        Genetically modified (GM) crops have never been cultivated commercially in Korea, it is necessary for a thorough assessment of the risks associated with their environmental release. We determined the frequency of pollen mediated gene flow from disease resistant GM rice (OsCK1) to non-GM rice (Nagdongbyeo) and weedy rice (R55). A total of 449,711 or 164,604 seeds were collected from non-GM and weedy rice, respectively which were planted around OsCK1. Resistance of the hybrids was determined by repeated spraying of herbicide and DNA analysis using specific primer to confirm hybrids. Though non-GM rice and weedy rice have similar flowering time, the hybrids were found only in non-GM rice and out-crossing ranged from 0.018% at 0.3 m to 0.013% at 0.6 m. All of hybrids were located within 0.6 m distance from the GM rice plot in southerly direction. The meteorological factors including temperature and relative humidity during flowering time were found to be the most important factors for determining rice out-crossing. It should be considered many factors like the local weather condition and flowering time to set up the safety management policy to prevent pollen mediated gene flow between GM and conventional crop.

        Soybean [Glycine max. (L.) Merr] is one of the most important legumes in the world. However, soybean varieties are sensitive to flooding stress and their seed yields are substantially reduced in response to the flooding stress. 192 soybean germplasm collection was screened to identify flooding tolerant germplasm at an early vegetative growth stage (V1). Soybean plants at V1 stage were waterlogged for 4 to 10 days. To evaluate flooding tolerance, survival rate were investigated as a time dependent manner. Jangbaegkong, Danbaegkong, Sowonkong, Socheong2 and Suwon269 showed flooding tolerance, while Shillog, T201, T181, NTS1116 and HP-963 showed flooding sensitivity. We also investigated effects of flooding stress on soybean morphology. The adventitious root development was greatly increased in flooding tolerant plants compared to it in flooding sensitive plants. In addition, root length and root number were analyzed. The significant reduction of root length and root number was observed in flooding sensitive plants. Thus, these results indicate that the morphological changes in roots are important for acclimation to flooding stress. Taken together, the relationship between the morphological changes in the roots and flooding tolerance may be useful in selecting a flooding tolerant soybean germplasm.

        Agastache rugosa, a member of the mint family (Labiatae), is a perennial herb widely distributed in East Asian countries. It is used in traditional medicine for the treatment of cholera, vomiting, and miasma. This study assessed the genetic diversity and population structures on 65 accessions of Korean mint A. rugosa germplasm based on inter simple sequence repeat (ISSR) markers. The selected nine ISSR primers produced reproducible polymorphic banding patterns. In total, 126 bands were scored; 119 (94.4%) were polymorphic. The number of bands generated per primer varied from 7 to 18. A minimum of seven bands was generated by primer 874, while a maximum of 18 bands was generated by the primer 844. Six primers (815, 826, 835, 844, 868, and 874) generated 100% polymorphic bands. This was supported by other parameters such as total gene diversity (HT) values, which ranged from 0.112 to 0.330 with a mean of 0.218. The effective number of alleles (NE) ranged from 1.174 to 1.486 with a mean value of 1.351. Nei's genetic diversity (H) mean value was 0.218, and Shannon's information index (I) mean value was 0.343. The high values for total gene diversity, effective number of alleles, Nei's genetic diversity, and Shannon's information index indicated substantial variations within the population. Cluster analysis showed characteristic grouping, which is not in accordance with their geographical affiliation. The implications of the results of this study in developing a strategy for the conservation and breeding of A. rugosa and other medicinal plant germplasm are discussed.

        In order to improve rice dough functionality, we cloned 4 kinds of high-molecular-weight glutenin subunit (HMW-GS) genes from bread wheat, ‘Jokyeong’. Among them, we first examined Dx5 gene to generate marker-free transgenic rice for advanced quality processing of bread and noodles. The GluB1 promoter was inserted into binary vector for seed specific expression of the Dx5 gene. Two expression cassettes comprised of separate DNA fragments containing only the high-molecular-weight glutein subunit (HMW-GS) protein (Dx5) and hygromycin phosphotransferase II (HPTII) resistance genes were introduced separately to tumefaciens EHA105 strain for co-infection. Each EHA105 strain harboring Dx5 or HPTII was infected to rice calli at 3: 1 ratio of Dx5 and HPTII, respectively. Then among 66 hygromycin-resistant transformants, we obtained two transgenic lines inserted both with Dx5 and HPTII gene to rice genome. We reconfirmed integration of the Dx5 and HPTII genes into the rice genome by Southern blot analysis. Wheat Dx5 transcriptsin rice seeds was examined with semi-quantitative RT-PCR. Finally, the marker-free plants containing only Dx5 gene were successfully screened at T1 generation. This result also provides that co-infection system with two expression cassettes could be efficient strategy to generate marker-free transgenic rice plants.

