        식품에서 발견되는 곤충이물은 제조단계와 유통, 소비단계 유입으로 대별된다. 식약처 발표에 따르면 2016년 5332건 이물신고 건수의 34.3% 인 1830건이 곤충이물이며 이중 조사, 판정 불가 56%, 제조단계 6.5%를 제외한 49.4%가 소비, 유통 단계에서 유입된 것으로 조사되었다. 유통, 소비단계에서의 곤충 이물 유입은 전 지구적인 기후변화와 소비자 트랜드 변화, 유통, 물류 시스템 변화 등의 요인과 결합되어 지속적으로 증가되고 있으나 제품 생산 후 제품이 물류센터, 특약점, 도, 소매점과 같은 다양한 환경에 노출되고 소비자 사용 시 다양한 원인으로 인해 발생되기 때문에 근본적인 원인 제거는 불가능 하다. 따라서 현재로써는 방충포장을 통해 곤충오염 리스크를 줄이는 방안이 가장 효과적일 것으로 판단된다. 본 연구에서는 유통 중 제품으로 유입되는 곤충을 제어하기 위해 생산비 증가와 식품 안전에 미치는 영향을 고려하여 천연식물 추출물을 활용한 방충박스포장소재를 개발하고 이를 약 770만 제품박스에 적용한 결과 소비자 해충클레임이 65%정도 감소하는 것으로 조사되었다.

        The aim of this study was to investigate what protein(s) of porcine epididymal fluid (pEF) are able to enhance the nuclear maturation of porcine germinal vesicle (GV) oocytes in vitro. Proteins of pEF were fractionated by affinity, ion exchange, and gel filtration chromatography. Porcine cumulus-oocytes complexes (COC) from follicles were cultured in tissue culture medium (TCM 199) containing various fractions obtained by chromatography. Porcine COCs were also cultured in TCM 199 containing various meiosis inhibitors and pEF. After 24 or 48 h culture, oocytes were examined for evidence of GV breakdown, metaphase I, anaphase-telophase I, and metaphase II. When porcine COCs were cultured in the medium with meiosis inhibitor such as, dibutyryl cAMP (dbcAMP) and forskolin (Fo), more than 80% of oocytes were unable to resume meiosis. However, porcine COCs supplemented with pEF were able to overcome the inhibitory effect of dbcAMP and Fo. Maturation rate of oocytes was significantly (p<0.05) increased in the media supplemented with cationic protein(s) during in vitro maturation than in those with anionic protein(s) (44.1% vs 20.0%). When oocytes were cultured in the TCM 199 with fractions obtained by gel filtration, the maturation rate of oocytes was significantly (p<0.05) higher in fraction 11 containing 18 kDa than other fractions. The present study suggests that 1) dbcAMP and Fo prevent the spontaneous maturation of oocyte after isolation from follicles, and that pEF contain a substance(s) that improves meiosis resumption in vitro of porcine COCs, 2) cationic 18 kDa protein(s) are responsible for promotion of MⅡ stage.

        The aim of this study is to investigate the effect of porcine epididymal fluid (pEF) on in vitro-maturation and subsequent fertilization of porcine follicular oocytes. Porcine cumulus-oocytes complexes retrieved from antral follicles were cultured in tissue culture medium (TCM)-199 supplemented with pEF of different concentrations. At 48 h after culture, development of oocytes to germinal vesicle (GV) breakdown, metaphase I, anaphase-telophase I, and metaphase II were examined. Significant (p<0.05) increase in the proportion of oocytes developed to MII stage was observed in oocytes cultured in pEF-containing TCM-199 than in oocytes cultured in pEF-free TCM-199 (46.2% vs 16.7%), which was a dose-dependent manner. Subsequently, the proportion of monospermic fertilization were significantly (p<0.05) increased in oocytes cultured in the TCM supplemented with pEF than those cultured in pEF-free TCM-199 (51.0% vs 24.1%). In the second series of experiment, the percentage of MII oocytes was significantly (p<0.05) increased after exposure of oocytes to pEF during the first 22 h period of culture than after exposure of oocytes to pEF during the next 24 h of culture, while no significant difference in the percentage of monospermy was observed. The results of this study demonstrate that pEF contains at least enhancing component(s) for nuclear maturation.

        We measured cobalt content in sorghum (Sorghum bicolor (L.) Moench.) by atomic absorption spectrophotometry: their absorbance and back ground values. These analyses were with AA-680 of Shimadzu. The mode was BGC; wave length, 240.8 ㎚, 240.7 ㎚, 240.6 ㎚, respectively. The recommended wave length is 240.7 ㎚ for Co analysis. It was done through several ratios among 4 factors; mean of absorbance(AM), standard deviation of absorbance (AS), mean of background(BM), standard deviation of background.(BS). The result was t㏊t 240.6 ㎚ was the favourable. And t㏊t of 240.8 ㎚ was better t㏊n t㏊t of 240.7 ㎚ for the Co analysis.

