        There are studies on the assessment of non-edible transgenic plants on soil microbial communities. In this research we evaluated the effect of virus-resistant trigonal cactus on soil microbial communities of the rhizosphere. Soil samples are collected and compared in genetically modified (GM) and non-GM trigonal cactus cultivation fields during vegetative growth period and post-harvest period. Biolog EcoplateTM was used to evaluate the functional diversity of soil microbial communities. There were no significant differences between the GM and non-GM soil samples collected during the vegetative growth period. However, we observed temporary difference in carbon substrate utilization. Principal component analysis showed that soil microbiota was influenced not by presence of GM or non-GM trigonal cacti, but rather by the cultivation period. Denaturing gradient gel electrophoresis fingerprinting revealed that virus-resistant trigonal cactus cultivation had insignificant effect on soil microbial communities including dominant rhizosphere bacteria, actinomycetes, and fungi. We found no clear evidence of GM trigonal cactus cultivation affecting the functional diversity of soil microbial communities.

        Human β- defensin-2 (hBD-2) is an antimicrobial peptide which is produced by epithelial cells after stimulation with microorganisms or inflammatory mediators. However, little is known as to whether the LPS and nicotine induces the expression of hBD-2 in periodontal l igament (PDL) cells. T he p urpose o f this s tudy was t o investigate t he r oles o f MAPKs pathway o f nicotine a nd L PS-induced hBD-2 expression in PDL cells. The maximal expression of hBD-2 was observed in nicotine 5 mM and LPS 1 μg/ml treated PDL cells. Nicotine and LPS induced the phosphorylation of p38, c-Jun N-terminal kinase 1 and 2 (JNK1/2), and extracellular signal-regulated kinases 1 and 2 (ERK1/2) MAPK. ERK1/2 inhibitor SB203580, p38 inhibitor PD98059, and JNK inhibitor SP600125, blocked the effects of nicotine and LPS on hBD-2 upregulation in PDL cells. These results collectively suggest that hBD-2 is up-regulated in nicotine and LPS-treated PDL cells through activation of p38, ERK and JNK MAPKs pathway. Our data regarding the up-regulation of hBD-2 may help us to understand the antimicrobial mechanism in periodontal disease

        Pleurotus citrinopileatus was successfully cultivated and commercially available in Korea. The antioxidant, xanthine oxidase, tyrosinase inhibitory activities and polyphenol contents of fruiting bodies of Pleurotus citrinopileatus extracted with acetone, hot water and methanol (hereinafter referred to Fr. Ace, Fr. HW and Fr. MeOH). The antioxidant activities on β-carotene-linoleic acid in the Fr. Ace, Fr. HW and Fr. MeOH were 96.12%, 94.21% and 96.52%, respectively at the concentration of 20 mg/ml. Xanthine oxidase inhibition activity in the Fr. Ace, Fr. HW and Fr. MeOH were 30.12%, 35.42% and 29.02%, respectively at the concentration of 5 mg/ml. Tyrosinase inhibition activity in the Fr. Ace, Fr. HW and Fr. MeOH were 58.78%, 49.25% and 63.29%, respectively at the concentration of 1.0 mg/ml. Total polyphenol contents in the Fr. Ace, Fr. HW and Fr. MeOH were 18.99 mgGAEs/g, 16.73 mgGAEs/g and 18.66 mgGAEs/g. These experimental results suggested that fruiting bodies of P. citrinopileatus contained good physio-chemical substances for promoting human health.

        Comparative effect of oyster mushrooms on plasma, fecal lipid profiles, liver and kidney functions were evaluated in hyper and normocholesterolemic rats. The feeding of hypercholesterolemic rats with 5% powder of fruiting bodies of oyster mushrooms i.e., Pleurotus ostreatus, P. sajor-caju and P. florida reduced the plasma total cholesterol level by 37%, 21% and 16%, respectively and triglyceride level by 45%, 24% and 14%, respectively. LDL/HDL ratio decreased by 64%, 45% and 41% for P. sajor-caju, P. ostreatus and P. florida fed rats, respectively. Mushroom feeding also reduced body weight in hypercholesterolemic rats. However, it had no adverse effect on plasma bilirubin, creatinin and urea nitrogen level. Mushroom feeding also increased the total lipid and cholesterol excretion through the feces. The present study reveals that feeding of 5% oyster mushroom powder do not have detrimental effects on the liver and kidneys rather may provide health benefits for the cardiovascular-related complication by decreasing the atherogenic lipid profiles.

        Methanol and hot water extracts of Daedalea bien-nis were investigated for its antioxidant activity, poly- phenol content and tyrosinase inhibitory effect. The antioxidant activity was evaluated using 1, 1-diphenyl -2-picrylhydrazyl (DPPH) scavenging assay. The antioxidant activities of hot water and 80% methanol extracts of Daedalea biennis fruiting body were 52.06%. and 92.62% at 1.5mg/ml concentration. Total polyphenol content of hot water and 80% methanol extracts were 12.26㎍ GAEs/mg and 13.69㎍ GAEs/mg. Tyrosinase inhibitory activity of water extract was 19.70% while 80% methanol extract was 15.75% at 1mg/ml concentration.

