        The rice weevil, Sitophilus oryzae, is one of the most common storage pests worldwide of grocery shops, flour mills and warehouses. This study investigated to assess the contact and fumigant toxicities of Juglnas regia and its active component against the S. oryzae adults. Using the two different bioassays against S. oryzae adults, the methanol extract of J. regia at 20 mg had the moderate (++) activity and weak (+) activity using direct contact and fumigant method, respectively. The methanol extract was partitioned with hexane, chloroform, ethyl acetate, butanol and water fraction. Ethyl acetate fraction obtained from the methanol extract showed strong (+++) and weak (+) activity at 10 mg using direct contact and fumigant method, respectively. Ethyl acetate fraction was purified by silica gel column chromatography and high performance liquid chromatography (prep-HPLC). The structure of active component was analyzed by EI-MS, 1H-NMR and 13C-NMR spectra, and was identified as C-21233. Purified C-21233 from ethyl acetate fraction had strong (+++) and moderate (++) activity using direct contact and fumigant method against S. oryzae adults. These results indicate that active component in J. reiga derived materials could be potential candidates as a contact and fumigant for managing S. oryzae adults. Further studies should be performed to the structure activity relationship of C-21233 and compared with its derivatives.

        The Black pine bast scale, Matsucoccus thunbergianae is one of the most serious in black pine, Pinus thunbergii forests in Korea. Since this pest was first reported in Goheung, Korea in 1963, which is gradually spread into neighboring regions and now occurs in many regions of the southern and eastern part of the Korean peninsula. The monitoring for distribution of M. thunbergianae was able to observed by naked eye egg sacs and pupa of male on the host until now. Therefore, this monitoring was very difficult in the low density of M. thubergianae. This experiment was conducted to use simple and practical moving cross-shaped flat trap for monitoring of M. thunbergianae. The monitoring of M. thunbergianae using the device was carried out to southern regions of the Korean peninsula. The first emergence of male showed mid. March in Namhae and late march in Busan, Jinju and Pohang. The peak of emergence showed late March in Namhae and early April in the other regions. When the number of M. thunbergianae intermediate nymph showed 58~59, 11~44 and 8~25 on 39.25 ㎠ bark area of the black pine, Pinus thubergii for 1 week, the number of captured its male adult was 58~83, 67~488 and 1~55 on the moving cross-shaped flat trap (10× 13㎝), respectively. The low density of M. thunbergianae was some few the number of capture, but there were no significant difference in its high density. Also, the number of captured its male adult was no significant in the different color (yellow, red, white and blue) of the moving cross-shaped flat trap.

        Paratlanticus ussuriensis enter prolonged diapause at an egg stage. Environmental conditions, such as temperature, can modify the diapause duration at initial diapause. Eggs enter initial diapause at 20℃, but continued early embryonic development at 30℃. Final diapause at a fully developed embryonic stage is obligatory regardless of temperature conditions. To determine temperature effects on diapause mechanism of P. ussuriensis eggs, we compared weights, DNA and RNA amounts of eggs incubated at either 20℃ or 30℃ for 50 days after oviposition. Both egg weight and total amount of DNA were constant at 20℃ but gradually increased at 30℃. However, total RNA level was rapidly increased at 15 days-old eggs at 30℃ and maintained high levels during further period whereas its level was constant at 20℃. In addition, we identified three heat shock protein 70 (hsp70a, hap70b, hsp70c) genes of P. ussuriensis and determined those expression levels at different temperature conditions. The levels of hsp70a and hsp70b was not detectable until 20 days after oviposition at both temperature conditions, but highly increased at 50 and 60 days when incubated at 30℃. In contrast, hsp70c level was rapidly peaked at 20 days after oviposition, which is the time of initial diapause entrance. Our results suggest that high temperature breakdown initial diapause and a certain hsp gene, such as hsp70c, may involve in the initial diapause mechanism of P. ussuriensis eggs.

        Cecidomyiidae and Sciaridae (Diptera: Sciaroidea) are mostly mycophagous, feeding on decaying plant materials and fungi. On the shiitake bedlogs, great number of larvae of the species cause serious damage, feeding on the mycelium of the shiitake mushroom. We confirmed five species emerged from the shiitake bedlogs, which are two cecidomyiids and three sciarids. Using the DNA barcording, the mitochondrial cytochrome c oxidase subunit I (COI) region (658 bp), the larvae and adult flies were identified, and Camptomyia corticalis was confirmed as a major pest on shiitake mushroom.

