Dendrobium moniliforme (DM) is a valuable and versatile herbal medicine with the anecdotal claims of antioxidant and anti-inflammation. In the present study, we investigated the whitening and anti-wrinkling effects of DM under various conditions with B16F10 melanoma cells and human dermal fibroblasts. The DM extract inhibited melanin contents and tyrosinase activity in a dose-dependent manner, compared with untreated group. Treatment of the DM extract effectively suppressed the α-MSH-stimulated melanin formation, tyrosinase activity and dendrite outgrowth. Moreover, the α-MSH-induced mRNA expressions of tyrosinase-related protein-1 (TRP-1) and tyrosinase-related protein-2 (TRP-2) and protein expression of tyrosinase were significantly attenuated by DM treatment. We also investigated the DM increased the production of type I procollagen and inhibited TNF-α-induced mRNA expressions of MMP-1, -3 in the human dermal fibroblast. These results indicate that DM may be a great cosmeceutical ingredient for its whitening and anti-wrinkle effects.

The transcriptomes of four ginseng accessions such as Cheonryang (Korean ginseng cultivar), Yunpoong (Korean ginseng cultivar), G03080 (breeding line of Korean ginseng), and P. quinquefolius (American ginseng) was characterized. As a result of sequencing, total lengths of the reads in each sample were 156.42 Mb (Cheonryang cultivar), 161.95 Mb (Yunpoong cultivar), 165.07 Mb (G03080 breeding line), and 166.48 Mb (P. quinquefolius). Using a BLAST search against the Phytozome databases with an arbitrary expectation value of 1E-10, over 20,000 unigenes were functionally annotated and classified using DAVID software, and were found in response to external stress in the G03080 breeding line, as well as in the Cheonryang cultivar, which was associated with the ion binding term. Finally, unigenes related to transmembrane transporter activity were observed in Cheonryang and P. quinquefolius, which involves controlling osmotic pressure and turgor pressure within the cell. The expression patterns were analyzed to identify dehydrin family genes that were abundantly detected in the Cheonryang cultivar and the G03080 breeding line. In addition, the Yunpoong cultivar and P. quinquefolius accession had higher expression of heat shock proteins expressed in Ricinus communis. These results will be a valuable resource for understanding the structure and function of the ginseng transcriptomes.

This study was designed to maximize extract yields of functional components in Angelica gigas Nakai by extrusion process. In addition, the optimizing condition of the extrusion process was established to increase the extraction of the functional components in A. gigas. The total polyphenol was increased by 40% compared to a control. And the optimizing condition was that the screw speed was 62.76 rpm, the amount of A. gigas powder was 34.79 kg/h, and the amount of water was 4.44 kg/h. The result of antioxidant activities of A. gigas extrusion, the inhibition of oxidization had the effect of 10.29 ~ 14.59% compared to a control. The content of decursin and decursinol angelate, which were index components, was 6.37%; it was increased by 16.64% compared to a control (5.31%) and showed a significantly difference (p < 0.05). And the optimizing condition was the screw speed 93.71 rpm, the amount of A. gigas powder was 28.67 kg/h, and the amount of water was 9.9 kg/h.

This study was conducted to find out the optimum composition of nursery soil for raising seedling of ginseng (Panax ginseng C. A. Meyer). Total 9 kinds of raw materials were used such as peat-moss, perlite, leaf mould, rice bran, gull's guano, castor-oil plant bark, palm bark, cow manure and chicken manure for optimum composition of nursery soil in ginseng. Occurrence of damping-off in ginseng was lowered about 50% in nursery soil type 1, 2 and 4 than in other types nursery soil in June, and occurrence rate of rusty root also lowest in nursery soil type 1. As the salinity of nursery soil increased, so did the occurrence of physiological disorder in ginseng seedling. The cause of salinity increasing in nursery soil has closely relation to NO3-N, P2O5 and Na+ content. Plant height, root length, diameter and weight were longer and heavier in nursery soil type 1 (mixing ratio of peat-moss, perlite and leaf mould was 50 : 20 : 30 based in volume) than in other types of nursery soil. So nursery soil type 1 was selected for raising seedling of ginseng. pH and electric conductivity (EC) of selected nursery soil type 1 was 5.55 and 0.13 dS/m. Contents of NO3-N and P2O5 were 21.0 and 40.0 mg/L, and K+ 0.36, Ca2+ 3.38, Mg2+ 2.01 and Na+ 0.09cmol+/L, respectively.

In order to find out anticancer activity of Korean pepper (Capsicum annuum L.), the cytotoxicity against 8 cell lines including 293 (normal kidney cells) and A-431 (epidermoid carcinoma cells) of extracts by extraction solvents and plant parts were investigated using MTT assay. Also the correlation between content of capsaicin known as anticancer ingredient and cytotoxicity of extracts from pepper were analyzed. The distilled water extracts from seed and germinated seed showed very high cytotoxicity against 6 cancer cell lines including A549 (lung carcinoma cells), AGS (stomach adenocarcinoma cells), HeLa (cervix adenocarcinoma cells), HepG2 (hepatoblastoma cells), HT-29 (colon adenocarcinoma cells), and MCF-7 (breast adenocarcinoma cells). But 80% ethanol and methanol extracts showed cytotoxicity against 293 and AGS. The RC50, that was, the concentration of sample required for 50% reduction of cell viability, of seed and germinated seed extracts against AGS were 33.4~389.1μg/mℓ and 63.9~1,316.7μg/mℓ, respectively, so anticancer activity was higher in seed than in germinated seed. In capsaicin contents, seed with high cytotoxicity and pericarp with a little cytotoxicity contained 47.4~1,260.0μg/g and 58.3~1,498.0μg/g, respectively. As these results, the correlation was not between cytotoxicity and capsaicin content.

