An efficient regeneration system was developed using leaf, petiole, and internode explants. Highly embryogenic callus was obtained following cultivation on MS basal nutrient supplemented with 2 mg/l 2,4-D. Globular, heart, torpedo and cotyledon shaped somatic embryo were produced from the surface of embryogenic callus. Direct shoot regeneration without intermediate callus formation has been achieved on MS medium supplemented NAA and BAP. The percentage of response varies with different concentration of auxin and cytokinin treated individually or in combination. The best shoot regeneration response (54.28%) and number of shoot per explant (12.67) were achieved on the medium supplemented with 0.1 mg/l NAA and 1 mg/l BAP. The regenerated shoot transformed into young plant when cultured into elongation and root induction medium. More than 90% of in vitro propagated plants could survive when transferred to the greenhouse for acclimation. This optimized regeneration system can be used for rapid shoot proliferation and genetic transformation.

From the EtOAc fraction of Eugenia caryophyllata, four compounds were isolated through activity-guided silica gel column chromatography, From the result of spectroscopic data including NMR, MS and IR, the chemical structures of the compounds were determined as 1-allyl-4-hydroxy-3-methoxybezene acetate (eugenol acetate, 1), 1-allyl-4-hydroxy-3-methoxybezene (eugenol, 2), 3β-hydroxyolean-12-en-28-oic acid (oleanolic acid, 3) and 2α, 3β-dihydroxyolean-12-en-28-oic acid (maslinic acid, 4). Compounds 3 and 4 were isolated for the first time from this plant. Also, compounds 1, 2 and 3 exhibited relatively high platelet aggregation inhibitory activity with the IC50 values of 0.24, 0.09 and 0.07 mM, respectively. Compound 2 significantly prolonged activated partial thromboplastin time (aPTT) with the value of 124±11.2 seconds as compared to the control with the value of 37.5±2.2 seconds at the concentration of 50 μg/ml. Compounds 1 and 3 revealed inhibitory activity on farnesyl protein transferase (FPTase) with the IC50 values of 0.49 and 0.24 mM and compounds 1 and 2 highly inhibited the growth of rat-H-ras cells with the Gl50 values of 6.63 and 5.70 μm, respectively.

The effects of angiogenesis inhibitor from the extract libraries of Korean and Chinese medicinal plants were investigated using a protein microarray chip. Protein chip was constructed by immobilization of integrin α5β1 on protein chip base plates and employed far screening active extracts that inhibit the integrin-fibronectin interaction from the extract libraries. The 100 extracts of medicinal plants were obtained from extract bank of National Institute of Crop Science, RDA. The 14 extracts among 100 extract libraries were shown efficient inhibition activity for the interaction between integrin-fibronectin. The medicinal plants of 14 extracts were Vitex negundo var. incisa (Lam.) C.B. Clarke, Epimedium koreanum Nakai, Cedrela sinensis A. Juss, Ipomea aquatica Forsk, Schisandra chinensis Baill, Pulsatilla koreana Nakai, Paeonia lactiflora Pall. var.hortensis Makino, Oenothera odorata, Allium chinense, Allium victorialis var. platyphyllum MAKINO, Polygonatum odoratum Druce var. pluriflorum Ohwi, Hosta lancifolia, Agrimonia pilosa L. var. japonica Nakai and Potentilla chinensis SER. The Paeonia lactiflora, Oenothera, and Agrimonia pilosa from these 14 extracts libraries were shown strong inhibition activity of integrin α5β1.

