        Sacbrood virus(SBV) causes a fatal disease(sacbrood) of honeybee larvae, which fail to pupate, change color and shape, and finally die. The complete nucleotide sequence of SBV has recently been determined, and with these data, we now report a Reverse Transcription-PCR(RT-PCR) test for the direct, rapid, and sensitive detection of these viruses. To detect the SBV infection in Korea, we collect beekeepers from various apiaries, which the RT-PCR technique was used. And we designed SBV specific primers in conserved region of the viral genome in the GenBank database. We confirmed the SBV amplicon using cloning and sequence. Homology between determined sequences of SBV korean strain and published virus sequences were 97% in DNA sequence, and 100% in amino acid sequence. We describe the first time that presence of sacbrood virus(SBV) in Korea honey bee colonies using RT-PCR. We also developed and validated a RT-PCR assay for the detection of SBV in Korea.

        Viruses of the honeybee, Apis mellifera L. are known to reside at low levels in colonies, typically showing no apparent signs of infection. Chronic paralysis virus(CBPV) is known to induce significant losses in honey bee colonies. The pathology is characterized by clusters of trembling, flightless, crawling bees and by individual bees, sometimes hairless, standing at the hive entrance. A minusstrand-specific RT-PCR was used to assess viral replication. This is the first report on the infection of CBPV in Korea. Using (-)RT-PCR, 27 apiaries in korea were screened for the honeybee viruses, with positive colonies being analysed for viral genetic diversity. We got 550-nt PCR product from CBPV genomic RNA. Nucleotide sequences were aligned to the complete CBPV genomic RNA sequence deposited in the GenBank database and was revealed 96%(AM-CBPV) identity, respectively.         Mortality of honeybees is a serious problem that beekeepers have to face periodically in Korea and worldwide. The presence of RNA viruses, in addition to other pathogens may be one of its possible causes. In this work, we detected Deformed wing virus(DWV), Israle Acute Paralysis Virus (IAPV), Black queen cell virus (BQCV), Cloudy wing virus(CWV), Kashmir bee virus(KBV), Sacbrood virus(SBV), Chronic bee paralysis virus(CBPV) in samples of korea honeybees with or without Varroa destructor and Nosema apis. The detection of viruses in all provinces, simultaneous co-infection of colonies by several viruses and the fact that 96.3% of the samples were infected with one or more virus, indicates they are widely spread in the region. Using uniplex and multiplex RT-PCR we screened honey bee colonies for the presence of several bee viruses, including DWV, IAPV, BQCV, KBV, CWV, and described the detection of mixed virus infections in bees from these colonies. Conclusively, investigated disease of the bee, and confirmed new virus that lead to bee disease, this is thought by valuable thing as data for development of beekeeping industry such as CCD(Colony Collapse Disorder)'s cause searching examination.

        The caterpillar-shaped Chinese medicinal mushroom (DongChongXiaCao) looks like a worm in the winter and likes a grass in the summer. The fruiting bodies of Cordyceps species have demonstrated various biological activities and used as traditional medicines in Asian countries. The price of wild Cordyceps have been ascending every year as their demand increase. Recently, the artificial cultivation of these fungi has been successfully established and they are currently produced on a rice or pupae media. This study was carried out to investigate the soluble sugar, amino acid and fatty acid profiles in the fruiting-bodies of Cordyceps militaris, C. pruinosa and Paecilomyces. tenuipes formed on the living silkworm (Bombyx mori) host. Soluble sugars such as glycerol, glucose, mannitol and sucrose were found in the fruiting bodies of Cordyceps militaris, C. pruinosa and P. tenuipes. Total soluble sugar content was 29.23 ㎎g-1in C. militaris, 8.61㎎g-1 in C. pruinosa and 24.00 ㎎g-1in P. tenuipes on dry weight basis. Total free amino acid content was 14.09 ㎎g-1in C. militaris, 34.60㎎g-1 in C. pruinosa and 17.09 ㎎g-1 in P. tenuipes. The content of oleic acid in fatty acids was above high more than 30% regardless of species.

        Treponema denticola is the best studied oral spirochete and numerous studies have shown that it is strongly associated with periodontitis and expresses several putative virulence factors. In this study, we report on a surface protein of T. denticola, Td92, which is homologous to Tp92 of Treponema pallidum, an agent of syphilis. Immunofluorescence assay and immunogold labeling with anti-Td92 Ab revealed that Td92 had surface-exposed epitopes. And Td92 was capable of binding to fibronectin and KB cells, an oral epithelial cell line. In addition, Td92 could enter the KB cells. These results indicate that Td92 is a fibronectin-binding protein which can bind to and internalize into the host cells, facilitating the virulence of T. denticola.

