        A winter forage tetraploid rye (Secale cereale L.) cultivar, ‘Daegokgreen’, was developed at the Department of Central Area Crop Science, NICS, RDA in 2016. The mutant line ‘CG11003-8-B’, which was induced from rye cultivar ‘Gogu’ (diploid) by colchicine treatment, was selected for its excellent agronomic performance and was placed in preliminary yield trials for one year, 2013. The line was designated “Homil59” and was tested for regional yield trials at the four locations in Korea from 2014 to 2016. Finally, the new cultivar was named as the ‘Daegokgreen’ (grant number 8274). The leaf of cultivar ‘Daegokgreen’ is wide, long and dark-green color. The cultivar also has a big-size grain with light-brown color. The heading date of cultivar ‘Daegokgreen’ was April 17 which was 2 days later than that of check cultivar ‘Gogu’. The tolerance to cold and wet injury, and resistance to powdery mildew and leaf rust of the new cultivar were similar to those of the check cultivar but the resistance to the lodging of the new cultivar was stronger than that of the check. The average roughage fresh and dry matter yield of the new cultivar after 10 days from heading were 37.0 and 7.7 MT ha-1, respectively, which were similar to those (38.4 and 8.0 MT ha-1) of the check cultivar. The roughage quality of ‘Daegokgreen’ was higher in crude protein content (8.9%) than that of the check cultivar (7.9%), while was similar to the check in total digestible nutrients (56.9%). This cultivar is recommended for fall sowing forage crops at all of crop cultivation areas in Korea.

        This experiment was performed to analyze the annual variation of growth traits of soybean to determine the information of less sensitive traits and stable cultivars based on the environmental conditions. Sowing was carried out on June 2 for each year and the test plot was arranged in three replicates by randomized complete block design with thirty soybean varieties including two landraces during the period 2014-2015. The weather conditions during the test period were quite different with extremely low precipitation and longer sunshine duration in 2015. The variation of characteristics related to growth period such as days of growth, days of maturity, days of flowering and 100-seed weight were less in spite of different environmental conditions. While the variation of the number of pods per plant was high. Considering growth and seed characteristics like the number of pods per plant, the number of seeds per pod, and 100-seed weight which are linked directly to the yield, cultivars such as Shingi, Daewonkong, Danbaegkong, and Daepung for beancurd and soypaste, Pungwon, Haepum and Shingang for soy-sprout and Seoritae for cooking with rice were more stable and are expected to have high yield in Suwon, the south-central part of South Korea. These results are useful for the selection of breeding resources to develop cultivars with high stability under changeable weather condition.

        This experiment was conducted to evaluate feed value and usability of soybean varieties as livestock forage. In this study, three soybean cultivars, OT93-26, Geomjeongsaeol, and Pungwon, were harvested at R5 (beginning seed development)- and R6 (full seed)-reproductive stages for analyzing feed value of soybean. Days to R5 stage harvest of OT93-26 among the three soybean cultivars was 55 days and the shortest while Pungwon took 103 days to reach at R6 stage. The R6-harvested soybeans had higher dry matter (DM) yields and crude protein (CP) content than the R5-harvested. However, both DM and CP were the highest in the R6-harvested Geomjeongsaeol. Contents of neutral detergent fiber (nDF) and acid detergent fiber (ADF) of Pungwon harvested at R5 were the highest whereas the R6-harvested Geomjeongsaeol had the lowest. Digestible dry matter (DDM), dry matter intake (DMI), and relative feed value (RFV) of the R6-harvested Geomjeongsaeol and Pungwon were higher than those of the R5-harvested, but in case of OT93-26, those at R6 stage were low rather than those at R5 stage. However, soybean could be used as alternative forage with high feed value for livestock. Taken together, Geomjeongsaeol could be used for developing new forage soybean varieties with high feed value, and R6 would be the optimum harvesting stage for yield and quality of forage soybean.

        Phosphorus (P) is an important structural component and plays critical roles in the process of energy transfer and signal transduction. Effect of low P on carbohydrate metabolism was investigated at the transcription level via transcriptome analysis using the rice 60K oligonucleotide DNA microarrays. Two-week-old rice seedlings were grown under a low (32 μM) or high (320 μM) P condition for two weeks and leaves from the seedlings were used for transcriptome analysis. Expression of genes involved in carbohydrate metabolic pathways (eg. glycolysis, sucrose degradation and starch synthesis and degradation) was most significantly affected under low P. Under low P, most genes involved in glycolysis were intensively down-regulated, genes of starch biosynthesis and degradation pathway were up- or down-regulated, and many genes involved in sucrose biosynthesis were intensively up-regulated. In leaves under low P, glucose and pyruvate levels decreased, but sucrose and starch levels increased. These results suggest that carbohydrate metabolism is adjusted primarily through comprehensive transcriptional modulation of genes involved in the carbohydrate metabolic super-pathway.

