Background : Irregular meals and insufficient exercise are major modern lifestyle-related risk factors for constipation. This study aimed to examine the effects of the aqueous extract of Dendrobium speciosum var. (DM) on the improvement of intestinal function and prevention of constipation in rats. Methods and Results : Constipation in rat was induced by loperamide (4 ㎎/㎏) injection for 5 days and rats were randomly assigned to the following groups: normal control rats (NOR), constipated rats (LOP induced) and constipated rats supplemented with dulcolax-S (POS), DMSG, 100 ㎎/㎏ (DMSG-100), 300 ㎎/㎏ (DMSG-300) and 500 ㎎/㎏ (DMSG-500). The DMSG groups showed increments in the frequency and amount of stools compared to the LOP group. Loperamide treatment markedly reduced the water content of feces, whereas DMSG administration significantly increased fecal water contents. Moreover, DMSG improved intestinal transit speed in constipated-rats. Alcian blue staining revealed increased mucus production by crypt cells and mucus contents in feces and the mucosal surface. Conclusions : In summary, DM extraction significantly improves intestinal function in rats, which indicates the effectiveness of this extract in the prevention and treatment of constipation.

Background : Ginger has been extensively used in foods and traditional medicines in Asian countries. Despite its frequent consumption in daily life, the mechanism of potential interactions between ginger components-drug has not been examined. To elucidate the mechanism of governing the effects of 6-shogaol, a primary constituent of dried ginger, on human cytochrome P450 (CYP) isoenzymes an incubation studies were carried out using pooled human liver microsome (HLM). Methods and Results : CYP isoenzyme specific substrate was incubated with multiple concentrations of inhibitor, HLM and cofactors. 6-shogaol showed a potent inhibitory effect on CYP2C9, CYP1A2 and CYP2C19 with half maximal inhibitory concentration (IC50) values of 29.20, 20.68 and 18.78 μM respectively. To estimate the value of the inhibition constant (Ki) and the mode of inhibition, an incubation study with varying concentrations of each CYP isoenzyme-specific probe was performed. 6-shogaol inhibited CYP2C9 and CYP2C19 noncompetitively (Ki = 29.02 and 19.26 μM respectively), in contrast, the inhibition of CYP1A2 was best explained by competitive inhibition (Ki = 6.33 μM). Conclusions : These findings suggest that 6-shogaol may possess inhibitory effects on metabolic activities mediated by CYP1A2, CYP2C9 and CYP2C19 in humans.

Background : This study aimed to investigate the antioxidant and antiproliferative activities of extract from different parts of water chestnut (Trapa japonica Flerow). Methods and Results : The total polyphenol content of pericarp and seed extract was 438.31 ㎎/g and 25.32 ㎎/g respectively. DPPH radical scavenging assay showed that the half maximal inhibitory concentration (IC50 values) of pericarp and seed extract were 5.28 ㎍/㎖ and 355.51 ㎍/㎖ respectively. In addition, the pericarp extract showed strong reducing power. In the MTT assay, the pericarp extract significantly inhibited the viability of A549, AGS, HeLa, PC-3, HCT116, HT29 and SW620 cell lines compared with the seed extract. Conclusions : These results suggest that T. japonica Flerow extracts have significant antioxidant and antiproliferative activity.

Background : Acer pictum is a deciduous tree belonging to the family Aceraceae. This study was conducted to collect basic data on mass propagation techniques by surveying the germination and growth characteristics of A. pictum seeds. Methods and Results : A. pictum seeds were harvested in 2014 on August 19, September 5, September 29 and October 31. The seeds were then sown on the same dates they were harvested. A portion of seeds harvested in August 2014 were stored at low temperatures and subsequently sown on March 11, 2015. The germination rate of A. pictum seeds was highest in seeds that were stored harvested on October 31. Mean germination time (MGT) was shortest and germination velocity (Rs) was fastest in seeds harvested on October 31. Root collar diameter, total root length, and dry weight were also highest from seeds collected October 31. Conclusions : A. pictum seeds harvested on October 31 had highest germination and growth rates compared with seeds harvested on earlier dates. It is expected that these results demonstrating optimum harvesting and sowing dates will be applicable to future seedling production for this tree species.

Background : This text was conducted to evaluate the distribution, shape characteristics and habitat condition of Crypsinus hastatus in Buyeo Korea. Methods and Results : In Buyeo, Crypsinus hastatus was distributed at an altitude of 31.2 m, a slope of 82.5% and a distance from river water of 27.2 m. The leaf blade was oblong, 2.7 ㎝ long, 1.2 ㎝ wide with a petiole length of 2.3 ㎝. The total population comprised 2,100 individuals, with the highest proportion being individuals with a leaf blade of length 0.5 - 1 ㎝. The temperature and light intensity of the growth point were lower, but the humidity higher, than that of growth around. The chemical properties of the soil were as follows: pH 4.42, organic matter 77.99 g/㎏, electrical conductivity 0.50 dS/m, nitrate nitrogen 20.50 ㎎/㎏ and available phosphate 165.7 ㎎/㎏. Exchangeable cation content of potassium, magnesium, sodium and calcium was 0.35, 0.70, 0.09 and 2.04 c㏖+/㎏, respectively. Conclusions : In Buyeo, Crypsinus hastatus was distributed in 5.3 ㎡ area of Nakhwaam, Naeseongri and Jeongamri. However, the population of the medicinal resources seemed to decrease owing to the poor environmental conditions of the habitat.

