The aim of this investigation was to examine the influence of extrusion on dietary fiber profile and the contentof bioactive compounds, rutin and quercetin in young sprout, whole seed, and matured stem of Tartary buckwheat.WSI(water soluble index) is increased by a function of both screw profile and process temperature, compared to control indifferent parts of Buckwheat. Also, WSI of ME is increased more than 5.2 times in grain, compared to that of control. Theeffect of precooking by extrusion on the dietary fiber profile of buckwheat flour was evaluated. Precooking by extrusion sig-nificantly increased SDF in flour, although in most cases extrusion decrease in TDF a little. The thermo-mechanical treat-ment undergone by the buckwheat flour during extrusion led to redistribute part IDF fraction to SDF, leading to an increasein the latter. The content of rutin was increased about two fold in extruded flour of sprout, compared to in control. Thisincrease maybe why these compounds are released from cell wall by high shear processing under high temperature.

The present study investigated an ethanol extract (HS0608) of a mixture of three medicinal plants of Curcumalongae radix, Phellinus linteus, and Scutellariae radix for possible neuroprotective effects on neurotoxicity induced by amyloid βprotein (Aβ) (25-35) in cultured rat cortical neurons and antidementia activity in mice. Exposure of cultured cortical neurons to10µM Aβ (25-35) for 36h induced neuronal apoptotic death. At 1-50㎍/㎖, HS0608 inhibited neuronal death, elevation of intra-cellular calcium concentration ([Ca2+]i), and generation of reactive oxygen species (ROS) induced by Aβ (25-35) in primary cul-tures of rat cortical neurons. Memory loss induced by intracerebroventricular injection of ICR mice with 15 nmol Aβ (25-35) wasinhibited by chronic treatment with HS0608 (25, 50 and 100㎎/㎏, p.o. for 7 days) as measured by a passive avoidance test. Fromthese results, we suggest that the antidementia effect of HS0608 is due to its neuroprotective effect against Aβ (25-35)-inducedneurotoxicity and that HS0608 may have a therapeutic role in preventing the progression of Alzheimer’s disease.

The low quality fresh ginseng was extracted by water at 80℃ and 240bar for 20min (HPE, High pressureextraction process). The cytotoxicity on human normal kidney cell (HEK293) and human normal lung cell (HEL299) of theextracts from HPE showed 28.43% and 21.78% lower than that from conventional water extraction at 100℃ in adding themaximum concentration of 1.0㎎/㎖. The human breast carcinoma cell and lung adenocarcinoma cell growth were inhib-ited up to about 86%, in adding 1.0㎎/㎖ of extracts from HPE. This values were 9-12% higher than those from conven-tional water extraction. On in vivo experiment using ICR mice, the variation of body weight of mice group treated freshginseng extracts from HPE of 100㎎/㎏/day concentration was very lower than control and other group. The extracts fromHPE was showed longer survival times as 35.65% than that of the control group, and showed the highest tumor inhibitionactivities compared with other group, which were 70.64% on Sarcoma-180 solid tumor cells. On the high performance liq-uid chromatogram (HPLC), amount of ginsenoside-Rg2, Rg3, Rh1 and Rh2 on fresh ginseng were increased up to 43-183%by HPE, compared with conventional water extracts. These data indicate that HPE definitely plays an important role ineffectively extracting ginsenoside, which could result in improving anticancer activities. It can be concluded that low qualityfresh ginseng associated with this process has more biologically compound and better anticancer activities than that fromnormal extraction process.

Hepatoprotective and antioxidant activities of Acer mono and A. okamotoanum were compared after beingextracted by low temperature and high pressure (LTHP) at 20 MPa and 60℃ for 15 minutes. Extraction yield of both A.mono and A. okamotoanum was increased about 40~43% by this extraction process. On scavenging activities, the bark of A.okamotoanum from this extraction process showed the highest activity as about 97%. This value was higher than that fromconventional water extraction and A. mono extracts. Both of A. mono and A. okamotoanum showed high ability on nitritescavenging, but decreasing tendency according to increasing of pH. On SOD-like test, A. okamotoanum had the highestactivity as 46.28% at 1.0㎎/㎖ concentration. A. okamotoanum extracted by LTHP also showed the highest activity as197.38% in adding 1.0㎎/㎖ concentration. Generally, the extracts from low temperature and high pressure extraction pro-cess are higher hepatoprotective and antioxidant activities than that from conventional water extraction. This study was carried out to select economical byproduct composts as the substitute for the traditionalorganic fertilizer, Yacto, in the cultivation of ginseng seedlings, and to investigate the application method of a selected com-post. Among tested byproduct composts, popped rice hull compost was the best substitute for Yacto, while the application ofdomestic animal manure composts resulted in red skinned roots of ginseng seedlings. Optimal mixing ratio of the poppedrice hull compost with virgin soil (fine sand) were 3~4:1 in bulk, showing the same root yield compared to that of conven-tional seedbed soil. When the popped rice hull compost was lower than 1±0.1% in nitrogen content, the expeller cake of oilseed was added to seedbed soil to rise nitrogen content until 1±0.1%.

