        Phasianus colchicus is not only a beautiful bird but also a great value in science and under the threat of endanger. Hence, the aim of this study was to isolate the pheasant male germ cells (mGCs) and then induce them into elongated sperm-like cells in vitro. The mGCs were purified and enriched by a two-step plating method based on the different adherence velocities of mGCs and somatic cells. The percentage of the c-kit positive cells and c-kit negative cells examined by flow cytometry analysis (FCA) was 92.87% and 2.57%, respectively. Subsequently, the mGCs were induced for 48h in DMEM/F12 medium supplemented factors such as retinol acid, testosterone and bovine FSH, followed by 5 weeks in culture. We found that some elongated sperm-like cells appeared initially in vitro under inducement of stimulated factors. The elongated sperm-like cells showed in the expression of changed morphology and post-transcriptional marker such as spermatid associated (SPERT), spermatid perinuclear RNA binding protein (STRBP), round spermatid basic protein 1 (RSBN1) and SPER1L. Moreover, in DNA content identified assay, induced cells showed that the 1C DNA population markedly increased in differentiated group but it was not change in undifferentiated group. Successful in vitro differentiation of pheasant testicular germline cells into spermatids appears to offer extremely attractive potential for the conservation of endangered birds and treatment of male infertility.

        Selection based on phenotype in the traditional manner does not help in tracking the selected gene. Molecular genetics has revolutionized the old established breeding techniques. A new epoch of molecular markers has been acquainted for genetic improvement of livestock. This study is engaged on the neoteric molecular markers used in various fields of livestock. DNA markers are more encouraging in selecting genomes that have recombination events close to the target gene. Molecular markers rely on DNA assay and are better than the morphological and biochemical markers. In this study DNA-based molecular markers developed during the past decagons for animal genome analysis are reviewed. Mapping of molecular markers provide a framework, required for its subsequent use in the selection procedure. Molecular techniques help in the utilization of genetic variability in breeding population. At the same time livestock genomes play important role in human genomics and their role for understanding human genomics cannot be overlooked. Recently, in the epigenetic and transcriptomic studies, RNA sequencing as a part of next generation sequencing has revolutionized the approach and ongoing trends of analysis. Keeping in mind the goals, these molecular techniques can be implemented successfully by following well defined, crisp and integrated strategy.

        The recent development in genetic assisted selection (combining traditional- and genome assisted selection method) and reproduction technologies will allow multiplying elite cow in Hanwoo small farm. This review describes the new context and corresponding needs for genome assisted selection schemes and how reproductive technologies can be incorporated to get more genetic gain for cow genetic improvement in Hanwoo. New improved massive phenotypes and pedigree information are being generated from commercial farm sector and these are allowing to do genetic evaluation using BLUP to get elite cows in Korea. Moreover cattle genome information can now be incorporated into breeding program. In this context, this review will discuss about combining the reproductive techniques (Multiple Ovulation Embryo Transfer; MOET) and genome assisted selection method to get more genetic gain in Hanwoo breeding program. Finally, how these technologies can be used for multiplication of elite cow in small farm was discussed.

        This study aims to identify DNA marker related to health index which is derived from fatty acid composition of Hanwoo meat. We investigated a genetic association between two SNPs (-867G>C and 878C>T) of SCD gene and health indexes. Two health index values (index of atherogenicity and index of thrombogenicity) were derived from a combination of fatty acid composition. Phenotypic correlation indicated that oleic acid (C18:1) was negatively correlated to index of atherogenicity (-0.84) and index of thrombogenicity (-0.91), respectively. Statistical analysis revealed that 878T>C SNP was significantly associated with IA (index of atherogenicity, p=0.012) and IT (index of thrombogenicity, p=0.006). There was no association between the regulatory SNPs (-867G>C and -877Gdel) and health indexes. Haplotype analysis detected 4 main haplotype (GdelT; 0.004, GdelC; 0.344, CGT; 0.350 and CGC; 0.261) in Hanwoo. The GdelT haplotype was significant on IA and IT. The effect of GdelT haplotype showed increasing IA and IT values, while GdelC haplotype has a decreasing IA and IT value in Hanwoo. In conclusion, the 878C>T SNP in the SCD gene seems to have an effect on this health index and might be implemented into animal breeding program.