The cost of conventional cultivation of ginseng (Panax ginseng C.A. Meyer) is very expensive, because shadow condition should be maintained during cultivation periods owing to inherently weak plant for high-temperature. Therefore, application of plant biotechnology may be possible to overcome these difficulties caused by conventional breeding of ginseng. Transgenic plants were produced via Agrobacterium tumefaciens Gv3101, both carrying the binary plasmid pBI121 mLPISO with nptII and Iso (isoprene synthase) gene. Integration of the transgenes into the P. ginseng nuclear genome was confirmed by PCR analysis using nptII primers and Iso primers. RT-PCR result also demonstrated the foreign isoprene synthase gene in three transgenic plant lines (T1, T3, and T5) which was expressed at the transcriptional level. When whole plants of transgenic ginseng were exposed to high temperature at 46℃ for 1 h, a non-transformed plant was wilted from heat shock, whereas a transgenic plant appeared to remain healthy. We suggest that the introduction of exogenous isoprene synthase is considered as alternative methods far generating thermotolerance ginseng.

A protocol for the production of transgenic Panax ginseng C.A. Meyer was established via Agrobacterium tumefaciens-mediated genetic transformation of direct somatic embryos. A number of conditions related to the co-cultivation were tested with respect to maximizing transformation efficiency. The results showed that pH of the co-cultivation medium (5.7), the bacterial growth phase (optical density; OD600 = 0.8), co-cultivation period (3 days), and acetosyringone concentration (100 μM) had positive effects on transformation. Selected plantlets were cultured on the medium at an elevated hygromycin level(30 mg/l). Integration of the transgenes into the P. ginseng nuclear genome was confirmed by PCR analysis using hpt primers and by Southern hybridization using hpt-specific probe. The transgenic plantlets were obtained after 3-month cultivation and did not show any detectable variation in morphology or growth characteristics compared to wild-type plants.

The protective effect of ethanol extract of Korean mistletoe (KM; Viscum album coloratum) on hydrogen peroxide (H2O2)-induced neurotoxicity was examined in primary cultured rat cortical neurons. H2O2 reduced viability of cortical neurons in a concentration-dependent manner. The addition of KM, over a concentration range of 10 to 100 μg/ml, concentration-dependently prevented the H2O2(100 μM)-induced neuronal cell death, as assessed by a 3-[4,5-dimethylthiazol-2-yl]-2,5-di-phenyl-tetrazolium bromide (MTT) assay and Hoechst 33342 staining. KM significantly inhibited H2O2-induced elevation of the cytosolic Ca2+ concentration ([Ca2+]c), which was measured by a fluorescent dye, fluo-4 AM. KM inhibited glutamate release into medium and generation of reactive oxygen species (ROS) induced by H2O2. These results suggest that KM may mitigate the H2O2-induced neurotoxiciy by interfering with the increase of [Ca2+]c, and inhibiting glutamate release and generation of ROS in cultured neurons.

Effects of Oplopanax elatus have known to various pharmaceutical therapies. However, chemical prosperities in the plant are rarely investigated. In order to detect biological activity, we evaluated the antioxidant activity of five fractions from methanolic extracts in each part of O. elatus. Also, contents of polyphenols and polysaccharides were measured. Five fractions were of sub-fractions using n-hexane, ethyl-acetate, n-butanol, and water from methanolic extracts. Ethylacetate and n-butanol fractions from stem and root exhibited strong antioxidant activity and high total phenolics content. On the HPLC analysis, ten free phenolics, including p-hydroxybenzoic acid, chlorogenic acid, caffeic acid, syringic acid, p-cou maric acid, ferulic acid, naringin, hesperidin, quercetin and trans-cinnamic acid, were identified from the fractions and were shown to different quantitative proportions. Furthermore, ethylacetate and n-butanol fraction had the highest amount of chlorogenic acids, one of the cinnamic acid derivatives possessing pharmacological properties. These results indicated that the fractions of O. elatus, as well as methanolic extracts, could be used as natural antioxidative ingredients.

