        More Roller-compacted concrete (RCC) is a dry concrete consisted of same materials as conventional concrete with different proportioning which requires compaction effort in order to reach its final form. Thus, both hydration and aggregate interlock play important roles in its strength augmentation. Flexural strength, an important factor in pavement design and fatigue cracking resistance, can be difficult to be obtained at in-situ and may be subjected to high variability. Even though its compressive strength is relatively high compared to conventional concrete with similar binder content, the relationship between compressive strength and flexural or tensile strength were not well defined. The goal of this research is to compare the relationship between compressive strength and flexural strength as well as the relationship between compressive strength and splitting tensile strength of RCC with those of conventional concrete using various equations suggested in other researches and also to determine new regression equations for estimating RCC’s flexural and splitting tensile strength. The positive result of RCC’s flexural strength was found; it was higher than majority of predicted values from conventional concrete for the same compressive strength. In contrast, RCC’s splitting tensile strength was relatively low compared to that of conventional concrete for the same compressive strength. Power equations were learned to be suitable for relationship between compressive and flexural strengths as well as relationship between compressive and splitting tensile strengths.

        Cryopreservation of boar semen is continually researched in reproductive technologies and genetic resource banking in breed conservation. For evaluating the boar semen quality, sperm motility (MOT) is an important parameter because the movement of spermatozoa indicates active metabolism, membrane integrity and fertilizing capacity. Various researches have been trying to improve the quality of semen post-thawed in boar. Recently, polymorphism (g.358A>T) of cluster-of-differentiation antigen 9 (CD9) gene reported to be significant association with MOT. Also, CD9 gene was expressed in the male germ line stem cells is crucial for sperm-egg fusion, and was therefore selected as candidate gene for boar semen. This study was conducted to evaluate the pig SNP (g.358A>T) of CD9 gene as a positional controlling for semen parameters of post-thawed boar semen. To results, the g.358A>T SNP of the CD9 gene was significantly associated with the traits such as MOT, curve linear velocity, straight line velocity, average path velocity and amplitude of lateral head displacement. Particularly, the g.358A>T SNP significantly has the highest association with MOT and animals with AA genotype (p<0.001). Therefore, we suggest that the g.358A>T in the intron 6 region of the porcine CD9 may be used as a molecular marker for Duroc boar Post-thawed semen quality, although its functional effect was not defined yet.

        Porcine parvovirus (PPV), a member of the genus Parvovirus, family Parvoviridae, is a significant causative agent in porcine reproductive failure, causing serious economic losses in the swine industry. PPV is a non-enveloped virus and its capsid is assembled from three viral proteins (VP1, VP2, and VP3). The major capsid protein, VP2 is the main target for neutralizing antibodies in PPV. When VP2 was expressed in large amounts, it assembled into virus-like particles (VLPs) similar in size and morphology to the original virions. In this study, we generated the recombinant Bombyx mori nucleopolyhedrovirus (BmNPV) to express the VP2 protein. Expression of the VP2 protein was analyzed by SDS-PAGE and Western blot. The recombinant VP2 protein of approximately 64 kDa was detected by both analyses. The formation of VLP by recombinant VP2 was confirmed through transmission electron microscopy examination. The purified VP2 protein assembled into spherical particles with diameters ranging from 20 to 22 nm.

        Biological properties of antimicrobial peptides (AMPs) of hemimetabolous insect are poorly characterized in innate immunity field. To investigate the biochemical properties of hemimetabolous insect’s AMPs, we purified the pyrrhocoricin-like AMP from the hemolymph of Riptortus pedestris and then named as riptocin. We successfully determined the primary protein structure and its cDNA sequence. Interestingly, the determined cDNA revealed that riptocin precursor is composed of 12 repeating units of active riptocins, which implied that riptocin precursor might require to be processed to generate active riptocins by several unidentified processing enzymes. In order to characterize the bio-processing mechanisms of riptocin precursor, we generated the antibody against active riptocin. Using quantitative PCR and Western blot analyses, we showed that gene of riptocin was started to express from the fatbody after three hours post bacterial infection. To address our hypothesis that active riptocin is generated from riptocin precursor by several processing enzymes, we need to obtain the riptocin precursor. Currently, we are expressing the recombinant riptocin precursor using in vitro translation system. Meanwhile, we investigated whether naive hemolymph (naive HL), which may contain precursor riptocin, can generate active riptocin when riptocin precursor was co-incubation with bacteria-challenged hemolymph (active HL), which may contain all processing enzymes. Actually, when naive HL was incubated with active HL, antimicrobial activity was dramatically increased, suggesting that processing enzymes in active HL may induce processing of riptocin precursor to generate active riptocins.

