This study is a basic study on the development of functional substances involved in obesity prevention, lipid metabolism, and immune regulation. Male Sprague-Dawley rats were fed a high-fat diet for 10 weeks. Allium monanthum extracts (AME) were administered orally to obesity-induced rats, and their lipid-lowering, antioxidative and various types of biological effects related to the immune system were examined. Blood free fatty acid and triglyceride concentrations decreased as the dose of AME increased. Total cholesterol and LDL cholesterol concentrations in the blood decreased as the dose of AME increased. The total cholesterol concentrations in the liver of the AME-treated groups were lower than the control group. The thiobarbituric acid reactive concentrations were lower in the plasma and liver of all AME-treated groups than the control group. Plasma AST and ALT activities did not show any significant differences among the treatment groups. IL-1β and IL-6 concentrations in the liver tended to decrease as the dose of AME increased. TNF-α and IL-10 concentrations did now show any significant differences compared to the control group. Lower expression levels of TNF-α, Apo-B and Apo-E genes were found in the AME-treated groups. Taken together, these results indicate that AME may show positive effects in lipid lowering, antioxidation and anti-inflammation.

Apple and pear are popular fruits consumed in Korea and are common fruit in daily diet. In order to compare the antioxidant activity of the apple and pear peels, total polyphenol contents, total flavonoid contents, ABTS+ free radical scavenging activity, and DPPH free radical scavenging activity were measured from hot water, ethanol, and methanol extracts of the two fruit peels. The total polyphenol and flavonoid contents were highest in 95% methanol extracts of the apple peelsand 70% ethanol extract of the pear peels, respectively. Total polyphenol contents of the pear peels were higher than that of apple peels, and total flavonoid contents of the apple peels were higher than that of pear peels. The apple and pear peels had the highest ABTS+· and DPPH free radical scavenging activity in 95% methanol extracts and 70% ethanol extracts, respectively. ABTS+· and DPPH free radical scavenging activity of pear peels was higher than that of apple peels, and the DPPH free radical scavenging activity of apple and pear peels were detected in hot water, 95% methanol, and 70% ethanol extracts, respectively. Ascorbic acid, a synthetic antioxidant used as positive control, had significantly higher scavenging activity than the apple and pear peels. In conclusion, the apple and pear peelshave great potential as natural antioxidants. Therefore, above results should be considered to provide the possibility for the development of high functional antioxidants.

The present study attempts to evaluate antioxidant activities of extracts from Abeliophyllum distichum. Nakai flower. The samples were collected in Jangyyeon-myeon, Goesan-gun, Korea and extracted with either hot-water or ethyl acetate (EtOAC). In DPPH, hydroxyl radical scavenging activity and Fe2+ chelating activity of EtOAC extracts were 93.41%, 98.43%, and 7.38%, while those of hot-water extracts were 86.93%, 41.33% and 47.68% at 200 μg/ml, respectively. In ΦX-174 RF I plasmid DNA cleavage assay, the protective effects of EtOAC and hot-water extracts against oxidative DNA damage were 82% and 17% at 200 μg/ml, respectively. Both extracts showed the protective effect of DNA migration by oxidative stress in intracellular DNA migration assay. Both extracts had no cytotoxity in NIH3T3 cells. Several polyphenolic compounds were identified such as 2-methoxy-benzoic acid, vanillic acid, phytol and pulegone by GC/MS. These results indicated that extracts of Abeliophyllum distichum Nakai flower showed antioxidant activities and protective activities against oxidative DNA damage and showed the possibility to be used as an effective natural antioxidants.