        In vitro maturation of porcine immature cumulus-enclosed oocytes can be enhanced by co-incubation with spermatozoa even before fertilization. The aim of this study was to determine whether the addition of spermatozoa into the culture medium can stimulate the meiosis resumption of porcine cumulus-enclosed oocytes arrested at germinal vesicle (GV). Cumulus-enclosed oocytes (CEOs) were collected from follicles of 3 to 5mm diameter. Porcine CEOs were cultured in tissue culture medium containing various meiosis inhibitors and spermatozoa. Oocytes were examined for evidence of GV and GV breakdown after 24 h culture. After 24 h culture 43.8% of oocytes cultured in only TCM 199 remained at GV stage whereas 56.2% of oocytes were able to resume meiosis. When porcine CEOs were cultured in the medium with meiosis inhibitor such as, dibutyryl cAMP (dbcAMP) and forskolin (Fo), more than 90% of oocytes were not able to resume meiosis. However, co-culture of porcine CEOs with spermatozoa was able to overcome the inhibitory effect of dbcAMP and Fo. Irrespective of the presence of 3-isobutyl-1-methylxanthine (IBMX), no difference was observed in the proportion of oocyte reached germinal vesicle breakdown (GVBD). The present study suggests that dbcAMP and Fo prevent the spontaneous maturation of competent oocyte in culture after isolation from follicles and that mammalian spermatozoa contain a substance(s) that improves meiosis resumption in vitro of porcine cumulus-enclosed oocytes.

Cumulus-oocyte complexes (COCs) were collected from follicles of 3- to 5-mm diameter. Porcine COCs were cultured in tissue culture medium containing spermatozoa. After 48 h culture, oocytes were examined for evidence of GV breakdown, metaphase I, anaphase-telophase I, and metaphase II. The proportion of oocytes reaching at metaphase II was significantly (P < 0.05) increased in the oocytes cultured in media containing spermatozoa compared to those in media without spermatozoa (52.3% vs 12.5%). No difference in the percentage of metaphase II was observed among the different periods of spermatozoa exposure and among the spermatozoa from different species. The proportion of oocytes reaching metaphase II was significantly different between high and low concentrations of spermatozoa. The present study suggests that manunalian spermatozoa contain a substance(s) that improves nuclear in vitro maturation of porcine cumulus-enclosed GV oocytes. Enhancing effect of spermatozoa for in vitro maturation of oocytes is a highly dose-dependent.

        최근 기후변화와 도시화로 인한 수재해 문제가 증가하고 있으며, 이에 대응방안인 저영향개발(Low-Impact Development, LID) 기법에 관한 연구가 확대되고 있다. LID 기법은 도시 내의 우수유출수를 저감시켜 다양한 수재해 문제를 친환경적으로 제어하고, 도시 개발 이전의 물순환 체계로 회복시키는 기술이다. 하지만 LID 기법에 관한 정량적 데이터가 부족한 실정이다. 따라서 본 연구에서는 저류형 LID 기술인 식생화분(Planter Box)의 Curve Number (CN)값을 산정하여, 물순환(침투, 유출, 월류수) 분석을 실시하였다. Planter Box의 물순환 분석에 관한 강우강도 시나리오(60.4 mm/hr, 83.1 mm/hr, 97.4 mm/hr, 108.2 mm/hr)는 부산시 확률강우강도표(2010)를 이용하여 선정하였다. 실험 결과는 건물화분3 (BPB-3)과 거리화분3(SPB-3)에서 우수저류율이 각각 43.5%∼52.9%, 33.4%∼39.0%로 나타났다. 또한 BPB-3에서 CN값은 평균 83이 산출되었고, Horton 침투능 곡선식 적용에 따른 우수유출효과는 17%∼96%로 나타났다.

        Purple acid phosphatase is important for phosphorus remobilization in plants, but its role in plant adaptation to low phosphorus availability is not known. The cDNA encoding O. sativa purple acid phosphatase (OsPAP1) has 1008 bp with an open reading frame of 335 amino acid residues. The amino acid sequence of OsPAP1 cDNA shows of 50-51% identity with other plant purple acid phosphatases. OsPAP1 was expressed in rice plants and in cell cultures in the absence of phosphate (Pi). The expression was organ-specific with the strongest expression in Pi-deprived roots. Functional expression of the OsPAP1 gene in the transgenic Arabidopsis line was confirmed by northern and western blot analysis. OsPAP1 overexpression lines had higher phosphatase activity than wild-type. Overexpression of OsPAP1 in Arabidopsis plants resulted in increased Pi accumulation under Pi sufficient condition. These results show that the OsPAP1 gene represents more efficient Pi uptake and can be used to develop new transgenic dicotyledonous plants.