        In order to investigate functional groups on the surface of Multi-walled Carbon Nanotubes (MWCNTs) induced by oxyfluorination, XPS (X-ray photoelectron spectroscopy) analysis was carried out. All core level spectra of MWCNTs were deconvoluted to several Pseudo-Voigt functions (sum of Gaussian-Lorentzian functions). Both O1s and F1s binding energy of oxyfluorinated MWCNTs shifted high value as increment of fluorine mixing ratio. The carbon-fluorine covalent bonding concentration increased as increment of fluorine mixing ratio. The shape and intensity of OF10-MWCNTs are similar with those of as-received MWCNTs. However, the intensity and binding energies of main peak position of OF20-MWCNTs and OF30-MWCNTs were dramatically increased by oxyfluorination.

        사용후핵연료 건식저장용기는 낙하사고조건에서 캐니스터의 건전성이 입증되어야 한다. 낙하사고조건은 캐니스터를 건식저장용기에 장입하기 위하여 저장용기의 상부에서 크레인으로 취급하는 도중에 캐니스터가 저장용기 내부의 받침대로 자유 낙하하는 조건이다. 저장용기 내부의 받침대는 이러한 조건에서 캐니스터의 구조적 건전성을 유지하도록 완충효과가 좋아야 한다. 본 연구에서는 다양한 저장용기 내부 받침대 에 대한 3차원 유한요소해석을 통하여 낙하사고조건에서 캐니스터의 구조적 건전성을 향상시킬 수 있는 구조를 결정하였다. 저장용기 내부 받침대는 탄소강으로 만들어진 원통 쉘의 내부에 콘크리트를 장입한 구조와 받침대 높이의 변화 없이 콘크리트 높이의 1/4정도에 탄소강과 폴리우레탄폼을 이용한 구조물을 사용하여 완충효과를 보완하고자 수정된 구조를 고려하였다. 완충체의 형상 및 구조를 결정하기 위하여 십자형상이나 원형의 탄소강 구조물을 받침대 상부에 위치하여 그 영향을 알아보았다. 이때 탄소강 구조물의 두께를 24 mm, 12 mm, 6mm로 변화를 주었다. 또한, 탄소강 구조물 사이에 충진하는 폴리우레탄폼의 밀도에 대한 영향을 알아보았다.

        This study was conducted to examine the effect of IRES controlled reporter gene on screening and production of recombinant human erythropoietin (EPO) proteins from cultured CHO cells. The cDNA was cloned for EPO from human liver cDNA. Using site-directed mutagenesis, we generated recombinant human EPO (rhEPO) with two additional N-glycosylations (Novel erythropoiesis-stimulating protein: NESP). Wild type hEPO and NESP were cloned into expression vectors with GFP reporter gene under regulatory control of CMV promoter and IRES so that the vectors could express both rhEPO and GFP. The expression vectors were transfected to cultured CHO-K1 cells. Under microscopy, expression of GFP was visible. Using supernatant of the culture, ELISA assay, immunocytochemistry and in vitro assay using EPO dependant cell line were performed to estimate biological activity to compare the production characteristics (secretion levels, etc.) between rhEPO and NESP. The activity of NESP protein, obtained by mutagenesis, was described and compared with its rhEPO counterpart produced under same conditions. Although NESP had less secretion level in CHO cell line, the biological activity of NESP was greater than that of rhEPO. These results are consistent with previous researches. We also demonstrated that rhEPO and GFP proteins expressed simultaneously from transfected CHO cell line. Therefore we conclude that use of GFP reporter gene under IRES control could be used to screen and produce rhEPO in cultured CHO cells.

        고리 원전 사용후핵연료 저장조의 저장용량을 확보하기 위하여 2002년부터 사용후핵연료 운반용기를 이용하여 400다발 이상의 PWR 사용후핵연료 집합체를 원전부지 내에 수송, 저장하였다. 이를 위하여 KN-12 운반용기, 관련장비 및 수송차량으로 구성되는 수송시스템을 구성하였다. KN-12 운반용기는 국내 원자력법 및 IAEA의 수송규정에 따라 설계, 제작되고, 정부로부터 인허가를 획득하였으며, 취급장비 역시 관련규정에 따라 구비하였다. 수송 저장작업은 2 대의 운반용기를 동시에 투입하여 수행하였으며, 모든 작업공정에 대하여 엄격한 품질관리 및 방사선 안전관리를 수행하여 수송 안전성을 확보하고 신뢰도를 제고하였다.