        The purpose of this study was to investigate the distribution of trig˗ ger points(TrPs) on athletes with various sporting background. To achieve the purpose, a study was carried out through a survey from 180 athletes involved in 6 selected sports at Yong-In University. Selected sports included Judo, Taekwondo, Kendo, Ssi-reum(Korean traditional wrestling), Boxing, and Golf. An interview type survey and physical examination were conducted with each thirty athletes from each of the selected sports groups. Technical statistic(SPSS 15.0) was used to analyze the distribution of TrPs on these athletes. The most common TrPs observed in muscles of Trapezius, Quadratus Lumborum, Quadriceps in Judo. In Taekwondo, it was on the trapez˗ ius and triceps surae. Kendo athletes had TrPs at sites of trapezius, brachioradialis and triceps surae. Ssirem athletes were found to have TrPs on trapezius, deltoid and quadrates lumborum. In boxers, TrPs appearing at trapezius and brachioradialis were observed. Finally, Golf players were seen to have TrPs at trapezius, quadrates lumbo˗ rum and brachioradialis. Hence, the analysis shows that there are significant differences of the distribution of TrPs according to the different sport items of the athlete.

        Potato (Solamum tuberosum 'Dejima') plantlets were investigated on culture type and initial quantity of inoculation in bioreactor and survival rate by hydroponics for mass production. rode stems (1 to 1.5cm in length) of potato plantlets multiplied in vitro were grown for 3 weeks in liquid Murashige and Skoog (MS) medium with sucrose 30 g L-1. When plantlets (80-node inoculation) were raised in 10L balloon type bubble (BB) bioreactor, the healthiest growth of plantlets was obtained from explants cultured in ebb & flow culture with medium supplied periodically 12 times per day. The suitable inoculation quantity of 20L BB bioreactor was 120 pieces of stem segments (mean 2.2g fresh weight) in ebb & flow culture. Number of nodal shoot was eight on the average. In controlled culture room, survival rate of plantlets at 7 days after stem cutting was above 70% when they were acclimatized by hydroponics grown in deep flow and solid medium culture. The highest survival rate of the stem cutting plantlets was in nutrient solution adjusted to EC 1.4dS·m-1. Stem cutting plantlets through one culture could be obtained 670~900, when plantlets were grown in ebb & flow culture during 3 weeks using a 20L bioreactor with initial 120 pieces of nodal segments. 11 is possible In do mass production of seedlings cultured in bioreactor and hydroponics.

        원형질체 융합에 의한 화합성 및 불화합성 종간 체세포잡종을 얻었다. 화합성 종간인 Pleurotus ostreatus 와 P. florida 의 융합체는 이질핵체 (heterokaryon) 를 형성하였고, 불화합성 종간인 P. cornucopiae + P. florida , P. ostreatus + Ganoderma applanatum, P. florida + Ganoderma lucidum, 그리고 P. ostreatus + Flammulina velutipes 는 합핵체(synkaryon) 를 형성하였다. 이질이핵체는 동일한 양상의 자실체를 형성하는데 비해 합핵체는 유사분열상의 꺽쇠연결체 형성, 한쪽 친과 유사한 자실체 형성, 비정상적 유전형질 분리 및 유전자재조합 현상을 나타내었다. 화합성 및 불화합성 계통간 융합체의 RAPD 분석결과 화합성 종간 융합체는 동일한 DNA 패턴을 나타내었고, 불화합성 종간 융합체는 한쪽 친과 유사한 DNA 양상이면서 비양친 DNA 밴드도 형성하였다. 합핵체의 패턴은 microgenome insertion type 과 macrogenome insertion type 으로 구분되었다. 합핵체의 자실체 발생은 융합 모균주 양친의 자가임성에 의존하는데 이는 느타리의 동형핵체 자가임성과 유사한 양상이었고, 교배형 전환과 관련이 있는 것으로 사료된다. 여기서는 이러한 관점에서 논할 것이다.

        Olive flounder Paralichthys olivaceus is one of the most widely cultured fish species in Korea. Although olive flounder receive attention from aquaculture and fisheries and extensive research has been conducted eye morphological change in metamorphosis, but little information was known to molecular mechanism and gene expression of eye development- related genes during the early part of eye formation period. For the reason of eyesight is the most important sense in flounder larvae to search prey, the screening and identification of expressed genes in the eye will provide useful insight into the molecular regulation mechanism of eye development in olive flounder. Through the search of an olive flounder DNA database of expressed sequence tags (EST), we found a partial sequence that was similar to crystallin beta A1 and gamma S. Microscopic observation of retinal formation correspond with the time of expression of the crystallin beta A1 and gamma S gene in the developmental stage, these result suggesting that beta A1 and gamma S play a vital role in the remodeling of the retina during eye development. The expression of crystallin beta A1 and gamma S were obviously strong in eye at all tested developing stage, it is also hypothesized that crystallin acts as a molecular chaperone to prevent protein aggregation during maturation and aging in the eye.
