Bovine rotavirus (BRV) is one of the common gastrointestinal diseases that can occur in calves, causing severe diarrhea. This study was conducted to investigate the effects of oral administration of single-domain antibodies, single variable domain of heavy chain of HCAb (VHH), on the prevention and treatment of rotavirus-induced diarrhea in calves. Thirty calves were divided into five experimental groups (negative control, positive control, group A, group B and group C). Except for the negative control group, the experimental groups were inoculated with BRV on the 4th day after birth. The VHH supplementation groups were fed with a substitute milk containing different concentrations of VHH (1%, 2.5% and 7.5%). Changes in body weight, clinical symptoms, serum antibodies, and virus detection in feces were observed for a total of 10 days in each experimental group. The results showed a higher survival rate in the VHH supplementation groups compared to the positive control group. Clinical symptoms caused by rotavirus were alleviated in the VHH supplementation groups. The detection rate of rotavirus in feces also decreased. Gross findings indicated that the severity of disease in the VHH supplementation groups was milder than that in the positive control group. Therefore, VHH could be proposed as a useful alternative for the prevention and treatment of rotavirus-induced diarrhea in calves.
This study evaluated cell viability and cytokine release in immortalized human oral fibroblasts (hTERT-hNOFs) and keratinocytes (IHOK) exposed to a dental-impregnated gingival retraction cord. To prepare the extracts, dental gingival retraction cords impregnated with aluminum chloride hexahydrate were immersed in a cell culture medium for 24 h at 37 °C. hTERT-hNOFs and IHOK were cultured for 24 h. The cell culture medium was removed and extracts of the dental gingival retraction cords were added. After incubation with the extract solution, cell viability was evaluated using an MTT assay. The levels of the cytokines IL-1α and IL-8 were measured in the supernatants of each cell type. The cell viability after exposure to the extract solution for 10 min exceeded 70 % in both cell types. The ET50 values for hTERT-hNOF and IHOK were 35.75 and 28.98 min, respectively. For IHOK, the IL-1α level was (5.35 ± 5.22) pg/mL at 10 min, (3.58 ± 5.38) pg/mL at 20 min, and (2.85 ± 4.28) pg/mL at 60 min of exposure (p > 0.05). The IL-8 level in IHOK was (67.16 ± 18.70) pg/mL at 10 min, (78.36 ± 7.50) pg/mL at 20 min, and (111.9 ± 26.10) pg/mL at 60 min of exposure (p > 0.05). Cytokine release was not observed from hTERThNOFs. Based on these results, cell viability and cytokine release were confirmed in cells exposed to the impregnated gingival retraction cord. In addition, the application of the extracts to hTERT-hNOF and IHOK during the actual contact time and determination of ET50 may be beneficial for evaluating the biocompatibility of dental-impregnated gingival retraction cords.
본 연구는 아세트아미노펜, 비타민 C, 무수 구연산으로 구성된 해열진통 및 염증 완화제, 피버닐산의 사료첨가가 구제역(FMD) 백신의 접종으로 야기 될 수 있는 돼지의 부작용에 대한 감소 효과를 확인하기 위해 실시되었다. 8-9주령의 돼지(LYD) 30마리를 공시, 대조군(n=10), 구제 역 백신 접종군(n=10; T1), 구제역 백신+피버닐 산 투여군 (n=10; 백신 접종 후 5일 간 0.2% 피버닐 산 경구 투여, T2), 3개 그룹으로 배치하여 시험을 진행하였다. 각 시험 구에서 총 14일 동안 체중, 혈액학치, 혈액화학치, FMD 항체 및 스트레스 지표의 변화를 관찰했으며, 총 증체량 은 T1(11.13±1.33kg)에 비해 T2(13.76±1.73kg)에서 유의적 으로 증가하였다(p<0.001). 혈액학치 및 혈액화학치 분석 결과, 백신을 접종한 두 처리군에서 유의한 차이는 없었 다. FMD 항체 역가는 T1(89.1±7.9)에 비해 T2(91.9±5.6)에 서 증가하는 경향을 보였으나 통계적 유의성을 나타나지 않았다. T2 그룹의 코르티솔, TNF-α 및 CRP와 같은 스 트레스, 염증 지표 물질은 구제역 백신 접종 후 3일에서 5 일 사이에 각각 310.31±44.11ng/mL, 59.05±11.88pg/mL, 29.05±2.84ng/mL으로 T1(361.58±58.74ng/mL, 74.47±19.26 pg/mL, 34.21±5.34ng/mL)보다 유의적으로 낮게 측정되 었다. 시험 14일째에 육안 및 조직병리학적 분석 결과, 주 사 부위의 염증 반응은 T2에서 적게 관찰되었으며 염증세 포 침윤 및 섬유조직이 증식된 육아종성 염증 또한 T2에 서 적게 관찰되었다. 본 연구결과로 미루어 보아 피버닐 산의 0.2% 경구 투여는 체중감소, 발열 등의 임상증상을 감소시키는 효과가 있으며, 접종 부위의 염증반응을 억제 하여 구제역 백신 접종의 부작용에 대한 스트레스를 경감 시키는 것으로 판단된다. 따라서 백신 접종 후 피버닐 산 의 0.2% 경구 투여는 돼지에서 구제역 백신 접종과 관련 된 부작용을 줄이는 데 효과적인 것으로 사료된다.
