        웰빙식품, 건강에 대한 관심 증대로 버섯의 소비량은 꾸준히 증가하고 있으나, 주로 생버섯으로 소비되 고 2차 가공식품의 기능성 구명으로 새로운 용도 개발 연구는 미진한 상황이다. 버섯의 고부가가치 창 출을 위해서는 버섯의 생산 및 단순가공형 산업에서 탈피하여 새로운 신기능성 구명 연구를 통해 새로 운 미래 바이오산업 확대를 위한 용도 창출을 위하여 본 연구를 수행하였다. 연구내용은 버섯류 35종 에 대한 전립선암세포내 PSA발현 억제 효능 및 이를 통해 확인된 버섯자원들의 함유성분 중 암에 관 련된 화합물들에 대한 구조적 상관관계를 분석하였다. 35종의 버섯류는 99% 에탄올 추출 및 물로 추출 후 동결건조하여 공시재료로 사용하였다. 그 결과 35종의 버섯중 느티만가닥(Hypsizigus marmoreus), 눈 꽃동충하초(Cordyceps takaomontana), 차가버섯(Inonotus obliquus), 동충하초(Cordyceps sinensis), 양송이 (Agaricus bisporus) 등에서 전립선암세포내 PSA 발현을 억제하는 효과를 보였다. 이들 버섯들은 면역력 을 높이는데 유효한 성분인 베타글루칸이 다량으로 함유되어 있어 면역력증진에 따른 전립선암세포의 증식 억제에 효과를 나타내고 있는 것으로 사료된다.

        본 연구에서는 녹각영지버섯 추출물의 항산화 효과 및 lipopolysaccharide (LPS)/interferon (IFN)-γ로 자극 된 RAW 264.7 대식세포에서 항산화 효과 및 항염증 효과를 조사하였다. 녹각영지버섯의 항산화 활성 을 확인하기 위하여 1,1-diphenyl-2-picrylhydrazyl (DPPH)과 2,2'-azino-bis-3-ethylbenzthiazoline-6-sulphonic acid (ABTS) radical 소거 활성 및 reactive oxygen species (ROS) 생성량을 측정한 결과, DPPH radical 및 ABTS radical 소거효과에 대한 IC50은 각각, 80.74, 52.28 ㎍/ml의 radical 소거능이 관찰되었으며 ROS 생성량은 농도 의존적으로 감소하는 것으로 나타났다. 그리고, RAW 264.7 대식세포를 이용하여 항산화 및 항염증 활성을 확인하기 위하여 ROS 및 nitric oxide (NO) 생성량을 측정한 결과, 활성산소 종 ROS와 염증 매개물질인 NO는 투여농도 의존적으로 감소하였다. 또한, 염증과 관련된 inflammatory cytokine의 발현을 농도 의존적으로 억제하여 항염증 활성을 나타내는 것을 확인할 수 있었다. 이상의 결과로부터 녹각영지버섯 추출물은 항산화 및 항염증을 지닌 기능성 소재로 활용이 가능할 것으로 사 료된다.

        본 연구는 국내 유통 방풍(S. divaricata, P. japonicum, G. littoralis) 한약재의 원산지 판별을 위해 수행 되었다. 이를 위 해, 형태적 특징비교 뿐만 아니라 엽록체(nrDNA-ITS2) 및 핵 DNA 바코드 유전자(cpDNA-matK, psbA-trnH, rpoB2와 rpoC1)를 이용한 염기서열 분석을 수행하였다. 방풍류 3종의 형태적 특징을 비교한 결과, 잎 모양과 거치의 형태에서 가장 큰 차이를 보인 반면, 건조약재의 경우 육안으로 구별하기에 어려움이 있었다. DNA 수준에서의 차이를 비교하기 위해 DNA 바코드 후보 유전자들을 이용한 방풍류 3종의 식물자원 에 대한 염기서열 분석을 수행한 후 판별 마커 개발 가능성이 있는 ITS2와 matK, psbA-trnH 세 primer를 선발하였다. 이 를 국내 유통 한약재에 적용한 경우 방풍은 S. divaricata와, 식방풍은 P. japonicum과, 해방풍은 G. littoralis와 동일한 것 으로 확인되었다. 중국산 방풍은 S. divaricata와 동일종으로 바르게 표기되어 유통되나, 국산 방풍은 P. japonicum과 동일 종으로 식방풍(갯기름나물)의 뿌리를 건조시켜 방풍으로 혼용 유통됨을 확인하였다. ITS2 구간의 염기서열 분석 결과, 식방 풍의 경우 두 그룹으로 나눠져 동일 종 내에 유전적 변이가 있음이 확인되었다. 따라서 본 연구결과 방풍류 3종 판별을 위 해 nrDNA-ITS2와 cpDNA-matK, psbA-trnH의 사용이 유용 할 것이며, 차후 방풍류 한약재의 판별을 위한 마커 개발 연 구의 기초 자료로서 활용될 수 있을 것으로 사료된다.

