        베트남 메콩 델타 지역은 베트남에서도 쌀 생산이 활발히 이루어지는 지역으로 꼽히고 있으나 기후온난화에 따른 고온 현상 증가로 쌀생산량 감소의 우려가 증대되고 있다. 이에 고온에 따른 벼 피해발생 현황 및 대응 기술에 대한 정보를 제공하고자 한다. 베트남의 평균기온은 2060년 까지 약 2.7oC 증가, 35oC 이상의 폭염은 2030년까지 2달 이상 길어질 것으로 예측되고 있으며 기온상승에 따라 기온이 높은 작기의 벼 생산량이 감소되고 있다. 벼 개화기 온도가 33oC 이상으로 높을 시, 화분의 감수분열에 영향을 미쳐 화분의 활력이 저하되며 수술에서 약의 개열이 감소되어 화분의 방출이 감소될 뿐만 아니라 화분이 암술의 주두에 부착하여 발아 시 발아율을 낮추어 불임 발생이 증가하게 된다. 베트남의 벼 연구소는 고온에 대비하여 베트남 벼 생산의 안정화를 위해서 개화기에 내열성을 가지는 품종 육성 뿐만 아니라 이른 아침에 개화할 수 있는 품종을 육성하고 있다. 또한 재배 적인 측면에서 고온 피해를 줄일 수 있도록 벼 재배시기를 조절하여 기존 3기작에서 고온피해가 큰 시기의 재배를 제외한 2기작으로 재배하는 반면 파종 및 이앙시기를 조절하여 벼의 생육량을 충분히 확보하여 2기작에 따른 수량감소를 최소화하는 연구가 필요하다. 더불어 벼를 재배하지 못하는 무더운 계절에는 농업인의 소득보존을 위해 논을 이용한 물고기 양식 기술 도입하는 것도 필요하며, 시비 방법 등의 개선을 통한 벼 생육량 조절, 물관리 기술 개발을 통한 포장 미세기상변화 등의 연구가 필요할 것으로 판단된다.

        The purpose of this study was to investigate the effect of perceived risk, community usage motive, and price sensitivity of customers who purchase children’s wear directly from overseas on customer satisfaction. Specifically, this study is aimed at verifying the moderating effect of price sensitivity when perceived risk and community usage motive significantly influence customer satisfaction. A survey was conducted among consumers who experienced directly purchasing children’s wear from overseas and engaging in communities. A total of 415 questionnaires were distributed, which 41 responses were insincere and excluded; thus, 374 responses were analyzed. Results of this study are as follows. First, the analysis on how perceived risk and community usage motive influence customer satisfaction reveals the significant influence of economic and social psychological risk on customer satisfaction. Moreover, informational, recreational, and social emotional usage motive have a significant influence on customer satisfaction. Second, the effect of perceived risk and community usage motive on customer satisfaction based on different levels of price sensitivity was verified. As a result, delivery and economic risks affect customer satisfaction for consumers with high price sensitivity. Moreover, recreational usage motive has a significant influence on customer satisfaction for high price sensitive consumers. In conclusion, perceived risk negatively impacts customer satisfaction. Therefore, it is essential to provide a system that can reduce the perceived risk of consumers who purchase children’s wear directly from overseas.

        This study investigates the effects of the type of advertisement, brand extension, and consumer innovativeness on the credibility of fashion advertisements. The factorial design is constructed as a 2(type of fashion advertisement: general vs. cross-media) × 2(consumer innovativeness: high vs. low) × 2(the type of fashion brand extension: similar vs. dissimilar) three-way mixed design in which consumer innovativeness is the between-subjects variable. Subjects of this study included 210 men and women in their 20s and 30s who resid in Seoul and had SNS experiences. Frequency analysis, credibility analysis, three-way ANOVA, and simple interaction analysis were conducted using the SPSS 20.0 statistics package. The results are as follows: First, the type of fashion advertisement had a significant effect on advertising credibility. Cross-media advertisements had a more positive effect than general advertisements on all of the dependent variables. Also, the type of brand extension and consumer innovativeness showed significant effect on advertising credibility and the interaction effect between the type of fashion advertisement and consumer innovativeness was significant. Lastly, the effect of different types of brand extensions on advertising credibility showed a significant difference according to consumer innovativeness. Thus, a marketing strategy using cross-media advertising is proven to be effective in gaining consumer trust for a fashion brand.