The objective of this research was to evaluate the ability of water and ethanol extracts from mulberry fruit (Morus alba L.) to influence the inhibitory activity of angiotensin converting enzyme (ACE) and xanthine oxidase(XOase). The total phenol contents and sixteen phenolic compounds were investigated in water and ethanol extracts. In order to understand the factors responsible for the potent antioxidant and antihypertensive ability of mulberry, it has been evaluated for anti-oxidative activity using Fenton's reagent/ethyl linoleate system and for free radical scavenging activity using the 1,1-diphenyl-2-picryl hydrazyl free radical generating system. The total phenol contents and total of phenolic compounds in ethanol extract showed higher levels than water extract in mulberry fruit six phenolic compounds (chlorogenic acid, narigin, syringic acid, quercetin, naringenin, kampferol) has a higher individual phenolic compound content in the 60% ethanol extraction than 80% ethanol extract. The inhibitory activity on angiotensin converting enzyme (ACE) were highest in 80% ethanol extract (9.0%). Also, activity of xanthine oxidase(XOase) inhibition appeared highest in 80% ethanol extracts and correlated well with the total phenolic content, which was modulated by the concentration of individual phenolic compounds. This result revealed, that strong biological activity was caused by specific phenol compound contents. Utilization of water and ethanol extracts from mulberry fruit are expected to be good candidate for development into source of free radical scavengers and anti-hypertentive activity

This study was carried out to investigate inhibitory effect of extracts from Artemisia capillaris Thumb. on maltase, sucrase, α-amylase, nonspecific α-glucosidase, and postprandial hyperglycemia. Methanol extract and organic solvent (n-hexane, ethyl acetate, butanol, aqueous) fractions from the medicinal herb were determined for the inhibitory activities against maltase, sucrase and α-amylase. The methanol extract from A. capillaris strongly inhibited maltase (57%) and α-glucosidase (72%) at the concentration of 100 μg/ml. Among the four fractions (n-hexane, ethyl acetate, butanol, aqueous) examined, the butanol fraction from A. capillaris showed potent inhibitory effects on maltase (73%), sucrase (33%), and α-amylase (75%) at the concentration of 100 μg/ml. The butanol fraction from Artemisia capillaris also exhibited significant reductions (20%) of blood glucose elevation in mice loaded with maltose. These results suggest that the extract from Artemisia capillaris can be used as a new nutraceutical for inhibition on postprandial hyperglycemia

Inhibitory effect of Scutellaria baicalensis ethanol extracts (SR) on chemical mediator release and immunoglobulin (Ig) production from Sprague-Dawley rats originated cells as type I allergic reaction was examined. SR showed concentration-dependent inhibition on basal and concanavalin A (ConA)-stimulated Ig production. In the mesenteric lymph node lymphocytes, the inhibitory effect of SR on the IgE production in the presence of Con A was stronger than these on IgA and IgG production. Moreover, tumor necrosis factor-alpha (TNF-α) production-inhibiting effect of SR in the presence ConA was observed. However, SR did not affect the production of interferon-γ. SR also inhibited histamine release from the peritoneal exudate cells stimulated with a calcium ionophore A23187. In the case of leukotriene B4, SR markedly inhibited it at the concentration of 100 mg/ml. From these results, ethanol extracts obtained from Scutellaria baicalensis may have an anti-allergic effect on the intestinal system of rats.

Good quality seedlings produced in the seedbed of Yangjik, traditional seedling cultivation, is one of the most important factors in determining the yield and quality of 4~6-year-old ginseng. This study was carried out to substitute Yacto, traditional organic fertilizer, for economical compost in the cultivation of seedling by fertilizing relatively little amount of compost into seedbed soil. Bulk density and solid phase were decreased in physical properties of seedbed soil, while air phase and porosity were increased by more addition of compost. When the amount of applied compost in seedbed soil was above 8l per Kan, the contents of nutrient were exceeded the range of optimal standard for ginseng cultivation. Chlorophyll content and stem length were increased by more addition of compost, while the length and the width of leaves showed the highest value at the application level of 8l per Kan. Heat injury was also increased distinctly above the application level of 8l per Kan. The number of first grade seedlings and usable seedlings, and fresh root weight per plant showed the peak at application level of 8l per Kan, respectively. Fertilizing the compost of 8l per Kan into seedbed soil was the optimal amount for producing the good quality seedlings.
