In order to evaluate the effect of seed weight on different aspect of safflower (Carthamus tinctorius L.) seed germination and growth characteristics. Quantity of sinapine leaked from seed was greater as the viability of seeds was dropped by the time elapsed of seed aging model and long storage condition in safflower (Carthamus tinctorius L.). The cultivar of safflower was Jin-Sun and the seeds that are separated to three different weights of small, middle, and large were used in this experiment. Large seeds revealed the highest germination percent, coleoptiles fresh weight, coleoptiles dry weight, radicle fresh weight and 1000 seed weights than other seed weight. Seed weight had little effect on yield while seed number exerted a positive influence. Interestingly, yield per plant and its major components, number of capsules and capsule weights, revealed a negligible relationship with oil content.

This study was carried out to survey the vascular plants in 6 regions including around Hopo-Bridge of Yangsan region at waterfronts of Nakdong river, Korea. Distributed plants were surveyed from April to November, 2011. The vascular plants in 6 regions (around Mulgeum-chuisujang, Mulgeum-station, Jeungsan-ri, Hopo-bridge, Kumoh-bridge and lower part of Yangsan-stream) were total 135 taxa of 3 divisions, 4 classes, 37 orders, 66 families, 116 genera, 114 species, 2 subspecies and 19 varieties. The most number of species is Campanulales of 14.1% in Dicotyledoneae of 81.5% among total species. Regional distribution of species is mainly around Mulgeum-chuisujang and Jeungsan-ri as 61% among total species, and other regions is in 24 ~ 44%. Ecologically, there were herbaceous plants 75% and woody plants 25%, and 9 species of aquatic plants in herbaceous plants. Medicinal (herbal) plants included in KP (The Korean Pharmacopoeia) and KHP (The Korean Herbal Pharmacopoeia) were 56 species including Platycodon grandiflorum (Jacq.) A. De Candolle, Peucedanum japonicum Thunberg and etc., and folk herb plants were 14 species including Metaplexis japonica Makino, Commelina communis Linné and etc. among total 135 species.

This study was conducted to determine the contents of heavy metal in commercial herbal medicines. The monitoring of lead, arsenic, cadmium and mercury was carried out on 100 samples with 10 kinds of herbal medicines. The contents of heavy metal were measured by inductively coupled plasma atomic emission spectrometry (ICP-AES) and mercury analyzer. The mean values of heavy metal contents for herbal medicines were Pb 1.87 mg/kg, As 0.39 mg/kg, Cd 0.26 mg/kg and Hg 0.021 mg/kg. Of the total samples, 19 samples (19%, 4 types) were violated the KFDA regulatory guidance of heavy metal in herbal medicines (9 samples on Pb, 10 samples on Cd). Among these 19 unsuitable samples, 16 samples were domestic and 3 samples were imported. Therefore, we need continuously monitoring to ensure confidence and safety for these herbal medicines.

Gastrodia elata, an achlorophyllous orchid plant, is rare medicinal plant. We investigated the genetic diversity in G. elata from 4 locations by using Inter-Simple Sequence Repeats (ISSR) markers. Shannon's information Index (S.I.) indicating genetic diversity ranged from 0.255 (Pocheon) to 0.322 (Muju) with the mean of 0.29. The level of genetic diversity was lower than other plant and most genetic diversity was allocated among individuals within populations (26.81%). The UPGMA dendrogram based on genetic distance failed in showing decisive geographic relationship. In the case of gastrodin (GA), the major components in G. elata, Sangju was highest. The ergothionine (ERG) was detected a lot of contents in Muju and Pocheon. In conclusion, our results is very important information for explaining relationship of genetic variation and functional substances without the effects of environment factors and developing genetic marker by ISSR in G. elata, which may be responsible for the development of breeds with a lot of functional substance in G. elata.