        The tribe Phylini (Miridae: Phylinae) is revised in the Korean Peninsula. A total of 65 species in 20 genera are confirmed to exist in this territory. Five genera and 20 species are recognized for the first time in Korea, including a new genus and 6 new species: Gen. nov. sp. nov.1; Psallus sp. nov.1, P. sp. nov.2, and P. sp. nov.3; and Moissonia sp. nov.1 and M. sp. nov.2. Herein, checklist for the Korean Peninsula is reconstructed with its geographical distribution, and short discussion.

        The cotton aphid, Aphis gossypii (Glover), is one of the most serious pest in seed potato and various vegetable cultivation. The imidacloprid-resistant strain (IR) was over 200-fold more resistant to imidacloprid compared to a susceptible strain (S) as judged by LC50 values. The IR showed cross resistances to other neonicotinoid insecticides. IEF and 2DE analyses revealed that general esterase isozyme patterns in IR were almost identical to those of S. Nevertheless, a significantly overexpressed protein spot was detected in IR. To identify differentially expressed genes in IR, comparative transcriptome analyses based on GS-FLX were conducted using total RNAs extracted from both IR and S strains, which generated ca. 290 Mb reads for each strain. Generally, most nicotinic acetylcholine receptor subunit genes, such as alpha 2 and beta 1, were more transcribed in S than in IR. In contrast, only alpha 5 subunit gene was 1.8 fold more expressed in IR. Seven ATP-binding cassette (ABC) transporter genes were newly identified in A. gossypii, among which only ABCC9 gene was highly expressed in IR. Therefore, this ABCC subfamily, a member of the MRP subfamily which is involved in multi-drug resistance, could be one of the main factors associated with imidacloprid resistance in A. gossypii.

        The faulty regulation of imprinting gene lead to the abnormal development of reconstructed embryo after nuclear transfer. However, the correlation between the imprinting status of donor cell and preimplantation stage of embryo development is not yet clear. In this study, to determine this correlation, we used the porcine spermatogonial stem cell (pSSC) and fetal fibroblast (pFF) as donor cells. As the results, the isolated cells with laminin matrix selection strongly expressed the GFRα-1 and PLZF genes of SSCs specific markers. The pSSCs were maintained to 12 passages and positive for the pluripotent marker including OCT4, SSEA1 and NANOG. The methylation analysis of H19 DMR of pSSCs revealed that the zinc finger protein binding sites CTCF3 of H19 DMRs displayed an androgenic imprinting pattern (92.7%). Also, to investigate the reprogramming potential of pSSCs as donor cell, we compared the development rate and methylation status of H19 gene between the reconstructed embryos from pFF and pSSC. This result showed no significant differences of the development rate between the pFFs (11.2±0.8%) and SSCs (13.3±1.1%). However, interestingly, while the CTCF3 methylation status of pFF-NT blastocyst was decreased (36.3%), and the CTCF3 methylation status of pSSC-NT blastocyst was maintained. Therefore, this result suggested that the genomic imprinting status of pSSCs is more effective than that of normal somatic cells for the normal development because the maintenance of imprinting pattern is very important in early embryo stage.

        Pleurotus citrinopileatus was successfully cultivated and commercially available in Korea. The antioxidant, xanthine oxidase, tyrosinase inhibitory activities and polyphenol contents of fruiting bodies of Pleurotus citrinopileatus extracted with acetone, hot water and methanol (hereinafter referred to Fr. Ace, Fr. HW and Fr. MeOH). The antioxidant activities on β-carotene-linoleic acid in the Fr. Ace, Fr. HW and Fr. MeOH were 96.12%, 94.21% and 96.52%, respectively at the concentration of 20 mg/ml. Xanthine oxidase inhibition activity in the Fr. Ace, Fr. HW and Fr. MeOH were 30.12%, 35.42% and 29.02%, respectively at the concentration of 5 mg/ml. Tyrosinase inhibition activity in the Fr. Ace, Fr. HW and Fr. MeOH were 58.78%, 49.25% and 63.29%, respectively at the concentration of 1.0 mg/ml. Total polyphenol contents in the Fr. Ace, Fr. HW and Fr. MeOH were 18.99 mgGAEs/g, 16.73 mgGAEs/g and 18.66 mgGAEs/g. These experimental results suggested that fruiting bodies of P. citrinopileatus contained good physio-chemical substances for promoting human health.

