Background: Prenatal exposure to infectious and/or inflammatory insults can increase the risk of developing neuropsychiatric disorder such as bipolar disorder, autism, and schizophrenia later in life. We investigated whether Valeriana fauriei (VF) treatment alleviates prepulse inhibition (PPI) deficits and social interaction impairment induced by maternal immune activation (MIA).Methods and Results: Pregnant mice were exposed to polyriboinosinic-polyribocytidilic acid (5㎎/㎏, viral infection mimic) on gestational day 9. The adolescent offspring received daily oral treatment with VF (100㎎/㎏) and injections of clozapine (5㎎/㎏) for 30 days starting on the postnatal day 35. The effects of VF extract treatment on behavioral activity impairment and protein expression were investigated using the PPI analysis, forced swim test (FST), open field test (OFT), social interaction test (SIT), and immunohistochemistry. The MIA-induced offspring showed deficits in the PPI, FST, OFT, and SIT compared to their non MIAinduced counterparts. Treatment with the VF extract significantly recovered the sensorimotor gating deficits and partially recovered the aggressive behavior observed in the SIT. The VF extract also reversed the downregulation of protein expression induced by MIA in the medial prefrontal cortex.Conclusions: Our results provide initial evidence of the fact that the VF extract could reverse MIA-induced behavioral impairment and prevent neurodevelopmental disorders such as schizophrenia.

Background: We investigated the optimal aqueous extraction conditions for recovery of high yields of total phenolic compounds from roots of Arctium lappa L. (burdock, Asteraceae), and we compared their antioxidant capacity.Methods and Results: The antioxidant activity of the extracts was tested using 2,2-diphenyl-1-picrylhydrazyl, 2,2’-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid)-diammonium salt, and oxygen radical absorbance capacity assays. In addition, the major phenolic compounds present in the extracts were determined by high performance liquid chromatography analysis. Our results suggest that the roasted burdock 100°C, 15 min extract exhibited the strongest radical scavenging activity and possessed the highest concentration of phenolic compounds. The polyphenol content of both dried burdock and roasted burdock significantly increased with increase in the extraction temperature and time.Conclusions: These results indicated a relationship between phenolic compound levels in burdock and their free radical scavenging activities. This suggests that phenolic compounds significantly increase the antioxidant potential of burdock extracts.

Background: Cylindrocarpon destructans (Zins) Scholten is an important pathogenic fungus that causes ginseng root rot in many ginseng growing areas in China. Although C. destructans have been studied worldwide, research on its chemotaxis towards ginseng (Panax ginseng C. A. Meyer) root exudates in the rhizosphere remains limited.Methods and Results: In this study, we collected ginseng root exudates with three different polarities from three-year-old ginseng roots, and performed chemotaxis and spore germination assays to investigate the ability of these exudates to induce the response in C. destructans. The results showed that, compared with other conditions, when C. destructans cultivated at 20°C and a pH of 6 exhibited a strong positive chemotactic response toward 2 ㎎/ℓ aqueous phase, 20 ㎎/ℓ butanol phase, and 0.2 ㎎/ℓ petroleum ether from ginseng root exudates, the chemotactic moving indexes were 0.1581, 0.1638 and 0.1441, respectively. In addition, the spore germination rate with optimal chemotactic parameters were 48%, 53%, and 41% in the aqueous phase, butanol phase and petroleum ether groups, respectiviely, which were significantly higher than that in the control group (23%) (p < 0.05). The mycelial growth rate with optimal chemotactic parameters increased with culture time, and the maximum growth rates in the aqueous phase, butanol phase and petroleum ether groups were 0.425, 0.406 and 0.364 respectively, on the 4th day. The optimal chemotactic parameters were 39.73 ㎎/50 ㎎/ℓ , 48.93 ㎎/50 ㎎/ℓ , and 31.43 ㎎/50 ㎎/ℓ , in aqueous phase, butanol phase and petroleum ether respectively, from ginseng root exudates, compared with 5.5 ㎎/50 ㎎/ℓ , in the control group.Conclusions: The present study revealed that certain ginseng root exudates containing chemical attractants act as nutritional sources or signals for C. destructans and support its colonization of ginseng roots.

