        Recent research on stem cell conditioned medium (CM) has been revealed that CM could influence on the embryo development when supplemented to in vitro culture medium. However, the optimal basal medium for CM production has not determined although it is the fundamental factor of CM. The purpose of this study is to examine the effect of human derived adipose stem cell CM (hASC-CM) with different basal medium on mice embryo development after parthenogenetic activation (PA). hASC-CM was collected from 2 kinds of serum free basal medium, DMEM and KSFM, respectively on day 5 from the culture of hASC isolated from human fat tissue. Intra-peritoneal injection of PMSG and hCG was conducted into 7-week-old ICR mice for superovulation. The oocytes were recovered from the oviductal ampulla, 18 h after hCG injection, and denuded using 0.1% hyaluronidase. PA of oocytes was conducted with KSOM media including strontium chloride. The parthenotes were in vitro cultured in 3 groups: 100% KSOM (Control), 75% KSOM + 25% DMEM or KSFM without FBS (DMEM or KSFM group) and 75% KSOM + 25% hASC-CM from DMEM or KSFM (DMEM-CM or KSFM-CM group). Cleavage rate was assessed after 2 days post IVC and blastocyst formation rate was evaluated after 6 days post IVC both using stereomicroscope. Total cell number of blastocysts was counted by Hoechst staining. 1way ANOVA from Graphpad prism 5 was used for statistical analysis and the values are presented as means ± standard error of mean. As a result, blastocyst formation rate of DMEM-CM group (16.09±3.32%, P<0.05) was significantly lower than control and DMEM group (34.43±2.89% and 34.49±5.34%, P<0.05) but cleavage rate and total cell number of blastocysts showed no significant difference among groups. In case of KSFM, there was no significant difference in cleavage rate, blastocyst formation rate and total cell number of blastocysts among the control, KSFM group and KSFM-CM group. The sort of basal medium used for the CM collection affected the development of parthenotes during in vitro culture differently. Therefore, further research should be conducted to find out the alternative basal medium of CM able to improve the embryo development.         국내에서 재배하는 주요 핵과류(복숭아, 자두, 매실, 체리)에 발생하는 깍지벌레는 뽕나무깍지벌레(Pseudaulacaspis pentagona), 말채나무공깍지벌레(Lecanium corni), 무화과깍지벌레(Coccus hesperidum), 가루깍지벌레류(Pseudococcus sp.) 4종이 발견되었으나, 뽕나무깍지 벌레는 2017년에 총 113과원 중 103과원(발생과원율 91.2%), 2018년에는 77과원 중 64과원(발생과원율 83.1%) 모든 핵과류에서 가장 많이 발생하여 우점종으로 확인되었다. 2017년도에는 5월 상순부터 부화를 시작하여 암컷성충 1마리가 평균 75.5개(47∼159개)의 알을 낳으며 모든 알이 부화하는데 약 19일이 소요되었으며 5월 20일경에 월동성충의 모든 알이 부화하였다. 그러나, 2018년에는 4월 하순부터 부화를 시작하여 (부화율 72%) 부화유충(1령)으로 이동 후 5월 중순부터 고착유충(2~3령), 6월 중순부터 2세대 성충이 활동하기 시작하였다. 부화유충기(1령)에 약제 살포를 하면 살충율이 100.0%이었으나, 고착유충기(2~3령)에는 살충율은 2.7%에 불과하였다. 부화약충기를 제외한 모든 단계에서 몸체가 밀납깍지로 덮여 있어 약제를 살포하여도 직접 접촉되지 않아 치사율이 낮았다. 따라서 반드시 부화약충기에 약제를 살포하여야 방제효과를 높일 수 있다.

