(clade XIII)and P. serotinus (clade XVI) were used each one strain in analysis, so they were clustered other groups.
Pleurotus has increased rapidly production and consumption because of highly nutritional value, natural healthy food and so on. The basic studies for Pleurotus need for development of mushroom industry. This study was to investigate the cultural characteristics among 15 strains of 6 species and to analyze their phylogenetic relationships. The cultural characteristics were investigated by mycelial growth activity at different media, temperature and pH. The optimum media for mycelial growth were YM and MCM in most species. The optimum temperature for mycelial growth were 25˚C and 30˚C. The optimum pH for mycelial growth were widly range from pH 5.1 to 7.4. Through the RFLP (restriction fragment length polymorphism) of IGS (intergenic spacer) I region in ribosomal DNA, it was analyzed phylogeny of interspecies and intraspecies. Each species was discriminated well as isolates within each species formed clade to be distinguished other species. P. florida was highly similar to P. floridanus, and P. flabellatus was P. cornucopiae. P. fuscus var. ferulae was highly similar to P. eryngii but discriminated different species in analysis of RFLP of IGS I region and showed different characteristics in mycelial culture. RFLP of IGS I region was useful of studying phylogenetic relationships of species and population.
김치를 10℃에서 8일간 숙성시켜 산도 0.6~0.7%로 숙성된 김치를 -70℃와 -20℃로 냉동하여 -20℃에서 저장하면서 배추 조직의 elasticity, hardness, 세포 조직의 변화, 드립양을 실험한 결과는 다음과 같다. -70℃에서 급속 동결한 것과 -20℃에서 완만동결한 냉동 김치중 배추조직의 elasticity는 냉동저장 15일까지 감소하다 일정하게 유지되었고 hardness는 거의 변화가 없었으며 냉동 방법에 따른 변화도 거의 나타나지 않았다. 투과 전자현미경으로 관찰한 결과 control의 경우 세포벽이 매우 두꺼우며 세포의 모양들이 잘 보존되어 있는 것을 볼 수 있었고 -20℃로 냉동 처리하여 해동시킨 세포벽들은 많이 손상되어 있음을 볼 수 있었으며 -70℃로 급속 냉동 시료의 경우 세포벽의 손상 정도가 -20℃로 냉동 처리한 시료보다 덜 파괴되어 있음을 볼 수 있었다. 한편 냉동 저장기간 동안 드립의 손실량의 변화는 -70℃로 냉동 처리한 시료의 드립양은 3~4%정도로 -20℃로 냉동 처리한 시료의 5~6%에 비해 적은 것을 알 수 있었다.
양념에 Leu. mesenteroides, Lac. plantarum의 젖산균과 멸치액젓 첨가가 김치 양념의 대장균군 제어, 숙성촉진여부 및 관능적 특성에 미치는 영향을 조사하였다. Leu. mesenteroides과 젓갈을 첨가한 경우 초기 숙성이 촉진되나 후기 산패는 완만하였으며 Lac. plantarum의 첨가시에는 초기 숙성에는 큰 차이가 없고 후기 산패가 현저하게 진행되었다. 대장균군은 산도 0.80% 이상에서 사별되었는데 Leu. mesenteroides의 경우가 숙성이 촉진되어 보다 신속히 사멸시켰다. 총균수와 젖산균수는 초기에 starter 첨가구는 106 CFU/mL 정도인데 반해서 starter를 첨가하지 않은 처리구에서 총균수는 106 CFU/mL, 젖산균수가 104 CFU/mL 이었고 숙성 후에는 첨가군이 높은 수준을 유지하였다. 하지만 대체적으로 스타터의 첨가가 숙성 및 대장균에 크게 영향을 주지 못하는 경향으로 동일하게 나왔다. 관능적 특성에서는 Leu. mesenteroides 첨가구가 Lac. plantarum의 경우보다 우수하였다. 한편, 시판 겉절이 김치는 산도가 0.09~0.18%, pH 5.46~5.89, 총균수 2.2×106~3.1×107 CFU/ml, 젖산균수 2.1×104~6.6×105 CFU/ml, 대장균군수 1.8×104~7.1×105 CFU/ml, 염분 1.89%~2.51%으로 스타터 첨가 겉절이에 비해 pH와 염도를 제외한 품질 특성에서 전반적으로 낮은 값을 나타냈다.
This study was conducted to investigate the difference of general feature and ginsenoside content of 6 years old ginseng root among different grade of roots. Total weight of a 1st grade-6 years old ginseng root was 115.1g and weight, length, diameter and specific gravity of main root were 64.68g, 8.39cm, 3.31cm and 0.96, respectively. Main root of 1st grade ginseng root was larger in size and specific gravity and more heavy than that of End or 3rd grade of the roots. Though crude saponin contents were not so different among the different grade of roots, but ginsenoside Rb1, Rg1 and Re content were higher in 1st grade of root than that of 2nd or 3rd grade of root. Those ginsenosides were located mainly in periderm and cortex.