This study was conducted to determine the contents of phenolic compounds and astragalosides in Astragalus membranaceus. Between the phenolic compound groups, flavonoids compounds (51.13%) had higher average concentrations than the phenolic acids groups. Among the 30 phenolic compounds, quercetin (353.11μg g-1) provided the highest concentrations. According to different cultivation year, 3-year-cultivated Astragalus (2612.57μg g-1) showed the highest concentrations of phenolic compounds. According to different harvest days, 6/5 harvesting Astragalus (2615.80μg g-1) showed the highest concentrations of phenolic acids. Comparison between the top and subterranean parts of harvested Astragalus plants cultivated for 2 years, 3 years, and 5 years showed big difference of total phenolic compounds in concentrations. Further, the top part had higher amounts of the total phenolic compounds than the subterranean part among all Astragalus. This tendency was similar to those of the top root and lateral root. The concentration of the phenolic compounds in the top root was higher than that of the lateral root.

In this study, kaempferol and its rhamnosides (α-rhamnoisorobin, afzelin, kaempferitrin) were isolated electively by bioconversion technology from Hibiscus cannabinus L. leaves to evaluate the anti-wrinkles effects and anti-microbial effects. In order to evaluate anti-wrinkles activity, reduction of expression matrix metalloproteinase-1 (MMP-1) protein and proliferation/ pro-collagen production were investigated. Kaempferol and α-rhamnoisorobin showed inhibition activity of MMP-1 generated to compared to positive control. In HaCaT cell proliferation assay, kaempferol and α-rhamnoisorobin significantly promoted cell proliferation in a concentration-dependent manner. In addition, procollagen synthesis assays (by HDF-N cell) showd that TGF-β induced procollagen production and also, all four kinds of experimental significantly promoted procollagen synthesis in a concentration-dependent manner. Kaempferol and α-rhamnoisorobin exhibited strong antimicrobial activities on five of microbes, Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Candida albicans and Aspergillus niger.

This study was carried out to select the appropriate medium (especially, carbon and nitrogen source, potassium phosphate and pH) for somatic embryogenesis in order to develop the rapid mass production system in suspension culture of Oplopanax elatus Nakai. Direct somatic embryos were obtained from root explants in the hormone free suspension culture (MS). Combination of NH4NO3 and KNO3 at the ratio of 1650 (mg/l) : 1900 (mg/l) obtained the better result to produce somatic embryo in suspension culture. MS medium supplemented with 170mg/l KH2PO4. The addition of 1 and 3% sucrose was effective for formation of embryogenic callus. Therefore, this report will be helped to improve the establishment for suspension culture in Oplopanax elatus Nakai.

'Usul' is a traditional medicinal herb, which has anti-inflammatory activities is distributed in India, Nepal, China, Korea, Japan. Korea pharmacopeia listed 'Usul' as either a species of Achyranthes japonica (Miq.) Nakai or A. bidentata Blume. Recent taxonomic studies in China and Japan delimited these taxa as two varieties, A. bidentata Blume var. bidentata and var. japonica Miq. A multivariate morphometric study of Achyranthes bidentata complex was undertaken to assess the entities of taxa that usefully could be recognized. Five quantitative characters were reviewed and analyzed with 293 specimens from Korea. The univariate analysis of inflorescence length, interval between florets, angle between floret and floral axis indicated that ranges among all taxa were continuous. However, quantitative characters of membrane size and the number of hairs within 4 were useful to identify two varieties. In PCA, the first three principal components accounted for 89.4% of the total variance. PCA revealed that var. bidentata showed distinctions in morphological attributes from var. japonica entity. Therefore, continued recognition at the infraspecifc level for these taxa is supported.