        Two sugary mutants, Hwacheong sugary-1 (Hsu1) and Hwacheong sugary-2 (Hsu2) were obtained by chemical mutagenesis from japonica cultivar, Hwacheongbyeo. Sugary mutants exhibited wrinkled and translucent grain with high soluble sugar content. In addition, amber-colored endosperm of sugary mutants was loosely packed due to abnormal starch granules compared to densely packed wild-type. Especially, the grain of Hsu2 mutant was less wrinkled than that of Hsu1, thus Hsu2 can be polished easily. Previous studies reveal that su1 mutant was resulted from mutation in gene for a debranching enzyme, isoamylase but the sequence of the mutated gene has not been identified. To identify the sequence of sugary genes, the map-based cloning strategy was applied. The genetic study revealed that the phenotype of Hsu2 mutant was controlled by two recessive genes. Interestingly, one of the genes was located on chromosome 8 at the position of isoamylase which was known as su-1. This indicates that mutation in isoamylase gene causes sugary endosperm characteristics. However we found different mutation points between the Hsu1 and Hsu2. The point mutation in Hsu1 was occurred at 10th exon whereas the other mutation related with Hsu2 was occurred at 15th exon. As mentioned above, the Hsu2 mutant has less wrinkled shape and less soluble sugar content than the Hsu1 mutant. Thus, we hypothesize that the other gene controlling Hsu2 mutant phenotype may have a role in weakening the effect of the su-1. Further study on the other gene associated with the Hsu2 phenotype is in progress.

        Low temperature stress is one of the major negative factors affecting vegetative and reproductive growth of rice. To better understand responses of rice plants to low temperature we analyzed transcriptome expression patterns in glumous flower of cold-tolerant japonica rice variety, Stejaree45, and cold-susceptible variety, HR19621-AC6 at booting stage under cold water irrigation. A total of 2,411 probes were differentially expressed by low temperature in glumous flowers of the two varieties. Some important genes involved in hormone biosynthesis showed variety-specific regulation. Expression of GA20ox3 and GA2ox, among the genes involved in GA biosynthesis, was regulated differentially in the two varieties. Among the genes involved in IAA biosynthesis, YUCCA1 and TAA1:1 showed variety-specific regulation. Among the genes involved in cytokinin biosynthsis and signaling, expression of LOG, HK1 and HK3 was significantly down-regulated only in the cold-susceptible variety. Among the genes involved in ABA biosynthesis, NSY and AAO3 were down-regulated only in the cold-tolerant variety. In general, genes involved in GA, IAA and cytokinin biosynthesis responded to cold temperature in such a way that capacity of those bioactive hormones is maintained at relatively higher levels under cold temperature in the cold-tolerant variety, which can help minimize cold stress imposed to developing reproductive organs in the cold-tolerant variety.

        An unbalance of rice productions and consumptions caused serious problems in both of agricultural area and grain market of korea. In recently, various efforts for rice processing products such as rice noodles and rice wines are in progress to overcome the unstable rice market. Among them, waxy rice is predominant items in processing rice. However, varietal features of starch viscosity are not considered in the processing industries and rice breeding field. In this study, 12 waxy rices and 2 cultivars Ilmibyeo(japonica) and IR72(Indica) were studied for physicochemical and amylogram to characterize the use of waxy rices. The amylose contents of waxy rices were from 7.1 to 8.1% with soft gel consistency and relatively low alkali digestion value(1.4% KOH) compare to Ilmibyeo. In the amylogram analysis(RVU) of waxy rices, unlikely normal rices, very fast peak time(about 3.5 min.) was obtained compare to that of 6.2 min. of Ilmibyeo. And 2 to 3 groups were classified based on peak viscosity and consitency of RVU. Wangchal, Odorokimochi and Hangangchal showed relatively high peak viscosity seemed not suitable for rice cake due to the hardness speed. And most of waxy rices developed in korea were believed to be suitable for oil fried cakes(Hankwa) because of a certain grade of starch degradation speed. And Mochiminori and Midoromochi originated from japan could be a good sources in waxy rice breeding program to improve the cooking properties especially in slow down of hardness speed with very low peak viscosity, hot viscosity and cool viscosity.