        국민의 건강과 웰빙에 대한 관심 증대 및 소득 증가로 한약재 소비를 비롯한 식품과 화장품 원료로서의 산업적 가치가 커지면서 약용작물의 수요는 점차 증가할 것으로 예상하며 한, 중 FTA 체결로 인해 약용작물 시장개방의 수입증가는 위협요인으로 작용할 것이다. 그러한 측면에 서 약용작물을 포함한 농식품산업을 미래 성장 동력산업 으로 육성하기 위해서는 종자품종육성과 생산유통기반의 정비와 확충, 산지이용규제 완화 등의 다양한 대책이 마련되어야 한다. 본 연구 결과의 활용방안을 살펴보면 다음과 같다. 첫째, 국가의 약용산업 육성 중장기 전략과 종자관리 체계의 구축에 필요한 약용작물의 데이터베이스를 구축 하고 향후 개발해야 할 중점기술을 제시해야 한다. 둘째, 우수한 약용작물 품종 개발 및 보급으로 농업생산성을 높여야 한다. 셋째, 약용작물 종자생산 및 보급에 필요한 제도정비가 필요하다. 넷째, 우량종자의 표준화 및 규격화 로 약용작물의 생산유통체계를 확립해야 한다. 현재 약용 작물의 소비행태는 직접적인 섭취보다 화장품, 의약품, 식품의 형태로 소비되는 패턴으로 바뀌고 있으며 규모가 점점 확대될 것으로 전망된다. 그러므로 향후 약용작물의 생산에서는 이러한 산업화 추세와 변화에 대응한 정책개 발과 제도적인 개선이 뒷받침되어야 한다.

        본 연구는 국내 유통 감초약재의 원산지 판별을 위해 수행되었다. 이를 위해 약재로 쓰이는 만주감초(Glycyrrhiza uralensis) 및 유럽감초(G. glabra)와 약재로 사용되지 않는 국내 자생종 개감초(G. pallidiflora) 식물의 형태적 차이 및 엽록체와 핵 DNA 구간의 염기서열의 차이를 구명하였다. 감초식물 3종의 형태적 특징을 비교한 결과, 잎과 뿌리에서 차이를 보였다. 특히 잎의 형태에 있어서 중국감초와 유럽감초는난형 혹은 타원형인 반면, 개감초는 엽장/엽폭비가 큰 피침형이었다. 또한 건조된 뿌리로 시중되는 감초 한약재의 경우, 중국산과 한국산은 백색인 반면 우즈베키스탄산은 황백색이었다.감초식물 3종을 대상으로 DNA 수준에서의 차이를 비교한 결과 rpoB2, rpoC1에서는 G. uralensis와 G. glabra가 구분되지않았으며, psbA-trnH의 경우 단 한 구간의 SNP에서 차이를 보였다. 반면 ITS2에 의해 증폭된 450bp의 염기서열을 비교한 결과, G. glabra는 98, 99, 100번째 위치에서, G. pallidiflora는 137, 161, 164, 203, 296위치에서 감초 종간판별을 위한 특이적 SNP가 확인되었다. 또한, phylogenetic tree 분석을 통해, 약재로 쓰이는 G. uralensis과 G. glabra는 약재로 쓰이지 않는 G. pallidiflora에 비하여 유전적 거리가 가까움을 확인하였다. 본 표준시료에서 얻은 결과를 유통 한약재 원산지 판별에 적용한 결과, 중국산과 한국산의 한약재는 G. uralensis와 염기서열이 일치하였고, 우즈베키스탄으로부터 수입된 한약재는 G. glabra와 염기서열이 일치됨을 확인하였다. 이에 감초의 생체 및 한약재의 원산지 판별을 위해 ITS2를 이용한 분자수준에서의 판별이 유용하다고 판단된다.

