In order to better understand the morphological differentiation of the two cultivated types of Perilla crop and their weedy types in Korea and Japan, we studied the variation of 62 accessions by examining 15 morphological characteristics. By using ANOVA (one-way analysis of variance), we determined that var. frutescens and var. crispa showed significant morphological differences in terms of plant height and seed weight. Furthermore, cultivated var. frutescens and var. crispa could also be clearly discriminated from one another using PCA (principal component analysis). Specifically, quantitative and qualitative characteristics such as plant height, seed weight, degree of pubescence, shape of leaf, color of leaf, fragrance of plant, color of flower, color of stem and seed size greatly contributed to differences seen in the positive and negative direction on the first axis. In our study, most accessions of cultivated var. frutescens and those of its weedy type could be clearly discriminated from one another, however, most accessions of cultivated and weedy types of var. crispa were not clearly discriminated by the ANOVA and PCA analyses. These results indicated that cultivated var. frutescens can be considered to be a domesticated form, while the cultivated var. crispa can not be considered to be a domesticated form in Korea and Japan. It is our belief that our results concerning the morphological variations among cultivated types of Perilla crop and their weedy types in Korea and Japan will help ensure the long-term success of breeding programs and maximize the use of the germplasm resources in Korea.

For the purpose of breeding a high-yield superior variety of Korean raisin tree (Hovenia dulcis var. koreana Nakai), whose value as an edible and medicinal resource is increasing, tree candidates for superior individuals were selected from its 11 habitats in Korea from 1996 to 1998. A clone bank preserve was created in 1998 with 70 clones proliferated by grafting; the fruition traits (e.g., the number of fructified laterals, the average number of bunches per fructified lateral, the average number of bunches per fruiting lateral, the fruitpetiole weight per individual, and the yield per individual) of 47 clones that had bloomed and borne fruit were investigated and analyzed in 2002; five upper-ranking clones whose yield per individual exhibited a 261% improvement against the total average were picked in 2005; and three clones, including ‘oong-Sung 1’ that showed a difference in their fruit petiole ripening stage, were finally selected in 2007 as high-yield new cultivars of Korean raisin tree.

The growth performance of Eastern white pine (Pinus storbus L.) was studied with six provenances in four plantations. All growth performances were significantly different among provenances and plantations. The most outstanding source in volume growth at age 39 is North Carolina in all plantations and this trends had been started from four years old seedlings. In plantations, the growth is best in ChunCheon plantation while that is worst in GunPo plantation. The Effect of plantations was thought to be larger than that of provenances growth of P. strobus. A pattern of growth by ages was different by among plantations and even ate age about 40, the growth was still vigorous. Annual precipitation, foggy days, altitude and sand contents in soil are positively correlated with growth and the correlations between by ages were very high.

Over 7 individual rice (Oryza sativa L.) plants per a line were sowed and sampled by pooled sampling method for genomic DNA extraction. The 5,400 flanking sequence tags (FSTs) were analysed by adaptor PCR and direct sequencing. FST analysis showed that the intragenic FSTs, the intergenic FSTs, and the original insertional sequences including hot spot covered 48.1% (2,597), 25.6% (1,383), and 25% (1,350), respectively. The 2,597 intragenic FSTs were used for genotyping to determine whether they are heterozygous or homozygous, and 1,393 core lines were selected. Among them, 422 knockout genes were distributed on chromosome 3, while 56 - 157 intragenic FSTs scattered on other chromosomes. Among 1,393 FSTs, known genes such as transcription factor covered 59.4% (827), while unknown genes such as expressed protein covered 40.6% (566). RT-PCR indicated that some core lines had no expression or decreased expression level in their knockout genes. It means that core lines are very useful knockout lines for functional genomic studies.

Blast resistance of one hundred and thirty-one rice cultivars bred in Korea was tested with thirty Korean isolates and twenty-two Philippines isolates using three screening methods. In the blast nursery conducted in Korea and in the Philippines, average disease index of rice cultivars were 4.6 and 2.2, respectively. Seventy-nine cultivars showed different resistance reaction in Korea and in the Philippines, and 19 cultivars showed the same resistant reaction in two locations. In the seedling test, Korean blast isolates displayed different levels of virulence. 93-093, a Korean isolate, was compatible with 90 cultivars whereas 97-057 showed a compatible reaction with 13 cultivars. Twenty-three cultivars showed high level of resistance against Korean and Philippines isolates but Chucheongbyeo, Heugnambyeo, and Manmibyeo showed susceptible reaction to all blast isolates. Through the sequential planting test in Korea and in the Philippines, Palgongbyeo and Seomjinbyeo displayed durable resistance, and Nagdongbyeo and Gihobyeo showed high level of disease infection over the planting time. These results indicate that blast isolates collected in two countries have different genetic background and number of compatible isolates should be considered in definition the durability of rice cultivar to rice blast.

