        A basidiomycetous wood rotting fungus Pleurocybella porrigens (angel wings) has been well known as an excellent edible mushroom and named as Sugihiratake, in Japanese common name based on its morphological and ecological features. This mushroom has been favorably consuming by Japanese people. In 2004, an outbreak of serious acute encephalopathy exclusively occurred in patients with chronic kidney diseases after the intake of this mushroom. Thereafter, many researchers have conducted to examine the factor of onset of the encephalopathy, but the exact factors that induced the encephalopathy still remain unclear. Matsumoto et al. (2005) reported the mushroom specimens from different geographical areas in Japan were grouped into two distinct clusters based on ITS rDNA sequences. Vegetative growth of P. porrigens is extremely slow even cultivation on natural media surveyed and its physiological characteristic gives a disadvantage for the development of various fields of researches on this mushroom. Thus, we attempted to find a medium accelerating the vegetative growth of P. porrigens for further research to elucidate the possible association between its chemical constituent(s) and the onset of encephalopathy. Fifteen isolates of P. porrigens collected from different geographical areas in Japan were cultivated on PDA. The growth of the isolates was extremely slow as we know and most tested isolates formed mycelia look like those often observed in the cultivation of basidiomycetous ectomycorrhizal fungi. Among them, we chose five isolates based on the ranking of their higher growth rates. The top five isolates were cultured in five kinds of liquid media, PD medium, MY medium, Carrot medium, Ohta’s medium for ectomycorrhizal fungi (Ohta, 1990) and Amazake medium (Ishihara, 2007), and on a solid medium, PDA, at 20℃ in the dark. The dry biomasses of the isolates cultured in the liquid media were determined after 6 weeks of the stationary cultivation whereas the growth rates of the isolates cultured on PDA were compared by the colony diameter after 8 weeks of the cultivation. Among the tested liquid media, PD medium was the most suitable medium for their biomass growth (17 mg dry mycelium/flask - 127 mg dry mycelium/flask) and followed by Ohta’s medium (13 mg dry mycelia/flask - 79 mg dry mycelia/flask), MY medium (10 mg dry mycelium/flask - 72 mg dry mycelium/flask), Amazake medium (8 mg dry mycelium/flask - 47 mg dry mycelium/flask) and Carrot medium (4 mg dry mecelium/flask - 18 mg dry mycelium/flask). The average biomass growths of the isolates cultured in the synthetic medium, Ohta’s medium were 20-92% of those of the isolates cultured in PD medium. No correlation was observed between the cultivation in PD medium (liquid medium) and that on PDA (solid medium). Extremely large biomass variation was observed among the same isolates cultured on the same kind of the liquid medium. Large variation in colony features associated with their growth rates was also observed among the isolates cultured on PDA. These results suggest that PD medium would be a suitable one for most researches and Ohta’s medium for researches requiring clarity of its chemical constituents. The above results also indicate that the Japanese population of P. porrigens has large variation in vegetative growth although we have not yet examined whether the tested isolates comprise cryptic species or not.

        작물육종 프로그램에 선충저항성은 매우 중요한 구성요소의 하나이다. 세계적으로 선충에 의한 작물의 수량 감소는 약 10% 정도로 추산되고 있으나 아시아 각국에서는 주요 작물별로 선충의 피해현황이나 저항성 작물 등에 대한 연구가 아직 미진한 실정이다. 본 연구에서는 한국, 베트남, 말레이시아 각국의 최근 선충저항성 관련 연구 현황을 분석하였다. 한국에서는 주요 경제작물 중 쌀, 콩, 고추, 오이, 참외 뿐만 아니라 장미, 카네이션, 국화 등 화훼작물에 대한 선충저항성 품종 선발 연구가 이루어졌다. 베트남에서 는 바나나, 커피, 쌀, 리치 등에 대한 선충저항성 검정연구와 식물 기생선충의 분포에 관한 연구가 이루어졌다. 말레이시아에서는 최근 파인애플에서 선충으로 인한 연작장해가 문제가 되고 있으나 저항성품종 개발 연구는 시작단계에 있다. 아시아 각국에서 선충에 대한 연구가 부진한 이유는 방제전략의 부재, 경제적인 종합방제 대책의 부족, 선충에 대한 정보 부족, 살선충제 가격 부담 등이 들 수 있다. 따라서 보다 경제적이면서 지속가능한 선충관리전략의 수립이 시급하다. 선충저항성 유전자원의 이용과 품종개발은 아시아 지역에서 경제적이고 효과적인 선충관리 대책의 하나가 될 수 있다.