The purpose of this study was to identify soil physical characteristics as guideline for high yield potential inginseng cultivated field which produced 6 years root. Harvest yields of ginseng to be divided by parent rock was in order ofphyllite and red shale 3.1㎏/3.3㎡ > granite and gneiss·schist 3.0㎏ > basalt 2.6㎏ > porphyry 2.2㎏ in upland and forestsoil. Also, with classified by topography, it was in order of foot slope and alluvial fan 3.2㎏/3.3㎡ > valley 3.0㎏ > low hill2.9㎏ > hill, lave flow and dilluvial terrace 2.8㎏ in survey tilth. Class determination of soil texture, it was in order of sandyloam 3.1㎏/3.3㎡ > loam and silt loam 3.0㎏ > clay loam 2.9㎏ > silt clay loam 2.8㎏ in survey tilth. Slope condition offarming land, in case of sloping (2~7%), it was 3.1㎏/3.3㎡ but deep sloping (15~30%) caused decreasing harvest yield. Indrainage classes (excessively, well and moderately well), there was no significantly different in harvest yields. Relationshipbetween harvest yield and soil series, Production sites as yielding 3.0㎏/3.3㎡ were seven sites, also it was contained 14 soilseries. Production sites as yielding 2.5~3.0㎏/3.3㎡ were eleven sites, it was contained 16 soil series. Production sites asyielding 2.0~2.5㎏/3.3㎡ were 10 sites, it was contained 4 soil series.

Volatile flavor compounds from the shoot and root of Angelica gigas Nakai were extracted by HE (Hydrodis-tillation extraction), SDE (Simultaneous steam distillation & extraction), and SFE (Supercritical fluid extraction system),and analyzed by GC-MS. The amount and the number of chemical components in essential oils from shoot and root by SFEwas the higher than those by other extraction methods. Respectively, thirty one constituents were identified from the essen-tial oil of the shoot and root by HE, twenty seven and twenty three constituents were identified from the essential oil of shootand root by SDE, thirty one and forty five constituents were identified from the essential oil of shoot and root by SFE. Theresult showed large differences in extraction methods and in plant parts of Angelica gigas Nakai. Also, the bioactive com-pounds in root part was identified as nodakenin and decursinol (11.95% and 8.42%, respectively) by SFE. These results sug-gested that SFE was the best extraction method for the increasing of extraction yield, the determination of volatilecomponents and the increasing of bioactive compounds in the shoot and root of Angelica gigas Nakai.

For the development of a new antidementia functional food or alternative drug using agricultural products,Job's tears (Coix lachrymajobi L.), which shows high acetylcholinesterase (AChE) inhibitory activity (55.1%) was selectedand the extraction conditions of AChE inhibitor were optimized. AChE inhibitor of Job's tears was maximally extractedwhen it was treated with 60% methanol at 40℃ for 6h. The AChE inhibitor of the methanol extracts was partially purifiedby systematic solvent extraction, thin layer chromatography, silica gel chromatography and reverse-phase HPLC and thepartial purified AChE inhibitor with inhibitory activity (IC50) of 0.608㎍ was obtained. The partial purified AChE inhibitorwas soluble in methanol and hexane, and insoluble in water. Its maximum absorption spectra was 230㎚ and also it was sta-ble in the range of 30℃ and 70℃ and pH 4.0-8.0 for 1h.

This study was to set the guidelines of soil chemical components in order to assure the safety and quality ofthe panax ginseng from physiological disorder. The disorder symptoms appeared on the leaf with yellow spot, atrophy, yel-low-brown spot, also showed red skin and rough skin of the root. Occurrence type of physiological disorder in cultivatedfield divided into two types:type I ‘such as, yellow spot’ consist of single disorder symptom; type II ‘such as, yellow spotand yellow-brown spot’ consist of two or more different disorder symptoms. The individual contribution of soil properties tothe occurrence type was as follows ; The yellow spot was affected by Na>NO3-N>salinity (EC) in soil. The same results wasobserved in red skin. Atrophy was affected by NO3-N>salinity (EC)>Ca>Mg. Rough skin was affected by P2O5>pH>Organic material > K. It showed positive associated to P2O5, pH and K, but negative associated to organic matter. Simulta-neous occurrence of two different disorder, including cases which yellow spot and yellow-brown spot, those were affected byNO3-N > salinity (EC) > Na > Mg. In the case of atrophy plus yellow-brown spot, those also were affected by in the order :NO3-N > salinity (EC) > Ca > Mg > Na. Red-rough skin was affected in the order : salinity (EC) > NO3-N > K > Na. Soilchemical components appear to be related to occurrence of physiological disorder, particularly in salinity (EC) and NO3-N.The salinity (EC) and NO3-N were negative related to plant growth. In addition, exchangeable cation capacity play criticalroles in attributing to complex occurrence of physiological disorder.