        Methanol and hot water extracts of Suillus granulatus were investigated for its antioxidant activity, polyphenol content and tyrosinase inhibitory effect. The antioxidant activity was evaluated using 1, 1- dipheny l-2-picrylhydrazyl (DPPH) scavenging assay. The antioxidant activities of hot water and 80% methanol extracts of Suillus granulatus fruiting body were 51.49%. and 90.58% at 1.5mg/ml concentration. Total polyphenol content of hot water and 80% methanol extracts were 33.21㎍ GAEs/mg and 27.97㎍ GAEs/mg. Tyrosinase inhibitory activity of water extract was 7.8% while 80% methanol extract was 20.88% at 1mg/ml concentration.

        Acinetobacter baumannii is usually considered an opportunistic pathogen that it responsible for a variety of nosocomial infections, such as, pneumonia, tracheobronchitis, meningitis, endocarditis, peritonitis, skin and soft tissue infections, and urinary tract infections. However, over the years, the organism has developed substantial antimicrobial resistance, and thus, the management of infections has become more difficult. The less common infective endocarditis is one of the more serious consequences of nosocomial bacteremia. Here, we report the successful treatment of the first case of multidrug-resistant A. baumannii endocarditis in a 33-year-old patient.

        CGMMV (cucumber green mottle mosaic virus) infects many cucurbitaceae species causing mosaic symptoms, yellowish leaf, and fruit deterioration. Several isolates of CGMMV from Korea, Israel, Japan, Greece and Spain have been characterized. In fact, CGMMV outbreaks have caused huge losses in the total yields of cucurbitaceous crops in Korea. Unfortunately, no genetic source is available for the resistance against CGMMV infection, and therefore, the breeding management offers no access to a solution yet. Thus, an alternative was to transform the viral gene such as CGMMV-CP into crops to induce tolerance against CGMMV. Here, we present the successful transformation of watermelon rootstock (gongdae) by Agrobacterium-mediated transformation with the CGMMV-CP gene. We obtained 29 independent T0 watermelon rootstock plants from the transformation of 7,887 explants. T0 plants were self-crossed and T1 plants were screened by the virus infection. Those gongdaes tolerant to virus were selected continuously, and BC2T1 and T3 generation were obtained. We have also obtained watermelons of BCF generation by crossing GM gongdae to watermelon line and those BCF watermelones were strongly tolerant to CGMMV.

        The aim of this study was to isolate chicken feather-degrading bacteria with high keratinolytic activity and to investigate cultural conditions affecting keratinolytic enzyme production by a selected isolate. A chicken feather-degrading bacterial strain CH3 was isolated from poultry wastes. Isolate CH3 degraded whole chicken feather completely within 3 days. On the basis of phenotypical and 16S rDNA studies, isolate CH3 was identified as Bacillus thuringiensis CH3. This strain is the first B. thuringiensis described as a feather degrader. The bacterium grew with an optimum at pH 8.0 and 37℃, where maximum keratinolytic activity was also observed. The composition of optimal medium for keratinolytic enzyme production was feather 0.1%, sucrose 0.7%, casein 0.3%, K2HPO4 0.03%, KH2PO4 0.04%, MgCl2 0.01% and NaCl 0.05%, respectively. The keratinolytic enzyme had a pH and temperature optima 9.0 and 45℃, respectively. The keratinolytic activity was inhibited ethylenediaminetetraacetic acid, phenylmethylsulfonyl fluoride, and metal ions like Hg2+, Cu2+ and Zn2+. The enzyme activated by Fe2+, dithiothreitol and 2-mercaptoethanol.

        Tocopherol belongs to the Vitamin E class of lipid soluble antioxidants that are essential for human nutrition. Tocopherols consist of a chromanol ring and a 15-carbon tail derived from homogentisate (HGA) and phytyl diphosphate, respectively. Condensation of HGA and phytyl diphosphate, the committed step in tocopherol biosynthesis, is catalyzed by HGA phytyltransferase (HPT). To increase tocopherol level in cucumber fruit, we performed Agrobacterium-mediated transformation with mdhpt gene, encoding HPT, isolated from apple (Malus domestica). We obtained two independent T0 cucumber plants from transforming 2,142 explants. Southern blot analysis of T0 showed that mdhpt was inserted in the genome. In order to test the tocopherol level, T1 cucumber fruits were screened by GC analysis. From 39 T1 fruits tested, some fruits showed 1.5-2 fold in α- and β-tocopherol content comparing to control. However, most of fruits contained lower levels of γ- and δ- tocopherol indicating a partition of metabolites in tocopherol biosynthesis pathway. We selected these T1 line and made a self-cross to obtain T2 seeds.