        The dynastid beetle Allomyrina dichotoma has been used as a herbal medicine. Recently, we performed de novo RNAsequencing of Allomyrina dichotoma and identified several antimicrobial peptide candidates based on in silico analysis.Among them, cationic antimicrobial peptide, Allomyrinasin, was selected and we assessed the anti-inflammatory activitiesof Allomyrinasin against mouse macrophage Raw264.7 cells. The results showed that Allomyrinasin decreased the nitricoxide production of the lipopolysaccharide-induced Raw264.7 cells. In addition, quantitative RT-PCR, ELISA and Westernblot analysis revealed that Allomyrinasin reduced cytokine expression levels in the Raw264.7 cells. Taken together, thesedata indicated that Allomyrinasin had anti-inflammatory activity in the lipopolysaccharide-induced Raw264.7 cells.

        Although the grasshopper Oxya chinensis sinuosa has long been used as a food in Korea, there is little data on itsfunctional effects. Thus we prepared and analyzed total RNA from the whole body of adult Escherichia coli non-immunizedand immunized Oxya chinensis sinuosa strains. Using an Illumina Hiseq sequencer, we generated a total of 66,555 pooledtranscriptome and singletons with and without Escherichia coli immunization, respectively. Then, we performed in silicoanalysis of the Oxya chinensis sinuosa transcriptome, using bioinformatics tools for screening putative antimicrobial peptides(AMPs) and 38 AMPs were finally selected and tested their antimicrobial activity of Gram positive, Gram negative bacteriaand antifungal activity with radial diffusion assay. As a result, 5 out of 38 AMPs showed the highest antimicrobial activityand antifungal activity against microbes and it also evidenced with no hemolytic activity.

        It is very difficult to get the information about semen quality analysis in transgenic pigs because of limited numbers and research facilities. Therefore, in the present study, we analyzed the semen quality of transgenic boars generated for xenotransplantation research. Briefly, the semen samples were collected from 5 homozygous α1,3-Galactosyltransferase knock-out (GalT-/-) transgenic boars and immediately transported to the laboratory. These semen samples were decupled with DPBS and conducted to analyze semen parameters by a computer-assisted semen analysis (CASA) system. The boar semen were examined all 12 parameters such as total motility (TM), curvilinear velocity (VCL), straight line velocity (VSL), average path velocity (VAP), and hyperactivated (HYP), etc. In results, among the 5 GalT-/- boars, three boars (#134, 144, and 170) showed normal range of semen parameters, but #199 and 171 boars showed abnormal ranges of semen parameters according to standard ranges of semen parameters. Unfortunately, #171 boar showed azoospermia symptom with rare sperm counts in the original semen. Conclusively, assessment of semen parameters by CASA system is useful to pre-screening of reproductively healthy boar prior to natural mating and artificial insemination for multiplication and breeding.

