        The whitebacked planthopper (WBPH) is one of the most serious insect pests of rice. The nymphs and adults suck the phloem sap which causes reduced plant vigor, stunting, yellowing of leaves. We employed a mapping population composed of 113 doubled haploid lines (DHLs), derived from a cross between resistant cultivar ‘Cheongcheong’ and susceptible cultivar ‘Nagdong’, to identify the quantitative trait loci (QTL) associated with WBPH resistance. The DH population was evaluated for resistance to WBPH using seed box screening test. The WBPH resistance of each lines or plants was estimated when susceptible check ‘Nagdong’ was killed using 1-9 scale according to Standard Evaluation System for Rice (IRRI, 1988). A linkage map was constructed with 119 polymorphic microsatellite markers using the Joinmap 4 software program to adjust for segregation distortion. Transgressive segregation was observed on WBPH resistance. Two significant QTLs conferring resistance to WBPH, qwbph6.1 and qwbph6.2, were detected in the regions of RM588-RM276 and RM527-RM528 on chromosome 6 with LOD scores of 3.7 and 2.4, respectively. They collectively explained 55% of phenotypic variation. These two QTLs have large effects on WBPH resistance and may be useful for establishment of marker assisted selection (MAS) in the rice breeding programs.

        We conducted a QTL analysis of grain quality traits using 117 BC3F4 and BC3F5 lines developed from a cross between Ilpumbyeo and Moroberekan. Genotypes of 117 BC3F5 lines were determined using 134 simple sequence repeat (SSR) markers. A linkage map constructed using 134 SSR markers was employed to characterize quantitative trait loci (QTL). The 117 BC3F4 and BC3F5 lines were evaluated for eleven grain quality traits in 2005 and 2006. A total of 18 QTLs were identified for eleven traits, and the phenotypic variance explained by each QTL ranged from 9.9% to 35.2%. Moroberekan alleles contributed positive effects in the Ilpumbyeo background at two QTL loci for 1,000 grain weight. Four QTLs, two for chalky rice and one each for 1,000 grain weight and head rice were consistently detected in two consecutive years indicating that these QTLs are stable. Clusters of QTLs were observed in three chromosome regions. One cluster harboring five QTLs including head rice and brown rice ratio near SSR markers RM190 and RM314 was detected on chromosome 6. Another cluster harboring grain weight and white belly was detected on chromosome 2. Increase in white belly at this locus might be due to the increase in grain weight due to the presence of the Moroberekan allele. The Moroberekan alleles at two QTL loci, gw3 and gw4 associated with increased grain weight might be useful in breeding programs to develop high-yielding cultivars.

        We conducted a QTL analysis of agronomic traits using 117 BC3F5 and BC3F6 lines developed from a cross between Ilpumbyeo and Moroberekan. Genotypes of 117 BC3F5 lines were determined using 134 simple sequence repeat (SSR) markers. A total of 832 Moroberekan chromosome segments with 410 homozygous and 422 heterozygous, respectively, were detected, and the genetic distance of introgression segments ranged from 0.5 cm to 112.1 cm. A linkage map constructed using 134 SSR markers was employed to characterize quantitative trait loci (QTL). The 117 BC3F5 and BC3F6 lines were evaluated for seven agronomic traits at two locations in 2006 and 2007 and at one location in 2007. A total of 26 QTLs were identified for seven traits including days to heading, and the phenotypic variance explained by each QTL ranged from 9.2% to 24.2%. Moroberekan alleles contributed positive effects in the Ilpumbyeo background at eleven QTL loci including panicle length and spikelets per panicle. Five QTLs, two for days to heading and one each for culm length, panicle length and spikelets per panicle were consistently detected in every occasions indicating that these QTLs are stable. Among them, two QTLs, spp6 for spikelets per panicle and pl6 for paniclel length were localized in the similar region. Increase in spikelets per panicle at this locus might be due to the increase in panicle length, because both traits were associated with increase in spikelets per panicle and panicle length due to the presence of the Moroberekan allele. These Moroberekan QTLs might be useful in breeding programs to develop high-yielding cultivars.