        The perilla leaf pyralid moth, Pyrausta panopealis Walker (Lepidoptera: Pyralidae), is a serious pest damaging to leaf perilla. In order to establish the life parameters of P. panopealis for eventual purpose of control, the developmental span of each stage were investigated under five temperature regimes (20℃ ~ 3 0℃). In addition, the larvicial efficacy of several on-the-market environment-friendly agricultural materials (EFAMs) was tested. The width of head capsule at each larval stage measured to be the mean of 0.21, 0.32, 0.47, 0.64, and 0.98 mm, respectively. The larval period of P. panopealis was longest at 20ºC as 27.0 days and shortened as temperature goes up to 30ºC as 11.3 days. Survivorship of the larval P. panopealis was the highest at 30ºC as 80%, whereas that of other temperatures ranged from 40% (20ºC) to 62.5% (27.5ºC), indicating that the P. panopealis appears to favor higher temperature. In addition to larval period, the duration of egg, prepupa, and pupa stages also shortened as temperature goes up, whereas the duration of adult stage increased as temperature goes up: from 4.1 days at 20 ºC to 6.1 days at 30ºC. After the perilla leaf pyralid moths were successfully stabilized in indoor environment the larvicidal efficacy of the ten EFAMs that were previously selected from the result of other moth species was tested aimed at 4th instar larvae for 48 hrs. Seven of the ten tested showed more than 90% of mortality within 12 hrs and reached nearly up to 100% within 24 hrs, but the remaining three showed less than ~70%.

        This study was performed to investigate the correlation between changes of feeding behavior of green peach aphid, Myzus persicae and residual effect of an insecticide, Pyrifluzuinazon, using EPG technique. Pyrifluquinazon was showed the insecticidal activity until three days (72h) after treatment, and the activity was high in nymph than adult of GPA. There was no difference among treatment methods. Lethal sign was observed the slimed the abdomen of GPA after 3 days that rises the insecticidal activity, and dieㅇ as being sticked and/or pulled out the needle. Residual efficacy from 1, 3, 5, 7 to 20 days after treatment, insecticidal activity was showed 70% in 50 ppm, recommended concentration, until 5 days. And waveforms relating to non-probe time and phloem phase time using EPA, feeding inhibition efficacy was showed during 5 days after treatment, but showed similar level with control after 13 days. These results show that the change of feeding behavior of GPA is correlated with the change of residual effect of pyrifluzuinazon.

        Our previous study conducted in a wasted mulberry plantation showed significant decline of leaf productivity of mulberry (Morns sp.) by cattle grazing in three years (Ogura et al., 2009). This means t㏊t improvement of understory vegetation is essential to use such wasted areas as grazing lands. In this study, the mixture of seeds (Dactylis glomerata, Lolium perenne, Poa pratensis and Trifolium repens, 15:15:5:2) was seeded on 18 Sep. 2007, in nine experimental plots (6 m × 6 m each) with different seeding rates (L: 37 ㎏/㏊, M: 56㎏/㏊ and H: 74㎏/㏊, three replications) in the wasted mulberry plantation described in the previous paper (paddock A) (Ogura et al., 2009). Two cows were grazed in the paddock for 42 and 54 days in 2007 and 2008, respectively. No fertilizer nor herbicides were applied in the paddock. The coverage of the sown species and weeds, and the proportion of bare area were recorded on 19 Sep. 2007, 16 Nov. 2007, 21 May 2008 and 21 May 2009. T. repens rapidly covered the understory; the coverage ranged 49.4-71.7% in 2008-2009 (Fig. 1). D. glomerata and L. perenne also increased to 10.6-18.3% and 20.6-33.9% in 2009, respectively, regardless to the seeding rates. In contrast, the proportion of bare area rapidly decreased. The quick establishment of the sown plants probably due t㏊t defoliation and trampling of cows suppressed the growth of native plants. In conclusion, the seeding rate of 3.7 ㎏/㏊ is sufficient to establish herbage plants in wasted mulberry plantations under cattle grazing.