This study evaluated whether fevernill, which is primarily composed of acetaminophen, vitamin C, and anhydrous citric acid, is effective in relieving stress caused by foot-and-mouth disease virus (FMDV) vaccination in calves. Three-month-old calves from a farm in Korea were randomly assigned to one of three groups (n = 10 per group: control [untreated], group A [FMDV vaccination], and group B [FMDV vaccination + addition of fevernill 0.2% to feed for five days after vaccination]). Body weights, antibody formation, clinical symptoms, serum parameters were measured in the 14 days following vaccination. Total weight gain and average daily gain during the experiment period were group A (12.00±1.15 kg, 0.86±0.08 kg/day) compared to group B (13.57±0.98 kg, 0.97±0.07 kg/day) was significantly improved (p<0.05). There was no significant difference in the formation of antibodies against the FMDV vaccine between group A and group B, though the antibody value of group B tended to be higher than that of group A. The pre-inflammatory cytokine TNF-α value was 74.47±19.26 pg/ml in group A and 59.05±11.88 pg/ml in group B on the 5th day of the experiment, which was significantly lower in group B than in group A (p<0.05). Also, cortisol concentrations were significantly lower in group B than in group A (p<0.05). In conclusion, the feeding of fevernil is judged to be helpful in mitigating the side effects caused by FMD vaccination, and thus it is thought to be able to prevent the decrease in productivity caused by vaccination.
본 연구는 한우의 전장유전체 내에 존재하는 단일염기 다형성(SNP)에 대해 DNA chip 분석을 통해 각 개체의 유 전자형을 파악하고, SNP 표지인자간 연관불평형의 크기를 알아보기 위해 실시하였다. 강원도에서 사육되고 있던 한 우 거세우 139두의 조직을 채취하여 DNA를 추출하였으 며, Bovine SNP50K BeadChip 분석을 이용하여 분석에 사 용 가능한 최종 35,769개의 SNP 표지인자를 얻어 연관성 분석을 실시하였다. 분석에 이용된 SNP 표지인자의 총 길 이는 2499.26Mb이었고, 전장 유전체에서 평균 대립유전자 빈도(MAF)는 0.273이었으며, SNP 표지인자간의 평균 거 리는 0.060과 0.082 사이에 분포하였다. 강원지역 내 한우 거세우를 이용하여 확인한 본 실험 결과는 기존의 전국의 한우를 대상으로 한 연구결과와 유사한 패턴을 보였다. 50Kb 이하로 인접한 거리를 갖는 SNP 표지인자들의 연관 불평형 값(r2)이 0.2 초과인 인자가 34%로 나타났다. 또한 SNP 표지인자간 거리가 멀수록 측정값이 떨어지는 지수형 식의 그래프를 보였다. 염색체별로 구분한 40kb 이하로 인 접한 SNP표지인자의 연관불평형 값(r2)은 0.2 이하로 나타 난 것이 총 7개(13, 15, 19, 23, 25, 28, 29번 염색체)였다. 종 합하여 볼 때, 본 연구 결과는 강원한우의 유전적 개량을 이 용한 기초자료로서 활용될 수 있을 것이며, 유전적 개량을 이용한 육종시스템에 도움을 줄 수 있을 것으로 기대된다.
The HNF4α (hepatocyte nuclear factor 4, alpha) is a hepatic transcription factor related to the lipid metabolism and regulation of insulin secretion in humans. The current study about commercial broiler reported that the A543G single nucleotide polymorphism (SNP) within HNF4α gene has an effect on fat deposition and wing yield in the chicken. This study was performed to investigate the association between the SNP within HNF4α gene and growth trait and to verify the applicability as a molecular marker for the improving the performance in Korean native chickens (KNCs). A total of 764 KNCs was collected from the livestock farm of Gyeongnam National University of Science and Technology in Korea and genotyped by PCR-RFLP. The body weight measured at birth, 2, 4, 6, 8, 10, 12, 14, 15, 16, 18, 20, 24, 28, 32, 36, and 40 weeks of age for the association analysis between chicken growth and the SNP genotype. Statistical analysis of the SNP with phenotypic traits was performed using SAS program. The KNC strain was classified by the genotypes of the A543G SNP. The frequencies of three genotypes were 0.47 (AA), 0.43 (AG) and 0.10 (GG), respectively. The SNP of HNF4α has highly significant association (p<0.001) with all-round growth in KNCs. These results suggest that the A543G SNP within HNF4α gene could be a genetic marker for the breeding in Korean native chickens.