        목 적 : 자기공명영상(MRI)의 발달은 소아신경계의 병리학적 과정을 이해하고 진단하는데 중요한 역할을 하고 있으며 그 검사 건수는 나날이 증가하고 있다. 하지만 이같은 장점에도 불구하고 소아의 Brain MR 검사는 환자에게 주의를 주거나 Sedation을 하여 검사를 진행하지만 움직임, 소음 등으로 인하여 실패하는 경우가 발생한다. 고해상력의 영상 획득이 가능한 3.0T의 경우 1.5T에 비해 SAR가 약 4배 증가하게 되며, 최근 미국 소아과학회에서는 어린이가 어른보다 전자파에 더 큰 영향을 받기 때문에 더욱 주의하여 관리해야 한다고 주장하고 있다. 이때, 기존의 TSE가 아닌 다양한 Refocusing FA를 사용하는 Hypere-TSE은 동등한 SNR을 유지하면서 SAR를 감소시키기 때문에 그 해결책이 될 수 있다. 또한, MR 검사 시의 소음은 Gradient의 빠른 Switching을 통해 발생하는데, 이러한 기존의 소음 감소를 위한 Gradient mode인 Whisper mode는 Scan time이 증가하는 등의 단점이 있었으나 최근 E11 Upgrade를 통해 도입된 Acoustic noise reduction는 이를 개선하여 동등한 Scan time과 향상 된 소음 감소 효과를 가져왔다. 이에 본 연구에서는 소아 Brain MRI 검사 시 Hyperecho와 Acoustic Noise Reduction을 이용하여 Image quality를 동등하게 유지하면서 소아를 더욱 안전하고 안정된 환경에서 검사할 수 있는 방법을 제안하고자 한다. 대상 및 방법 : 장비는 SIEMENS사의 3.0T SKYRA(Germany)와 32ch Head coil을 사용하였고, Spherical Phantom D165(Siemens, Germany) 및 만 15세 이하의 소아 환자 20명(남자 12, 여자 8, 평균연령 6세)을 대상으로 하였다. Hyper-TSE와 Acoustic noise reduction을 사용했을 때와 사용하지 않았을 때의 T1 Dark fluid(TR: 2000, TE: 9, TI: 900, Matrix: 384*307, FOV: 200*200, NEX: 1) T2WI(TR: 6000 TE: 103, Matrix: 448*269, FOV: 200*200, NEX: 2), T2 FLAIR(TR: 9000, TE: 84, TI: 2500, Matrix: 320*192, FOV: 200*200, NEX: 1)에서 비에스코리아사의 디지털 소음 측정기를 통하여 Isocenter에서 150㎝ 거리에서의 소음을 측정하였고, SAR와 SNR은 Siemens NUMARIS/4 - syngo MR E11로 Gray matter, White matter, Eye ball을 관심 영역 0.02㎠ 측정하여 IBM SPSS Statistics v22.0을 통해 정량적 분석을 하였다. 결 과 : 정량분석 결과 Hyper-TSE와 Acoustic noise reduction을 사용했을 때는 사용하지 않았을 때에 비해 SAR가 T1 Dark fulid에서 -26%, T2에서 -33%, T2 FLAIR에서 –33%로 감소되었으며, 소음은 T1 Dark fulid에서 -14%, T2에서 -16.6%, T2 FLAIR에서 –16.1% 감소하였고, SNR은 T1 Dark fulid에서 GM: 17%, WM: 15%, Eye ball: 12%, T2에서 GM: 10%, WM: 14%, Eye ball: 14%, T2 FLAIR에서 GM: 3%, WM: 9%, Eye ball: 7%로 증가한 것으로 나타났다. CNR은 T1 Dark fulid에서 14%, T2에서 3%, T2 FLAIR에서 –19%로 증감된 것으로 나타났다. 비모수 대응표본 Wilcoxon T검정을 실시한 결과 대부분의 값에서 유의한 차이(p<.05)가 있는 것으로 나타났다. 결 론 : Hyperecho를 사용하게 되면 SNR을 유지하면서 SAR를 상당히 감소시킬 수 있다는 것과 Acoustic noise reduction을 사용할 경우 동일하거나 약간의 검사 시간 증가와 함께 많이 조용하게 된다는 이론적 사실을 바탕으로 안전하고 안정된 환경을 만들기 위해 노력하였다. 이번 연구를 통해 Hyperecho 및 Acoustic noise reduction을 사용할 경우 기존의 Sequence와 비교하여 SAR와 소음은 감소시키면서 SNR과 CNR 값은 동등하거나 증가된 영상을 얻을 수 있고 이를 통하여 소아 환자에게 더욱 안전하고 안정된 환경을 제공하고 효과적이며 진단 가치 높은 영상을 얻을 수 있으리라 사료된다.

        RNA Sendai virus (SeV) vector system has no risk of being integrated into the host genome. Sendai virus (SeV) vectors expressing pluripotent factors have been used to produce integration-free induced pluripotent stem cells (iPSCs) with high efficiency from various cell types in human and mouse. In this study, we generated iPSCs from pig ear fibroblast cells using the SeV vector expressing 4 human factors (POU5F1, SOX2, C-MYC, and KLF4). Colonies were emerged at Day 14 of transduction and expressed the classical pluripotency markers (POU5F1, NANOG, and SOX2) and surface marker (SSEA1). Furthermore, they showed a domed shape and could passage over 40 times under 2i (CHIR99021 and PD0325901)-LIF and MEF feeder culture condition having in vitro differentiation ability into 3 germ layers. Next, we examined the ability of six feeder free culture conditions to maintain piPSCs in a pluripotent state. piPSCs were plated on Matrigel coated dishes in different media: 1. CM: control media (LIF culture media); 2. CM-F: CM+100 ng Fetuin-A; 3. CM-N: CM+100 ng Nanog-TAT; 4. CM-2i: CM+3 uM CHIR99021+1 uM PD0325901; 5. CM-2iN: CM-2i+100 ng Nanog-TAT; 6. CM-2iN+100 ng Fetuin-A. However, piPSC could not maintain the typical self-renewal morphology on feeder free conditions regardless of culture media tested here. Further, expression of pluripotency-related genes (Oct4, Nanog and Klf4) of piPSCs cultured on feeder free conditions could not be compared with that of iPSCs cultured on MEF feeder plate. Our results suggest that integration free pluripotent stem cell from pigs could be generated by SeV vector system and maintained their pluripotency under 2i-LIF and MEF feeder culture condition, but further optimization of culture conditions may be required.