        목적: 안경광학과 전공 학생과 전문가를 대상으로 스마트 기반 국가시험에 대한 인식에 대하여 알아보고자 하였다. 방법: 안경광학 전공 학생, 교수, 임상전문가(안경사)로 구성된 171명을 대상으로 지필평가방식과 스마트 기반 멀티미디어 문항이 포함된 평가방식(SBT)에 대한 인식을 조사하기 위하여 설문조사를 실시하였다. 결과: 안경관련 전문가 집단의 SBT에 대한 인식은 다른 보건의료직종의 전문가 집단보다 낮은 것으로 나 타났지만, 안경사국가시험에서 지필평가와 SBT 평가 두 시험 체제는 차이가 없을 것이라는 의견이 44.4%, SBT 평가체제가 더 좋다는 의견이 37.4%로 지필평가(31.0%) 보다 높게 나타났다. 스마트 기반 시험의 가장 긍정적인 측면은 시험에 소요되는 시간 단축을 최대 장점으로 인식하는 것으로 나타났으며, 멀티미디어 문항의 긍정적인 측면은 임상현장에서의 수행능력을 효과적으로 평가할 수 있다는 것으로 나타났다. 또한, 교수집단의 경우 응시자의 임상수행능력을 보다 정확하게 평가할 수 있다, 졸업 후 임상현장에서 업무를 수행 하는 과정에 도움이 된다, 수행평가가 용이하다는 점에서 임상전문가보다 더 긍정적으로 반응하였다. 결론: 지필시험에서 사용하고 있는 선택형 시험문항은 문제해결능력보다는 직무를 수행하는데 필요한 관련 지식을 기억하거나 이해하고 있는지를 주로 평가하고 있어, 해당 직무의 실제성 또는 현실성을 반영하는 시험이 되기 위해서는 본 연구결과를 반영한 SBT 체제로의 전환이 필요한 것으로 나타났고, 이를 위한 타당성 연구가 지속적으로 이루어져야 할 것이다.

        The objective of this study was to investigate the effects of freeze dried placenta supplementation on reproductive performance, colostrum and plasma biochemical composition in pregnant sows. Eleven Landrace × Large white sows were fed with corn-soybean meal diets supplemented with or without 1% freeze dried placenta powder from 10 days before their expected farrowing dates until 10 days postpartum. The colostrum protein content was significantly higher(P=0.043) in the treatment group than in the control group. Compared to the control group, the immunoglobulin G(IgG) concentration in the colostrum was significantly higher(P=0.004) in the treatment. In day 25 piglets plasma, the IgG concentration was higher(P=0.184) in the treatment than the control. The mortality rate was lower(P=0.102), and the piglet weight gain was higher(P=0.35) in the treated group. Overall, the treatment group showed greater levels of protein and IgG concentration in the colostrum, when compared to control group. Therefore, the freeze dried placenta supplementation on pregnant sows can enhance its colostrum composition, hence decrease the mortality and increase the growth rate of piglets.

        West Nile Virus (WNV) is transmitted by infected mosquitoes. Vector mosquitoes usually acquire these pathogens fromfeeding on an infected host, and transmit the pathogens to a naive host during feeding events. To understand the virustransmission dynamics and to survey WNV throughout country, the present study has been conducted. We collected mosquitoesat urban parks in Seongnam, Wonju, Gunsan, Daegu, and Tongyeong using CDC light trap with Dry ice from April toSeptemper in 2017 (mosquito collecting is on going). Among collected mosquitoes, blood-fed mosquitoes were conductedblood meal identification assay and the other mosquitoes were subjected to virus detection using real-time PCR method.A total of 2,290 mosquitoes representing 6 genera and 15 species were collected. The most dominant species was Culexpipiens complex (42.1%) followed by Aedes albopictus (15.1%), Ae. vexans nipponii (14.6%), Ochlerotatus koreicus (9.8%),Cx. orientalis (6.5%), and Armigeres subalbatus (4.4%). The blood meal source were of mammal (93.3%), and birds (6.7%).So far, no WNV has been detected in any mosquitoes.

        The members of the genus Flavivirus are noteworthy, as they cause infectious diseases in humans, such as Zika, denguefever, yellow fever, West Nile, and Japanese encephalitis. Due to the increased awareness of the public health risk posedby flavivirus-infected mosquitoes, mosquito collections were performed in six urban parks of South Korea, as the parksare designated for human recreation but also provide suitable habitats for mosquitoes. We examined the diversity andabundance of mosquito species and conducted molecular diagnostics for the detection of flavivirus infections. Monthlycollections were carried out in each park from March to August in 2017. A total of 4,851 mosquitoes (5 genera and13 species) were collected using BG-sentinel traps and then investigated for flavivirus infections. Pathogenic flavivirusinfections causing human diseases were not observed in the field-collected mosquitoes. However, insect-specific flavivirus(ISF) infections were detected in several mosquito pools. ISF has been previously known to enhance or suppress the replicationof medically important flaviviruses in co-infected mosquito cells. In this study, partial sequences of ISF were analyzed.However, further studies are needed in order to determine its genetic characterization and biological function in vivo.