This study was conducted to develop the DNA markers for identification of the strawberry cultivars in Korea and Japan. We developed fifteen cleaved amplified polymorphic sequence (CAPS) markers based on the Fragaria gene sequences. Among them six CAPS markers showed polymorphism exclusively in one cultivar. Five CAPS markers (ANR-MspI, ANRBamHI, ACO-HinfI, DFR-AseI, FGT-MspI) provided enough polymorphism to identify eight Korean strawberry cultivars except for ‘Maehyang’ and ‘Sunhong’. To complement the fifteen CAPS markers, we selected another fifteen sequence-related amplified polymorphism (SRAP) and one of them, me1/em5_460bp marker, made it possible to discriminate between ‘Maehyang’ and ‘Sunhong’. Therefore, application of the five CAPS markers and one SRAP marker were sufficient to identify the nineteen Korean and Japanese strawberry cultivars. These markers could be used practically for cultivar identification of Korean and Japanese strawberry.

Rice blast resistance is considered one of the most important traits in rice breeding and the disease, caused by Magnaporthe grisea Barr, has brought significant crop losses annually. Moreover, breakdown of resistance normally occurs in two to five years after cultivar release, thus a more durable resistance is needed for better control of this disease. We developed a new variety, Keumo3, which showed strong resistance to leaf blast. It was tested in 2003 to 2007 at fourteen blast nursery sites covering entire rice-growing regions of South Korea. It showed resistance reactions in 12 regions and moderate in 2 regions without showing susceptible reactions. Durability test by sequential planting method indicated that this variety had better resistance. Results showed that Keumo3 was incompatible against 19 blast isolates with the exception of KI101 by artificial inoculation. To understand the genetic control of blast resistance in rice cultivar Keumo3 and facilitate its utilization, recombinant inbred lines (RIL) consisting of 290 F5 lines derived from Akidagomachi/Keumo3 were analyzed and genotyped with Pizt InDel marker zt56591. The recombination value between the marker allele of zt56591 and bioassay data of blast nursery test was 1.1%. These results indicated that MAS can be applied in selecting breeding populations for blast resistance using zt56591 as DNA marker.

We conducted a QTL analysis of agronomic traits using 117 BC3F5 and BC3F6 lines developed from a cross between Ilpumbyeo and Moroberekan. Genotypes of 117 BC3F5 lines were determined using 134 simple sequence repeat (SSR) markers. A total of 832 Moroberekan chromosome segments with 410 homozygous and 422 heterozygous, respectively, were detected, and the genetic distance of introgression segments ranged from 0.5 cm to 112.1 cm. A linkage map constructed using 134 SSR markers was employed to characterize quantitative trait loci (QTL). The 117 BC3F5 and BC3F6 lines were evaluated for seven agronomic traits at two locations in 2006 and 2007 and at one location in 2007. A total of 26 QTLs were identified for seven traits including days to heading, and the phenotypic variance explained by each QTL ranged from 9.2% to 24.2%. Moroberekan alleles contributed positive effects in the Ilpumbyeo background at eleven QTL loci including panicle length and spikelets per panicle. Five QTLs, two for days to heading and one each for culm length, panicle length and spikelets per panicle were consistently detected in every occasions indicating that these QTLs are stable. Among them, two QTLs, spp6 for spikelets per panicle and pl6 for paniclel length were localized in the similar region. Increase in spikelets per panicle at this locus might be due to the increase in panicle length, because both traits were associated with increase in spikelets per panicle and panicle length due to the presence of the Moroberekan allele. These Moroberekan QTLs might be useful in breeding programs to develop high-yielding cultivars.

In this work, we analyzed the activity of control enzymes of flower color biosynthesis, chalcone synthase (CHS) and flavanone 3-hydroxylase (FHT) using biochemical and enzymological methods in Lilium longiflorum and 11 Lilium cultivars. The results obtained are as follows ; Naringenin (NAR) was synthesized in all Lilium cultivars tested by the catalytic activity of CHS which used malonyl-CoA and 4-coumaryol-CoA as substrates. Substrate-specific activity of CHS was observed because eridictiol (ERI), which uses caffeoyl-CoA as a substrate, was not detected in tested cultivars. In next step, dihydroflavone product was synthesized by FHT using flavanones as a substrate. FHT synthesized dihydrokaempferol (DHK) by using NAR as substrates. A remarkable activity of FHT was observed in other 11 cultivars.

‘Gangbaek’ is a japonica rice variety developed and registered by the rice breeding team of Department of Rice and Winter Cereal Crop, NICS, RDA in 2006. ‘Gangbaek’ was derived from a cross between ‘Suweon345’ with good grain quality and ‘DV85’ resistant to bacterial blight, K3a. F1 plants were grown in the greenhouse in winter of 1992/1993 and backcrossed with ‘Suweon345’ as the recurrent parent. Plants resistant to K3a race of bacterial blight (BB) were selected from BC1F1 to BC4F1 and used as parents in the backcrossing processes. This variety has about 120 days growth duration from transplanting to harvesting in west-southern coast and Honam plain of Korea. It is about 69 cm in culm length and tolerance to lodging. In reaction to biotic and abiotic stresses, it shows moderately resistance to blast, and resistance to bacterial blight pathogen, K1, K2, K3 and K3a, but susceptible to other major diseases and insect pests. The milled rice of ‘Gangbaek’ exhibits translucent, relatively clear non-glutinous endosperm and midium short grain. It has lower amylose content of 18.6% and protein content of 6.4% compared with ‘Nampyeongbyeo’. The milled rice yield performance of this variety is about 5.28 MT/ha in local adaptability test for three years. This cultivar would be adaptable to the bacterial blight-prone area in the south-western coastal and Honam plain of Korea.