        Insect pollinators of the endanger orchid Cypripedium japonicum were surveyed and identified during two years, as a part of a conservation project of the orchid at Jukyeup-san and Hwaak-san (Mt.), South Korea. In total 40 individuals of 16 species in 4 families were identified. The dominant family was Halictidae, and Lasioglossum exiliceps Vachal visited the most frequently C. japonicum during the surveys. The average visiting frequency was 2.5 individuals per hour and the highest 4.3, from 12:00 – 13:00 in a day. After 15:00 insects did not visit the flowers at all. However, all of the visiting insects were found to not carry a pollinium or pollens of the orchard on their bodies; pollen carryover by any of the visiting insects did not occur at all. The orchid seems to require certain pollinators in particular body thickness due to its unique pollination mechanism. The orchid has two exit route openings, around 1 cm in diametre, where the entrapped insects can exit and an anther is situated just in front of each opening. It was inferred that a pollen carrier should be around 1 cm in body thickness. Therefore, the candidate species as the proper pollen carriers can be Tetralonia nipponensis Perez, Xylocopa appendiculata circumvolans Smith and Bombus consobrinus Dahlbom among the surveyed visitors.

        In this experiment, a highly porous scaffold of biphasic calcium phosphate (BCP) was prepared using the spongereplica method. The BCP scaffold was coated with 58S bioactive glass (BG) and sintered for a second time. The resulting scaffold was coated with gelatin (Gel) and cross-linked with [3-(3-dimethyl aminopropyl) carbodiimide] and N-Hydroxysuccinamide (EDC-NHS). The initial average pore size of the scaffold ranged from 300 to 700μm, with more than 85 % porosity. The coating of BG and Gel had a significant effect on the scaffold-pore size, decreasing scaffold porosity while increasing mechanical strength. The material and surface properties were evaluated by means of several experiments involving scanning electron microscopy (SEM), energy-dispersive X-ray (EDX) and X-ray diffraction (XRD). Cytotoxicity was evaluated using MTT assay and confocal imaging of MC3T3-E1 pre-osteoblast cells cultured in vitro. Three types of scaffold (BCP, BCP-BG and BCP-BG-Gel) were implanted in a rat skull for in vivo evaluation. After 8 weeks of implantation, bone regeneration occurred in all three types of sample. Interestingly, regeneration was found to be greater (geometrically and physiologically) for neat BCP scaffolds than for two other kinds of composite scaffolds. However, the other two types of scaffolds were still better than the control (i.e., defect without treatment).

        The Riptortus-Burkholderia symbiosis is a newly emerging insect-bacterium symbiotic system. This symbiosis system has a good merit as an experimental model system to produce the non-symbiotic (apo) and symbiotic (sym) host insect. In recent reported papers, the symbionts play important biological roles for the host insects. Meanwhile, juvenile hormone (JH) is one of major hormone synthesized corpora allata(CA) to control many physiology of insect. However, the study for cross-talk mechanism between symbionts and host hormones to control important physiological phenomenon of insects is almost none. In this study, we found that Riptortus speed up adult emerging and increase egg laying on presence of symbiont Burkholderia. Also we found that hexamerin proteins, which were controlled the expression by JH, were accumulated in sym-Riptortus hemolymph compare with apo-Riptortus. According as combined results, we hypothesized that the gut symbiont Burkholderia can control JH titer to conclude out beneficial effects such as development and reproduction of R. pedestris. To verify this hypothesis, we examined measurement of JH titer, expression of hexamerins as JH response genes and RNAi for hexamerin protein during whole Riptortus life on presence or absence of symbiont Burkholderia. All results demonstrated that gut symbiont controlled JH titer of Riptortus. Controlled JH amount by symbiont Burkholderia in host midgut regulated hexamerin protein expression for speeding up adult emerging and increasing egg production.