        The current investigation was undertaken to evaluate the antioxidant activities, tyrosinase inhibitory effects on the fruiting bodies of P. ostreatus extracted with acetone, methanol and hot water. The antioxidant activities were performed on β-carotene-linoleic acid, reducing power, 1,1-diphenyl-2-picrylhydrazyl free radical scavenging, and ferrous chelating abilities. In addition to this, phenolic acid and flavonoids contents were also analyzed. Methanolic extract of P. ostreatus showed the strongest β-carotene-linoleic acid inhibition as compare to others exracts. At 8 mg/ml, acetonic extract showed a high reducing power of 1.54. The scavenging effects on 1,1-diphenyl-2-picrylhydrazyl radicals, acetonic extract was effective than other extracts. The strongest chelating effect (85.66%) was obtained from the acetonic extract at 1.0 mg/ml concentration. Antioxidant activities of the extracts from the fruiting bodies of P. ostreatus were increased with the increasing concentration. After application of reverse phase high performance liquid chromatography, coupled to a diode array detector and electrospray ionisation mass spectra, six phenolic compounds namely, gallic acid, protocatechuic acid, naringenin, hesperetin, formononetin and biochanin were identified from acetonic extract. Tyrosinase inhibition of acetonic, methanolic, and hot water extracts of P. ostreatus were increased with the increasing of concentration. Results revealed that methanolic extract showed good, while acetonic and hot water extracts showed moderate activities of the tyrosinase inhibition at the concentration tested. This study suggests that fruiting bodies of P. ostreatus can potentially be used as a readily accessible source of natural antioxidants.

        The antioxidant, xanthine oxidase, tyrosinase inhibitory activities and polyphenol contents of the fruiting bodies of Pleurotus cornucopae extracted with acetone, hot water and methanol (hereinafter referred to Fr. Ace, Fr. HW and Fr. MeOH). The antioxidant activities in the Fr. Ace, Fr. HW and Fr. MeOH were 93.23%, 89.55% and 92.58%, respectively at the concentration of 2.0 mg/ml. Xanthine oxidase inhibition activity in the Fr. Ace, Fr. HW and Fr. MeOH were 45.84%, 46.50% and 45.60%, respectively at the concentration of 5 mg/ml. Tyrosinase inhibition activity in the Fr. Ace, Fr. HW and Fr. MeOH were 52.11%, 50.12% and 55.81%, respectively at the concentration of 1.0 mg/ml. Total polyphenol contents in the Fr. Ace, Fr. HW and Fr. MeOH were 18.99 mgGAEs/ g, 16.73 mgGAEs/g and 18.66 mgGAEs/g. These experimental results suggested that fruiting bodies of P. cornucopae contained good physio-chemical substances for promoting human health.

        Lentinus lepideus is an edible mushroom, belongs to the family Tricholomaceteae and order Agaricales. The purpose of this study was to evaluate the antioxidant activities, tyrosinase inhibitory effects on the fruiting bodies of L. lepideus extracted with acetone, methanol and hot water. The antioxidant activities were performed on β-carotene-linoleic acid, reducing power, 1,1-diphenyl-2-picrylhydrazyl free radical scavenging, and ferrous chelating abilities. In addition to this, phenolic acid and flavonoids contents were also analyzed. Hot water extract of L. lepideus showed the strongest β-carotene-linoleic acid inhibition as compare to others extracts. At 8 mg/ml, methanolic extract showed a high reducing power of 1.21. The scavenging effects on 1,1-diphenyl-2-picrylhydrazyl radicals, acetonic and methanolic extracts were effective than hot water extract. The strongest chelating effect (87.50%) was obtained from the methanolic extract at 1.0 mg/ml concentration. Antioxidant activities of the extracts from the fruiting bodies of L. lepideus were increased with the increasing concentration. After application of reverse phase high performance liquid chromatography, coupled to a diode array detector and electrospray ionisation mass spectra, six phenolic compounds namely, chlorogenic acid, vanillin, naringin, naringenin, formononetin and biochanin were identified from acetonic extract. Tyrosinase inhibition of acetonic, methanolic, and hot water extracts of L. lepideus were increased with the increasing of concentration. Results revealed that acetonic and methanolic extract showed good, while hot water showed moderate activities of the tyrosinase inhibition at the concentration tested. This study suggests that fruiting bodies of L. lepideus can potentially be used as a readily accessible source of natural antioxidants.

