Background: The present study was carried out to asses whether mulberry leaves (MLs) have the potential to inhibit the mutagenic effect of cigarette smoke condensates (CSCs). Methods and Results: ML powder was extracted with 70% ethanol, and a yield of 35.1% by weight was obtained. The 70% ethanol extract of ML was further extracted sequentially using diethyl ether, chloroform, butanol, dichloromethane and water. The crude 70% ethanol extract of MLs and its solvent fractions did not show any mutagenic effect when tested at concentrations up to 1 ㎎/ plate against Salmonella typhimurium TA98. In contrast, the crude 70% ethanol extract showed an inhibitory activity against the mutagenicity of CSCs in the presence of S-9 mixture. Among the solvent fractions, the diethyl ether fraction showed the highest inhibitory activity, which increased in a dose-dependent manner, inhibiting mutagenesis by approximately 97.1% at a concentration of 1㎎/plate. Conclusions: In this study, we found that a crude 70% ethanol extract of MLs and the diethyl ether fraction themselves are potentially not mutagenic, but inhibit the mutagenic effect of CSCs.

Background: Dehisced ginseng seeds need to be stored at cold temperatures for around 3 months to break their physiological dormancy, and thus, to aid in gemination. In the presence of high moisture in such an environment, seed spoilage and pre-germination may lower seed quality and productivity. To improve seed quality during cold-stratification, the effects of seed dehydration and temperature were tested. Methods and Results: In early December, dehisced ginseng seeds were dehydrated at 4 different levels and stored at 2℃- 2℃, and –20℃ for 3 months. Germination was carried out on the filter papers moistened with distilled water; emergence of root, shoot, and seed spoilage were assessed. Seed viability was examined by the tetrazolium test. More than 90% of the seeds stored at 2℃ and –2 ℃ without drying or endocarp dehydration germinated, but seeds that were dehydrated to have a moisture content (MC) below 31% showed poor germination and lost their viability. In addition, the seeds stored at –20℃ failed to show effective germination. Conclusions: Seed storage after endocarp dehydration might help to improve seed quality and increase seedling's ability to stand during the spring-sowing of ginseng.

Background: In Korea, 6-year-old ginseng root is economically more important than 4 or 5-year-old roots. In general, the root age is determined by counting the number of stem vestiges. However, this method does not accurately estimate ginseng root age. Methods and Results: In this study, the stem vestige counting method was used to survey a total of 18,395 fresh ginsengs cultured in 2014, and 2015, to determine the accuracy of this method. The proportion of 6-year-old roots, with more than four stem vestiges, was 46.1% in 2014. For the cultivar Chunpoong cultivated in Eumseong and Goesan countries in 2015, the proportion of more than four stem vestiges was 55.9%, and 43.5%, respectively. The proportion of more than four stem vestiges for the Gumpoong cultivated in Eumseong and Yangpyeong countries was 67.0%, and 35.1%, respectively, whereas that for the cultivar Yunpoong was 36.0% and 61.0%, respectively. Moreover, it was confirmed that differences in the levels of Rg1 will enable root age determination. Conclusions: Root age determination by the stem vestige test was found to differ depending on the environmental and cultivation conditions. To determine the age of ginseng roots, a comprehensive method, such as counting stem vestiges and evaluating differences in ginsenoside levels, should be applied.

Background: The current regulations regarding the storage of medicinal herbs do not define the storage temperature; instead, but the only requirement is that the plastic bags used for storing the medicinal herbs should be tightly sealed during storage. The practice of storing medicinal herbs at room temperature (28℃), causes many problems, including growth of insect pests on the stored medicinal herbs. Thus, it is necessary to understand the effect of insect pests on the quality of medicinal herbs stored at room temperature (28℃) for the improving the relevant regulations. Methods and Results: Cnidium rhizome and Angelica radix were infested with Lasioderma serricorne F. adults and incubated at 28℃ for 2.5 and 5 months. The population of L. serricorne was established rapidly, and left many holes, cadavers, and feces on the stored medicinal herbs, thereby greatly damaging the product appearance and hygiene. In addition, active ingredients of the medicinal herbs were significantly decreased, probably reducing their medicinal quality as well. Conclusions: These results indicate that the current regulations are not sufficient to prevent the establishment of storage insect pests and guarantee the quality and hygiene of stored medicinal herbs. Therefore, it is necessary to devise proper storage protocols and upgrade the current relevant regulations to maintain the quality of medicinal herbs during storage.