This study was carried out to investigate solid-state fermentation method using cellulolytic lactic acid bacteria Lactobacillus plantarum TO-2100 in order to increase piperidine alkaloid contents in mulberry leaves. Piperidine alkaloid, one type of which include 1-deoxynojirimycin (1-DNJ), is known to inhibit α-glycosidase activities. Using this strain, the optimal solid-state fermentation conditions on mulberry leaves powder were found as the following: initial moisture content, temperature and relative humidity were 20%, 30~35℃ and 60 ~ 70%, respectively, and the fermentation time was 72 hrs. The piperidine alkaloid contents in the fermented mulberry leaves were 2.86% on dry powder, which is 7-fold increase from that of non-fermented mulberry leaves. The 1-deoxynojirimycin contents after applying preparative thin layer chromatography were 2.02% on dry powder, which is 8 times higher than that of non-fermented mulberry leaves. α-Glycosidase activities was inhibited by 65.7 ~ 84.7% with 3 ~ 5% treatments of hot-water extracts of the fermented mulberry leaves, compared to 16.2 ~ 40.2% with 3 ~ 5% treatments of hot-water extracts of non-fermented mulberry leaves. Therefore, the results suggest that solid-state fermentation method does indeed increase of piperidine alkaloid contents on mulberry leaves.

The production of adventitious roots derived from root explant of Echinacea angustifolia and its secondary metabolite content were assessed in different types and levels of auxin. The induction of adventitious roots from root explant cultured in Murashige and Skoog solid medium supplemented with 1.0 mg/L indole -3-butyric acid (IBA) attained highest as 20.87 mg fresh weight and 3.07 mg dry weight per culture but root suspension culture at the same concentration of IBA enhanced biomass production as 3.07 g fresh weight and 0.38 g per culture after 4 weeks in culture. 3.0 mg/L α-naphthalene acetic acid (NAA) treatment had similar effect on root biomass production as 3.07 g fresh weight and 0.38 g per culture with liquid suspension culture, whereas adventitious roots exposed to over 3.0-5.0 mg/L IBA or 5.0 mg/L NAA were less responsive by reducing the number of adventitious roots and/or changing root morphology such as short and thick. The content of secondary metabolites such as phenolic, flavonoids and total caffeic acid in adventitious roots cultured on MS medium supplemented with 1.0 mg/L IBA were attained highest as 27.20, 9.60. 10.67 mg/g dry weight, respectively. Overall, the best production of root biomass and secondary metabolites were given by 1.0 mg/L IBA.

Damages of medicinal crops by thrips and identification of thrips species were investigated by periodic surveys on major medicinal crops, Platycodon grandiflorum, Schizandra chinensis, Codonopsis lanceolata, Lycium chinense, Bupleurum falcatum, Ledebouriella seseloides, Angelica gigas, Glycyrrhiza uralensis, Liriope platyphylla, Atractyloides sp., once or twice a month in Suwon, Eumseong, Jecheon, Pyeongchang, Chyungyang from May to October in 2012. Thrips and their damages were found at all of the medicinal crops investigated, mostly occurring at early growing stages of each medicinal crop and causing apical meristem region withered, thereby inhibiting the growth of plants. For species identification of thrips collected from each medicinal crops, DNA from each individual was extracted and ITS2 and COI regions were amplified by PCR. As a result, Frankliniella intonsa, Thrips tabaci, and Megalurothrips distalis have been identified as dominant species on medicinal crops in open field, whereas Frankliniella occidentalis was restricted to Lycium chinense in which protected cultivation. This study suggests that the occurrence of thrips species on medicinal crops seems to be affected by host specificity and environmental factors such as cultivation types.

We have established adventitious root culture systems of Codonopsis lanceolata, Codonopsis pilosula and Codonopsis ussuriensis. Root segments of C. lanceolata were the best explants for induction of adventitious roots and the number of adventitious root for explant was highest on solid medium with 0.5mg/l NAA and produced 18.8±1.9 roots per explant. Root segments of C. pilosula were the best explants for induction of adventitious roots and the number of adventitious root for explant was highest on solid medium with 1.0mg/l NAA and produced 8.5±1.8 roots per explant. Leaf segments of C. ussuriensis were the best explants for induction of adventitious roots and the number of adventitious root for explant was highest on solid medium with 0.5mg/l NAA and produced 7.8±0.4 roots per explant. In liquid culture, the best production of adventitious root (fresh weight) was obtained in 1/2 MS medium with 1.0mg/l NAA. This study demonstrated for the first time to produce adventitious roots in C. pilosula and C. ussuriensis.