        The Riptortus (stinkbug) has a specialized symbiotic organ, M4 midgut, to harboring symbiont Burkholderia. M4 midgut is located in abdomen and surrounded with insect hemolymph. Recently our group demonstrated that symbiotic Burkholderia showed different physiology after adapting in M4 gut compare with in vitro cultured Burkholderia. And population of symbiotic Burkholderia in the M4 midgut is regulated by special organ. However, the molecular mechanism to prevent spreading and migrating symbiont bacteria to other host tissues from symbiotic organ is not clear. Therefore, we assumed that symbiont Burkholderia are susceptible to host humoral immunity after established infection in M4 midgut to prevent spreading and migrating into the other host tissues through Riptortus hemolymph. To prove this assuming, we tested the susceptibility and survival rate of symbiont Burkholderia in hemolymph of Riptortus in vitro and in vivo. We also examined the susceptibility of symbiont Burkholderia using purified antimicrobial peptides (AMP), pyrrhocoricin-like, thanatin-like and defensin-like AMPs. Finally, we tested inducing ability for AMPs by systemic infection of symbiotic Burkholderia. Gene expression of purified AMPs was not different after systemic infection of both symbiont and in vitro cultured Burkholderia. Surprisingly, in vitro cultured Burkholderia resisted on bacteria injected hemolymph and purified AMPs but symbiont Burkholderia were highly susceptible in bacteria injected hemolymph and purified AMP. These results suggest that symbiont Burkholderia can't survive in the hemolymph after escaping symbiotic organ. Moreover, humoral immunity of host Riptortus is important to prevent spreading and migrating symbiont Burkholderia into the other host tissue or organ from symbiotic organ.

        Insect constitute the largest and most diverse group of animals on world and also serve as the hosts or nutrient sources. In addition, several insects have a strong influence on people's emotion. To utilize the preference and interest of insects in the field of mental healthcare, a survey study was conducted with individual living in Korea. As results, the most people had a high preference and interest of insect, but some were disagreeable to the insect itself. The preference and interest of insect were high on male, adult and practician experienced insect-related events than female, student and non-practician, respectively. The most favored insects were familiar or pet insects such as Papilio xuthus, Lucanus maculifemoratus, Allomyrina dichotoma and Lampyridae. These results may be useful to develop a healing program for mental healthcare using insects. Further research is needed to determine the effects of these insect in the mental therapy for this purpose.

        Polyhedrin is the major component of the nuclear viral occlusions produced during replication of the baculovirus Autographa californica multicapsid nucleopolyhedrovirus (AcMNPV). To enhance the production efficiency of foreign protein in baculovirus expression system, the effects of various polyhedrin fragments were investigated by fusion expressing them with the enhanced green fluorescent protein (EGFP). Recombinant viruses were generated to express EGFP fused with polyhedrin fragments based on the previously reported minimal region for self-assembly and the KRKK nuclear localization signal (NLS). The marked increase of EGFP by these fusion expressions was confirmed through protein and fluorescence intensity analyses. Among the fusion-expressed protein in nucleus and cytoplasm, the most hyper-expression was observed in the fusion of amino acids 19 to 110 and 32 to 59 of polyhedrin. Also these fragments, some degradation of only the fused polyhedrin was observed in the fusion of amino acids 19 to 85 and 32 to 85. The production of E2 protein, which is a major antigen of classical swine fever virus, was dramatically increased by fusion expression with polyhedrin amino acids 19 to 110, and its preliminary immunogenicity was verified using experimental guinea pigs. The production of luciferase was approximately two folds increased by fusion expression with polyhedrin amino acids 32 to 59, and its activity was measured using Luminometer. This study suggests a new option for higher expression of useful foreign recombinant protein using the partial polyhedrin fusion expression in baculovirus.

        Grapevines are cultivated globally, and are also damaged by a large number of insect pests especially including leafhoppers. Leafhoppers are one of the important insect pests on various crops including grape. Among them, the genus Arboridia Zachvatkin (Hemiptera: Auchenorrhyncha: Cicadellidae: Typhlocybinae) was known as insect pests against grape farming. Eighty species were described in the genus in the world, and eight species were recorded in the Korean Peninsula so far, which are: Arboridia apicalis, A. kakogawana, A. koreacola, A. maculifrons, A. nigrigena, A. okamotonis, A. silvarum, A. suzukii. Among them, A. apicalis, A. kakogawana, A. maculifrons, A. nigrigena, A. suzukii have been known as important insect pests on grapes. In this talk, we propose a new species from the Korean Peninsula, and confirmed two new records: A. agrillacea and A. lunula. Additionally we present differential diagnoses of the Arboridia species damaging on grapevines for the field of applied biology.

        In this study, the synthesis of nickel nanoparticles and copper nanospheres for the potential applications of MLCC electrode materials has been studied by plasma arc evaporation method. The change in the broad distribution of the size of nickel and copper nanopowders is successfully controlled by manifesting proper mixture of gas ambiance for plasma generation in the size range of 20 to 200 nm in diameter. The factors affecting the mean diameter of the nanopowder was studied by changing the composition of reactive gases, indicating that nitrogen enhances the formation of larger particles compared to hydrogen gas. The morphologies and particle sizes of the metal nanoparticles were observed by SEM, and ultrathin oxide layers on the powder surface generated during passivation step have been confirmed using TEM. The metallic FCC structure of the nanoparticles was confirmed using powder X-ray diffraction method.

