        Oral bacterial infections substantially affect the development of various periodontal diseases and oral cancers. However, the molecular mechanisms underlying the association between Fusobacterium nucleatum (F. nucleatum ), a major periodontitis (PT)-associated pathogen, and these diseases require extensive research. Previously, our RNAsequencing analysis identified a few hundred differentially expressed genes in patients with PT and peri-implantitis (PI) than in healthy controls. Thus, in the present study using oral squamous cell carcinoma (OSCC) cells, we aimed to evaluate the effect of F. nucleatum infection on genes that are differentially regulated in patients with PT and PI. Human oral squamous cell carcinoma cell lines OSC-2O, HSC-4, and HN22 were used. These cells were infected with F. nucleatum at a multiplicity of infection of 100 for 3 hours at 37℃ in 5% CO2. Gene expression was then measured using reverse-transcription polymerase chain reaction. Among 18 genes tested, the expression of CSF3, an inflammation-related cytokine, was increased by F. nucleatum infection. Additionally, F. nucleatum infection increased the phosphorylation of AKT, p38 MAPK, and JNK in OSC-20 cells. Treatment with p38 MAPK (SB202190) and JNK (SP600125) inhibitors reduced the enhanced CSF3 expression induced by F. nucleatum infection. Overall, this study demonstrated that F. nucleatum promotes CSF3 expression in OSCC cells through p38 MAPK and JNK signaling pathways, suggesting that p38 MAPK and JNK inhibitors may help treat F. nucleatum-related periodontal diseases by suppressing CSF3 expression.

        Preserving intact genetic material and delivering it to the next generation are the most significant tasks of living organisms. The integrity of DNA sequences is under constant threat from endogenous and exogenous factors. The accumulation of damaged or incompletely-repaired DNA can cause serious problems in cells, including cell death or cancer development. Various DNA damage detection systems and repair mechanisms have evolved at the cellular level. Although the mechanisms of these responses have been extensively studied, the global RNA expression profiles associated with genomic instability are not well-known. To detect global gene expression changes under different DNA damage and hypoxic conditions, we performed RNA-seq after treating human cervical cancer cells with ionizing radiation (IR), hydroxyurea, mitomycin C (MMC), or 1% O2 (hypoxia). Results showed that the expression of 184–1037 genes was altered by each stimulus. We found that the expression of 51 genes changed under IR, MMC, and hypoxia. These findings revealed damage-specific genes that varied differently according to each stimulus and common genes that are universally altered in genetic instability.

        In the present work, a new hydrogen added argon heat treatment process that prevents the formation of hydrides and eliminates the dehydrogenation step, is developed. Dissolved hydrogen has a good effect on sintering properties such as oxidation resistance and density of greens. This process can also reduce costs and processing time. In the experiment, commercially available Ti-6Al-4V powders are used. The powders are annealed using tube furnace in an argon atmosphere at 700oC and 900oC for 120 min. Hydrogen was injected temporarily during argon annealing to dissolve hydrogen, and a dehydrogenation process was performed simultaneously under an argon-only atmosphere. Without hydride formation, hydrogen was dissolved in the Ti-6Al-4V powder by X-ray diffraction and gas analysis. Hydrogen is first solubilized on the beta phase and expanded the beta phases’ cell volume. TGA analysis was carried out to evaluate the oxidation resistance, and it is confirmed that hydrogen-dissolved Ti-6Al-4V powders improves oxidation resistance more than raw materials.

        Sea Buckthorn (Hippophae rhamnoides L.) has been used in traditional medicine for the treatment of cough, indigestion, circulatory problems and pain. The associated anti-inflammatory effect of this agent is achieved via the inhibition of Nf-kB signaling, a property that has been demonstrated to effectively control the symptoms of various skin disorders, including atopic dermatitis. Accordingly, the purpose of this study was to assess the efficacy of Sea Buckthorn in reducing the production of lipopolysaccharide (LPS) activated nitric oxide (NO) by inhibiting the Nf-kB pathway, as measured by the symptoms of atopic dermatitis (AD) occurring secondarily to inflammation and immune dysregulation. Our data demonstrate that Sea Buckthorn significantly decreased the LPS-induced production of NO (p〈0.001). Atopic dermatitis was induced by repeated application of 2,4-dinitrochlorobenzene to the dorsal skin of mice. Topical application of 5% Sea Buckthorn extract improved the symptoms of AD, specifically reducing disease severity scores, scratching behaviors and epidermal thickness. When compared to the control group, animals treated with Sea Buckthorn exhibited increased serum IL-12 levels and decreased serum TNF-α, IL-4 and IL-5 levels. Such a modulation of biphasic T-helper (Th)1/Th2 cytokines may result in a reduction in serum IgE levels. Our findings suggest that mechanism of action of Sea Buckthorn in the treatment of AD is associated with a marked anti-inflammatory effect as well as an inhibition of Th2-mediated IgE overproduction via the modulation of biphasic Th1/Th2 cytokines. Such results suggest that topical Sea Buckthorn extract may prove to be a novel therapy for AD symptoms with few side effects.

        There are many species as bioenergy crops and have different cold sensitivity in each. Cold-tolerant camelina and rapeseed, -sensitive jatropha, were used to investigate the cold stress response. Various physiological parameters such as leaf length, width, electrolyte leakage, stomatal conductance and chlorophyll fluorescence were measured to determine the growth rate treated with cold (2℃) for 5 days. Cold treated jatropha was damaged seriousiy but camelina and rapeseed were withstand. In order to investigate the cold-response on plasma membrane H+-ATPase activity isolated from leaves and roots of camelina, rapeseed and jatropha crops were exposed to cold stress. There were an increase in the activity of leaves and roots plasma membrane in cold-tolerant crops (camelina, rapeseed) while decreased the activity in cold-sensitive crop (jatropha). By western-blot analyses, the protein expression of plasma membrane H+-ATPase isolated from leaves and roots of camelina and rapeseed was increased in the presence of cold stress, but not in jatropha. These results may suggest that increased plasma membrane H+-ATPase of crops are closely related with cold-tolerant.

        Aluminum (Al) toxicity in plants is one of the major limitations to crop growth on acid soils. The Al-induced change of H+-ATPase expression has been regarded as an important mechanism for Al tolerance in soybean. To investigate whether translational regulation of plasma membrane H+-ATPase is involved in the response to Al stress, we conducted western - blot of this protein. The results show that western - blot of plasma membrane H+-ATPase in the "Sowon" (Al tolernace) significantly increased in translational expression level, while citric acid (50 μM) with Al (50 μM) treatment has not effected. In contrast, Al sensitive cultivar "Poongsannamool" inhibited expression level of plasma membrane H+-ATPase with Al treatment. Two - dimensional gel analysis were performed to determine the protein induction patterns of control and Al (50 μM, 24 h) treated soybean. There are many changes of plasma membrane proteins in both cultivars under Al stress. Especially "Sowon" was significantly enhanced the expression of the plasma membrane H+-ATPase in Al treatment. But protein expression of "Poongsannamool" was less than "Sowon". These results suggest that the regulatory role of plasma membrane H+-ATPase may involved the tolerance mechanism in soybean roots. At the present, transcriptional level of H+-ATPase is under investigation.