        선박의 충돌회피조선에 있어서 조종성능은 중요한 요소이다. 일반적으로 사용되는 조종성능은 심수역을 대상으로 작성되며, 천수역을 항주하는 선박의 조종성은 일반적으로 선회성은 저하되고 침로안정성 또는 추종성은 향상된다. 이러한 선회성의 변화는 충돌회피에 있어 위험을 초래할 수 있다. 본 연구에서는 천수역의 조종성능을 반영함과 동시에 충돌위험정도를 쉽게 파악할 수 있도록 하기 위한 가변안전경계영역의 새로운 적용기법을 제시하였다. 수학적 수치시뮬레이션 검증을 통하여 새롭게 제안된 기법의 유용성을 확인하였다. 따라서 선박충돌위험에 대한 충분한 충돌회피 조선을 지원할 수 있을 것으로 기대된다.

        해양사고를 감소시키기 위한 노력에도 불구하고, 해양사고는 증가하는 추세이다. 해양사고는 선박, 인적 그리고 자연조건의 3개 조건으로 분류할 수 있다. 본 연구에서는 1997년부터 2006년 사이의 해양사고의 자료를 이용하여 해양사고를 조건 별로 분석하였고, SPSS의 분산분석기법을 사용하여 그 차이의 통계적 의미를 분석해 보았다. 항목별로 일원배치 분산분석의 결과 시간대에서는 20~04 시간대와 그 외 시간대와는 집단간에 유의한 차이를 보였다. 속력에서는 5~10 노트 대에서, 초인거리는 1 마일 이하에서 유의한 차이를 보여 해양사고의 예방을 위해 위에 열거한 시간대, 속력 그리고 초인거리에 유의하여야 할 것으로 판단되었다.

        본 논문에서는 고투자율 등방성 자기물질을 이용한 공진형 마그네토미터(Resonant-type Magnetometer, RM)의 설계와 개발에 관하여 기술하였다. 먼저, 자기 물질에 감은 코일의 인덕턴스 L과 등방성 고투자율 자기 물질에서 나타나는 투자율 u(H) 사이의 기본이론을 정립하였다. 다음으로, L 의 변화를 간단한 슈미트 트리거 회로를 이용하여 주파수로 획득할 수 있는 RM회로를 구현하였다. RM의 측정능력을 평가하기 위한 선회실험을 통하여 RM의 지구자장 성분 측정 가능성을 확인하였다.

        Bisphenol A (BPA), a known endocrine disruptor, induces toxicity in cells and in experimental animals. Ginseng extracts were evaluated to determine whether they can inhibit BPA-induced toxicity. The antioxidant activity of fresh ginseng extract (WGE), dried white ginseng extract (DGE), and dried red ginseng extract (RGE) was measured using the DPPH assay. WGE and RGE increased DPPH free radical scavenging activity. Cell viability was measured in HepG2 cells following treatment with BPA and ginseng extracts using the MTT assay. DGE and RGE increased HepG2 cell viability following treatment with 200 μM BPA. RGE reduced levels of biochemical markers of liver damage, aspartate aminotransferase (AST) and alanine aminotransferase (ALT) that increased in mice following treatment with BPA. In addition, the regeneration and proliferation of damaged liver cells were significantly increased in RGE-treated mice. Moreover, RGE inhibited hepatic fibrosis in the surrounding area and in the central vein of the liver microstructure. RGE also significantly inhibited BPA-induced cytotoxicity. In addition, RGE protected liver damage and regenerated liver tissues in BPA-treated animals. These results show that RGE may represent a potential candidate drug for the treatment and prevention of liver damage caused by environmental toxins.