        Drought is one of the detrimental factors that impair plant growth and productivity. In this study, we applied annealing control primer (ACP)-based reverse transcriptase PCR (polymerase chain reaction) technique to identify differentially expressed genes (DEGs) in maize leaves in response to drought stress. Two-week-old maize seedlings were exposed to drought (DT) by suspending water supply. DEGs were screened after 3 days of DT-treated samples using the ACP-based technique. Several DEGs encoding 16.9 protein, antimicrobial protein, hypothetical protein NCLIV_068840, thioredoxin M-type were identified in maize leaves under drought stress. These genes have putative functions in plant defense response, growth and development. These identified genes would be useful for predictive markers of plant defense, and growth responses under drought stress in plants.

        Aedes albopictus is one species of mosquito transmitting flavivirus causing Dengue, Zika, and West Nile fever. Although it is an important disease vector, the genetic study of Ae. alpopictus populations has not been undertaken yet in South Korea. Here, we investigated the genetic variation of 99 Ae. albopictus individuals collected from 29 sites in nine provinces in 2016, through mitochondrial COI gene analysis. Haplotype analyses revealed seven haplogroups in South Korea. The main haplogroup, comprising 76 individuals (77.8%), was genetically identical to the one from Nagasaki. Two groups from Jeju Island (11) and the southern coast of South Korea (nine) were closely related to different Ae. albopictus strains from Kumamoto and Guangdong/Fujian, respectively. However, the others (four) were distinct from these two countries. No geographic divisions of populations were found in the study regions. The results suggest the possibility that the currently prevalent Ae. albopictus in South Korea, represents a part of the descendants that originated from nearby countries. However, more comprehensive investigations are needed to explain its movement routes.

        Dengue is the most important arboviral disease in tropical and subtropical areas where 2.5 billion people are at risk of infection. Aedes albopictus will be a major vector transmitting dengue virus in Korea, where this virus overseas inflow is possible. We collected mosquitoes in Jeju, Busan, Gunsan, and Incheon using BG sentinel trap from April to October in 2016. Collected mosquitoes were conducted virus detection using real-time PCR method and analysis bloodmeal source of Ae. albopictus. A total of 15 species comprising 7 genera were identified and 4,854 female mosquitoes collected. The most dominant species ratio (SR) was 52.9% (Culex pippins complex) followed by 20.3% (Ae. albopictus), 10.8% (Ae. vexans nipponi) and Ochlerotatus dorsalis (3.8%). Dengue virus was not detected. Bloodmeal source of Ae. albopictus was mammals (80.9%) followed by birds (18.6) and amphibians (0.5).

        We performed a survey for flavivirus infection and distribution of Aedes albopictus that known as Zika and Dengue virus vector using black–light trap and BG-sentinel trap around urban area in Korea. Mosquitoes were collected in 27 cities during March to November (twice a month) year 2016. Total numbers of mosquitoes collected 102,102 including 19 species 8 genera during collecting period. Total 21,467 Ae. albopictus was collected that 20,961(24.3%) by BG-sentinel trap and 506 (3.2%) by Black-light trap in urban area. Trap index(trap/night) of Ae. albopictus was showed highest in Hamyang (TI:992.3) and lowest in Taebaek (TI:0.3) there was only collected by Black-light trap. A total of 894 pools from all collecting Ae. albopictus were performed a Flavivirus detection. Flavivirus was not detected during study period. This study may provide basic information for surveillance of imported diseases (include Zika virus) and vectors in Korea.

        The tobacco cutworm, Spodoptera litura(Fabricius), is a major pest of tomato and frequently demands control measures. The timing of insecticide application is a key factor in determining its efficiency, so an experiment was designed to investigate this. Application of insecticide was based on three criteria: (i) the number of trap-caught moths in a Delta-type trap with a commercial sex pheromone lure placed in the center of the target area, soon after plant emergence; (ii) the percentage of plants exhibiting pinhole-type damage (10% or 20%) and (iii) the percentage of plants exhibiting shot hole-type damage (10% or 20%) compared to a check plot without any control measures. We found that the number of trap-caught moths was, compared to the other methods, the best means of deciding on insecticide application in tomato plant to control the tobacco cutworm. Using pheromone traps, we obtained the best performance of the insecticide Shinnago, causing > 90% larval mortality. Without insecticide application, tomato yield reduction due to the tobacco cutworm larva damage was 27%.

