        Clover cyst nematode (Heterodera trifolii, CCN) is one of the important plantparasitic nematodes in the Republic of Korea, causing serious damage to Kimchi-cabbage in Gangwon province since 2017. Soil fumigants are the preferred choice for managing CCN. However, cabbage fields in Gangwon province have a slope, making it challenging to apply soil fumigants and use plastic mulch. Consequently, alternative materials should be developed for managing CCN. Nematicidal cover crops and resistant cultivars are among the alternatives. We conducted resistance screening of 82 white mustard (Sinapis alba) resources from a genebank at RDA against CCN for use as nematicidal cover crops or breeding materials. In the first assay (1st, 2nd, and 3rd pot experiments), 15 white mustard resources were selected, while others were susceptible to CCN. To confirm the reproducibility of resistance to CCN for 13 selected resources, the second assay was performed. As a result, five white mustard resource (IT297309, IT297312, IT302951, IT302953, IT302954) demonstrated resistance to CCN. These findings indicate the potential use of these five white mustard resources as nematicidal cover crops or for breeding cultivars of Kimchi-cabbage in Republic of Korea.

        담양습지는 하도 내에 발달하는 하천습지로, 국내 처음으로 습지보호구역으로 지정되었으며 유네스코 무등산권 세계지질공원 후보지역으로 알려져 있다. 담양습지 인근은 영산강 상류지역으로 현재는 비교적 넓은 평야지대가 분포한다. 담양습지 주변의 퇴적환경을 해석하고자 시추코어를 획득하였고, 퇴적상, 연대측정(AMS, OSL), 입도분석, 지화학 분석 등을 수행했다. 또한 습지 주변의 기존 시추 코어 자료를 사용하여 종합적인 퇴적환경 변화를 해석했다. 담양습지가 분포하는 영산강 상류 일대의 평야 지역은 후기 플라이스토세 동안 형성된 하안단구 퇴적층이 비교적 하도와 먼 지역에 분포하고 있다. 홀로세 퇴적층은 자갈층이 평야 일대에 걸쳐 넓게 분포하고 있어 홀로세 중기 이후 해수면이 안정화 된 다음에 망상하천의 형태로 퇴적된 것으로 해석했다. 그리고 현재와 같은 사행하천으로 전이되면서 주로 모래가 우세한 퇴적물이 유입되었으며, 하도 주변으로는 범람원 환경이 조성되었다. 또한 화분분석 결과를 근거로 약 6천년 전 후에는 온난 습윤한 환경이었으며 이후 습지 퇴적층이 발달한 것으로 추정했다. 담양습지가 분포하는 하도 일대의 트렌치 조사 결과 하부에는 원마도가 좋은 자갈층이 분포하고 있으며, 자갈층 상부를 모래층이 덮고 있다. 담양습지는 1970년대 담양호가 건설된 이후 모래층 상부에 머드층이 퇴적되면서 형성된 것으로 추정된다.

        Thrips worms and root-knot nematodes occur in a variety of crops, and have shown a great deal of damage to farm income every year, and the damage is increasing every year. In order to solve these problems, a variety of biological materials are used in Korea to develop a control agent. However, there are very few products available that can satisfy the consumer's satisfactory control effect, efficacy, formulation stability and pesticide compatibility. In order to propose a biological control solution to these problems, this study was conducted to develop the optimal bioprocess technology and formulations suitable for the material by transferring the Aspergillus nigerF22 strain, which is effective for root-knot nematodes, at Chonnam National University. This study was conducted to investigate the efficacy of Aspergillus niger F22 20% suspension concentrate (Productname:NEMAFREE), which has excellent efficacy on root nematodes. The packing test result showed about 70-90% control effect. Soil fumigation and disinfection treatments after 4 days of planting were effective. In addition, we have developed a product to control the under powder pupa using Beauveria bassiana ERL836, an insect pathogenic microorganism, which has excellent control effect against resistant insect pupa. The main purpose of this study was to investigate the effect of insect pests on the under poor control of the pupa in the soil. In the pavement test, more than 70%(GR) formulation, which can be treated withch emical pesticides, and it is confirmed that synergy effect is in the control of Thrips worm.