Background: Cisplatin is one of the most extensively used chemotherapeutic agents for the treatment of cancer, including bladder, and ovarian cancers. However, it has been shown to induce nephrotoxicity, despite being an outstanding anti-cancer drug. In this study, we investigated the protective effect of dopaol β-D-glucoside (dopaol) on cisplatin-induced nephrotoxicity. Methods and Results: To confirm the protective effect of dopaol on cisplatin-induced nephrotoxicity, HK-2 cells were treated with 20 μM cisplatin and 80 μM dopaol. Cisplatin increased apoptosis, caspase-3 activity and mitochondrial dysfunction; however pretreatment with 80 μM dopaol successfully attenuated apoptosis, caspase-3 activity and mitochondrial dysfunction. To evaluate the protective effect dopaol on cisplatin-induced nephrotoxicity in vivo, we used an animal model (balb/c mice, 20 ㎎/㎏, i.p. once/day for 3 day). The results were similar to those obtained using HK-2 cells; renal tubular damage and neutrophilia induced by cisplatin reduced following dopaol injection (10 ㎎/㎏, i.p. once/day for 3 day). Conclusions: These results indicate that dopaol treatment reduced cisplatin-induced nephrotoxicity in vitro and in vivo, and can be used to treat cisplatin-induced nephrotoxicity. However, further studies are required to determine the toxicity high dose dopaol and the signal pathways involved in its mechanism of action in animal models.

Background: The production method of ginseng seedlings for ginseng cultivation is very important to ensure healthy rooting system as well as high quality, and yield of the resultant plants. This study was carried out to compare the growth characteristics of 2- year-old ginseng plants that were produced from seedlings grown in self soil nursery (SSN), nursery soil (NS) or hydroponic culture (HC). Methods and Results: The shading prop used was composed of four-layered 4 polyethylene (blue 3 + black 1) shade screen. The management of main field was done by inserting oil cake (1,200 ㎏/10 a) and then allowing Sudan grass to grow for a year. Seedling transplantation was carried out on April 6. Root growth was measured on October 25. Root weight was observed to be excellent at 6.0 g, following SSN transplantation. Root length was 21.2 ㎝ for HC seedlings, but these plants had a physiological disorder (i.e., rusty root), in 83.5% plants of this treatment. The ratio of PD/PT (protopanaxadiol saponins / protopanaxatriol saponins) was higher in NS seedlings. Plant analysis revealed that Fe content was lower in HC seedlings with high rustiness. The growth of 2-years-old ginseng was different following these varying seedling cultivation methods, but seedlings from NS were not different from those grown in SSN. Conclusions: For the propagation of 2-year-old ginseng plants, NS seedlings may be a good substitute for SSN seedlings.

Background: The application of crop rotation systems may reduce the occurrence of soil-borne diseases by releasing allelochemicals and by subsequent microbial decomposition. Methods and Results: For reduction of ginseng root rot by the crop rotation system, after harvesting 6-year-old ginseng, fresh ginseng was grown along with continuous cultivation of sweet potato, peanut, and bellflower. Growth of 2-year-old ginseng was significantly inhibited in the continuous cultivation than in the first cultivation. Sweet potato, peanut and bellflower cultivations assisted in obtaining normal yields of ginseng in the first year after the harvest of 6-year-old ginseng. Salt concentration, potassium and sodium contents were gradually decreased, and, organic matter was gradually increased through cirp rotation. Phosphate, calcium and magnesium contents were not altered. The density of the root rot fungus was gradually decreased by the increase in crop rotation; however it was decreased distinctly in the first year compared to the second and third year. The severity of root rot disease tended to decrease gradually by the increase of crop rotation. Conclusions: Short-term crop rotation for three years promoted the growth of ginseng, however root rot infection was not inhibited significantly, although it was somewhat effective in lowering the density of the root rot pathogen.