마늘이 김치의 숙성과 품질에 미치는 영향을 조사하고 아울러 cyclodextrin과 sorbitol의 마늘냄새 masking 효과를 실험하였다. 김치 숙성 초기의 산도는 마늘 첨가량이 많을수록 대조구 비해서 높았고 환원당 함량은 마늘 첨가량이 많을수록 높았으나 숙성말기에는 거의 비슷한 수준을 유지하였다. 미생물수의 경우 숙성초기에는 마늘 첨가에 의해 성장이 촉진되었으나 이후에 그 수준을 유지한 후에는 대조구에 비해서 오히려 억제되었다. 관능검사 결과는 온도에 관계없이 마늘을 첨가하지 않은 김치가 모든 관능적인 면에서 유의적으로 나쁘게 평가되었으며 마늘을 2~3% 첨가한 김치가 유의적으로 가장 좋게 평가되었다. Cyclodextrin 또는 sorbitol을 1%씩 첨가한 결과 마늘맛을 masking하는데는 효과가 없었고 관능적으로도 나쁘게 평가되었다.
This study was conducted to investigate the changes in lipid components of red pepper under various processing conditions such as drying, exposing to lights, extraction and storage conditions. The material used in this study were Chungyang cultivars which contained the largest amount of capsaicinoids and Dabok cultivars which is most popular among the various cultivars produced in Korea. Total lipid content of Chungyang was 15.7% and those of Dabok, Udeungsang and Hongilpum were about 17%. The neutral lipid content in free lipid of red pepper was 78.5~80.3% Phospholipid and glycolipid content in bound lipid were 53.8~56.9% and 35.3~38.7% respectively. The major fatty acids of lipid were linoleic, palmitic and oleic acid. Linoleic acid was presented mainly in pericarp, seed and placenta, whereas the most of palmitic acid was presented in stem. Lipid content of cut and whole red pepper were decreased 24.7~28% and 18.1~21.5% by drying for 48hrs at 65℃ and 95℃. And lipid content was also decreased 3.5~3.6% when the red pepper powder was exposed to 15,000 lux of incandescent light for 30 days at 40℃ and 4~4.9% to fluorescent light. The lipid was extracted the highest content of 50.5~51.7% by acid solution(pH2) and the lowest content by neutral solution, and the higher the salt concentration, the greater the lipid was extracted. During storage at 4℃, 25℃, 40℃, for 30 days the lipid content was not much changed but linoleic acid content was decreased during storage at the same conditions.
This study was conducted to investigate the standardization of kimchi and related products. There are about 339 kinds of kimchi and related products in Korea. Half of those various kimchi was 50% of the total products, and the other groups are in the order of Jangachi (high salted vegetable pickles, Saengchae (a salad type kimchi), Kakduki(reddish kimchi), Keotjulyi(nonfemented kimchi), Pickles, Seobakji (kimchi based on raddish, fish and other ingredients) and Shickgae (lactic fermented fish products). About 200 ingredients (chinese cabbage, raddish, cucumber, other vegetables, fruits, seaweeds, fish, meat etc.) were used for those products. Tongbaechu kimchi (whole headded chinese cabbage kimchi) and Possam kimchi (wraped kimchi with chinese cabbage leaves) were made by about 30 ingredients, respectively. Kakduki, Oyster-kakduki, Chonkak kimchi (kimchi based on small raddish with leaves) and Tongchimi (raddish kimchi with brine soup) were made by raddish and more than 10 ingredients, but the subsidiary ingerdients were less than that of chinese cabbage kimchi. Other products such as Yulmu kimchi and Oisobaki (a cucmber kimchi) were also dicussed.
About 760 papers and patents of the traditional fermented food in Korea were collected which were published during 1917-1988, and then the papers were classified, outlined and reviewed. Study of traditional fermented food was about 10% of total study on Korean Foods, and that of alcoholic beverage was 50%, soy sauce and related product was 30% and kimchi was less than 20% of total study of traditional fermented foods. Though considerable amount of studies were accomplished before the World War II, but a few study was accomplished during the Korean war. The research activity is, hitherto, increasing gradually, but the systematic studies were rarely done. The studies to be investigated were processing quality of raw materials, classification, standardization, sensory quality of the product and so on.
김치의 표준화를 위한 기초자료로서 1976년부터 1987년까지 간행된 김치관계자료 896건을 수집하여 종류와 사용하는 재료들을 검토한 결과 김치류 및 절임류의 명칭은 총 186종에 달하고 그중 김치류 106종, 깍두기류 16종, 동치미류 10종, 소박이 11종, 겉절이 및 생채류 18종, 장아찌 23종, 기타 식해류 2종이었다. 사용하는 재료는 총 118종에 달하고, 주재료별로는 무우가 34%, 배추가 14%, 기타 채소가 30%를 차지하고 동물성 재료와 해조류가 15%를 차지하고 있다. 통배추김치, 보쌈김치, 백김치 등 배추를 주원료로 하는 김치의 재료는 배추, 소금, 마늘, 고추, 생강, 파, 미나리 등을 사용하고 그밖에 젓갈류, 갓, 깨, 굴 등 30종 이상의 재료를 사용하는데 비하여 무우를 주재료로 하는 깍두기, 동치미 등은 무우, 소금, 고추, 마늘, 파, 생강 등을 주재료로 하는 것은 배추김치류와 같지만 부재료의 종류가 20종으로 훨씬 단순하였다. 그밖에 열무김치, 오이소박이 등에 사용되는 재료도 검토하였다.