        Background: The study was conducted to elucidate the extraction conditions under which white ginseng has cognition-improving efficacy.Methods and Results: Extracts from white ginseng under different solvent and temperature conditions were analyzed for ginsenoside content and inhibitory effect on N-methyl-D-aspartate (NMDA) receptor and acetylcholinesterase. The total ginsenoside contents and amounts of ginsenoside Rb1 plus ginsenoside Rg1 from the 1st extracts (prepared with EtOH/H2O as solvent) were higher than those from the 2nd extracts (extracted with H2O after the 1st EtOH/H2O extraction). The contents in the 1st and 2nd extracts produced at 80°C were also higher than those obtained at 50°C. Samples from the 1st extraction at 80°C indicated higher inhibitory activities on NMDA receptors-whose excessive activation is thought to mediate the calcium-dependent neurotoxicity associated with several neurodegenerative diseases-than those from the 2nd extraction. Among the samples prepared at varying temperatures, the extract prepared at 50°C showed the highest suppression activity on NMDA receptors. Note, however, that the extracts from the 2nd extraction at 50°C inhibited acetylcholinesterase-whose inhibition could be a therapeutic strategy for neurodegenerative diseases with cognitive deficits and memory malfunction-more effectively than those from the 1st extraction.Conclusions: To enhance the cognition-improving activity of white ginseng extract, it is suggested that the extracts be utilized after being combined the 1st extracts (made with EtOH/H2O solvent) and the 2nd extracts (prepared with H2O) at low temperature.

        Background : Cynanchum wilfordii and Cynanchum auriculatum belong to the Asclepiadaceae family and appear morphologically similar. In order to discriminate them, it is needed to find the presence of sap and the leaf shapes: C. auriculatum has a blade ovate leaf comparing to C. wilfordii. However, in the herbal medicine market, they have been handled as cut and dried roots. Due to their similar morphology, it is limited to distinguish the roots of C. wilfordii and C. auriculatum. Recently in Korea, it has been a critical issue to misuse these two roots in the herbal market and food industry. Thus, it is required to establish a robust tool for the discrimination and quality control of them. Methods and Results : To separation and characterization of flavor compounds, C. wilfordii and C. auriculatum samples were analyzed by head space solid phase micro extraction (HSS) fiber coupled with gas chromatography-mass spectrometry using Rtx-5MS (30 m x 0.25 mm x 0.25 μm) column. As a result, We have identified compounds of a few hundred in aliphatic aldehydes and aliphatic alcohols, alkenes and acids, aromatic compounds, aromatic compounds containing nitrogen & sulfur, etc,. In particular, The aliphatic and aromatic compounds had been clearly separated on the second dimensional direction by using two-dimensional GC. Conclusion : The volatile flavor compounds of C. wilfordii and C. auriculatum could easily analyzed without pre-treatment with improved resolution and sensitivity using HSS-GCxGC-TOFMS. We have identified compounds of a few hundred in C. wilfordii and C. auriculatum sample. And It was more accurately qualitative confirmed with separation of GCxGC and TSD. We have confirmed the PCA and PLS-DA Plot that was classified depending on C. wilfordii and C. auriculatum through multivariate statistical analysis of the identified flavor compounds.

        Background : Ginseng, an important traditional medicinal plant still used in rats with bone fractures or dislocation to promote connective tissue repair and to reduce inflammantion. We investigated the effect of ginseng on the proliferation rate of rat bone. Methods and Results : We investigated the effect of ginseng extracts on blood biochemical parameters, bone density and bone inorganic components etc. and data were analyzed by one-way ANOVA. In the results of our study, the level of albumin and HDL, Ca, P, Mg, and estradiol in blood, and the content of Ca, P, ash in femur were significantly increased in ginseng treated group than in OVX group, and the level of ALP, AST, ALT, blood glucose, total cholesterol, triglyceride, LDL, creatinine, osteocalcin, and N-terminal telopeptide were significantly decreased in red ginseng treated group than in OVX group (p < 0.01). Conclusion : From these results, we knew that within the normal level, ginseng extracts improved liver and kidney function, component of glucose and lipid in blood, bone densith, bone ash and inorganic components in femur, and index related with bone metabolism.