The blood meal source were of mammal (70.4%), birds (29.0%) and amphibian (0.6%). WNV was not detected in any mosquitoes.

        We developed a type of sausage made of chicken breast and liquid egg whites for consumers interested in weight management. To determine the quality of the product, its chemical characteristics, fatty acid composition, free amino acid contents, and nucleotides contents were evaluated during 4 weeks of storage. Sensory evaluation was conducted by both general consumers and body-builders. The sausage was proposed as a fat-free product as fat content was 0.12% based on the Korean Indication Standard of Animal Origin Food. Protein content was 13.42% and calorie value was 61.50 kcal/100 g of the sausage. In sensory evaluation, the mixture of chicken breast and egg whites stuffed into the same casing had an adverse effect on taste, color, texture and overall acceptance while the product that contained egg white stuffed separately into the outer casing enclosing the chicken breast (double layer) improved these attributes. The developed double-layer sausage can last for at least 4 weeks of storage without quality deterioration of flavor-related compounds, such as fatty acids and nucleotides.

        Pheromone biosynthesis activating neuropeptide (PBAN) produced in the subesophageal ganglion is known to stimulate pheromone production in the pheromone gland. A cDNA isolated from female adult heads of Maruca vitrata encodes 197 amino acids including PBAN, designated as Mvi-PBAN, and four other neuropeptides (NPs): diapause hormone (DH) homologue, α-NP, β-NP and γ-NP. All of the peptides are amidated in their C-termini and shared a conserved motif, FXPR(or K)L-NH2 structure. Mvi-PBAN consists of 35 amino acids as previously reported (Chang and Ramasamy, 2014). RT-PCR analysis revealed that Mvi-PBAN cDNA was expressed in all examined body parts. Nucleotide sequence analysis of RT-PCR products indicated the Mvi-PBAN sequence was identical in all examined body parts of both sexes. These results suggest that Mvi-PBAN expression is maintained in examined stages or tissues.

        The present research investigated copper and cadmium stress-induced differentially expressed genes (DEGs) using annealing control primers (ACP) with the differential display reverse transcription polymerase chain reaction technique in alfalfa (Medicago sativa L. cv. Vernal) leaves. Alfalfa leaves were subjected to 250 μM of copper and cadmium treatment for a period of 6 h. A total of 120 ACPs was used. During copper and cadmium treatment, 6 DEGs were found to be up or down regulated. During copper stress treatment, 1 DEG was up-regulated, and 3 novel genes were discovered. Similarly, during cadmium stress treatment, 1 DEG was up-regulated and 5 novel genes were identified. Among all 6 DEGs, DEG-4 was identified as the gene for trans-2,3-enoyl-CoA reductase, DEG-5 was identified as the gene for senescence-associated protein DIN1 and DEG-6 was identified for caffeic acid O-methyltransferase. All the up-regulated genes may play a role in copper and cadmium stress tolerance in alfalfa.

        This study aimed to investigate the effects of Dandelion (Taraxzcum coreanum) supplementation on milk yield, milk composition and blood characteristics in lactating dairy cows. Eight lactating dairy cows were divided into two groups (control: TMR supplementation, treatment: TMR with Dandelion supplementation). The milk yield, milk fat, lactose, solids not fat (SNF) and somatic cells counts (SCC) were not significantly different between the control group and the treatment group, whereas milk protein, milk urea nitrogen (MUN) and free fat acid (FFA) were significantly higher in the treatment group compared to the control (p<0.05). The blood components of the treatment group were compared with those of the control group and only aspartate aminotransferase (AST) appeared significantly high (p<0.05). The other blood components were not significantly different in the two groups. Blood corpuscle components in the groups were not significantly different. Especially, all blood corpuscle components in the treatment group were within the normal range. However, the white blood cells (WBC), lymphocytes (LYM) and hematocrits (HCT) in the control group exceeded the normal range. Based on the above results, the addition of Dandelion to feed increased milk protein, MUN and FFA, but did not significantly affect the composition of the blood and corpuscle in Holstein milking cows.