        Flexible transparent conducting films (TCFs) were fabricated by dip-coating single-wall carbon nanotubes (SWCNTs) onto a flexible polyethylene terephthalate (PET) film. The amount of coated SWCNTs was controlled simply by dipping number. Because the performance of SWCNT-based TCFs is influenced by both electrical conductance and optical transmittance, we evaluated the film performance by introducing a film property factor using both the number of interconnected SWCNT bundles at intersection points, and the coverage of SWCNTs on the PET substrate, in field emission scanning electron microscopic images. The microscopic film property factor was in an excellent agreement with the macroscopic one determined from electrical conductance and optical transmittance measurements, especially for a small number of dippings. Therefore, the most crucial factor governing the performance of the SWCNT-based TCFs is a SWCNT-network structure with a large number of intersection points for a minimum amount of deposited SWCNTs.

        The purpose of this study is to identify the effective lumbar area exercise program to prevent falls of the elderly by dividing 14 female elders over 65 living in Daegu into two groups such as resistance exercise group and resistance & stabilization exercise group, and applying a 60 minute exercise program three times a week for 12 weeks. The followings are the results. First, both of resistance exercise and resistance & stabilization exercise brought improvement of lumbar strength and there was no significant difference between two groups. Second, there was no significant difference in lower limb strength in resistance exercise group but resistance & stabilization exercise group showed significant increase in lower limb flexor strength. To make a summary of the above results, it can be said that 12 weeks’ resistance & stabilization exercise is effective to the lumbar strength and lower limb strength of the elderly. Accordingly, if combining resistance exercise and stabilization exercise appropriately, we can build a successful preventive program which even the elderly who cannot perform resistance exercise easily can follow.

        The purpose of this study is to identify the bridge exercise posture for the efficient exercise by comparing the muscle activity of the lower limbs according to the changes in muscle length because of knee angle in bridge exercise. The subjects of this study were 9 randomly selected males in their 20s living in D city from those who satisfied inclusion criteria. The measured muscles were Vastus medialis oblique, Vastus lateralis, Semitendinosus, Biceps femoris, Gluteus maximus, Gluteus medius, Tensor faciae latae, and Adductor longus. Data were analyzed through paired comparison test. In the result, ST, BF, and TFL muscle activities were high when knee joint flexion angle was 90°Although in most cases higher muscle activity was shown at 90°than 60°there was no statistical significance. Interestingly, it was lower at 90°than 60°in VL. In ST, BF, and TFL, it was significantly higher at 90°han 60°(p<.05). Conclusively, knee angles in bridge exercise may affect the muscle activity, and in particular when the activity of two joint muscles such as semimenbranosus muscle, biceps femoris muscle, and tensor fasciae latae muscle increase as the angle gets higher. Therefore, it is considered that this study will provide helpful tips to develop muscular strength enforcement program for the patients with damages in the lower limbs through bridge exercise in clinical situations.

        The insecticidal activity of 120 plant essential oils and control efficacy of six experimental spray formulations (SF) containing the oils (SF-0.25, 0.5, 1, 2.5, 5 and 10% sprays) against both nymph and adult of citrus flatid planthopper, Metcalfa pruinosa, was examined using a direct contact application. Reponses varied according to dose (1000 mg litre-1 and 500 mg litre-1). Based on 24 h exposure in leaf dipping assay at 1000 mg litre-1 strong mortality was observed in more than 19 essential oils among 130 was screened. At 500 mg litre-1 very strong mortality (100%) was observed in cinnamon technical, cinnamon green leaf, cinnamon #500, cassia tree, citronella java and penny royal followed by origanum, thyme white, grape fruit, savory, fennel sweet, aniseed and cinnamon bark (93.3- 80%) showed considerable moratality against nymphs of M. pruinosa. The moderate mortality was found in thyme red, tagette, calamus, lemoneucalptus and geranium (73.3-60%). The other oil has low or very low mortality against M. pruinosa. The oil applied as SF-10% sprays provided 100% mortality against adult M. pruinosa. In particular cinnamon technical showed very strong (>SF-0.5= 100%) effect followed by cinnamon #500 (>SF-2.5=100%), cinnamon green leaf (>SF-2.5=100%) and penny royal (>SF-2.5=100%) respectively. Global efforts to reduce the level of highly toxic synthetic insecticides in the agricultural environment justify further studies on the active essential oils as potential larvicides for the control of M. pruinosa populations as direct spray with contact action.