        Background : Reactive oxygen species (ROS), whether produced endogenously as a consequence of normal cell functions or derived from external sources, pose a constant threat to cells living in an aerobic environment as they can result in severe damage to DNA, protein, and lipids. The effects of Valeriana fauriei extract and fractions on hydrogen peroxide-induced neuronal cell damage are studied. Methods and Results : Oxidative stress plays an important role in the pathological process of neurodegenerative diseases. Valeriana fauriei extract (VFE) and EA fractions (VFEA) was investigated total phenolic contents using method. VFE of total phenolic contents had 2.54 ± 0.01 mg/g, also, VFEA had a 18.78 ± 0.03 mg/g. High phenolic content of the VFEA is expected to better the inhibition of oxidative stress. VFE and VFEA were experimented to inhibit ROS induced 200 μM 3-morpholinosydnonimine (SIN-1). VFE of inhibit SIN-1 induced-ROS dose dependently and signficantly. In addition, VFEA inhibition was also dose dependant and significant. Moreover, Treatment of SH-SY5Y and SK-N-SH cells with VFEA significantly reduced hydrogen peroxide-induced generation of intercellular ROS. Conclusion : From the above results, we may suggest that VFEA might have useful as a material for functional food and pharmaceutics for the pathological process of neurodegenerative diseases.

        Background : Five medicinal herbs have been selected from the preliminary screening for in vitro anti-allergy activity (in RBL-2H3 cells). The present study is conducted to investigate the inhibitory effects of the medicinal herbs on allergic inflammation in other kind of cells. Methods and Results : Cells treated with five extracts prepared from Betula costata Trautv. (aerial part), Camellia sinensis L. (aerial part), Polygonatum stenophyllum Maxim. (root), Pyrus pyrifolia var. culta (Makino) Nakai (leaf), and Rehmannia glutinosa (Gaertn.) Libosch. ex Steud. (root) were measured for mRNA levels of TNF-α on HaCaT keratinocytes stimulated by TNF-α /INF-γ and for mRNA levels of IL-2 in Jurkat T cells mediated by PMA/A23187. Pre-treatment with the five extracts reduced the mRNA levels of TNF-α in HaCaT cells and mRNA levels of IL-2 in Jurkat T cells. In particular, the leaf extract of Pyrus pyrifolia var. culta (Makino) Nakai significantly and dose-dependently decreased the mRNA levels of TNF-α and IL-2. To determine the toxicity of the extracts from the selected medicinal herbs to HaCaT cells and Jurkat T cells, the viabilities of the cells treated with several concentrations of the five extracts were measured by MTT assay. Extracts of Polygonatum stenophyllum Maxim. (root), Pyrus pyrifolia var. culta (Makino) Nakai (leaf), and Rehmannia glutinosa (Gaertn.) Libosch. ex Steud. (root) (up to 250 ㎍/㎖) did not show cytotoxic effects on HaCaT cells and Jurkat T cells. On the other hand, 250 ㎍/㎖ of extracts of Betula costata Trautv. (aerial part) and Camellia sinensis L. (aerial part) reduced cell viability in both cells. Conclusion : These results demonstrate that the leaf extract of Pyrus pyrifolia var. culta (Makino) Nakai has an anti-inflammatory effect by inhibiting pro-inflammatory cytokine expression. Therefore, the leaf of Pyrus pyrifolia var. culta (Makino) Nakai can be a useful resource for the development of anti-allergy/anti-inflammatory materials.

        Background : Ginseng has been commonly used as a traditional oriental medicine for its wide spectrum of medicinal properties, including anti-inflammatory, antitumorigenic, adaptogenic and anti-aging properties. 20(S)-Protopanaxadiol (PPD), a intestinal metabolite of ginsenosides, is one of the active ingredients in ginseng. In this study, we have found inflammation-related genes regulated by 20(S)-PPD in mouse bone marrow-derived macrophage (BMDM) to elucidate the role of 20(S)-PPD in inflammatory signaling pathways. Methods and Results : We examined cell viability of BMDM cells after treatment of 20(S)-PPD and found that 20(S)-PPD has no cytotoxicity up to 10 uM. BMDM cells treated with none or 10uM 20(S)-PPD were used for RNA extraction and microarray analysis. It was found that 2 inflammation-related genes are upregulated and 4 genes are downregulated by 20(S)-PPD. Conclusion : These results can give clues to elucidate the role of 20(S)-PPD in inflammatory signaling pathways.