After application of reverse phase high performance liquid chromatography, coupled to a diode array detector and electrospray ionisation mass spectra, four phenolic compounds namely, naringenin, hesperetin, formononetin and biochanin were identified from acetonic extract. Tyrosinase inhibition of acetonic, methanolic, and hot water extracts of L. edodes were increased with the increasing of concentration. Results revealed that acetonic and methanolic extracts showed good, while hot water showed moderate activities of the tyrosinase inhibition at the concentration tested. This study suggests that fruiting bodies of L. edodes can potentially be used as a readily accessible source of natural antioxidants.

The strongest chelating effect (88.10%) was obtained from the methanolic extract at 1.0 mg/ml concentration. Antioxidant activities of the extracts from the fruiting bodies of P. eryngii were increased with the increasing concentration. After application of reverse phase high performance liquid chromatography, coupled to a diode array detector and electrospray ionisation mass spectra, ten phenolic compounds namely, gallic acid, protocatechuic acid, chlorogenic acid, vanillin, ferulic acid, naringin, naringenin, hesperetin, formononetin and biochanin were identified from acetonic extract. Tyrosinase inhibition of acetonic, methanolic, and hot water extracts of P. eryngii were increased with the increasing of concentration. Results revealed that acetonic and methanolic extracts showed good, while hot water showed moderate activities of the tyrosinase inhibition at the concentration tested. This study suggests that fruiting bodies of P. eryngii can potentially be used as a readily accessible source of natural antioxidants.

        To develop new nutraceuticals from mushrooms, nutritional properties of two kinds of Hypsizygus marmoreus fruiting bodies were investigated. Hypsizygus marmoreus (white) contained 21.63% H. marmoreus (brown) contained 27.34% of crude protein, 55.75% of total sugar, and 2.69% crude lipid and 7.77% ash. It also contained 10.04 mg% Na, 19.28 mg% Ca, 9.30 mg% Fe and its calorie value was 356.57 Kcal per 100 g. Free sugar and amino acid content of H. marmoreus (brown) were determined.

        Comparative effect of oyster mushrooms on plasma, fecal lipid profiles, liver and kidney functions were evaluated in hyper and normocholesterolemic rats. The feeding of hypercholesterolemic rats with 5% powder of fruiting bodies of oyster mushrooms i.e., Pleurotus ostreatus, P. sajor-caju and P. florida reduced the plasma total cholesterol level by 37%, 21% and 16%, respectively and triglyceride level by 45%, 24% and 14%, respectively. LDL/HDL ratio decreased by 64%, 45% and 41% for P. sajor-caju, P. ostreatus and P. florida fed rats, respectively. Mushroom feeding also reduced body weight in hypercholesterolemic rats. However, it had no adverse effect on plasma bilirubin, creatinin and urea nitrogen level. Mushroom feeding also increased the total lipid and cholesterol excretion through the feces. The present study reveals that feeding of 5% oyster mushroom powder do not have detrimental effects on the liver and kidneys rather may provide health benefits for the cardiovascular-related complication by decreasing the atherogenic lipid profiles.

        World wide mushroom productions have been increased to 10-20% and more various mushrooms have been attempted to cultivate. Similar trends were also observed in Korea. More diverse mushroom varieties such as Pleurotus eryngii, Hericium erinaceus and Agrocybe aegerita have been attempted to cultivate in larger areas. In these days, 235 varieties of 33 different mushroom species have been cultivated. However only 50 varieties were protected by the law. Since 1960s, mushroom industry had been considered as one of the major export industries in Korea and mushroom production has been rapidly increased. In 2008, total mushroom production was about 198,209 ton, which were about 800 billion won (one trillion won if include mushroom factory products). Major cultivated species are Pleurotus ostreatus, P. eryngii, Flammulina velutipes, Lentinula edodes and Agaricus bisporus, which cover 90% of total production. We believed that the protection of mushroom vavarieties by the law is one of the main problems to be solved. Also, more studies to develop better mushroom production methods with low cost and improve the distribution structures are certainly required. In these days, the export of mushroom has been increased in Korea. We imported more mushrooms than exported for last 4 years, however, it has been changed that export is getting bigger in these days. Because we developed the liquid spawning and automatic cultivation systems, which lead the reduction of mushroom production cost and resulted in the increasement of export. If we develop better post-harvest system, it is certain that the mushroom export should be more important in the future. As an alternative way, some Korean companies are planning to build the plants in main export countries. Mushroom industry has promising future because mushroom is good for your health.