        The present study was conducted to investigate the effect of caffeine and theophilline on sperm motility during in vitro fertilization (IVF). Briefly, commercial boar semen was centrifuged and resuspended (5x107 sperms/ml) with fertilization medium (mTBM) supplemented with 2 mM caffeine (Caf), 5 mM theophylline (The), 2 mM caffeine + 5mM theophylline (Caf + The), and not supplemented control. The semen parameters of the four groups were analyzed by computer-assisted semen analysis (CASA, Medical Supply Co. Ltd) system at 6 h as time for IVF at 38.5 C under 5 % CO2 in air. The groups were examined 11 semen parameters such as total motility (TM), curvilinier velocity (VCL), straight-line velocity (VSL), average-path velocity (VAP), and hyperactivated (HYP), etc. A total motility of control group (41.3 %) at 6 h showed significantly (P<0.05) higher than those of other groups (Caf, 37.2; The, 35.2; Caf + The, 30.1 %). Results from many other sperm parameters indicated that the theophylline and caffeine negatively affected on sperm motility during IVF. These results suggested that the supplementation of caffeine and/or theophylline was not essential for IVF in pigs. To prove this suggestion, further studies are needed to analyze fertility and embryonic development after IVF with or without the supplementation of caffeine and/or theophylline.

        Although piglets have been delivered by embryo transfer (ET) with in vitro produced (IVP) embryos and blastocysts, a success rate has still remained lower level. Unlike mouse, human, and bovine, it is difficult to a production of piglets by in vitro fertilization (IVF) because of an inappropriate in vitro culture (IVC) system in pig. Therefore, the present study was conducted to investigate whether minimized exposure time in IVC can improve the pregnancy and delivery rates of piglets. Immediately after IVM, the oocytes were denuded and co-incubated with freshly ejaculated boar semen for 3.5 to 4 hours at 38.5 ˚C under 5% CO2 in air. To avoid long-term exposure to in vitro state, we emitted IVC step after IVF. After that the presumptive zygotes were transferred into both oviducts of the surrogate on the same day or 1 day after the onset of estrus. Pregnancy was diagnosed on day 28 after ET and then was checked regularly every month by ultrasound examination. The 3 out of 4 surrogates were determined as pregnant (75%) and a total of 5 piglets (2 females and 3 males) were delivered at 118.3 ± 2.5 days of pregnancy period. In conclusion, a short-term exposure time may be an important factor in the production of IVP-derived piglets. It can be apply to the in vitro production system of transgenic pig by IVF, cloning, and pronuclear microinjection methods.

        To overcome the hyperacute immune rejection during pig-to-non-human primates xenotranasplantation, we have produced and bred α-1,3-galactosyltransferase knock-out (GalT —/—) pigs. In this study, the somatic cells and tissues from the GalT —/— pigs were characterized by an analysis of the expression of Galα-1,3-Gal (α-Gal) epitope. Briefly, ear fibroblast cell lines of 19 homozygous GalT —/— pigs were established and cryopreserved. The expression of α-Gal epitope in the cells was measured by fluorescence activated cell sorter (FACS) analysis using BS-I-B4 lectin. Also, the homozygous (GalT —/—) cells and tissues samples were immunostained with BS-I-B4 lectin for analysis of α-Gal epitope expression. The results showed that the expression of α-Gal epitope in GalT —/— cells (0.2 %) were significantly (p< 0.05) down-regulated to the range of cynomolgus monkey fibroblast (0.2 %) cells compared to heterozygous (GalT —/+) (9.3 %) and wild type (GalT +/+) (93.7 %) fibroblast cells. In the immunostaining results, while the expression of α-Gal epitope was detected a partly in GalT —/+ cells and mostly in GalT +/+ cells, it was almost not detected in the GalT —/— cells. Also, immunostaining results from various tissues of the GalT —/— pig showed that the expression of α-Gal epitope was not detectable, whereas various tissues from GalT +/+ pig showed a strong expression of α-Gal epitope. Our results demonstrated that α-Gal epitope expressions from GalT —/— pigs were successfully knocked out to prevent hyperacute immune rejection for further study of xenotransplantation.

        This study compared the effects of the initial head position (i.e., a HHP versus a relaxed head position) of subjects with and without a FHP on the thickness of the deep and superficial neck flexor muscles during CCF. The study recruited 6 subjects with a FHP and 10 subjects without a FHP. The subjects performed CCF in two different head positions: a HHP, with the head aligned so that the forehead and chin formed a horizontal line, and a relaxed head position (RHP), with the head aligned in a self-selected comfortable position. During the CCF exercise, the thickness of the longus colli (LCo) and the thickness of the sternocleidomastoid (SCM) were recorded using ultrasonography. The thickness of each muscle was measured by Image J software. The statistical analysis was performed with a two-way mixed-model analysis of variance. The thickness of the SCM differed significantly (p<.05) between the subjects with and without FHP. According to a post hoc independent t-test, the change in thickness of the SCM increased significantly during CCF in the subjects with FHP while adopting a HHP compared to that in the subjects without FHP. The change in thickness of the SCM was not significantly different between the two positions in subjects without FHP, and there was no significant change in thickness of the LCo muscle during the CCF exercise according to the initial position in both subjects with and without FHP. The results suggest that CCF should be performed in RHP to minimize contraction of the SCM in subjects with a FHP.