Background: Many menopausal women suffer from health problems including metabolic diseases such as dyslipidemia and osteoporosis. Thus they need natural products and functional foods particularly highly nutritional food products, that can help alleviate these diseases. This study was carried out to determine the effect of Drynariae Rhizoma water extract on the lipid and bone metabolism of ovariectomized Sprague-Dawley rats.Methods and Results: The animals were randomly divided into six dietary groups comprising SHAM-operated rats, OVX rats (normal diet), and OVX-DR rats (Drynariae Rhizoma extract). After 8 weeks, plasma, liver, and fat samples were collected to analyze the lipid metabolism, plasma Ca, alkaline phosphatase (ALP), osteocalcin and C-terminal telopeptide (CTx) concentrations, which are biochemical makers of bone metabolism. The left femurs of rats were also collected for histological analyses. OVX counteracted menopause induced body weight gain, as well as increases in triglycerides, total cholesterol, and free fatty acids. The Drynariae Rhizoma group showed low levels of triglycerides, high HDL-cholesterol, and decreased lipogenesis based on activity of the lipid-regulating enzymes (fatty acid synthase and malic enzyme). Decreased serum levels of ALP and osteocalcin were observed in Drynariae Rhizoma group.Conclusions: The results of this study show that Drynariae Rhizoma extract may effectively regulate hyperlipidemia and improve bone density.

Background: The study was conducted to elucidate the extraction conditions under which white ginseng has cognition-improving efficacy.Methods and Results: Extracts from white ginseng under different solvent and temperature conditions were analyzed for ginsenoside content and inhibitory effect on N-methyl-D-aspartate (NMDA) receptor and acetylcholinesterase. The total ginsenoside contents and amounts of ginsenoside Rb1 plus ginsenoside Rg1 from the 1st extracts (prepared with EtOH/H2O as solvent) were higher than those from the 2nd extracts (extracted with H2O after the 1st EtOH/H2O extraction). The contents in the 1st and 2nd extracts produced at 80°C were also higher than those obtained at 50°C. Samples from the 1st extraction at 80°C indicated higher inhibitory activities on NMDA receptors-whose excessive activation is thought to mediate the calcium-dependent neurotoxicity associated with several neurodegenerative diseases-than those from the 2nd extraction. Among the samples prepared at varying temperatures, the extract prepared at 50°C showed the highest suppression activity on NMDA receptors. Note, however, that the extracts from the 2nd extraction at 50°C inhibited acetylcholinesterase-whose inhibition could be a therapeutic strategy for neurodegenerative diseases with cognitive deficits and memory malfunction-more effectively than those from the 1st extraction.Conclusions: To enhance the cognition-improving activity of white ginseng extract, it is suggested that the extracts be utilized after being combined the 1st extracts (made with EtOH/H2O solvent) and the 2nd extracts (prepared with H2O) at low temperature.

Background: Sorghum is a major cereal food crop used in many parts of the world. It has been grown on a subsistence level by farmers, under various conditions of environmental stresses in the semi-arid tropics of Africa and Asia. This plant has received significant attention because of its ability to reduce cholesterol in the blood, and its anti-dementia, antioxidant, and antimicrobial properties. It is possible to develop a functional and commercially viable sorghum variety by using superior cultivars of sorghum. The objective of this study was to build a database of superior sorghum accession.Methods and Results: We used 250 sorghum accessions collected from different geographical bioregions in Korea. We determined various agronomic characters including germination rate and ear length of these accessions. To determine the antioxidant capacity, we measured the 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity, 2,2’-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid)-diammonium salt radical scavenging activity, total phenolic contents, and total flavonoid contents. Accession 189 showed higher germination (> 80%) than the other accessions. Higher 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity was observed in 11-SB-078 (RC50; 1.89 ± 2.88 μg/ml), and higher 2,2’-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid)-diammonium salt radical scavenging activity was recorded in 11-SB-116 (RC50; 35.48 ± 2.42 μg/ml) than in the other accessions. The ear length ranged from 15 ㎝ to 48 ㎝, the total phenolic contents ranged from 3 ㎎• GAE/g to 77 ㎎• GAE/g, and total flavonoid contents ranged from 0.09 ㎎• QE/g to 1.07 ㎎• QE/g.Conclusions: Among 250 sorghum accessions, we selected 10 with both superior agronomic characters and highly functional food quality.

