Haploid system by anther culture allows the development of homozygous lines when doubled. The response of anther culture to Basta (glufosinate) resistance was investigated on transgenic plants (cv. Anjungbyeo) in order to identify inheritance of bar gene associated with Basta. Most of the regenerated transgenic plants were sterile, and only a few plants produced viable seeds (A1) in the greenhouse. The bar gene was analysis by PCR in basta resistant transgenic plant (TA0). The transgenic seeds (A1) were significantly germinated in Basta solution compared with non-transformed seeds. As a result of anther culture, in regenerated haploid plants, segregation ratio was 1:1 in five of eight cross combinations. In diploid plants, segregation ratio was 1:1 in seven of eight cross combinations. Although there was some differences in the cross combinations, most of the combinations had 1:1 segregation ratio which supports the theory. The difference may be a result of the small sample size or the difference of anther culture response caused by genotypic difference. Hence, when many cross combinations were anther-cultured the results would support the theory.

Fifty-two Korean japonica rice cultivars were analyzed for leaf blast resistance and genotyped with 4 STS and 26 SSR markers flanking the specific chromosome sites linked with blast resistance genes. In our analysis of resistance genes in 52 japonica cultivars using STS markers tightly linked to Pib, Pita, Pi5(t) and Pi9(t), the blast nursery reaction of the cultivars possessing the each four major genes were not identical to that of the differential lines. Eight of the 26 SSR markers were associated with resistant phenotypes against the isolates of blast nursery as well as the specific Korean blast isolates, 90-008 (KI-1113), 03-177 (KJ-105). These markers were linked to Pit, Pish, Pib, Pi5(t), Piz, Pia, Pik, Pi18, Pita and Pi25(t) resistance gene loci. Three of the eight SSR markers, MRG5836, RM224 and RM7102 only showed significantly associated with the phenotypes of blast nursery test for two consecutive years. These three SSR markers also could distinguish between resistant and susceptible japonica cultivars. These results demonstrate the usefulness of marker-assisted selection and genotypic monitoring for blast resistance of rice in blast breeding programs.

In rice, tillering is an important trait determining yield. To study tillering at the agricultural and molecular aspects, we have examined a spontaneous rice mutant that showed reduction in the number of culms. The mutant was derived from a F6 line of the cross of Junambyeo*4 / IR72. It could produce, on average, 4 tillers per hill in the paddy field while wild-type plants usually have 15. Except the reduced culm numbers, they also show pale green phenotypes. The phenotypes of this mutant were co-segregated as the monogenic Mendelian ratio (χ2 = 0.002, p = 0.969). In order to locate a gene responsible for the rcn phenotype, the mutant with the japonica genetic background was crossed with Milyang21 of the indica background. Bulked segregant analysis was used for rapid determination of chromosomal location. Three SSR markers (RM551, RM8213, and RM16467) on chromosome 4 were genetically associated with the mutant phenotype. Each of the 217 F2 plants was genotyped with simple sequence length polymorphisms. The data showed that RM16572 on chromosome 4 was the closest marker that showed perfect co-segregation among the F2 population. We suggest the new rcn gene studied here name as rcn10t because there was no report which exhibit a rcn phenotype with a pleiotropic effect of pale green (chlorophyll deficiency), and mapped at same position on chromosome 4.

This study was carried out to get the genetic information on the germination rate and fat acidity after 12 weeks’storage at 35 . Germination rate decreased with longer storage period, ℃ while fat acidity increased. Germination rate was higher in the order of Koshihikari, Hanmaeum and Unkwangbyeo after 12 weeks’storage at 35℃. Fat acidity of Koshihikari and Hanmaeum was gradually increased, but that of Unkwangbyeo was dramatically increased after 4 weeks’storage at 35℃. The genetic mode on germination rate and fat acidity using the joint scaling test was shown to be additive-dominance gene effects. Additive([d]) effect was higher than dominance effect([h]) in all crosses. The average degree of dominance, [h]/[d], was less than 1, indicating that germination rate and fat acidity was partially dominant. Germination rate and fat acidity based on frequency distribution of 2 crosses were quantitative characters expressed by polygenes.