Background : We studied the anti-oxidant activity and anti-inflammatory effects of Spiraea fritschiana Schneid extract (SFSE). Methods and Results : The SFSE was prepared using methanol and was evaluated for its total phenol and flavonoid content, DPPH (1,1-diphenyl-2-picrylhydrazyl) free-radical scavenging activity, reducing power, and effect on nitric oxide (NO) production, and cell viability by using real-time polymerase chain reaction (PCR). The total phenol content was 212.78 ㎍• gallic acid equivalent (GAE)/㎎ and the total flavonoid content was 66.84 ㎍• quercetin equivalent (QE)/㎎. The extract showed antioxidant activity (DPPH free-radical scavenging activity) with RC50 value of 76.61 ㎍/㎖. The reducing power of the extract was Abs 0.58 at 250 ㎍/㎖. Cell viability was determined using the MTT 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. To evaluate anti-inflammatory activity, we examined the inhibitory effects on lipopolysaccharide-(LPS)-induced NO production in RAW 264.7 cells. The NO inhibition rate was 90% at 200 ㎍/㎖ SFSE. At the same concentration, the expression of pro-inflammatory genes such as inducible nitric oxide synthase (iNOS) and cyclooxygenase (COX)-2 also decreased. Conclusions : Our results suggest that SFSE is a novel resource for the development of foods and drugs that possess anti-oxidant and anti-inflammatory activity.

Background : The white jelly mushroom (Tremella fuciformis), one of the most popular edible fungi, has medicinal properties. However, the effects of T. fuciformis in skin whitening or anti-wrinkle efficacy has not been defined to date. The aim of the present study was to investigate the effects of T. fuciformis extracts on whitening and anti-wrinkle efficacy in skin cells. Methods and Results :We prepared T. fuciformis extracts with water. The extracts (80℃) contained 12.11 ㎎/g polyphenol and 8.54 ㎎/g flavonoid concentration. T. fuciformis extracts markedly decreased melanin contents and tyrosinase activity in α-MSHstimulated melanocytes (B16F10 cells). In addition, the mRNA expression of melanin formation factors, such as microphthalmiaassociated transcription factor (MITF), tyrosinase-related protein-1 (TRP-1) and tyrosinase-related protein-2 (TRP-2) were significantly down-regulated in α-MSH-stimulated melanocyte. Furthermore, T. fuciformis extracts increased the synthesis of type I procollagen and reduced mRNA expression of matrix metalloproteinase 1 (MMP-1) in the human dermal fibroblast (HDFn cells). These data indicated that T. fuciformis extracts induce repression of cellular melanogenesis and protect against wrinkles caused by UVB-stimulated damage. Conclusions : Thus T. fuciformis extracts could be a cosmetic candidate for skin whitening and anti-wrinkle effects.

A new extraction method-heated ultrasonic extraction was qualitatively and quantitatively analyzed for the extraction of major ginsenosides from ginseng extract; this new high-performance liquid chromatography (HPLC) method was compared with the official extraction method of Korean industrial standards and standard for health functional food. Methods and Results : Ginsenoside compounds were analyzed for 35 minutes by the new HPLC analysis method using a Halo® RP-Amide column. The new HPLC analysis method was validated by the measurement of intra-day and inter-day precision, accuracy, limit of detection (LOD), and limit of quantification (LOQ) of each ginsenoside. The correlation coefficients (r2) for the calibration curves of the ginsenoside compounds were over 0.9997 in terms of linearity. The heated ultrasonic extraction method using ultrasonication for 30 minutes at 50℃ yielded higher amount of ginsenosides than the extraction method of the Korean industrial standards owing to the enhancement of extraction efficiency. Conclusions : Compared to the other extraction methods, the heated ultrasonic extraction method yielded a higher amount of ginsenoside Rb1 than Rg1 index compounds for the quality evaluation of ginseng roots.

Plant breeding requires the collection of genetically diverse genetic resources. Studies on the characteristics of Platycodon grandiflorum resources have not been carried out so far. The present study was carried out to discriminate P. grandiflorum based on morphological characteristics and genetic diversity using simple sequence repeat (SSR) markers. Methods and Results :We collected 11 P. grandiflorum cultivars: Maries II, Hakone double white, Hakone double blue, Fuji white, Fuji pink, Fuji blue, Astra white, Astra pink, Astra blue, Astra semi-double blue and Jangbaek. Analyses of the morphological characteristics of the collection were conducted for aerial parts (flower, stem and leaf) and underground parts (root). Next, the genetic diversity of all P. grandiflorum resources was analyzed using SSR markers employing the DNA fragment analysis method. We determined that the 11 P. grandiflorum cultivars analyzed could be classified by plant length, leaf number and root characteristic. Based on the genetic diversity analysis, these cultivars were classified into four distinct groups. Conclusions : These findings could be used for further research on cultivar development using molecular breeding techniques and for conservation of the genetic diversity of P. grandiflorum. Moreover, the markers could be used for genetic mapping of the plant and marker-assisted selection for crop breeding.