        IL-34 (NIL) developed by introgressing chromosomal segment substitution from an accession of Oryza minuta (2n=48, BBCC, Acc. No. 101141) into the O. sativa subsp. japonica cv. Hwaseongbyeo, showed significantly higher number of spikelets per panicle (SSP) than the recurrent parent Hwaseongbyeo. QTL analysis in F2 generation derived from the cross between IL-34 and Hwaseongbyeo revealed that ssp7, a QTL was located in the pericentromeric region of chromosome 7. The frequency distribution of spikelets per panicle followed 3:1 ratio for single locus segregation. The additive effect of the O. minuta allele at the QTL was 23 spikelets per panicle, and 43.6% of the phenotypic variance could be explained by the segregation of marker RM21596. To clarify whether ssp7 could be dissected genetically, we carried out fine-scale mapping with 3,700 F2 plants derived from the cross between IL-34 and Hwaseongbyeo using markers flanking spp7. 186 F2 plants having informative recombination breakpoints within the region flanked by two SSR markers RM500 and RM21615 were identified and used for fine mapping of ssp7. ssp7 was mapped between the SSR markers RM21596 and RM418 which was approximately 441kb in length based on the physical map of the region. Of great interests, the QTL region also had effects on primary branch number (PB), grains per panicle (SP) and grain yield (YD). These results are very useful for transferring or pyramiding ssp7 by molecular marker assistant selection in rice breeding programs.

        A population of 120 double-haploid lines derived from Samgang/Nagdong was used for analyzing the developmental behaviors of plant height and tiller number by conditional mapping via composite interval approaches at five growth stages of rice. The traits were normally distributed with transgressive segregations in both directions. A total of 4 QTLs associated with plant height were identified in 40th/initial, 50th/40th and 60th/50th periods, respectively. The qph7.1 was observed in 50th/40th and 60th/50th periods, with increasing effects from Samgang and Nagdong alleles, respectively. This indicated that an individual gene or genes at the same genomic region might have opposite genetic effects at various growth stages. Five QTLs associated with tiller number were identified in four stages except 60th/50th stage. Four of five increasing effects of QTLs were from Nagdong alleles. Six of nine QTLs for two traits were detected before 60 days. It was evident that the expression of QTLs for plant height and tiller number was more active at early stages than at maturity stage. Furthermore, QTLs identified at early stages showed opposite additive effects on plant height and tiller number. These results provided a possible genetic explanation for the negative correlation between two traits.

        Most soybean cultivars have ovate leaflets, although a few cultivars have narrow or lanceolate leaflets. Narrow leaflet cultivars tend to have more seeds produced per pod than ovate leaflet cultivars, suggesting that the narrow leaflet trait is tightly linked to or cosegregating with the trait controlling number of seeds produced per pod (nspp). The objective of this study was to construct a high resolution map of a chromosomal region controlling narrow leaflet trait and nspp in soybean. A BC3F2 population from a cross between 'Sowonkong' and 'V94-5152' was used. Sowonkong have narrow leaflet and 4-seeded pod and V94-5152 have ovate leaflet and do not have 4-seeded pod. The plants of F2 populations showed a segregation ratio of 3:1 of ovate leaflet to narrow leaflet and then leaflet genotypes were obtained from F2:3 population of each F2 individual. The narrow leaflet-containing plants showed Sowonkong-like pod trait and the ovate leaflet-containing plants V94-5152-like pod trait. The results suggested that, in our mapping population, a single gene controls inheritance of the narrow leaflet character and the narrow leaflet trait is tightly linked to or co-segregating with the trait controlling nspp. Subsequently, we mapped the narrow leaflet locus near Sat_105, Satt270 and SM315 on soybean linkage group I that corresponds with the previously known ln locus. Work is ongoing to construct a fine molecular genetic linkage map on this MLG I region and to find a linkage relationship between ln and nspp. Our results should facilitate further elucidation for the relationship between ln/nspp and yield.