To identify low temperature-induced genes of wheat chromosome substitution line 5D, suppression subtractive hybridization (SSH) was performed with mRNAs from leaf samples that treated with low temperature (4℃). A cDNA library was constructed using mRNA isolated from wheat chromosome substitution line 5D leaves treated with low temperature (4℃). The nucleotide and deduced amino acid sequences of the putative gene products were compared. wfr-9 and wfr-32 showed identity over 90% related to vernalization gene. Other two genes, wfr-77 and wfr-83 which is related to freezing-resistant gene have also identity over 90%. This result suggest that those genes may be transcribed into antifreeze proteins which are accumulated within leaf apoplasts, when wheat chromosome substitution line 5D is acclimated during low temperature treatment.

In the previous study, 141 BC3F2 lines from a cross between the Oryza sativa cv. Milyang 23 and O. glaberrima were used to identify favorable wild QTL alleles for yield component traits. In this study, we carried out QTL analysis of four grain morphology as well as four yield component traits using 141 BC3F5 lines from the same cross and compared QTLs detected in two different generations. The mean number of O. glaberrima segments in the 141 BC3F5 lines ranged from 1 to 13 with 2.69 and 5.71 of the average means of homozygous and heterozygous segments, respectively. There was a three-fold difference in the number of QTLs detected for four traits commonly evaluated in two generations (seven QTLs in the BC3F5 vs 21 in the BC3F2 population). The percentages of the phenotypic variance explained by QTLs in the BC3F5 population were similar to or less than those in the BC3F2 population. This is probably due to the difference in the genetic composition of two populations and the environmental effects. The locations of the QTLs commonly detected in both generations were in good agreement except for one QTL for spikelets per panicle. The yield QTL, yd3 was colocalized with the spikelets per panicle, spp3. Yield increase at this locus is due to the increase in spikelets per panicle, because both traits were associated with increase in spikelets per panicle and yield due to the presence of an O. glaberrima allele. Clusters of QTLs for grain morphology traits were observed in two chromosome regions. One cluster harboring five QTLs near SSR markers RM106 and RM263 was detected on chromosome 2. This population would serve as a foundation for development of the introgression line population from a cross between Milyang 23 and O. glaberrima.

Knowledge of genetic diversity and of the genetic relationships among elite breeding materials has had a significant impact on the improvement of crops. In maize, this information is particularly useful in i) planning crosses for hybrid and line development, ii) in assigning lines to heterotic groups and iii) in plant variety protection. We have used the SSR technique to study the genetic diversity and genetic relationships among 76 Korean waxy corn accessions, representing a diverse collection from throughout Korea. Assessment of genetic diversity among members of this group was conducted using 30 microsatellite markers. Among these 30 microsatellite markers, we identified a total of 127 alleles (with an average of 4.2 and a range of between 2 and 9 alleles per locus). Gene diversity at these 30 microsatellite loci varied from 0.125 to 0.795 with an average of 0.507. The cluster tree generated with the described microsatellite markers recognized two major groups with 36.5% genetic similarity. Group I includes 63 inbred lines, with similarity coefficients of between 0.365 and 0.99. Group II includes 13 inbred lines, with similarity coefficients of between 0.45 and 0.85. The present study indicates that the 30 microsatellite loci chosen for this analysis are effective molecular markers for the assessment of genetic diversity and genetic relationships between Korean waxy corn accessions. Specifically, this study's assessment of genetic diversity and relationships between a set of 76 Korean waxy corn inbred lines will be helpful for such activities as planning crosses for hybrid and line development and association mapping analyses of maize breeding programs in Korea.