Agastache rugosa, a member of the mint family (Labiatae), is a perennial herb widely distributed in East Asian countries. It is used in traditional medicine for the treatment of cholera, vomiting, and miasma. This study assessed the genetic diversity and population structures on 65 accessions of Korean mint A. rugosa germplasm based on inter simple sequence repeat (ISSR) markers. The selected nine ISSR primers produced reproducible polymorphic banding patterns. In total, 126 bands were scored; 119 (94.4%) were polymorphic. The number of bands generated per primer varied from 7 to 18. A minimum of seven bands was generated by primer 874, while a maximum of 18 bands was generated by the primer 844. Six primers (815, 826, 835, 844, 868, and 874) generated 100% polymorphic bands. This was supported by other parameters such as total gene diversity (HT) values, which ranged from 0.112 to 0.330 with a mean of 0.218. The effective number of alleles (NE) ranged from 1.174 to 1.486 with a mean value of 1.351. Nei's genetic diversity (H) mean value was 0.218, and Shannon's information index (I) mean value was 0.343. The high values for total gene diversity, effective number of alleles, Nei's genetic diversity, and Shannon's information index indicated substantial variations within the population. Cluster analysis showed characteristic grouping, which is not in accordance with their geographical affiliation. The implications of the results of this study in developing a strategy for the conservation and breeding of A. rugosa and other medicinal plant germplasm are discussed.

Korean ginseng has been used for thousands of years as an important medicinal plant. Lime-Bordeaux mixture (LBM) was made with copper sulfate and quicklime, which was sprayed instead of pesticides in ginseng field. Net photosynthesis (PN) was compared between Treatment and Non-treatment of LBM in 3 Year Old Ginseng. PN in control plot recorded 2.94μmol (CO2) m-2s-1 at the first day of experiment, which was similar until the last day of experiment. However, The PN in LBM recorded 2.23μmol (CO2) m-2s-1, which was lower than that in control plot. As time goes by, The PN in LBM was gradually increased up to 3.21μmol (CO2) m-2s-1 and finally, it was similar with that in control plot at 7th day as a 3.20μmol (CO2) m-2s-1).

The objective of this study was determined to evaluate α-amylase, α-glucosidase, pancreatic lipase inhibition in vitro and DPPH radical scavenging activity of the several Korean resources plants. The α-amylase inhibitory activity of Salicornia herbacea, Erythronium japonicum (flower) and Phragmites communis (root) in water extract showed relatively high 62.8%, 66.5% and 69.3%, respectively. The α-amylase inhibitory activity of Citrus junos (pericarp) and Cornus officinalis in methanol extract was found to have an effect with 32.8% in Citrus junos (pericarp) and 60.9% in Cornus officinalis. Corylopsis coreana in both water and methanol extract had the highest α-glucosidase inhibitory activity of 81.7% and 89.5%, while the extract of Portulaca oleracea, Ficus carica and Citrus junos was not measured α-glucosidase inhibitory activity at given experiment concentration. Depending on the extraction solvent and the plant species, it was observed that there was a significant difference in α-glucosidase inhibitory activity. The pancreatic lipase inhibitory activity showed relatively higher in the methanol extract than water extract except pericarp of Citrus junos. The DPPH radical scavenging activity of selected plants was much difference between measured plant species, and showed that the increase was proportional to the concentration. These results suggested that selected plants had the potent biological activity on carbohydrate, lipid Inhibitory activity and antioxidant activity, therefore these plant resources could be a good materials to develop medicinal preparations, nutraceuticals or health functional foods for diabetes or obesity.

The research concerned of the regeneration of plants from embryos obtained from anther cultures of ginseng (Panax ginseng C. A. Meyer). The aim was to determine the influence of the regeneration medium on the efficiency of the regeneration process. We conducted to determine the optimum conditions such as cold pretreatment, plant growth regulators and carbon sources on anther culture of P. ginseng. Highest callus formation rate was obtained when flower buds pretreated at 4℃ for 1 day. Among the treated growth regulators with various degrees of concentration in Murashige and Skoog's (MS) medium, 4.53 μm of 2.4-dichlorophenoxyacetic acid and 4.44 μm of 6-benzylaminopurine gives the most responsive callus with the frequency of 73.89% and 129.53 g of fresh weight. When we used 3-9% of sucrose and maltose among the different kinds and various concentrations of carbohydrates, callus was formed highest 67.29% in the medium with 3% of sucrose. Shoots induced from callus supplemented with 28.9 μm of gibberellic acid and rooted in Gamborg's B5 medium supplemented with 14.7 μm of indole-3-butyric acid.

