본 연구는 NaCl 및 jasmonic acid(JA) 처리 시 스피아민트(Mentha spicata)의 생육 및 생리적 반응 변화를 조사하고자 실시하였다. 대조구, JA(20μM JA 전처리), JA+NaCl(20μM JA 전처리+ 50mM NaCl), NaCl(50mM NaCl) 처리 등 4처리구로 구분하여 0.5배 Hoagland 용 액에서 3주간 수경 재배한 후 생장반응과 엽록소, 비타민 C, 프롤린, 무기물, DPPH 라디칼 소거 항산화 활성등을 측정하였다. 스피아민트의 초장, 엽장, 엽폭 및 생 체중은 대조구에 비해 모든 처리구에서 감소하였고, 특히 NaCl 처리구에서 현저히 감소하였다. 반면에 건물중 은 JA+NaCl 혼용처리구를 비롯하여 모든 처리구에서 증가하였다. 엽록소 a와 b 함량은 JA 처리구가 가장 높았으나 비타민 C와 항산화 활성 및 지상부의 프롤린 함량은 염 스트레스로 생육이 가장 낮았던 NaCl 처리구에서 가장 높게 나타났다. 이온 흡수의 균형 정도를 알 수 있는 K/Na 비율은 JA 단독 처리 시 증가하였고, NaCl 처리 및 JA+NaCl 혼용 처리구에서는 Na+ 흡수 증가로 인하여 K/Na 비율이 낮아졌다. 이러한 결과는 낮은 염 스트레스나 JA처리 같은 적절한 스트레스 처리가 항산화 활성과 정유를 포함하는 이차대사물질의 축적을 유도 함으로써 스피아민트의 품질을 향상시킬 잠재적인 가능성을 보여준다.
This study was performed to investigate attraction effect of spearmint oil and to confirm their EAG response against lantern fly, Lycorma delicatula nymphs and adult. In dose responses to spearmint oil, 2nd ∼ 4th nymphs and adult were significantly attracted at a dose of 5㎕. Attraction effect was showed highest (84.8%) at a 4th instar nymph, and mild effect (approximately 60%) was observed at 2nd∼3rd instar nymphs, but the others are not showed the significance. At a dose of 10㎕, 4th instar nymph and adult were significant and only 4th instar nymph was significantly attracted at a dose of 2.5㎕ but the others are not. Carvone, a constituent of spearmint oil, exhibited significantly attraction effect on nymphs and adult except 1st and 2nd instar nymphs. But limonene did not show any attraction effect. The attraction assay of L. delicatula to the mixtures of constituents appeared to be efficient additively. In EAG response to spearmint oil which exhibited attraction effect, antennae of 4th instar nymph and adult responded to only carvone. In conclusion, attraction effect of spearmint oil was more effective in 4th instar nymph and adult than 1st ∼ 3rd instar nymphs.
This study was performed to investigate attraction effect of six plant essential oils and to confirm their electrophysiological response against Lycorma delicatula. Among the tested oils, spearmint oil (94.1%) significantly attracted L. delicatula at a dose of 1.25㎕/cm2 by using an olfactometer. In dose response to spearmint oil, a dose of 2.5㎕/cm2 was very effective. GC-MS analysis revealed that the active components responsible for the effective attraction effect of spearmint oil were carvone (70.6%) and limonene (54.8%). Of the two active components, carvone was more significant than limonene with reference to attraction activity against L. delicatula. Analysis by GC-EAD showed, major components of spearmint oil that elicited response in L. delicatula antennae, indicating the potential role of the essential oil as attractant that determine the choice of the attraction material. In the field test, spearmint oil exhibited attraction effect up to 5 days. This effect was different in accordance with test places and treatment dose.
Fumigant toxicity of four plant essential oils (muguet flower, patchouli, pennyroyal, spearmint) were tested against the adults of westem flower thrips, Frankliniella occidentalis. Among them, pennyroyal and spearmint oils showed 100% mortality at 1 ㎕/45 ㎖ air dose. Through the constituent analysis using GC and GC-MS, we confirmed that main constituents of pennyroyal oil were pulegone(100.0%), and spearmint oil were β-myrcene(3.5%), limonene(12.1%), carvone(85.4%). Among them, carvone and pulegone showed 100% fumigation at 1 ㎕/45 ㎖ air dose, respectively. It can concluded that two oils aloe potential control agents against F. occidentalis.
본 연구는 시설하우스의 주요 해충인 온실가루이와 담배가루이에 대한 spearmint oil의 훈증독성 및 접촉독성을 조사하였으며, gas chromatograph (GC)와 gas chromatograph mass spectrometry(GC/MS)를 이용하여 speamint oil의 주요 구성성분을 분석하였다. 훈증독성 시험 결과, 온실가루이에 대해서 spearmint oil은 air의 약량처리에서 의 살충활성을 나타내었으며, 5와 air의 약량처리에서 각각 와 의 살충활성을 나타내었다. 또한 담배가루이에 대해서 spearmint oil은 air의 약량처리에서 각각 의 살충활성을 나타내었으며, 5와 air의 약량에서도 각각 100, 의 높은 살충활성을 나타내었다. 그러나 온실가루이와 담배가루이 성충에 대한 spearmint oil의 접촉독성은 살충효과가 이하로 낮았다. Spearmint oil을 GC와 GC/MS로 분석한 결과, limonene (), -terpinene(), -cymene(), 3-octanol(), carvone()이 주요 구성성분으로 나타났으며, 주요성분인 carvone 은 air 약량에서 의 높은 훈증독성을 나타내었다.
Mentha spicata, Ocimum basilium L., and Rosemarinus officinalis L. were selected as subjects for the study, and analysis of rosmarinic acid contents by HPLC analysis and the fragrance components of each herb were compared by GC-MS analysis. It was prepared and cultivated in a herb farm located in Sacheonjin-ri, Gangneung-si in 2022 (Fig. 1) before flowering, collected in the summer, stored at minus 20℃, and dried 50 g each. The analysis was requested to the NICEM analysis room of Seoul National University to obtain the test results. Through the above study, HPLC showed that the content of rosmarinic acid was 555.53 mg/Kg, which was absolute, 27.12% in rosemary for 1,8-cineol in GC-MS, and 32.44% in linalool in sweet basil. For eugenol, 23.11% in sweet basil and 19.92% in rosemary were found in α-pinene. Through this study, it was verified that spearmint, sweet basil, and rosemary contain a large amount of major ingredients that act on concentration or brain activation.
IPP isomerase (Iso) and Limonene synthase (Limo) are important enzymes in terpenoids biosynthesis pathway. The wild type and each metabolically engineered (Iso and Limo) transgenic spearmint (Mentha spicata Linne) plants were compared for their growth patterns and the contents of essential oil in in vitro culture media. The profile of terpenoid metabolites was obtained from the essential oil of the metabolically engineered transgenic spearmint, which was extracted using a modified SDE method, by GC-MS analysis. The growth of wild spearmint was more profuse in B5 culture medium than in other media. Significant differences in leaf and root growth patterns were observed between metabolically engineered transgenic and wild type spearmint plants. The leaves of the transgenic spearmint plants were slightly elongated but were dramatically narrower than those of wild type spearmints. The content of essential oil of transgenic spearmint was different slightly depending on the target terpenoid genes. The content of essential oils in Limo transgenic plants was higher than that of Iso, except for transgenic plant in B5 medium. The transgenic spearmint produced more terpenoids than the wild type. Iso spearmint extracts showed eleven terpenoids and a phenylpropane, while Limo spearmint extracts contained nine terpenoids. However, extracts from the wild type showed the presence of only four terpenoids.