Curcuma longa L. leaf was extracted by water at 60˚C for 12 hours after being treatment of Ultra High Pressure under 500 MPa for 5-15 minute. The high pressure extraction for 15 minute (HPE15) was measured the highest extraction yield of 25.48% (w/w), compared to those from conventional extraction methods. The HPE15 showed the lowest cytotoxicity as 11.97% in adding 1.0mg/mℓ of concentration. Also, HPE15 was measured the highest inhibition of hyaluronidase as 44.48% in adding 1.0mg/mℓ. In addition, The production of NO from macrophages was measured as 7.06μM in adding 1.0mg/mℓ of HPE15, which was lower than the those from others processes. Finally, HPE15 significantly reduced up to 649.44pg/mℓ of ProstaglandinE2 production from UV-irradiation. These results suggest that the Curcuma longa Linne leaf extract from high pressure process might enhance the skin immune activities possibly by high elution of active components than other processes.

This study was carried out in order to investigate the early growth and root development characteristics of Peucedanum japonicum container seedlings. The experiment was performed by containers (128 and 200 cavities containers) and shading levels (0%, 35%, 50% and 75% shading). Germination rate of P. japonicum seeds was getting higher in the lower shading level and the highest in the full sunlight (71.9%). Plant height was the highest under 50% shading of all containers. Fresh weight and dry weight of the shoot (leaves + stem) were the highest under 50% shading of all containers, too. Meanwhile, fresh weight and dry weight of the root per plant were the highest under the full sunlight of 128 cavities container such as 0.34 g and 0.03 g, respectively. Total root length, root project area and root surface area were higher under the full sunlight of 128 cavities container such as 234.5 cm, 6.6cm2 and 20.8cm2, respectively and the next higher was under 35% shading such as 201.7 cm, 5.9cm2 and 18.4cm2, respectively. A case of root volume was the highest with 0.15cm3 under the full sunlight. As a result of the surveying the whole experiment, it is concluded that the shoot and root of P. japonicum seedling grow nicely by maintaining 35% shading.

To control the disease of root rot in ginseng nursery, inorganic sulfur solution of 0.1%, 1.0%, and 2.0% were irrigated by amount of 10ℓ per 3.3m2 before sowing. On the last ten days of July, Fusarium solani and F. oxysporum were similarly detected by 44.8% and 43.8%, respectively, while Cylindrocarpon destructans was low detected by 4.4% in the diseased seedling. The more sulfur's concentration was increased, the more soil pH was decreased. Soil pH was decreased from 5.87 to 4.59 by the irrigation of sulfur solution of 1.0%. The more sulfur's concentration was increased, the more electrical conductivity (EC) of soil was increased. EC was increased from 0.27 dS/m to 1.28 dS/m by the irrigation of sulfur solution of 1.0%. Irrigation of sulfur solution was effective on the inhibition of damping-off caused by Rhizoctonia solani in ginseng seedling. Control value for damping-off by the irrigation of sulfur solution of 1.0% and 2.0% were 75.7%, and 78.5%, respectively. Growth of leaf was inhibited by the irrigation of sulfur solution of 2.0%. Root weight per 3.3m2 showed the peak in sulfur solution of 1.0%, while survived-root ratio and root weight per plant were decreased in the level of 2.0%. Survived-root ratio of seedling in sulfur solution of 1.0% was distinctly increased by 4.7 times compare to the control, but control value for root rot was relatively low as 49.2%. Mycelium growth of C. destructans, F. solani, and R. solani were distinctly inhibited by the increase of sulfur's concentration in vitro culture using PDA medium.

Salt stress is one of the major abiotic stresses affecting the yield of ginseng (Panax ginseng C. A. Meyer). The objective of this study was to identify bio-marker, which is early responsive in salt stress in ginseng, using proteomics approach. Ginseng plants were exposed to 5 ds/m salt concentration and samples were harvested at 0, 6, 12 and 18 hours after exposure. Total proteins were extracted from ginseng leaves treated with salt stress using Mg/NP-40 buffer and were separated on high resolution 2-DE. Approximately 1003±240 (0 h), 992±166 (6 h), 1051±51 (12 h) and 990±160 (18 h) spots were detected in colloidal CBB stained 2D maps. Among these, 8 spots were differentially expressed and were identified by using MALDI-TOF/TOF MS or/and LC-MS/MS. Ethylene response sensor-1 (spot GL 1), nucleotide binding protein (spot GL 2), carbonic anhydrase-1 (spot GL 3), thylakoid lumenal 17.9 kDa protein (spot GL 4) and Chlorophyll a/b binding protein (spot GL 5, GL 6) were up-regulated at the 12 and 18 hour, while RuBisCO activase B (spot GL 7) and DNA helicase (spot GL 8) were down-regulated. Thus, we suggest that these proteins might participate in the early response to salt stress in ginseng leaves.