        In the previous study, 141 BC3F2 lines from a cross between the Oryza sativa cv. Milyang 23 and O. glaberrima were used to identify favorable wild QTL alleles for yield component traits. In this study, we carried out QTL analysis of four grain morphology as well as four yield component traits using 141 BC3F5 lines from the same cross and compared QTLs detected in two different generations. The mean number of O. glaberrima segments in the 141 BC3F5 lines ranged from 1 to 13 with 2.69 and 5.71 of the average means of homozygous and heterozygous segments, respectively. There was a three-fold difference in the number of QTLs detected for four traits commonly evaluated in two generations (seven QTLs in the BC3F5 vs 21 in the BC3F2 population). The percentages of the phenotypic variance explained by QTLs in the BC3F5 population were similar to or less than those in the BC3F2 population. This is probably due to the difference in the genetic composition of two populations and the environmental effects. The locations of the QTLs commonly detected in both generations were in good agreement except for one QTL for spikelets per panicle. The yield QTL, yd3 was colocalized with the spikelets per panicle, spp3. Yield increase at this locus is due to the increase in spikelets per panicle, because both traits were associated with increase in spikelets per panicle and yield due to the presence of an O. glaberrima allele. Clusters of QTLs for grain morphology traits were observed in two chromosome regions. One cluster harboring five QTLs near SSR markers RM106 and RM263 was detected on chromosome 2. This population would serve as a foundation for development of the introgression line population from a cross between Milyang 23 and O. glaberrima.

        In rice, tillering is an important trait determining yield. To study tillering at the agricultural and molecular aspects, we have examined a spontaneous rice mutant that showed reduction in the number of culms. The mutant was derived from a F6 line of the cross of Junambyeo*4 / IR72. It could produce, on average, 4 tillers per hill in the paddy field while wild-type plants usually have 15. Except the reduced culm numbers, they also show pale green phenotypes. The phenotypes of this mutant were co-segregated as the monogenic Mendelian ratio (χ2 = 0.002, p = 0.969). In order to locate a gene responsible for the rcn phenotype, the mutant with the japonica genetic background was crossed with Milyang21 of the indica background. Bulked segregant analysis was used for rapid determination of chromosomal location. Three SSR markers (RM551, RM8213, and RM16467) on chromosome 4 were genetically associated with the mutant phenotype. Each of the 217 F2 plants was genotyped with simple sequence length polymorphisms. The data showed that RM16572 on chromosome 4 was the closest marker that showed perfect co-segregation among the F2 population. We suggest the new rcn gene studied here name as rcn10t because there was no report which exhibit a rcn phenotype with a pleiotropic effect of pale green (chlorophyll deficiency), and mapped at same position on chromosome 4.

        Karyotypic analysis and FISH (fluorescence in situ hybridization) with 45S and 5S rRNA genes were carried out in Persicaria tinctoria H Gross. The somatic metaphase chromosomes were ranged from 2.25 μm to 1.50 μm in length. Chromosome number was 2n = 4x = 40 with the basic number of x = 10. The chromosome complement of the species consisted of 16 pairs of metacentrics (chromososomes 1,2,3,4,6,7,8,9, 10, 11, 12, 13, 15, 18, 19 and 20) and 4 pairs of submetacentrics (chromosome 5, 14, 16 and 17). The karyotype formula was K(2n) = 4x = 32 m + 8 sm. In FISH analysis, three pairs of 45S rRNA gene loci on the terminal region of submetacentrics (chromosomes 5, 16 and 17) and two pairs of 5S rRNA gene loci on the centromeric region of metacentrics (chromosomes 9 and 11) were detected, respectively.