        Background : Recently, ginseng (Panax ginseng C.A. Meyer.) berry has been used as a health-promoting supplements. Also, Mulberries (Morus alba L.) fruit have been used in traditional herbal medicine to treat and prevent diabetes. In this study, we measured the cytotoxicity after fermentation of the extracts in Panax Ginseng Berry and Mulberry Fruit. Methods and Results : The extracts were prepared by decoction for 3 hours in distilled water (100 g/L). The dried extract was then dissolved in phosphate-buffered saline (PBS) in preparation for use. Cell viability was examined by an MTT assay. RAW 264.7 cells were seeded at 1 × 104/mL densities in 96-well plates. Each grouping had a non-treated group as the control. The extracts were added to each well and incubated for 24 hours at 37°C and 5% CO2. The MTT solutions (5 mg/mL) were added to each well, and the cells were cultured for another 2 hours. The supernatant was then discarded, and 100 μL of dimethyl sulfoxide was added to each well. The optical density was read at 540 nm. Conclusion : Probiotics and prebiotics modulate the composition of human and domestic animal gut microbiota. The beneficial effects may result from suppression of harmful microorganisms or stimulation of organisms which contribute in a positive way to the nutrition and health of human and domestic animal. Recently, fermentation using microorganisms for the production of more effective compounds has been extensively studied. In particular, the novel pharmacological effects of a new compound generated by fermentation have been reported. Some previous studies have demonstrated that Fermented herbal medicine extract showed better bioactivity than normal herbal Plants extract when used at the same concentration.

To breed a new variety named as a dwelling “ Woogang” of Achyranthes japonica N. the review by fostering december 2015 to be registered. Conclusions : Leaf type is the broad lancet, flowering time is new variety two days than Jangheung native variety delayed. New variety of stem length and main root length are check variety (89㎝, 13.5㎝) of 9㎝ and 2㎝ tall respectively. Seed yields has native variety as much as 35 % compared to the many into 43.5 kg/10a. The ecdysteroids contents of roots was 1.2 times higher compared to check variety (668.1ppm). The dried root yields of the new variety are 285kg/10a a many native variety than 20%.

        Background : The purpose of this study was to evaluate methods to high quality medicinal vegetables, thereby increasing farm income. This study investigated the effects of different covering material and weed eliminating on stable foliage production in Peucedanum japonicum Thunberg. Methods and Results : method of weed elimination with covering material were conducted under four condition compose to Non covering(4~6 times by manual weed control), Barley straw cutting covered with 700㎏․10 a−1, Rice straw cutting covered with 500㎏․10 a−1, Rice husks covered with 1,000㎏․10 a−1. Fermented mixed oil cake compost applied at 380㎏․10 a−1, 60% of basal fertilizer in March 14, 40% of top dressing were four times application in April 14 ~July 15. Sowing dates were March 25, 2015. Plants were spaced 15 ㎝ apart in rows 15 ㎝ apart with open cultivation. Conclusions : Rice husks covering < Barley straw covering < Barley straw covering cultivation this order, aerial part as a result were plenty amount of growth. Weed the rates was 12 to 29% lower incidence. Green leaf yields is uncovering control plot (915 kg․10 a−1) compared to 2% from covering rice husks, barley straw and rice straw covering increased to 18%, 20%, respectively.

        Background : The purpose of this study was to evaluate methods to high quality medicinal vegetables, thereby increasing farm income. This study investigated the effects of different organic fertilizer and applicated cultivation on stable foliage production in Peucedanum japonicum Thunberg. Methods and Results : method of application with organic fertilizer were conducted under nine condition compose to control(non application), N-P2O5-K2O (20-12-10)㎏․10 a−1, Hyphal incubation compost(HIC) applied at 851㎏․10 a−1, Hangawi organic compost(HOC) applied at 547㎏․10 a−1, Fermented mixed oil cake compost(FMOC) applied at 380㎏․10 a−1, 60% and 40% of basal fertilizer in March 12, top dressing were four application in April 12, May 12, June 13 and July 12, respectively. Sowing dates were March 23, 2015. Plants were spaced 15 ㎝ apart in rows 15 ㎝ apart with open cultivation. Conclusions : chemical fertilizers < hyphal incubation compost < hangawi organic compost < fermented mixed oil cake compost application, top dressing than 60% in order of 40% additional fertilizer, aerial part as a result were so many such amount of growth as number of leaf per plant, etc., Green leaf yields is untreated control plot (875 kg․10 a−1) compared to 12% from using chemical fertilizers, HIC application 18% higher, HOC application 20% higher, and FMOC application increased to 28% of the most.