        Mulberry (Morus sp.) plantations have been deteriorating in Japan due to decline of silk industry. In mountainous areas, large part of the mulberry plantations has been wasted. Cattle grazing in the wasted mulberry plantations is a practical system to reuse the wasted areas for food production, because mulberry leaves are highly nutritive (Ezenwa and Kitahara, 2001), productive (Kitahara et al., 2002) and palatable to cattle (Ogura et al., 2008). In this study, the productivity and quality of mulberry leaves were investigated for 3 years in a wasted mulberry plantation grazed with beef cows, in Minamisanriku town, Miyagi, Japan. Cattle grazing started in September 2005 (paddock A, 44 a) and June 2006 (paddock B, 96 a). For each paddock, two pregnant cows were grazed from June to August-October in 2006-2008. In the paddocks A and B, four mulberry trees were chosen and the leaf mass and chemical composition were measured in mid-June and late August (pre-grazing leaf mass) during 2006-2008. A non-grazed paddock was also measured in 2007-2008. Leaf productivity of mulberry trees significantly decreased with cattle grazing; pre-grazing leaf mass changed from 246.8 g to 10.4 g DM/tree and 286.2 g to 17.6 g DM/tree in the paddock A and B, respectively (Fig 1). In contrast, 149.2-365.1 g DM/tree of leaves was produced in the non-grazed paddock. Chemical composition was stable during the three years in all paddocks. It is concluded that cattle grazing gives severe damage to leaf production of mulberry trees in three years.

        The objective of this study was to develop a reliable and reproducible descriptive analysis procedure for Korean style sweet pumpkin gruel (Hobakjuk). The sensory attributes of the sweet pumpkin gruel were developed and defined, the sample preparation method was standardized, and the sensory evaluation procedure for a sample was established. Seven types of sweet pumpkin gruel (five ready-to-eat type vs. two ready-to-heat type) were selected to be analyzed. Panel training and descriptive analysis were carried out with these 7 samples. A total of 12 sensory attributes (2 aroma/odor, 5 taste/flavor, 4 texture/mouthfeel, and 1 aftertaste attributes) were developed to describe the sensory characteristics of the sweet pumpkin gruel. The definition and reference standards for each sensory attribute were determined to clearly understand each attribute. In the main experiment, trained panelists evaluated the sensory characteristics of the 7 gruel samples based on a fifteen-point intensity scale using the developed attributes. The results were statistically analyzed by analysis of variance, principal component analysis, and cluster analysis. The results showed that the 7 sweet pumpkin gruel samples significantly diffieredin their intensities of all attributes except for sweet pumpkin aroma and viscosity. The ready-to-eat style samples were distinctly characterized by their sweet pumpkin aroma and flavor, whereas the ready-to-heat style samples were markedly characterized by their low intensity of gelatinized starch and pumpkin flavor retention.

        A full genomic DNA microarray technique was employed to investigate the effects of Dongchunghacho on aortal and hepatic gene expression in apolipoprotein E knockout mice fed a high-fat/high-cholesterol diet. Male 8- week - old ApoE-/- mice were randomly divided into two groups, control(high cholesterol group; HC) and supplementation of Dongchunghacho (SD). All of the mice were fed a high-fet/high cholesterol diet with or without Dongchunghacho supplemented by 1% for 6 weeks. At first, lipid profile of the Dongchunghacho was measured by biochemical analysis. No differences were observed in serum triglyceride and total cholesterol levels between the two groups. Antigenotoxic effect of the Dongchunghacho was measured by the single cell gel electrophoresis assay (Comet assay) and quantified as % fluorescence in tail. Dongchunghacho supplementation decreased significantly leukocytic DNA damage and also there was a tendency of reduction in hepatic DNA damage in Dongchunghacho group compared with the control group. In up regulated genes in liver and aorta of the mice, genes with 0 to 2- fold difference in expression level between the two group (HD and SD) was very much more in liver than in aorta, on the contrary, those with 2-fold to 16-flod difference increased greatly rather in aorta than in liver. Also, almost the same results were observed in down regulated genes in liver and aorta between the two groups. These results suggested that supplementation of Dongchunghacho might be helpful in preventing leukocytic DNA damage induced by high fat diet, and has a more crucial roles in aortal gene expression.