The experiment was conducted to investigate the potential for environmentally friendly cultivation of cv. Haryeong, which was bred by the National Institute of Highland Agriculture in 2006, through evaluation of the late blight field resistance at the farmer's practices level. cv. Haryeong showed higher field resistance to late blight as 590.3 in mean Area Under the Disease Progress Curve (AUDPC) during the cultivation period than cv. Superior as 1377.7 in AUDPC. However there was no observed different trend among the experimental treatments. Mean of tuber yield of cvs. Haryeong and Superior were 2,870 kg/10a and 2,301 kg/10a, respectively, and in all experimental treatments Haryeong was 25% higher than those of Superior. Mean of dry matter content of Haryeong and Superior were 18.1% and 16.1%, respectively, and there were no significant differences among the experimental treatments. cv. Haryeong was confirmed as a suitable cultivar for environmentally friendly cultivation in this experiment. In the future, additional research on fertilization and disease management will be required.

Estimates of genetic gain (in volume growth) and diversity (expressed as status number, Ns) were determined in a clonal seed orchard of Pinus koraiensis. The genetic thinning was based on clonal breeding values (represented by general combining ability) obtained from progeny tests, clonal fertility estimated by strobilus production, and clonal size variation determined by the ramet numbers per clone. Parental GCA values for volume growth were calculated, based on height and diameter at breast height measured from field trials. Clonal fertility was estimated from the assessments of strobilus production over twelve years from 1991 to 2003, and used for the calculation of status number. There are 179 clones and 5,268 ramets in 12ha area of P. koraiensis clonal seed orchard. Genetic gain and diversity estimates were determined under assumptions of 30% pollen contamination and inferior genetic value of contaminating pollen. Genetic gain increased as thinning rates were set from 10% to 60%. However, for the higher thinning intensities, the increase of genetic gain was not remarkable. Genetic thinning by means of truncation selection resulted in a greater genetic gain but a large decrease in status number. Status number was represented around 40 clones for 10% through 60% thinning intensities, but for the higher thinning intensities, it was a bit fluctuated. Based on the present results, it could be concluded that thinning rate should not be stronger than 60% to optimize genetic gain while conserving genetic diversity. Consequently 50% or 60% thinning rate might be appropriate for genetic thinning in the clonal seed orchard of P. koraiensis. The effect of pollen contamination on the genetic gain and the consequence of genetic thinning for seed production in the clonal seed orchard, and seed orchard management scheme were also discussed.

Amino acid composition in grain of Korean native rice core collections was evaluated to identify the useful genotypes for quality enhancing rice breeding and value-added food development. Large variations were found in the contents of 16 amino acids among 388 genotypes tested with 14.9% of the minimum coefficient of variation (CV), and the contents of methionine and histidine showed the highest CV and the second high, respectively. The average values of total amino acid, total essential amino acid, lysine and threonine contents of Korean native rice collections were almost similar with those of cultivating japonica rice, Saechucheong. Among 388 native core collections Baekkokna, Daesona and Saducho were identified as high amino acid germplasm in total amino acid, total essential amino acid, lysine and threonine contents. And Seoksanjo and Sodujo were identified as low amino acid collections. Rice collections showing purple apiculus color tended to distribute in lower serine, proline, glutamic acid, glycine, alanine, valine, leucine, phenylalanine, arginine, and total amino acid contents compared with those of yellowish apiculus color collections. The glutinous rice collections showed the tendency of higher contents of serine, glutamic acid, alanine, valine and leucine compared with those of non-glutinous collections.

An efficient transformation method for soybean [Glycine max (L.) Merr.] using meristematic tissues of germinating seeds has been established. The embryonic axes were excised from germinating seeds of Korean soybean cultivar, Iksannamulkong and 0.5-2 cm long segment containing meristematic tissues were prepared by cutting hypocotyl region. The explants were inoculated with Agrobacterium tumefaciens strain LBA4404 harboring a binary vector with the bar gene as a selectable marker gene and a β-glucuronidase (GUSINT) reporter gene, and then co-cultured for 7 days on co-cultivation medium (CCM). The meristematic tissues were cultured on shoot induction medium (SIMP6) supplemented with 0.4 mg/l N6-benzylaminopurine (BAP) and 0.1 mg/l indolebutyric acid (IBA) in the presence of 6 mg/l L-phosphinotricin (PPT) for 2 weeks and the surviving explants were transferred to shoot elongation medium (SEMP6). Transformation was confirmed by Southern blot analysis and the transformation efficiencies ranged from 1.48 to 2.07%. The new modified transformation method was successfully implemented for obtaining several transgenic lines with SMV-CP gene. It is expected that this method could efficiently be used for the transformation of recalcitrant soybean cultivars.