The antioxidant enzyme and DPPH radical scavenging activity with variations in drying methods of diploid and tetraploid in Platycodon grandiflorum were determined. Antioxidant enzyme activities were measured as superoxide dismutase (SOD), catalase (CAT), peroxidase (POX), and ascorbate peroxidase (APX). The roots of Platycodon grandiflorum were freeze-dried, indoor-dried, hot-air dried, and microwave dried. The root extract of P. grandiflorum have shown the highest SOD enzyme activity of 92% in tetraploid of freeze-dried and indoor-dried while diploid of microwave dried showed the lowest SOD enzyme activity of 47.5%. The activity of CAT showed higher values in the root of tetraploid than in the diploid of P. grandiflorum in all drying methods. The APX activity showed relatively higher values in the root extract of freeze-dried both the diploid and tetraploid, but the difference in comparison with other extracts was not significant. The POX activities according to drying methods of diploid and tetraploid in P. grandiflorum showed relatively high values in freeze-dried and indoor-dried compared with other drying methods, and the POX activity between the diploid and tetraploid was not significant difference in each drying method. The DPPH radical scavenging activity with variation in drying methods of diploid and tetraploid in P. grandiflorum was the highest in the freeze-dried, and was higher in tetraploid than diploid in all the concentrations. In conclusion, the root of P. grandiflorum had the potent biological activities in both diploid and tetraploid. In particular, the tetraploid root of P. grandiflorum showing high antioxidant enzyme activity could be good materials for development of source of functional healthy food.

Morus Folium (Sang-yeop in Korean) is one of the most important Oriental medicinal plants. In Korea, both M. alba and M. cathayana are regarded as the botanical sources for Morus Folium. In order to discriminate M. alba and M. cathayana from their adulterant, M. tricuspidata, mitochondrial NADH dehydrogenase subunit 7 (nad7) intron 2 region was targeted for molecular analysis with universal primers. DNA polymorphisms, including SNP sites, insertions, and deletions, were detected among these three species sequencing data. Based on these DNA polymorphisms, specific primers were designed for the three species respectively. Multiplex PCR was conducted for molecular authentication of M. alba, M. cathayana, and M. tricuspidata with specific primers. The present results indicate that it is possible to identify Morus Folium from its adulterant using mitochondrial nad7 intron 2 region. The established multiplex-PCR system was proved to be effective for identification of Morus Folium. The results indicate that mitochondrial introns can be used for inter-specific polymorphic study, and the described method can be applied for molecular identification of medicinal materials.

This study has assessed the anti-oxidative activities and cytotoxic effects of Lithospermum erythrorhizon ethanol extract and measured the effects of tyrosinase inhibition activities with a goal of estimating the usage of the medicinal plant as an ingredient of cosmetics. First, to perform a basic test on the extract, pH and UV-spectrum were measured. According to the measurement, the extract had control functions at pH 5.5, and maximum absorbance occurred at 530nm. In particular, DPPH (1-1-diphenyl-2-picryl-hydrazyl)-inhibiting activity (IC50) and polyphenol content were 149.81 μg/mL and 51.28±2.52 mg/mL respectively. In addition, as extract concentration increased, tyrosinase inhibition activities improved as well. In raw 264.7 cell-based MTT assay, cell survival rates were 98% at 1000 ppm and 153% at 100 ppm. Therefore, it's been confirmed that there is almost no cytotoxin. According to the test results above, it appears that the Lithospermum erythrorhizon ethanol extract would be effective in anti-oxidation and application as a cosmetic ingredient.
