        Korean Native Pig (KNP) has a uniform black coat color, excellent meat quality, white colored fat, solid fat structure and good marbling. However, its growth performance is low, while the western origin Yorkshire pig has high growth performance. To take advantage of the unique performance of the two pig breeds, we raised crossbreeds (KNP × Yorkshire to make use of the heterotic effect. We then analyzed the liver transcriptome as it plays an important role in fat metabolism. We sampled at two stages: 10 weeks and at 26 weeks. The stages were chosen to correspond to the change in feeding system. A total of 16 pigs (8 from each stage) were sampled and RNA sequencing was performed. The reads were mapped to the reference genome and differential expression analysis was performed with edgeR package. A total of 324 genes were found to be significantly differentially expressed (|log2FC| > 1 & q < 0.01), out of which 180 genes were up-regulated and 144 genes were down-regulated. Principal Component Analysis (PCA) showed that the samples clustered according to stages. Functional annotation of significant DEGs (differentially expressed genes) showed that GO terms such as DNA replication, cell division, protein phosphorylation, regulation of signal transduction by p53 class mediator, ribosome, focal adhesion, DNA helicase activity, protein kinase activity etc. were enriched. KEGG pathway analysis showed that the DEGs functioned in cell cycle, Ras signaling pathway, p53 signaling pathway, MAPK signaling pathway etc. Twenty-nine transcripts were also part of the DEGs, these were predominantly Cys2His2-like fold group (C2H2) family of zinc fingers. A protein-protein interaction (PPI) network analysis showed that there were three highly interconnected clusters, suggesting an enrichment of genes with similar biological function. This study presents the first report of liver tissue specific gene regulation in a cross-bred Korean pig.

        The relationship of several candidate genes polymorphisms with breeding values of economic traits were investigated in Korean Native Pigs. Record (2001-2006) of 546 Korean native pigs were obtained from National Institute of Animal Science, Korea having data on average daily gain (ADG), age at 70 kg (D70 kg) and backfat thickness (BF). The data's obtained were analyzed by the DF-REML (Derivative-Free Restricted Maximum Likelihood) program of Boldman using a single-trait animal model to analyse the genetic parameters. The analysis of restriction fragment length polymorphism (RFLP) was conducted on 68 Korean native pigs (KNP) using single nucleotide polymorphism (SNP). Different genotype frequencies of 5 candidate genes such as MC4R, PRKAG3, FABP3, ESR and PRLR3 were observed in KNP. Significant relationship of AA and AB genotype between MC4R polymorphic site and breeding value for average daily gain (ADG, p<0.05) was observed. PRKAG3 polymorphic sites were also found to be significantly related to breeding values for ADG, AA, AB genotype (p<0.05) and also, for Backfat thickness (BF), days to 70 kg and BB genotype (p<0.05). In conclusion, selection method would be more effective if it encompasses significant genotype for performance traits and that would further aid in the selection of seed stock in KNP.

        본 연구는 후보 유전자의 경제형질에 미치는 염기 변이 효과를 검증하기 위해 국내에서 사육된 듀록 품종 96두와 한국 재래 돼지 86두를 활용하였다. 검증에 활용된 4개의 후보 유전자는 MC4R, PA-KAG3, FABP3 그리고 ESR 유전자였다. 각 후보 유전자들의 유전자형 분석 결과 두 집단 간에 유전적 특성의 차이가 분명히 나타나고 있음을 확인하였다. MC4R 유전자의 A 대립 유전자는 두 집단 모두에서 성장 형질과의 유의한 연관성이 검출되었고,

        Molecular genetic markers were genotyped used to detect chromosomal regions which contain economically important traits such as growth traits in pigs. Three generation resource population was constructed from a cross between the Korean native boars and Landrace sows. A total of 193 F2 animals from intercross of F1 were produced. Phenotypic data on 7 traits, birth weight, body weight at 3, 5, 12, 30 weeks of age, live empty weight were collected for F2 animals. Animals including grandparents (F0), parents (F1), offspring (F2) were genotyped for 194 microsatellite markers covering from chromosome 1 to 18. Quantitative trait locus analyses were performed using interval mapping by regression under line-cross model. To characterize presence of imprinting, genetic full model in which dominance, additive and imprinting effect were included was fitted in this analysis. Significance thresholds were determined by permutation test. Using imprinting full model, four QTL with expression of imprinted effect were detected at 5% chromosome-wide significance level for growth traits on chromosome 1, 5, 7, 13, 14, and 16.

        The Korean native pig (KNP) have been considered as animal models for animal biotechnology research because of their relatively small body size and their presumably highly inbred status due to the closed breeding program. However, little is reported about the use of KNP for animal biotechnology researches. This study was performed to establish the somatic cell nuclear transfer (SCNT) protocol for the production of swine leukocyte antigens (SLA) homotype-defined SCNT KNP. The ear fibroblast cells originated from KNP were cultured and used as donor cell. After thawing, the donor cells were cultured for 1 hour with 15 μM roscovitine prior to the nuclear transfer. The numbers of reconstructed and parthenogenetic embryos transferred were 98 ± 35.2 and 145 ± 11.2, respectively. The pregnancy and delivery rate were 3/5 (60%) and 2/5 (40%). One healthy SLA homotype-defined SCNT KNP was successfully generated. The recipient-based individual cloning efficiency ranged from 0.65 to 1.08%. Taken together, it can be postulated that the methodological establishment of the production of SLA homotype-defined cloned KNP can be applied to the generation of transgenic cloned KNP as model animals for human disease and xenotransplantation researches.