Many important traits have been tagged allowing plant breeders to apply marker assisted selection (MAS) in rice. PCR itself is simple to set up, and requires little hands-on time. However, a crucial limiting step of MAS programs is the reliable and efficient extraction of DNA which can be performed on thousands of individuals. In this study, We describe a modification of the DNA extraction method, in which cetyltrimethylammonium bromide (CTAB) is used to extract DNA from leaf tissues for suitable MAS in rice. We followed the standard 2% CTAB extraction method in all the procedure. In addition we used the 1.2 ml 8-strip tube instead of 1.5 ml E-tubes to fit the 8-multichannel pipette and employ the 96 well plate to use the swing bucket centrifuge. Our modified CTAB DNA extraction method offers several advantages with respect to traditional and simple methods. 1) adult leaf samples collected in paddy field are applicable. 2) 96 leaf samples can be homogenized only one-time by using tungsten carbonate bead and 96well block. 3) semiautomatic loading method using 8-multichannel pipette from DNA extraction to electrophoresis of PCR products. 4) our system can extract about 400 leaf samples per day by only one technicion. Therefore, this method could be useful for marker assisted breeding in rice.

Amylogram properties such as peak viscosity, hot viscosity, cool viscosity, breakdown, pasting consistency and setback were investigated by the Rapid Visco Analyzer and interpreted by the relationship among amylogram properties according to the varietal groups classified by the rapidity of grain filling (RGF) which was calculated by the percentage of grain weight at 15 days after heading to 40 days after heading. The 164 rice recombinant inbred lines from the cross of Milyang23 and Gihobyeo were used to get the basic information regarding the amylogram properties. The used recombinant inbred lines could be grouped into 4 varietal groups such as slow maturing (less than 40% of RGF), mid-slow maturing (41-60% of RGF),mid-fast maturing (61-80% of RGF), and fast maturing (more than 81% of RGF) groups based on the RGF. The peak viscosity and setback showed regular tendency according to the varietal groups classified by the RGF. Positive significant correlations were found between pasting consistency and setback, however negative significant correlations were found between breakdown and setback in all varietal groups.

“ongjin2”is a new japonica rice cultivar developed from the cross between Milyang165 with short culm and lodging resistance and F1 plant of Iksan438, HR14018-B-1-1 and Iksan435 with high palatability at Honam Agricultural Research Institute (HARI), NICS, RDA in 2005. This cultivar has short grain shape and about 143 days growth duration from direct seeding to harvesting under Korean climate condition. The milled kernels of “ongjin2”is translucent with non-glutinous endosperm. It has about 19.3% of amylose content and better palatability of cooked rice compared with “ampyeongbyeo” This cultivar shows high resistance reaction to the bacterial blight pathogene race from K1 to K3, blast and stripe virus but susceptible to insect pests. “ongjin2”yields about 5.71 and 5.74 MT/ha under the wet direct seeding and the transplanting at standard fertilizer level. “ongjin2”would be adaptable for the southern plain area of Korea.

A new rose variety, was selected from the progenies of a cross between “osai”and “okimeky”in 2006 after evaluation trials for three years (2004-2006). “altz”was developed because of many petals and powdery mildew resistance. A standard type with large sized flower, “altz”shows in 11.0 cm flower diameter. “altz”produces 130 stems・m-2 in a year. It has red-white color flower and 49.6 petals per flower. The stem length of cut flower was long with 69.3 cm. Vase life of the variety could be as long as 9.6 days.

