This study was conducted to investigate the weight of crown part effect on growth and bolting response of Ostericum koreanum Kitagawa. Bolting rate with crown part weight was represented 0%, 0%, 4.1%, 12.9%, 25.1%, 34.9% corresponding to each crown part weight 5 g, 10 g, 20 g, 30 g, 40 g, 50 g. The heavier the crown part weight was the higher the direction rate rose and the flowering time came earlier. The dried root amount was the highest at 30 g and crown part number, the length of root, and crown diameter showed all the highest figures at 30 g. The relation between amount per 10 g and outcrop weight increased on secondary regression formula as weight increased and reached its peak at 29.7 g and then decreased over 29.7 g.
The bramble cultivated widely in South Korea, which is known as Bokbunja-ddal-gi, is regarded as having originated from Korean native Rubus coreanus. This study was carried out to obtain basic phylogenetic information on Korean cultivated bramble (KCB) by comparing the internal transcribed spacer (ITS) regions with those of R. coreanus, blackberry (R. lanciniatus), black (R. occidentalis) and red (R. idaeus) raspberry. Sequences of the ITS 1 suggest that some KCB accessions share a significant similarity with both R. occidentalis and R. coreanus in the ITS 1 region. The ITS 2 sequences of the three KCB accessions clustered more closely to those of two R. occidentalis accessions than to those of R. coreanus. These results suggest that there exist variations in the sequences of ITS among KCB accessions and KCB accessions are more closely related to black raspberry than R. coreanus in the ITS regions.
A variety of herbs and plants have been traditionally used in oriental folk medicine for the treatment of inflammatory diseases. In our attempt to search for anti-inflammatory agents from natural products, we investigated 64 methanol extracts from 42 medicinal plants belonging to 10 families which were evaluated for inhibitory activities of NO production in lipopolysaccharide (LPS)-stimulated macrophage RAW 264.7 cells. Among them, 16 extracts exhibited inhibitory activities of NO production (IC50 values ranging from 59.6 to 94.7 μg/ml). Only the extract from aerial parts of Hosta lancifolia (H. lancifolia) did not exert cytotoxic effects at the concentrations tested. The extract from H. lancifolia decreased the mRNA and protein levels of inducible nitric oxide synthase (iNOS) and pro-inflammatory cytokines in activated macrophage RAW 264.7 cells in dose-dependent manner. The results suggest that the extract may contain bioactive compounds that suppress expression of pro-inflammatory cytokines, which may prove beneficial with regard to the development of natural agents for prevention and treatment of inflammatory diseases.
The chemical composition of essential oil from the perennial herbaceous plants (Houttuynia cordata, Filipendula glaberrima, Peucedanum japonicum, and Ainsliaea acerifolia) was determined by GC/MS spectrometric analysis with the aid of NBS, Wiley Library and RI indice searches. The major constituents identified were α-phellandrene (18.97%), γ-terpinene (12.32%), decanal (8.72%), 1-decanol (10.92%), decanoic acid (12.12%), and 2-undecanone (12.32%) from H. cordata, farnesol (2.83%), l-α-terpineol (2.72%), benzenmethanol (2.03%), (Z)-3-hexen-1-ol (4.32%), and T-muurolol (2.07%) from F. glaberrima, α-phellandrene (14.25%), endobornyl acetate (3.84%), heptanal (47.52%), octanal (2.65%), (E,E)-2,4-decadienal (2.75%), and octanoic acid (4.52%) from P. japonicum, and geyrene (9.74%), β-cubebene (11.15%), berkheyaradulen (22.32%), β-elemene (6.21%), (-)-A-selinene (4.85%), benzaldehyde (4.52%), and benzenacetaldehyde (3.40%) from A. acerifolia.
This study was conducted to assess the response of LED (Light-emitting diode) irradiation on the growth characteristics and saponin contents of Panax ginseng C. A. Meyer. LED irradiation showed a positive effect for most of the parameters studied. The content of chlorophyll a in leaves was increased by 4.9~36.5%, under LED and fluorescent light conditions compared to the control. The content of chlorophyll b was also increased by 44.4~55.6% under blue and red LED compared to the control except under the red plus blue LED condition. The shoot and root weight were increased by 20~60% and 14.8~59.3%, respectively under LED and fluorescent light conditions compared to the control. The total saponin content was increased by 1.8% under blue LED compared to the control, while total saponin content was decreased by 8.8~11.5% under red LED, red plus blue LED and fluorescent light conditions.
