Inonotus obliquus is one of the immune-regulatory substances and is recognized to play the role in the metabolic process of inflammation, allergy and immuntiy. The purpose of this study was to evaluate the effects of water extracts of Inonotus obliquus (IOW) on the liver lymphocyte immune function in the Sprague-Dawley male rats treated with carbon tetrachloride (CCl4) to induce liver damage. Rats were fed with each experimental diet and water for 4 weeks. We found that effects of IOW on interferon-gamma (IFN-γ), signal transducer and activator of transcription 1 (STAT1), phospho-signal transducer and activator of transcription 1 (pSTAT1) and GATA-binding protein 3 (GATA-3) were decrease in vivo. Interleukin-4 (IL-4), STAT6, pSTAT6 and T-box expressed in T-cells (T-bet) decreased significantly lower in CCl4+IOW group than the CCl4 group. Our data indicated that cytokine protein production were increased in CCl4 group and CCl4+IOW group. As a result of this study, we assume that IOW fed could regulate the immuno-modulating functions through regulate the cytokine production capacity activated by liver damage.

Several presowing seed treatments have been used to facilitate the seed germination and seedling emergence. The study was performed to design a model of presowing seed treatments in Taraxacum platycarpum. Germination and seedling growth responses to temperature, drying, chilling, GA3 and priming were investigated to know the optimum germination conditions and presowing treatment. The optimum temperature condition for germination and seedling growth were 15~20℃ at dark condition. The seeds exposed to infra red light, however, did not germinate. The moisture content of seeds dried at 35℃ for 3.5 hours was returned to the level of unimbibed seeds. The germination of seeds dried under dark condition was better than those dried under infra red light. Prechilling treatment imposed for 6 weeks showed higher germination rate than GA3 treatment or priming. In field test, the highest field emergence was observed in the seeds dried after prechilled for 6 weeks and followed by drying followed by GA3 〉 low temperature 〉 KNO3 priming. Imbibition with 0.1 mM GA3 solution for 2 hours then drying at 35 for 3.5 hour under dark condition was effective to the practical presowing treatment for Taraxacum seeds.

This study was conducted to investigate the effects of Lycii fructus and Astragalus membranaceus mixed extracts on immunomodulators and prevention in a streptozotocin-induced diabetes rat model. A total of 28 male rats were divided into four dietary groups and fed a commercial diet (A), commercial diet plus induced diabetes by a streptozotocin (STZ) injection (B), induced diabetes by STZ plus medicinal crop extracts(I&H®) diet (C), and medicinal crop extracts (I&H®) diet (D). Immunoblotting analyses revealed cytokine expression, and ELISA analyses revealed immunoglobulin E and nitric oxide production. As a results, the tumor necrosis factor-α (TNF-α) and inducible nitric oxide synthase (iNOS) as a inflammatory cytokine were decreased. Interleukin-6 (IL-6) and signal transducer and activation of transcription 3 (STAT3) cytokine related in diabetes expression through JAK/STAT3 pathway were also decreased. Furthermore, immunoglobulin E and nitric oxide production were decreased in the serum and lens, respectively. These results suggest that Lycii fructus and Astragalus membranaceus mixed extracts provide positive effects on immunomodulators and prevention in diabetes and eye disease complications.

This study was carried out to investigate the physicochemical characteristics of 3-year-old ginseng (for Samgyetang product) cultured by various seeding density in direct-sowing culture. Ginsengs were cultured by the seeding density, 275, 300, 330 352 and 396 seeds per Kan, 180×90cm area. Survived rate (82.1%) were the highest in plot of 352 seeds sowed, length and leaf width were high in plot of 300 and 352 seeds. Root yield grain was increased with increase of the seeding density in direct-sowing culture except 352 seeds sowed. Average root weight and diameter were the highest in plot of 352 seeds sowed, 31.6 g and 18.4 mm, respectively. Crude saponin and each ginsenosides content were the highest in plot of 275 seeds sowed. Rg1 content was decreased, Rc and Rb2 content were increased with increase of the seeding density. Total soluble sugar content was the highest in plot of 330 seeds sowed and the lowest in plot of 396 seeds sowed, and oligo- and disaccaride content were high in plot of 330 and 352 seeds sowed. Reological characteristics of ginsengs cultivated according to various seeding density, hardness and springness were high and maximum fracture force was low with decrease of the seeding quantity.

Glycyrrhiza uralensis (Leguminosae) is a well-known herbal medicine that has long been valued as a demulcent to relieve inflammatory disorders. To compare the influence of different solvents on the anti-inflammatory efficacy of G. uralensis, we measured the inhibition of pro-inflammatory mediators such as NO, TNF-α, and PGE2 in lipopolysaccharide (LPS)-stimulated mouse macrophage RAW 264.7 cells by extracts produced using different solvents (water, methanol, ethanol, or n-hexane). The results showed that methanol was the most effective solvent for the inhibition of both NO and PGE2 production in RAW 264.7 cells. However, there was no difference among the extracts for inhibition of TNF-α. Further study must be performed for the analysis of correlation between the anti-inflammatory activity of extracts produced using different solvents and the content of major bioactive compounds in G. uralensis, such as glycyrrhizin and liquiritin. The present study suggests that methanol may be a more appropriate solvent of G. uralensis than other solvents (water, ethanol, and n-hexane) to yield the greatest anti-inflammatory activity for food additives and medicine.

Machilus thunbergii was an important medicinal resource and distributed widely in China. In this study, the bioactivities of dichloromethane fraction (DF) and water fraction (AF) from Machilus thunbergii methanol extract were investigated. Total phenolic contents of DF and AF were 57.90 mg Gal/g and 189.92 mg Gal/g, and total flavonoid contents were 17.34 mg Que/g and 58.38 mg Que/g respectively. The EC50 for DPPH radical scavenging activity of DF and AF were 24.37 μg/ml and 2.10 μg/ml. Reducing power and hydroxyl radical scavenging activities of AF were higher than those of DF. The α-glucosidase inhibitory activity of AF (IC50 = 1.13 μg/ml) was higher than that of DF (IC50 = 5.34 μg/ml). The cell viability was showed that only the DF had anti-proliferation effect on human cancer cell HT-29. These results suggested that both the DF and AF extract of Machilus thunbergii were potential materials for anti-diabetes and functional food for their radical scavenging activity.

The application of PCR analysis on the herbal medicine Moutan Cortex (Paeonia suffruticosa Andrews) was evaluated by the comparison of the genetic relationship based on the DNA sequence with Paeoniae Radix (Paeonia lactiflora Pallas) following development of specific primers. Moutan Cortex and Paeoniae Radix were distinguished through the PCR analysis based on the internal transcribed spacer (ITS-PCR) from nuclear ribosomal DNA region. The 294 bp PCR products both of Moutan Cortex and Paeoniae Radix was amplified by MIF1 and MIR1. And a Moutan Cortex specific 225 bp PCR amplification product was amplified by MIF2 and MIR1 primers. The 225 bp sequence could be successfully amplified from Mortan Cortex of dried herbal preparations. PCR analysis based on ITS (ITS-PCR) may be an efficient tool for the discrimination of Moutan Cortex.