This study was undertaken to develop a technique of discrimination using SSR makers in boxthorn cultivars.Forty one boxthorn cultivars, which were collected from Korea and China, were evaluated by 10 SSR markers. Total of 61alleles were detected, ranging from 3 to 13 with an average of 6.1 alleles per locus. The averages of gene diversity and PICvalues were 0.482 and 0.428, with a range from 0.25 (GB-LCM-022 and GB-LCM-087) to 0.83 (GB-LCM-167) and from0.24 (GB-LCM-022 and GB-LCM-087) to 0.81 (GB-LCM-167), respectively. Five markers out of 10 markers, GB-LCM-022, GB-LCM-075, GB-LCM-104, GB-LCM-167 and GB-LCM-217, were selected as key markers for discrimination inboxthorn cultivars. All of boxthorn cultivars were individually distinguished by the combination of five SSR markers.

This study was carried out to investigate the correlation between root diameter and ginsenoside compositionof Panax ginseng C. A. Meyer cultivar Yunpoong. Dry matter ratio of main root was a little higher than that of lateral rootand fine root, and that was higher by the increase of root diameter in the same root parts. Total ginsenosides composition ofmain and lateral roots increased by the decrease of root diameter, especially in lateral root. Similar resulted in fine root, butthere was no significant difference where root diameter was below 2.5㎜. Except for ginsenoside-Rg1, other ginsenosidescomponent, PDs, PTs and total ginsenosides had highly negative correlation with the root diameter within whole root, mainroot+lateral root and lateral root+fine root, while Rg1 had positive correlation with the root diameter.

This study was conducted to find out some conditions for optimum cultivation of Angelica gigas Nakai by theinvestigation of root yield and decursin content from different seeding sizes and accumulative temperature. Accumulativetemperature from April to October was 4,309℃ on altitude 100m , 4,242℃ on 250m, 3,662℃ on 530m, 3,435℃ on 730m,and altitude 530~730m was less 650~870℃ than altitude 100m cultivation areas of A. gigas. Seedling stand rate wasincreased from 86.4% to more than 90% as accumulative temperature decreases, and was increased in above 7㎜ of seed-ling size, and 10% in non-mulching more than PE film mulching. Yield was increased as accumulative temperaturedecreases and in PE film mulching as 310.2㎏/10a. Also, Yield was increased in 7~9㎜ than seedling diameter 5~7㎜ as313.0㎏/10a. Decursin content of primary roots was increased as altitude rises, ie, as 2.55% on altitude 100m, 3.33% on250m, 5.51% on 530m, and 6.24% on 730m. Decursinol angelate content appeared little than decursin content as 1.08%on altitude 100m, 1.37% on 250m, 1.99% on 530m, 2.38% on 730m, and as altitude was heightened, content was increased.

This study was conducted to find out environment-friendly disease control method to Alternaria blightcaused by Alternaria panax of ginseng. For this study, 150 methanol extracts from medicinal plants were evaluated and theextract of Impatiens balsamina showed most strong antifungal activity against A. panax. The methanol extract of I. balsam-ina showed also stable antifungal activity against other ginseng pathogens such as Botrytis cinerea and Fusarium sp., whentreated by heat or pH. In vivo, Alternaria blight incidence rate was low of 13% with the treatment of I. balsamina methanolextract compared to 35% of the non-treatment. The antifungal compound of I. balsamina was purified and identified byusing a silica gel column chromatography, TLC and ESI-LC/MS/MS analysis. The compound which showed strong antifun-gal activity was identified as 2-methoxy-1,4-naphtoquinone (C11H8O3).

Achyrantes japonica (AJ) has been used to treat edema and arthritis in the traditional Korean medicine. Toelucidate the anti-inflammatory and anti-nociceptive effects of ethanol extract of AJ, the carrageenan-induced pawedema using a plethysmometer and thermal hypersensitivity using the plantar test were measured. Ibuprofen was used asa control drug. Treatment with AJ (200㎎/kg p.o.) significantly reduced paw edema, compared to the carrageenan -treated rats. In the plantar test, the thermal withdrawal latency in AJ - treated group was significantly increased than thecarrageenan - treated group. The results indicate that AJ could have be the anti-inflammatory and anti-nociceptive prop-erties.
