        The family Crambidae, so called as Grass moths, have been known as a difficult group in identification due to their similar wing patterns among the allied species. Some species are well known as pests to the agricultural and ornamental plants of economic importance. Therefore, this study was carried out to provide the DNA barcode and related information for distinguishing the complex species among Crambidae. In this study, we extracted and analyzed the DNA barcodes from 185 specimens of 76 species in the family Crambidae. Based on the result, one species of the genus Schoenobius, Pagyda, Neoanalthes, Palpita, Ecpyrrhorrhoe and Paratalanta area reported for the first time from Korea.

        As a result of our taxonomic study of the genus Epistrophe (Walker) in Korea, we recognized a total of 16 species including seven putative new species (code names are temporarily given from A to F). We also conducted a DNA barcoding analysis of 25 nominal species and 130 samples of Epistrophe. In our barcoding analysis, despite insufficient bootstrap support, Epistrophe formed a cluster with two genera, Epistrophella and Leucozona, topologically supporting their close relationships. Our analysis also showed some interesting results worthy of attention: First, at least two distinct cryptic species were found within the E. grossulariae cluster. Second, the taxon identified as E. nitidicollis seems to represent at least three distinct species. Third, three BOLD-deposited European samples listed as E. olgae showed almost identical barcode sequences with the European E. nitidicollis.

        In recent years, macroalgal bloom occurs frequently in coastal oceans worldwide. It might be attributed to accelerating climate change. “Green tide” events caused by proliferation of green macroalgae (Ulva spp.) not only damage the local economy, but also harm coastal environments. These nuisance events have become common across several coastal regions of continents. In Korea, green tide incidences are readily seen throughout the year along the coastlines of Jeju Island, particularly the northeastern coast, since the 2000s. Ulva species are notorious to be difficult for morphology-based species identification due to their high degrees of phenotypic plasticity. In this study, to investigate temporal variation in Ulva community structure on Jeju Island between 2015 and 2020, chloroplast barcode tuf A gene was sequenced and phylogenetically analyzed for 152 specimens from 24 sites. We found that Ulva ohnoi and Ulva pertusa known to be originated from subtropical regions were the most predominant all year round, suggesting that these two species contributed the most to local green tides in this region. While U. pertusa was relatively stable in frequency during 2015 to 2020, U. ohnoi increased 16% in frequency in 2020 (36.84%), which might be associated with rising sea surface temperature from which U. ohnoi could benefit. Two species (Ulva flexuosa, Ulva procera) of origins of Europe should be continuously monitored. The findings of this study provide valuable information and molecular genetic data of genus Ulva occurring in southern coasts of Korea, which will help mitigate negative influences of green tide events on Korea coast.

        동물플랑크톤 군집 연구에 DNA 바코딩과 같은 DNA 분 석 기법의 적용은 분류형태학을 기반으로 하는 전통적인 종 동정 시 발생할 수 있는 문제 (e.g. 개체의 표현형 가소성에 의한 오동정, 유사종 및 자매종, 유생 시기의 종 동정의 어려움)를 보완할 수 있다. 최근 DNA 시퀀싱 기술의 발전으로 다양한 수생태계의 동물플랑크톤 군집은 물론, 육안 및 현미경을 통해 구분하는 데 한계가 있는 동물플랑크톤의 위 내용물에 대한 DNA 기반 군집 분석 또한 가능하게 되었으며, 이는 동물플랑크톤의 섭식 먹이원 분석을 통한 생물학적 상호 작용을 이해를 돕는다. 본 논문은 동물플랑크톤 연구에 DNA 분석 기법이 활용된 사례 (e.g. DNA 바코딩을 이용한 계통분 류학적 연구, 메타바코딩을 이용한 군집 분석, 위 내용물 분석)를 소개하고 분석 방법을 요약하여, 최종적으로 향후 이를 활용하고자 하는 연구자들에게 연구 접근성을 높일 수 있도록 방법론적인 기초 지식을 제공하고자 하였다.
