        호장근, 적하수오, 대황, 알로에 등과 같은 생약재의 주요 약리 활성 성분인 emodin은 항산 화, 항균, 항염, 항암, 간보호 등에 효능이 있는 것으로 보고되었다. 본 연구에서는 emodin의 피부 질 환 및 기능성 소재로서의 활용 가능성을 알아보기 위해 염증 개선과 피부장벽기능 개선 관련 활성을 확 인하였다. human keratinocyte인 HaCaT 세포에 대하여 항염효과를 관찰하기 위해 cytokine억제능은 ELISA kit로, 단백질 발현은 western blot으로 확인하였다. TNF-α (10 ng/mL)/IFN-γ (10 ng/mL)로 활성화된 HaCaT 세포에서 emodin을 농도별(5, 10, 20, 40) μM로 처리한 결과 TNF-α, IL-1β 및 IL-6의 생성량은 emodin의 농도가 증가함에 따라 감소됨을 확인하였다. 염증관련 단백질인 iNOS, COX-2 발현량에 대한 실험결과에서도 emodin 20 μM 농도에서 control 대비 iNOS는 48%, COX-2 는 29%가 저해됨을 확인하였다. 피부장벽기능 개선의 지표로써 filaggrin, involucrin, loricirn의 mRNA 발현 정도와 filaggrin, involucrin, loricirn의 생성량을 확인한 결과 에모딘 농도에 의존적으로 증가하는 우수한 결과가 얻어졌다. 특히 20 μM 농도에서 대조군 대비 2배의 생성량 증가를 보인 filaggrin은 천 연보습인자인 NMF의 형성에 관계되는 단백질로 각질층의 보습에 중요한 역할을 하는 것으로 알려져 있다. 결론적으로 emodin의 피부 질환 및 기능성 소재로서의 활용 가능성 중 염증 개선 및 피부장벽기 능 개선 소재로 활용될 수 있음을 확인하였으며 향후 추가적인 연구가 수행되면 그 활용 범위가 더 넓 어질 수 있을 것으로 사료된다.

        Aloe-emodin (AE) is the major bioactive component in aloe and known to exhibit anti-inflammatory activities. However, it has not been elucidated whether its anti-inflammatory potency can contribute to the elimination of obesity. The aim of the current study is to investigate the effect of AE on toll-like receptor 4 (TLR4) pathways in the presence of lipopolysaccharide (LPS) in 3T3-L1 adipocytes. 3T3-L1 adipocytes were treated with AE (0-20 μM) for one hour, followed by LPS treatment for 30 min and then, adipokine mRNA expression levels were measured. Next, TLR4-related molecules were measured in LPS-stimulated 3T3-L1 adipocytes. AE significantly decreased the mRNA expression of the tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), and monocyte chemoattractant protein-1 (MCP-1) in a dose-dependent manner. Moreover, AE suppressed TLR4 mRNA expression. Further study showed that AE could suppress the nuclear factor-κB (NF-κB) and phosphorylation of extracellular receptor-activated kinase (pERK). The results of this study suggest that AE directly inhibits TLR4/NF-κB/ERK signaling pathways and decreases the inflammatory response in adipocytes.

        This study was conducted to investigate the physicochemical properties of the ethanol extracts and soluble dietary fiber from Cassia tora L. seed. The proximate composition of Cassia tora, soluble solid contents, color intensity and contents of emodin and rhein of Cassia tora extract, molecular mass distribution, sugar contents and viscosity of soluble fiber from Cassia tora were analyzed. Cassia tora contains 12.6% of moisture, 5.2% of ash, 13.4% of crude protein, 7.2% of crude fat, 8.8% of insoluble fiber and 48.3% of soluble fiber. The effects of extract condition on soluble solid contents, color intensity and contents of emodin and rhein of Cassia tora extract were investigated. The soluble solid contents were higher in 70% or 50% ethanol extracts than those in 100% ethanol extracts and showed highest value in grind sample extracts. In Hunter's color value, 100% ethanol extracts and whole Cassia tora sample extracts were higher in L and b value, but on the contrary, were lower in a value, than those of the other. The highest emodin and rhein contents were observed in 70% and 50% ethanol extracts, respectively, and showed higher value in room temperature extracts than in heating extracts. The molecular mass of soluble fiber from Cassia tora seed was estimated by gel filtration chromatography. Most soluble fiber(80%) exhibited a molecular mass range of between 50~2000 kDa. The major sugars of soluble fiber from Cassia tora seed were identified as xylose, mannose and galactose. The apparent viscosity of 0.5% soluble fiber from Cassia tora seed was 33 mPas showing a higher value than pectin or xanthan gum.

        Emodin is a bioactive compound isolated from the root and rhizomes of Rheum plamatum L. (polygenaceae), which is known as a traditional Chinese and Japanese medicine. In the present study, the effect of emodin on YD-15 mucoepidermoid carcinoma cells and its molecular mechanism were investigated. This study shows that emodin significantly inhibits the growth of YD-15 cells. Activation of caspase-3 and PARP is triggered by emodin and it increases sub-G1 population and the number of YD-15 cells with nuclear condensation and fragmentation. In addition, we found that emodin significantly decreases myeloid cell leukemia 1(MCL-1). These results suggest that MCl-1 is an important molecule for emodin-induced apoptosis. Taken together, emodin inhibits cell viability and induces apoptosis via down-regulation of MCL-1 and it can be a new potent anticancer drug candidate for the treatment of mucoepidermoid carcinoma