        Low temperature stress is one of the major negative factors affecting vegetative and reproductive growth of rice. To better understand responses of rice plants to low temperature we analyzed transcriptome expression patterns in glumous flower of cold-tolerant japonica rice variety, Stejaree45, and cold-susceptible variety, HR19621-AC6 at booting stage under cold water irrigation. A total of 2,411 probes were differentially expressed by low temperature in glumous flowers of the two varieties. Some important genes involved in hormone biosynthesis showed variety-specific regulation. Expression of GA20ox3 and GA2ox, among the genes involved in GA biosynthesis, was regulated differentially in the two varieties. Among the genes involved in IAA biosynthesis, YUCCA1 and TAA1:1 showed variety-specific regulation. Among the genes involved in cytokinin biosynthsis and signaling, expression of LOG, HK1 and HK3 was significantly down-regulated only in the cold-susceptible variety. Among the genes involved in ABA biosynthesis, NSY and AAO3 were down-regulated only in the cold-tolerant variety. In general, genes involved in GA, IAA and cytokinin biosynthesis responded to cold temperature in such a way that capacity of those bioactive hormones is maintained at relatively higher levels under cold temperature in the cold-tolerant variety, which can help minimize cold stress imposed to developing reproductive organs in the cold-tolerant variety.

        Ascorbate peroxidase (APX) plays a crucial role in the detoxification of hydrogen peroxide. APX activity is maintained significantly higher in the paraquat­treated leaves of the paraquat-tolerant Rehmannia glutinos. This study was conducted to understand structural and regulatory characteristics of APX gene in R. glutinosa. A putative APX cDNA clone (RgAPX1) was isolated from a leaf cDNA library using a partially sequenced expressed sequence tag clone. RgAPX1 is consisted of 1148 bp nucleotides and contains an open reading frame encoding a 250 amino acid-long polypeptide. Deduced RgAPX1 amino acid sequence shares higher sequence similarity to cytosolic APXs. RgAPX1. expression was higher in the leaf than in the flower and root. Southern blot result indicates the presence of one or two RgAPX1-related genes in R. glutinosa genome. RgAPX1 transcription was affected differentially by various stresses and phytohormone. The results indicate that RgAPXl is constitutively expressed in most tissues and its expression is modulated for the immediate and efficient detoxification of H2O2 under normal and stress conditions.

        Phosphorus (P) is a macronutrient playing important roles in many plant processes. Significant interest has been devoted to search and utilize genotypic variations in P use efficiency in rice but with little effort to understand its physiological and biochemical bases. In this study, we examined responses to P deprivation of some primary and secondary traits in 3-week-old seedlings of the three genotypes, Sobi-byeo (japonica), Dasan-byeo (japonica ~times indica) and Palawan (indica). In general, percent weight due to root was increased up to 26%, but amounts of root protein and proteins secreted from roots were decreased by 11 to 19% and 31 to 51 %, respectively, by 3 to 21 days of P deprivation in the three genotypes. Interestingly, however, responses of Palawan to short-term P deprivation were contrasting to those of Dasan-byeo and Sobi-byeo in seedling weight and contents of shoot protein, chlorophyll and anthocyanin. Seedling weight was not decreased, but shoot protein content was decreased in P-deprived seedlings of Palawan. Contents of chlorophyll in leaves and anthocynin in roots were increased in Dasan-byeo and Sobi-byeo, but decreased in Palawan. The results suggest that responses of protein and pigment synthesis to P deficiency are different in modem and traditional varieties and the difference may at least in part be due to the selection for high yield under highly fertilized conditions.

        The content and composition of tocopherol were analyzed in peanuts produced in Korea. The tocopherol assay was consisted of an extraction with n-hex-ane:isopropanol mixture followed by high pressure liquid chromatography with fluorescence detection. Tocopherol content was higher in leaves and seeds but lower in seed coats and shells. α -/~gamma - Tocopherol ratio was as low as 0.53 in seeds and as high as 22.5 in shells. Tocopherol content in seeds of nine varieties ranged from 142 to 220 ng/mg dry weight, and the α -/~gamma -tocopherol ratio from 0.40 to 0.75. Tocopherol content decreased by 18% but α -/~gamma -tocopherol ratio increased by 44% in roasted seeds. The results indicate that the level of tocopherol in Korean peanut varieties is moderate and the ratio of α - to ~gamma -tocopherol is low.
