This study investigated the culture characteristics of Cryptoporus volvatus, whichis grow naturally in Korea, to determine the suitable environmental conditions for its cultivation. The physiological characteristics of the mycelia were assessed according to the cultivation conditions, to determine the optimal conditions for artificial cultivation. The visual characteristics of the hyphae of Cryptoporus volvatus KACC52303 included an irregular and uneven surface and a fuzzy or cotton-like texture. Under the microscope, its microstructure showed pre-chlamydospore formation, but no clamps were seen. The appropriate culture temperature was found to be a medium/high temperature of approximately 25–30oC, and the optimal pH was found to have a wide range from weakly acidic (pH 4) to neutral (pH 7). In the optimal nutrient source experiment, hyphal growth was shown to be fair in a mixed medium with 2.5% dextrin as the carbon source and 0.1% yeast extract as the organic nitrogen source. Among the various amino acids, organic acids, and inorganic salts tested, the fastest hyphal growth was observed in the presence of leucine, acetic acid or gluconic acid, and KCl or KH2PO4, respectively. The column test showed that the best mycelial growth occurred in a mixed medium of 80% pine sawdust, 10% rice bran, and 10% corncob sawdust.
야생 잎새버섯 추출물을 이용하여 심혈관계 질환과 관련된 기능성 식품 개발을 위한 기초자료를 얻기 위해 준비한 물 추출물과 유기용매 분획물의 혈전용해활성과 트롬빈저해활성, acetylcholinesterase 저해활성, 항산화활성을 확인하였다. 10 mg/mL의 농도에서 물 추출물과 물 분획물은 각각 0.93 plasmin unit와 0.73 plasmin unit의 높은 혈전용해활성을 나타냈으며, 부탄올 분획물은 79.60%의 트롬빈저해활성을 나타냈다. 클로로포름 분획물은 85.88%의 높은 acetylcholinesterase 저해활성을 나타냈고, 물 추출물의 항산화 활성은 39.81%로 비교적 낮게 나타 냈다. 부분 정제된 효소를 포함하고 있는 물 분획물은 섬유소원의 Bβ chain를 모두 분해시켰지만, Aα chain은 느리게 분해시키고 γ chain 과는 반응하지 않았다. 실험 결과로부터 야생 잎새버섯 추출물은 높은 혈전용해활성과, 트롬빈저해활성, acetylcholinesterase 저해활성을 나타내 심혈관계 질환 예방을 위한 제약과 기능성식품 개발에 이용 가능할 것으로 기대된다.
Auricularia auricula-judae is an edible mushroom, which is known as wood ear, free ear, black ear mushroom, and free jelly fish. This study was carried out to obtain the basic information for mycelial culture conditions of Auricularia auriculajudae. According to colony diameter and mycelial density, the media for suitable mycelial growth were PDA and MCM. The optimum temperature for mycelial growth was 25~30oC. Carbon and nitrogen sources were mannose and malt extract, respectively. The optimum C/N ratio was in the range of 10 to 1 with 2% glucose. Other minor components for the optimal growth were thiamine-HCl and biotin as vitamins, succinic acid and lactic acid as organic acids, and MgSO4·7H2O and KH2PO4 as mineral salts.
Ganoderma applantaum belonged to Polyporaceae of Basidiomycetes and has been known as one of the popular medicinal mushrooms due to high antitumor and anti-diabetes activity. This study was carried out to obtain the basic information for mycelial culture conditions of Ganoderma applantaum. According to colony diameter and mycelial density, the media for suitable mycelial growth of them were shown in PDA and MEA. The optimum temperature for mycelial growth was 25~30℃The colonies are white, flocculose and underside of the plates is whitish to cream. Carbon and nitrogen sources were mannose and malt extract, respectively. The optimum C/N ratio was 10:1 to 5:1with 2% glucose concentration, vitamin was thiamine- HCl, organic acid was succinic acid, and mineral salt was MgSO4ㆍ7H2O. Present experiments were conducted to determine the possibility of artificial culture with sawdusts of G. applantaum. The pH value which was 6.0 of oak sawdust and6.4 of ricebran. Mycelial density on oak sawdust of G.applantaum spp. was higher than Ganoderma lucidium (a control). Wt. of dried fruiting body (g) of G. applantaum. (origin culture: GBEA-01, strain name: SamJung(三淨)) showed that 16.0 g on oak sawdust bottle cultivation (850㎖), 7.8 g on oak sawdust and G. lucidum. This present study was undertaken to investigate comparative anti-tumor activity of water extracts of G. lucidum ASI 7125 (GLE), G. applantaum SamJung (GAE) in vitro. The anti-tumor activity in the present study was evaluated by sulforhodamine B (SRB) and microtetrazolium (MTT) assay in terms of cell survival level. The results showed that DW, GLE and GAE inhibited proliferation showing both tumor cells.
This study was carried out to investigate chemical ingredient of Amillaria tabescens. Methanol extract of Amillaria tabescens was analyzed by column chromatography, thin layer chromatography, HPLC, mass spectrophotometer. ATB201 was isolated by prep-HPLC from butyl alcohol fraction. From the physico-chemical data, including NMR, MS, the chemical structure of the compound was determined to be an uracil. (Fig. 1). The molecular weights of uracil was 112.

This study was carried out to investigate amino acid contents of golden mushroom and pink mushroom. The amino acid analysis was followed by AccQ-Tag method and HPLC on gradient conditions. Seventeen amino acids were analyzed and sixteen amino acids were found in golden mushroom ; fifteen amino acids in pink mushroom respectively. Among total amino acid in golden mushroom, cystein content was the highest and glycine, glutamic acid, proline were followed. Among total amino acid in pink mushroom, cystein was the highest and glycine, lysine, methionine were followed. As shown in Fig.1, 2, concerning amino acids, cystein was dominant. and alanine was detectied in golden mushroom but pink mushroom.