Camptotheca acuminata, a native of South China is a well known natural source of monoterpene-indole alkaloid camptothecin(CPT), one of the most promising anti-tumoural compounds. This study was conducted to optimize plant growth regulators and culture conditions on plantlets regeneration through organogenesis from callus of Camptotheca acuminta. Callus were induced from various explants of in vitro germinated plantlets of C. acuminta using WPM medium containing 0.2 ㎎/L 2,4-D. Hypocotyl segments were exhibited higher embryogenic callus than the other explants. Shoot buds formation from embryogenic callus was affected by plant growth regulators, pre-treated dark condition and liquid culture. Organogenesis was optimal in WPM liquid medium containing 0.5 ㎎/L BA. The dark pre-treatment for 2 weeks before the solid culture was effective for organogenesis. The regenerated shoots were rooted in WPM medium with 0.2 ㎎/L NAA and successfully acclimated in green-house conditions.
Isatis tinctoria var. yezoensis (Ohwi) Ohwi (Cruciferae) is one of major natural dyeing crops in the world and also have used as a medicinal plant in Korea. We evaluated seed purity in F1-hybrid accessions using amplified fragment length polymorphism (AFLP) markers. One hundred sixty seeds from the male and female harvests were subsequently screened for seed purity with ten primers. The 13 accession-specific bands and many variable AFLP bands scored for accessions. Especially, E-AAC/M-CAA and E-AAG/M-CAT were presented clear hybrid bands for F1 hybrids. F1 hybrids maintained higher average level of genetic diversity compared with their correspondent parents. Self-inbred seeds from the female and male harvests were revealed 8.0% and 5.0%, respectively. The AFLP may lead to a better insight in to the hybrid seed purity test in I. tinctoria var. yezoensis.
This study was conducted to assess water movement in paddy-upland rotation soil scheduled for ginseng cultivation through the measurement of infiltration and permeability of soil water. Soil sample was divided with four soil layers. The first soil layer (to 30cm from top soil) was loamy sand, the second and the third soil layers (30~70 ㎝) were sand, and the fourth (〈 120 ㎝) was sandy loam. The soil below 130 ㎝ of fourth soil layer was submerged under water. The shear strength, which represents the resisting power of soil against external force, was 3.1 kPa in the first soil layer. This corresponded to 1/8 of those of another soil layer and this value could result in soil erosion by small amount of rainfall. The rates of infiltration and permeability depending on soil layers were 39.86 cm hr-1 in top soil, 2.34 cm hr-1 in 30~70 ㎝ soil layer, 5.23 cm hr-1 and 0.18 cm hr-1 in 70~120 ㎝ soil layer, with drain tile, and without drain tile, respectively. We consider that ground water pooled in paddy soil and artificial formation of soil layer could interrupt water canal within soil and affect negatively on water movement. Therefore, we suggest that to drain at 5 m intervals be preferable when it makes soil dressing or soil accumulation to cultivate ginseng in paddy-upland rotation soil to reduce failure risk of ginseng cultivation.
This study was performed to compare the effects of immuno-modulating activities of Rhodiola sachalinensis A. Bor. fractionized by consecutive solvent separation. The Cytotoxicity of all fractionized extracts on human kidney cell (HEK293) was lower than crude extracts. Generally, the butanol and chloroform extracts showed less cytotoxicity on about 10.07% and 9.67% than the crude extracts. For human immune B and T cell growth, chloroform fraction showed the highest cell growth compared to the control. The secretion of cytokines (IL-6, TNF-α) on human B and T cells were increased by adding chloroform extracts. Also, NK cell growth was significantly improved up to nearly 30% by adding the supernatant of B cell medium grown with the chloroform fraction. It was also found that chloroform fraction could yield higher nitric oxide production from macrophage than untreated control cells. Differentiation of HL-60 cells was increased up to 131.9% after treatment with chloroform fraction extracts, compared to the control. These results indicate that the chloroform fraction of R. sachalinensis have high immune activation activity than others fractions and the crude extracts, implying that this chloroform fractions could be used a new functional material.
The purpose of this study evaluated the immunoregulatory effect of phellinus linteus ethanol (PLE) extracts on liver damage on carbon tetrachloride (CCl4) induced in rats. Four-week old Male Sprague-Dawley rats were divided into the three experimental groups randomly; Control group, CCl4 group, CCl4 + PLE group. We found that effect of PLE on IFN-γ, STAT1 and pSTAT1 was decrease in vivo. Several genes were demonstrated to be IL-4 inducible prior to the discovery of STAT6. IL-4, STAT6 and pSTAT6 decreased significantly lower in CCl4 + PLE than the CCl4 group. Our data indicated that cytokine protein production were increased in CCl4 group with CCl4 + PLE group. In our data indicate that IgA levels in MLN lymphocytes were low, while IgE was high in CCl4 + PLE group compared with CCl4 group. Therefore, the results of this study show that PLE can be proposed to protect the liver against CCl4-induced immunoregulatory activity in rats.