        Oxalic acid has a nematicidal activity against the root-knot nematode Meloidogyne incognita. High producer of oxalic acid was isolated, and then named as Aspergillus niger F22. Oxalic acid production was investigated under various temperatures from 20 – 33oC and rotational speeds in 5 L jar fermenters. Yield of oxalic acid increased with decreasing temperature. The highest yield was obtained at 23oC, showing the yield of oxalic acid of 8.7 g/L, whereas oxalic acid production was least at 33oC. At 20oC, the yield was lower than that of 23oC. At a rotational speed of 300 rpm, serious oxygen depletion was present from 48 - 72 h, resulting in low productivity of 26.2 mg /L·h. When a rotational speed was set at 600 rpm, dissolved oxygen tension was over 40% and oxalic acid production increased up to approximately 55%. Viscosity during the culture differed with temperatures. Viscosity increased with the increment of temperatures. When A. niger F22 was cultured at 23oC, viscosity was 810 cP, which was favorable for oxalic acid production.

        To find an alternative for synthetic pesticides, methanol extract from plant samples were tested for their insecticidal activity against insect. The extract of Asiasarum sieboldii had strongly insecticidal activity against Plutella xylostella. Roots of A. sieboldii were extracted with methanol, and the concentrated extract was partitioned with n-hexane, ethylacetate, n-buthanol and H2O. The highest activity was shown in the hexane fraction. Activity-guided fractionation led to the isolation of two amides from hexane fration through the repeated silica gel column chromatographic separations. From the interpretation of spectropic data including NMR, MS, IR, the chemical structures of compounds were determined as dodeca-2E,4E,8Z,10Z-tetraenoic acid isobutylamide and dodeca-2E,4E,8Z, 10E-tetraenoic acid isobutylamide. These compounds showed insecticidal activity on P. xylostella by 96.7% at 100ppm. The liquid formulation controlled on cabbage effectively. The extract and compounds from A. sieboldii showed insecticidal activity against Nilaparvata lugens. As a naturally occurring pesticide, A. sieboldii could be useful as a new botanic insecticide.

        A new Bacillus thuringiensis isolate 19-22 (Bt 19-22) exhibited high anti-fungal activity against barley powdery mildew (Blumeria graminis f. sp. hordei). The cry gene content of Bt 19-22 comprised cry1Aa, cry1Ab, cry1Ac and cry1D which have high insecticidal activity against lepidopteran larvae. We tried to confer a dipteran insecticidal activity to Bt 19-22 for constructing a recombinant strain which has multiple functions, anti-fungal and dual insecticidal activity. The insecticidal cry11Aa gene of B. thuringiensis was constructed under cry1Ac promoter in an E. coli-B. thuringiensis shuttle vector (pPro11A). The plasmid, pPro11A was introduced into Bt 19-22 isolate by electroporation and four transformants which had different cry gene contents were identified by PCR with cry11Aa and cry1-type specific primers. Among them, a Bt 19-22 transformant (11A/19-22 No. 7) expressed Cry11A protein (approximately 70 kDa) successfully without change of its inherent characteristics such as Cry protein expression and antifungal activity. The insecticidal activity of 11A/19-22 No. 7 was checked against Plutella xylostella and Culex pipiens. These results suggests that the recombinant strain shows dual insecticidal activity against lepidopteran and dipteran larvae as well as antifungal activity.

        A new Bacillus subtilis isolate showed high anti-fungal activities (more than 80% control efficacy) against several plant diseases such as rice blast (Magnaporthe grisea), tomato gray mold (Botrytis cinerea), tomato late blight (Phytophthora infestans) and wheat leaf rust (Puccinia recondita). We tried to confer an insecticidal activity to this B. subtilis isolate for constructing a recombinant strain which has dual functions, anti-fungal and insecticidal activity. The insecticidal cry1Ac gene of B. thuringiensis was constructed under its own promoter in a minimal E. coli-B. thuringiensis shuttle vector (pHT1K-1Ac). The plasmid, pHT1K-1Ac was introduced into B. subtilis isolate by electroporation and the transformant was confirmed by PCR with cry1Ac specific primers. B. subtilis transformant produced a parasporal inclusion in the cells as in B. thuringiensis and the size of that protein was appox. 130 kDa. The insecticidal activity of the transformant was checked against lepidopteran pest, Plutella xylostella. This result suggests that this recombinant B. subtilis strain shows the possibility of controlling harmful insect pests as well as plant fungal diseases simultaneously at one crop, and both culture broth and harvested cells of this strain can be used as individual biological control agents separately for integrated crop protection.