Background: This study was conducted to investigate the effects of germination temperature, storage container and storage temperature on Scrophularia buergeriana and Scrophularia takesimensis seeds.Methods and Results: Seed lengths of both species were 0.8 ㎜, while seed width differed, with S. buergeriana measuring 0.5 ㎜ and S. takesimensis measuring 0.4 ㎜. The seeds of S. buergeriana were packaged in paper containers under room temperature (15°C), cold temperature (4°C), and freeze temperature (−20°C). These seeds exhibited around 80% germination rate at temperatures between 15°C and 30°C. The germiantion rate of S. takesimensis, on the other hand, differed significantly at different germination temperatures. Seeds of S. takesimensis which were packaged in vinyl and paper containers and stored under room and cold temperatures, exhibited around 80% germination rate at 15°C. However, the germination rate of freeze-stored seeds were decreased to lower than 20% at germination temperatures of 15°C, 25°C and 30°C germiantion conditions. The rate of germination showed a low positive to a significantly negativie correlation with the other factor that were determined to evaluate the germination performance.Conclusions: This study elucidates the most suitable germination and storage conditions to increase the germination rate for the two species of Scrophularia buergeriana and Scrophularia takesimensis needs to be stored in paper containers under cold temperature and requires a temperature of 20°C for germination. On the other hand, S. takesimensis in vinyl containers need to be stored at room temperature and those in paper containers at cold temperature, and a temperature of 15°C is required for germination.

Background: Salvia has been widely cultivated for use in flavoring and folk medicines in many countries, including Korea and China. In this study, we investigated the anti-inflammatory activity and the underlying active compounds of Salvia extract and its fractions.Methods and Results: The anti-inflammatory activity was measured by assessing the inhibition of cysteinyl leukotriene production in rat basophilic leukemia (RBL)-2H3 mast cells. Salvia plebeia R. Br. was found to have the most potent inhibitory activity on leukotriene production than S. japonica and S. chanroenica had. Fifty percent ethanol extracts of S. plebeia R. Br. were successively partitioned with n-hexane, methylene chloride, ethyl acetate, 1-butanol and water. The ethyl acetate (EtOAc) fraction showed stronger anti-inflammatory activity than other solvent fractions did. The EtOAc fraction was subjected to silica gel column chromatography elution with a chloroform and methanol gradient system (100 : 1 → 1 : 1) yielding 10 fractions. Three kinds of fractions (chloroform:methanol = 20 : 1, 10 : 1 and 5 : 1) showed high inhibitory activity on leukotriene production. We confirmed the major compounds with anti-inflammatory activity from S. plebeia R. Br.Conclusions: In this study, the major components of S. plebeia that showed leukotriene production inhibitory activity were isolated using solvent extraction and silica gel column chromatography. Rosmarinic acid, hispidulin and luteolin were identified as the major compounds with anti-inflammatory effect.

Background: In recent years, adjuvants have received increasing attention owing to the development of purified subunit and synthetic vaccines which are poor immunogens and require additional adjuvants to evoke an immune response. Therefore, immunologic adjuvants have been developed and tested. Plant polysaccharides have been recognized as effective biological response modifiers with low toxicity.Methods and Results: In this study, the polysaccharide from the aboveground part of Astragalus membranaceus Bunge containing immunomodulating arabino-3,6-galactan was evaluated for its hemolytic activity and adjuvant potential in the specific cellular and humoral immune responses to ovalbumin. The polysaccharide from the aboveground part of Astragalus membranaceus Bunge was co-immunized with the purified Vi capsular polysaccharide of Salmonella typhi vaccine in mice. The polysaccharide from the aboveground part of Astragalus membranaceus Bunge did not induce any hemolytic activity or side effects at doses up to 500㎍/㎖. The concanavalin A-, lipopolysaccharide-, and ovalbumin-induced splenocyte proliferation and serum ovalbumin-specific IgG, IgG1 and IgG2b antibody titers in immunized mice were significantly enhanced by AMA. Pharmacological data revealed that the polysaccharide from the aboveground part of Astragalus membranaceus Bunge increased antigen-specific antibody levels in immunized mice. The polysaccharide from the aboveground part of Astragalus membranaceus Bunge-adjuvanted purified Vi capsular polysaccharide of Salmonella typhi vaccine improved the proliferation of splenocytes and macrophages as well as stimulated cytokine production.Conclusions: These results suggest that the polysaccharide from the aboveground part of Astragalus membranaceus Bunge-adjuvanted vaccines enhanced humoral and cellular immunity and that the polysaccharide from the aboveground part of Astragalus membranaceus Bunge is a safe and efficacious adjuvant candidate suitable for use in prophylactic and therapeutic vaccines.