We bred a new green-kerneled glutinous rice variety that can be cultivated in the whole area of Korea, because only one native green-kerneled glutinous rice cultivar, “aengdongchalbyeo” has been cultivated in the southern coastal area due to its late heading. The seeds of “aengdongchalbyeo”were irradiated with 200 Gy of gamma ray in 1995. A promising mutant variety, “ogwonchalbyeo”(“onnong 17” was selected through line selection and regional yield trials. In particular, the new variety revealed at the earlier mid of August compared to that of “aengdongchalbyeo” the early of September, and it was considerably tolerant to a field lodging due to its shortened culm length. Also, “ogwonchalbyeo”had a higher ripened grain ratio and 1,000 grain weight compared to the original variety. The brown grain yield of the new variety was about 5.40 MT/ha, which was 11.3% higher than that of the original variety, in the regional yield trials at 3 different fields during 2000∼2001. The brown and milled grains of the new rice variety contained 20 to 65% higher amount of total amino acids, respectively than that of the original and two checks. For chlorophyll -a, -b and total chlorophyll, the new variety showed nearly two-fold higher than the checks, and for the carotenoid, it had 5.3 –7.6 times higher amount. These results showed that the new variety can be cultivated as a special green-kerneled glutinous rice with high functional compounds.

“Dami”is a new japonica rice variety developed and registered by the rice breeding “Jeokjoong”, a white winter wheat (Triticum aestivum L.) variety was developed from the cross “Keumkang”/ “Tapdong”. “Jeokjoong” is an awned, semi-dwarf and soft white winter wheat, similar to “Keumkang” (check variety). The heading and maturing date of “Jeokjoong” were similar to “Keumkang”. Culm and spike length of “Jeokjoong” were 78 cm and 7.5 cm, similar to “Keumkang”. “Jeokjoong” had lower test weight (800 g) and lower 1,000-grain weight (40.1 g) than “Keumkang” (811 g and 44.0 g, respectively). It had resistance to winter hardiness, wet-soil tolerance and lodging tolerance. “Jeokjoong” showed moderate to scab in test of specific character although “Keumkang” is susceptible to scab. “Jeokjoong” had lower flour yield (69.2%) and ash content (0.36%) than “Keumkang” (72.0% and 0.41%, respectively) and similar flour color to “Keumkang”. It showed lower protein content (8.9%) and SDS-sedimentation volume (36.8 ml) and shorter mixograph mixing time (3.5 min) than “Keumkang” (11.0%, 59.7 ml and 4.5 min, respectively). Amylose content and pasting properties of “Jeokjoong” were similar to “Keumkang”. “Jeokjoong” had softer and more elastic texture of cooked noodles than “Keumkang”. Average yield of “Jeokjoong” in the regional adaptation yield trial was 6.19 MT ha-1 in upland and 5.33 MT/ha in paddy field, which was 19% and 16% higher than those of “Keumkang” (5.21 MT/ha and 4.58 MT/ha, respectively). “Jeokjoong” would be suitable for the area above the daily minimum temperature of -10℃ in January in Korean peninsula.team of Honam Agricultural Research Institute, NICS, RDA in 2006. Nokwon, a new cultivar for vegetable soybean, was developed from the cross between Keunolkong and Hyangnam-1 and released in 2006. The pedigree of Nokwon, designated as Milyang 153 in 2003, was SS96425-2B-11-4-1-1-1. Nokwon, used as a vegetable soybean was characterized by dark green pod, large seed, very short plant height, and lodging resistance. Nokwon has determinate growth habit, white flowers, gray pubescence, oval leaf shape and brown pods at maturity. The mature seeds have a greenish yellow seed coat with brown hilum and yellow cotyledon. In Korea, Regional Yield Trials (RYT) for vegetable soybean from 2004 to 2006, Nokwon shows strong tolerance to soybean mosaic virus and lodging in fields. Fresh pods of Nokwon harvested at the beginning of August, and stem height was 11cm shorter than 45 cm of Hwaeomputkong. In the same tests, fresh pod of Nokwon (11.4 ton/ha) yielded 14% higher than Hwaeomputkong (10.0 ton/ha). Nokwon had 5.9 cm fresh pod length, 13.1 mm fresh pod width, 75.4 g seed weight per 100 green seed, 39.4% green seed protein content, and 17.3% green seed oil content.
